Autoimmune keratitis in mycobacterium tuberculosis.

PURPOSE: To report a case of autoimmune keratitis in a patient with mycobacterium tuberculosis (MBT). METHODS: An 84-year-old male with pulmonary tuberculosis (TB) was admitted with chronic, non-healing bilateral ulcerations of the inferior peripheral cornea associated with stromal and subconjunctival nodules. RESULTS: Clinical examination revealed circumscribed peripheral corneal ulceration with whitish nodules in adjacent stromal and subconjunctival tissue. Microbiological cultures of the corneal tissue were negative for MBT and other microbial pathogens; however, enzyme-linked immunosorbe

[Immune response to Mycobacterium tuberculosis].

Infections with Mycobacterium tuberculosis (MTB) induce complex immune responses involving an orchestrated interplay of innate and adaptive immune mechanisms. Why the immune system fails to eradicate the pathogen and at best achieves control of infection in the latent stage, still remains an unsolved mystery even more than 100 years after the discovery of MTB by Robert Koch. This article provides an overview of the current state of the art in the constantly evolving field of tuberculosis (TB) immunology. This review focuses on a change of paradigm proposing that in the latent stage MTB is anyt

Mycobacterium tuberculosis complex in atherosclerosis.

In recent years, the results of some studies have revealed the possible potential role of several infectious agents in the inflammatory mechanism of atherosclerosis. The detection of specific antibodies against microorganisms such as and as well as Chlamydia pneumoniae and cytomegalovirus as well as antibodies directed to heat shock proteins in the sera of atherosclerotic patients and the presence of genomic material in atheromatous plaques all provide evidence supporting the presumptive role of infectious agents in atherosclerosis. There are some findings that can be accepted as clues for the

Iron and Mycobacterium tuberculosis infection.

BACKGROUND: iron is known to play a role in the susceptibility to and outcome of several infections. In view of the increasing worldwide problem of tuberculosis, it may be important to ascertain whether this is also the case with this infection. OBJECTIVES: (1) to review studies conducted in vitro, in experimental animals, and in humans that provide evidence that iron status may influence the occurrence and outcome of tuberculosis. (2) To perform an in vivo study in mice, examining the effect of iron loading on experimental infection caused by a virulent strain of Mycobacterium tuberculosis.

[Endophthalmitis caused by mycobacterium tuberculosis].

A 60-year-old woman developed an anterior uveitis of unknown etiology in her right eye. Topical treatment was corticosteroids led to a regression of the inflammation, but three months later a severe relapse occurred, followed by endophthalmitis and blindness. At this time a previously unknown pulmonary tuberculosis was diagnosed. In histologic sections of the enucleated eye numerous acid-fast bacilli were demonstrated. Diagnostic and therapeutic considerations are discussed.

Aerobiology and Transmission of Mycobacterium tuberculosis.

Despite vast changes in human social structures through evolution-from small hunter-gatherer bands to densely populated megacities-Mycobacterium tuberculosis (Mtb) has persisted within human populations for millennia, underscoring the bacterium's exceptional transmissibility. This paper examines the aerobiological foundations of Mtb transmission, tracing the bacillus from expulsion in respiratory aerosols through environmental survival to deposition and infection in new hosts. It considers how host, mycobacterial, and environmental factors interact to influence transmission efficiency, highlig

Mycobacterium tuberculosis as a model system.

Over the past 30 years, the study of Mycobacterium tuberculosis has become a vibrant field that supports basic investigation into mycobacteria, wide-ranging studies of M. tuberculosis pathogenesis, and drug and therapeutic development. In this minireview, we highlight the multiple ways in which M. tuberculosis is a model. M. tuberculosis has emerged as a model for prokaryotic biology, revealing basic mechanisms of gene expression, genomic integrity, and mutagenesis that expand prokaryotic dogmas established in earlier model organisms. Next, we highlight the experimental systems used to model M

Mycobacterium tuberculosis of the elbow joint.

