Occurrence of keratoconjunctivitis apparently caused by Mycoplasma gallisepticum in layer chickens.

Natural cases of keratoconjunctivitis, apparently caused by Mycoplasma gallisepticum (MG), in layer chickens are described. The disease occurred in a commercial flock consisting of 36,000 pullets (Babcock), first appearing around 30 days of age. Clinically, affected chickens showed unilateral or bilateral swelling of the facial skin and the eyelids, increased lacrimation, congestion of conjunctival vessels, and respiratory rales. Some of the severely affected chickens closed their eyes. The morbidity reached 27.8%, and it was estimated that approximately 10% died from reduced feed intake due t…

Extracellular HMGB1 as Inflammatory Mediator in the Progression of Mycoplasma Gallisepticum Infection.

High-mobility group box 1 (HMGB1), a member of damage-associated molecular patterns (DAMPs), is involved in the immune regulation of several infectious diseases. Mycoplasma gallisepticum (MG) infection is proved to cause an abnormal immune response, but the role of HMGB1 in MG-induced chronic respiratory disease (CRD) is unclear. In this study, we found that HMGB1 was released from the nucleus to the extracellular in macrophages upon infection with MG. Extracellular HMGB1 bound to TLR2 activating the NF-κB pathway triggering a severe inflammatory storm and promoting the progression of MG infec…

Lymphocytic infiltration in the chicken trachea in response to Mycoplasma gallisepticum infection.

A prominent feature of disease induced by Mycoplasma gallisepticum is a lymphoproliferative response in the respiratory tract. Although this is also seen in other mycoplasma infections, including Mycoplasma pneumoniae, the phenotype of the lymphocytes infiltrating the respiratory tract has not been determined. In this study, the numbers and distribution of lymphocytes in the tracheas of chickens infected with a virulent strain of M. gallisepticum were examined. Three groups of chickens were experimentally infected with M. gallisepticum and three unchallenged groups were used as controls. One i…

Prevalence and antimicrobial susceptibility of Mycoplasma gallisepticum and Mycoplasma synoviae isolated from the central peninsular Malaysia.

Mycoplasma gallisepticum (MG) and Mycoplasma synoviae (MS) have been recognized and listed by the World Organization for Animal Health (WOAH) as the two most pathogenic avian mycoplasma species. These organisms can cause a wide range of symptoms in birds, including acute and chronic respiratory diseases, synovitis, air sacculitis, severe weight loss, eggshell abnormalities, and reduced egg production. In Malaysia, previous studies have mainly focused on seroprevalence, and prevalence based on polymerase chain reaction (PCR) with molecular characterization, which have confirmed the presence of …

In vitro evaluation of various antimicrobials against field mycoplasma gallisepticum and mycoplasma synoviae isolates in Egypt.

Among many avian mycoplasmas, Mycoplasma gallisepticum (MG) and Mycoplasma synoviae (MS) are recognized as the main etiological agents of respiratory diseases and infectious synovitis in chickens and turkeys causing tremendous economic losses worldwide. Therefore, proper treatment is promoted for the control of these diseases. This study was the first in Egypt to evaluate the in vitro efficacy of various antimicrobials against field MG and MS isolates recovered from chicken and turkey flocks using both conventional broth microdilution and quantitative real-time polymerase chain reaction assays…

Cytoskeletal asymmetrical dumbbell structure of a gliding mycoplasma, Mycoplasma gallisepticum, revealed by negative-staining electron microscopy.

Several mycoplasma species feature a membrane protrusion at a cell pole, and unknown mechanisms provide gliding motility in the direction of the pole defined by the protrusion. Mycoplasma gallisepticum, an avian pathogen, is known to form a membrane protrusion composed of bleb and infrableb and to glide. Here, we analyzed the gliding motility of M. gallisepticum cells in detail. They glided in the direction of the bleb at an average speed of 0.4 microm/s and remained attached around the bleb to a glass surface, suggesting that the gliding mechanism is similar to that of a related species, Myco…

Mycoplasma gallisepticum uses itaconate-associated mitochondrial inhibition to suppress host immunometabolism.

Many pathogens actively suppress early host immune responses to enhance their fitness. Mitochondria function as key regulators of immune activation, yet whether pathogens suppress host immunity by manipulating mitochondrial metabolism in vivo remains largely unknown. During an innate immune response, the metabolite itaconate increases in abundance and acts as an immunomodulator, due to its inhibition of succinate dehydrogenase (SDH), a key mitochondrial regulator of cellular immunity. We hypothesized that Mycoplasma gallisepticum (MG), a recently emerged pathogen of wild songbirds, most notabl…

Targeted mutagenesis of Mycoplasma gallisepticum using its endogenous CRISPR/Cas system.

New, more efficient methods are needed to facilitate studies of gene function in the mycoplasmas. CRISPR/Cas systems, which provide bacteria with acquired immunity against invading nucleic acids, have been developed as tools for genomic editing in a wide range of organisms. We explored the potential for using the endogenous Mycoplasma gallisepticum CRISPR/Cas system to introduce targeted mutations into the chromosome of this important animal pathogen. Three constructs carrying different CRISPR arrays targeting regions in the ksgA gene (pK1-CRISPR, pK-CRISPR-1 and pK-CRISPR-2) were assembled an…

Detection of Mycoplasma gallisepticum in House Finches ( Haemorhous mexicanus) from Arizona.

