The increasing prevalence of drug-resistant Mycobacteriumtuberculosis (Mtb) and nontuberculous mycobacteria (NTM) underscores the need for novel strategies. Mycobacteriophages are attracting growing interest for therapeutic and diagnostic applications. Here, we describe four new subcluster A3 phages (Cat, Goose1D, Goose1X, and Shep) and investigate their genomic diversity and lytic activity. The phages share >98% nucleotide identity with known A3 members but carry distinct deletions within the lysogeny module: Cat and Goose1X have different repressor-loss variants, and Goose1D lacks the integr

Johne's disease (JD), a chronic, infectious enteritis caused by Mycobacterium avium subsp. paratuberculosis (MAP), affects wild and domestic ruminants. There is no cure or effective prevention, and current vaccines have substantial limitations, leaving this disease widespread in all substantial dairy industries causing economic, and animal welfare implications. Mycobacteriophages (MPs) have been gaining interest in recent years and are proposed as a promising solution to curtailing MAP infection. Using a well-validated infection model, we have demonstrated the preventative potential of MPs to

Evolution of drug-resistant mycobacterial infections warrants renewed efforts in identifying more efficient preventive and control strategies as well as alternative treatment options. Interest in phage therapy is regaining significant traction, especially in cases of failed conventional therapy. However, phage therapy faces challenges, including the identification of a suitable therapeutic phage, phage delivery, phage resistance, and host immunity. This article reviews existing clinical literature on the therapeutic use of mycobacteriophages as adjuncts to antibiotics in the treatment of drug-

The ever-increasing incidence of antimicrobial resistance has revived interest in phage-derived therapeutics against bacterial infections. Bacteriophage-encoded endolysins, which can destroy peptidoglycan of the host cell wall to release progeny, show potential as alternatives to traditional antibiotics. Owing to the highly complex cell wall of mycobacteria, most mycobacteriophages employ a two-component system, comprising a canonical endolysin (LysA) and a complementary enzyme, a specific mycolyl-arabinogalactan esterase referred to as Lysin B (LysB). In this study, LysB from mycobacteriophag

Mycobacteriophage Mcgavigan could be a promising candidate for use as a preventative agent against infections with Mycobacterium avium subsp. paratuberculosis. Bioinformatic analysis of the Mcgavigan genome revealed the presence of an operon containing a "Bxb1-like" repressor. The operon may have been acquired by the phage through horizontal gene transfer with a Bxb1-like mycobacteriophage in its evolutionary past. We sought to investigate the function of the acquired repressor as a potential regulator of lysogeny or as a source of heterotypic superinfection immunity. Recombineering with CRISP

Mycobacteriophages are phages that infect mycobacteria resulting in their killing. Although lysis is the primary mechanism by which mycobacteriophages cause cell death, others such as abortive infection may also be involved. We took recourse to perform immunofluorescence and electron microscopic studies using mycobacteriophage D29 infected Mycobacterium smegmatis cells to investigate this issue. We could observe the intricate details of the infection process using these techniques such as adsorption, the phage tail penetrating the thick mycolic acid layer, formation of membrane pores, membrane

Patients infected with mycobacteria, particularly Mycobacterium abscessus, frequently experience antibiotic treatment failure. Phage therapy, used under compassionate care, offers a promising alternative and has already demonstrated clinical efficacy. However, phage-bacteria interactions and their coevolution remain critical factors that can compromise therapeutic success. Trehalose polyphleates (TPPs), complex lipids of the outer membrane, play a key role in the adhesion of the therapeutic mycobacteriophage BPsΔ33HTH_HRM10, and loss of TPP synthesis confers bacterial resistance to the phage.

Infection by multidrug-resistant Mycobacterium abscessus is increasingly prevalent in cystic fibrosis (CF) patients, leaving clinicians with few therapeutic options. A compassionate study showed the clinical improvement of a CF patient with a disseminated M. abscessus (GD01) infection, following injection of a phage cocktail, including phage Muddy. Broadening the use of phage therapy in patients as a potential antibacterial alternative necessitates the development of biological models to improve the reliability and successful prediction of phage therapy in the clinic. Herein, we demonstrate th

Inhaled therapy of tuberculosis (TB) by a Dry Powder Inhalation (DPI) comprising mycobacteriophage D29 and TM4 was non-inferior to oral anti-tuberculosis therapy (ATT) with isoniazid and rifampicin in a mouse model of infection with Mycobacterium tuberculosis (Mtb). No pharmaceutical phage product of mycobacteriophages is approved for large-scale production. We scaled up preparation and downstream processing of phages, developed DPI formulations, and established methods for determining identity, purity, assay, stability, and critical quality attributes (CQA). We carried out cell-based assays o

Tuberculosis caused by Mycobacterium tuberculosis (Mtb) is one of the most dangerous diseases globally. Mtb poses a heavy death toll, especially in low-resource settings, where inadequate diagnostic capabilities greatly hinder treatment and prevention. Here, we present a rapid and cost-effective bacilli-capturing method that uses magnetic nanoclusters conjugated with mycobacteriophages. The mycobacteriophages provide Mtb recognition functionality, and the binding of the nanoparticles with attenuated Mtb H37Rv and Mycobacterium bovis Bacillus Calmette-Guérin (BCG) was visualized by electron mic