Mycobacterium tuberculosis(TB) affecting the elbow joint is rarely reported in the developed world. We present the case of an 85-year-old Caucasian female who complained of a chronically discharging and painful wound across her left elbow during her admission for an ischaemic stroke. This was initially deemed to be either a bursitis or local manifestation of amyloid by her general practitioner and dermatologist respectively prior to admission. She was commenced on flucloxacillin by the medical team for presumed cellulitis with minimal response. A synovial fluid sample and repeated wound swabs

A child with complicated Mycobacterium tuberculosis.

Tuberculosis (TB) is one of the leading causes of morbidity and mortality worldwide, with ever increasing resistance to commonly used antituberculous drugs. Drug-resistant TB was recognized shortly after the introduction of an effective therapy in the late 1940s, the use of streptomycin, which was the first widely used antituberculosis drug. Patients who received this drug usually had marked and rapid clinical improvement, but treatment failures were common after the first three months of therapy. Most children are infected by household contacts who have TB, particularly parents or other caret

Mycobacterium tuberculosis is resistant to streptolydigin.

Drug resistant strains of Mycobacterium tuberculosis (Mtb) undermine tuberculosis (TB) control. Streptolydigin is a broadly effective antibiotic which inhibits RNA polymerase, similarly to rifampicin, a key drug in current TB chemotherapeutic regimens. Due to a vastly improved chemical synthesis streptolydigin and derivatives are being promoted as putative TB drugs. The microplate Alamar Blue assay revealed that Streptococcus salivarius and Mycobacterium smegmatis were susceptible to streptolydigin with minimum inhibitory concentrations (MICs) of 1.6 mg/L and 6.25 mg/L, respectively. By contra

The pathology of Mycobacterium tuberculosis infection.

Mycobacterium tuberculosis is an old enemy of the human race, with evidence of infection observed as early as 5000 years ago. Although more host-restricted than Mycobacterium bovis, which can infect all warm-blooded vertebrates, M. tuberculosis can infect, and cause morbidity and mortality in, several veterinary species as well. As M. tuberculosis is one of the earliest described bacterial pathogens, the literature describing this organism is vast and overwhelming. This review strives to distill what is currently known about this bacterium and the disease it causes for the veterinary pathologi

Detection and diagnosis of Mycobacterium tuberculosis.

Tuberculosis continues to be a major health problem worldwide, affecting millions of people each year. Accurate and rapid diagnosis is key to controlling the disease, yet the traditional tests for TB produce results that are either inaccurate or take too long to be definitive. A fast and reliable diagnostic method that would differentiate between active and latent TB infection is also lacking. The current routine diagnostic tests for TB - chest x-ray, tissue culture, tuberculin skin test (TST) and acid-fast staining - all have their limitations. A chest x-ray alone is inconclusive; a tissue cu

Mycobacterium tuberculosis monoarthritis in a child.

A child with isolated Mycobacterium tuberculosis monoarthritis, with features initially suggesting oligoarthritis subtype of juvenile idiopathic arthritis, is presented. This patient illustrates the need to consider the possibility of tuberculosis as the cause of oligoarthritis in high-risk pediatric populations even in the absence of a tuberculosis contact history and without evidence of overt pulmonary disease.

A nosocomial outbreak of Mycobacterium tuberculosis.

BACKGROUND: The national incidence of tuberculosis (TB) is increasing, and hospitals are a site of transmission. We investigated a nosocomial outbreak of TB at a 160-bed community hospital in South Carolina that highlights the central role that primary care physicians must play to control this epidemic. METHODS: We reviewed medical records to identify potential source cases. We retrospectively evaluated exposures to suspected source patients and the subsequent tuberculin reactivity of the 38 hospital employees who had a previous negative tuberculin skin test and were assigned to the ward wher

A replication clock for Mycobacterium tuberculosis.

Few tools exist to assess replication of chronic pathogens during infection. This has been a considerable barrier to understanding latent tuberculosis, and efforts to develop new therapies generally assume that the bacteria are very slowly replicating or nonreplicating during latency. To monitor Mycobacterium tuberculosis replication within hosts, we exploit an unstable plasmid that is lost at a steady, quantifiable rate from dividing cells in the absence of antibiotic selection. By applying a mathematical model, we calculate bacterial growth and death rates during infection of mice. We show t