In 1994, an endemic poultry pathogen, Mycoplasma gallisepticum (MG), was identified as the causative agent of a novel disease in house finches ( Haemorhous mexicanus). After an initial outbreak in Maryland, MG spread rapidly throughout eastern North American populations of house finches. Subsequently, MG spread slowly through the northern interior of North America and then into the Pacific Northwest, finally reaching California in 2006. Until 2009, there were no reports of MG in the southwestern United States east of California. In August 2011, after reports of house finches displaying conjunc…

Arbutin improves lung injury in chicks induced by Mycoplasma gallisepticum infection.

Mycoplasma gallisepticum (MG) is a pathogen commonly found in poultry that can cause avian-associated respiratory diseases. Arbutin (AR) possesses various pharmacological activities, including anti-tumor, anti-inflammatory, apoptotic regulation, and anti-oxidative stress effects, however, its impact and mechanisms against inflammatory damage caused by MG infection remain unclear. The results demonstrated that after AR intervention, the MG-induced symptoms such as pulmonary wall thickening, lung tissue congestion and hemorrhage, inflammatory cell infiltration, and HD11 cell swelling were signif…

Minimal inhibitory concentration of seven antimicrobials to Mycoplasma gallisepticum and Mycoplasma synoviae isolates from six European countries.

Mycoplasma gallisepticum and Mycoplasma synoviae are bacterial pathogens that cause disease in poultry, adversely affecting their health and welfare, and are a financial burden on producers. This manuscript describes the results of the MycoPath project that is the first international antimicrobial susceptibility programme for mycoplasma pathogens isolated from poultry. Improved comparative analysis of minimal inhibitory concentration (MIC) results from participating countries was facilitated by using one laboratory determining all MICs. Chicken and turkey isolates were obtained from France, Ge…

Studies of PPLO infection. I. The production of cerebral polyarteritis by Mycoplasma gallisepticum in turkeys; the neurotoxic property of the Mycoplasma.

Turkey poults injected intravenously with suspensions of Mycoplasma gallisepticum develop a fatal neurologic disease associated with polyarteritis affecting almost exclusively the cerebral arteries. The incubation period depends on the dose of organisms. With high doses (10(10) to 10(11) mycoplasmas) the birds become ill and die within a few hours; with lower doses (10(6) to 10(8)) neurologic manifestations appear after 7 days. The rapid onset of neurologic signs after high doses indicates the presence of a toxin in the mycoplasma, but efforts to extract toxin from disrupted organisms or to de…

Assessment of the pathogenicity of Mycoplasma gallisepticum and the immunogenicity of its membrane proteins in geese.

INTRODUCTION: Mycoplasma gallisepticum (MG) imposes heavy losses on the goose industry in China, yet its goose-specific biology is poorly understood. In elucidation of the biology, the pathogenicity and immunogenicity of MG in geese were analysed. MATERIAL AND METHODS: Goslings were divided into three groups and either infected with the goose isolate MG-GD01/22, infected with chicken-derived MG R(low) or left untreated as controls. Clinical scores were recorded, and morbidity/mortality, air-sac lesions and tracheal and lung histopathology evaluated. Isolate membrane proteins were separated by…

Current status of vaccine research, development, and challenges of vaccines for Mycoplasma gallisepticum.

Mycoplasma gallisepticum (MG) is an important avian pathogen that causes significant economic losses in the poultry industry. Surprisingly, the limited protection and adverse reactions caused by the vaccines, including live vaccines, bacterin-based (killed) vaccines, and recombinant viral vaccines is still a major concern. Mycoplasma gallisepticum strains vary in infectivity and virulence and infection may sometimes unapparent and goes undetected. Although extensive research has been carried out on the biology of this pathogen, information is lacking about the type of immune response that conf…

Expression and immunological characteristics of the surface-localized pyruvate kinase in Mycoplasma gallisepticum.

The widespread avian pathogen Mycoplasma gallisepticum is a causative agent of respiratory disease. The wall-less prokaryotes lack some tricarboxylic acid cycle enzymes, therefore, the glycolysis metabolic pathway is of great importance to these organisms. Pyruvate kinase (PK) is one of the key enzymes of the glycolytic pathway, and its immunological characteristics in Mycoplasma are not well known. In this study, the M. gallisepticum pyruvate kinase fusion protein (PykF) was expressed in a pET system. The full-length of the gene was subcloned into the expression vector pET28a(+) to construct …

Mycoplasma gallisepticum experimental infection and tissue distribution in chickens, sparrows and pigeons.

The most effective approaches to control the spread of Mycoplasma gallisepticum include strict biosecurity measures, continuous surveillance and eradication of infected flocks. The rapid expansion of the poultry industry worldwide in restricted geographical areas and severe economic losses due to M. gallisepticum outbreaks make it crucial to identify and better control the vectors responsible for the transmission of the disease. In the present study we evaluated the susceptibility of sparrows and pigeons to M. gallisepticum and the tissue distribution of M. gallisepticum in these species as co…

Facial cellulitis induced in chickens by Mycoplasma gallisepticum bacterin and its treatment.

Ten-to-14-day-old replacement chickens were vaccinated subcutaneously in the neck with a Mycoplasma gallisepticum bacterin. A few days later, 40% of the flock developed cellulitis of the head. Birds were submitted for diagnostic investigation between 4 and 6 wk of age. Histology revealed lesions in the subcutis consistent with oil-emulsified vaccine/bacterin reaction. No significant bacteria were isolated from the lesions. Birds were treated with 1 mg of methyl prednisol acetate intramuscularly. Improvement was noted as early as 12 hr posttreatment. One week after treatment only 1%-2% of the f…