INTRODUCTION: The growing prevalence of multidrug-resistant Mycobacterium tuberculosis and nontuberculous mycobacteria (NTM) highlights the urgent need for alternative therapeutic approaches. Mycobacteriophages, viruses that selectively infect mycobacteria, have emerged as promising tools. Here, we report the isolation and characterization of a new subcluster K4 phage, Yasnaya_Polyana, with a focus on its regulatory motifs and engineered lytic variant.
METHODS: The phage was isolated by enrichment on Mycobacterium smegmatis mc(2)155, followed by genome sequencing and functional annotation. St

Conventional methods for determining drug susceptibility of Mycobacterium tuberculosis require several weeks to obtain results, limiting their usefulness; automated methods and those based on molecular biology techniques have been able to reduce the turnaround time, but their high cost and need for sophisticated equipment restrict their use in developing countries. The goal of the present study was to evaluate the diagnostic accuracy of a rapid (3-4 days) low-cost test based on the use of mycobacteriophage D29 to determine the susceptibility of strains of M. tuberculosis to rifampicin (RIF) an

UNLABELLED: There is an urgent need for rapid, affordable diagnostic tools capable of detecting baseline and treatment-emergent resistance among patients with drug-resistant tuberculosis (DR-TB). Reporter mycobacteriophages provide a phenotypic solution enhanced by brighter luminescent reporters compatible with simple readouts. We applied the TM4::GeNL reporter mycobacteriophage for rapid phenotypic drug susceptibility testing (pDST) on serial Mycobacterium tuberculosis isolates from six DR-TB patients, including four with treatment-emergent resistance and two with baseline bedaquiline (BDQ)-r

Bacteriophages represent a vast reservoir of genetic diversity; however, functional annotation remains a major challenge, as most predicted gene products lack detectable similarity to characterized gene families. Experimental approaches such as systematic overexpression screens provide an avenue for identifying phage genes that influence bacterial physiology. Here, we report an overexpression screen of all 123 predicted protein-coding genes from Cluster L1 mycobacteriophage LeBron, the first representative of this cluster to undergo genome-wide functional analysis. Tuberculosis, caused by Mycobacterium tuberculosis, is responsible for over two million deaths per year worldwide. Due to its long doubling time (18 h), the microbiological detection of M. tuberculosis by conventional methods takes up to one month, unless the number of bacilli in the biological sample is high enough. Thus, drug resistance assessment requires at least one month for obtaining the primary culture and another month to determine its susceptibility to antimycobacterial drugs. Moreover, for a long time, the lack of genetic tools for mycobacteria has been a barrier for undertaking stu

Lytic enzymes encoded by bacteriophages are conventionally understood to serve a single function in the phage life cycle: to enable progeny release through host cell lysis. Here, we identify an unexpected dual functionality in LysB, a lytic cassette protein of mycobacteriophage D29, which exerts opposing effects on phage propagation depending on the stage of infection. In our model, during the early phase of phage expansion, LysB's activity is limited to infected cells, hydrolyzing their outer membrane to facilitate efficient virion release. However, as the infection progresses, LysB accumulat

Mycobacterium leprae, the causative agent of leprosy, has never been cultured in vitro, posing significant challenges for genetic manipulation and drug discovery. Current antileprosy drug screening methods relying on microscopic count, radiorespirometry, and qPCR are time consuming and require the use of radioactive elements. We demonstrate mycobacteriophage-mediated introduction of foreign DNA using the broad-host range mycobacteriophage TM4 and the application of the luciferase reporter mycobacteriophage (LRM) for drug screening. Mycobacteriophage infection of M. leprae was shown using TM4 e

Emergence of antibiotic resistance in pathogenic Mycobacterium tuberculosis (Mtb) has elevated tuberculosis to a serious global threat, necessitating alternate solutions for its eradication. D29 mycobacteriophage can infect and kill several mycobacterial species including Mtb. It encodes an endolysin LysA to hydrolyze host bacteria peptidoglycan for progeny release. We previously showed that out of the two catalytically active domains of LysA [N-terminal domain (NTD) and lysozyme-like domain], NTD, when ectopically expressed in Mycobacterium smegmatis (Msm), is able to kill the bacterium nearl

The Mycobacteria are a genus of Actinobacteria that include human pathogens such as Mycobacterium tuberculosis (TB). Active TB disease can spread by airborne transmission to healthcare workers and to their community. The HHMI SEA-PHAGES program has contributed to discovering bacteriophages that are able to infect M. smegmatis MC2 155, a close relative of M. tuberculosis. This collection of diverse Mycobacteriophages is an excellent resource for trialling bacteriophage-sourced enzymes in novel applications. Herein we measured the ability Mycobacteriophage endolysins to lyse their host strain wh
