Molecular responses to cadmium (Cd) stress were studied in mycorrhizal and non-mycorrhizal Pisum sativum L. cv. Frisson inoculated with Glomus intraradices. Biomass decreases caused by the heavy metal were significantly less in mycorrhizal than in non-mycorrhizal plants. Real-time reverse transcriptase-polymerase chain reaction showed that genes implicated in pathways of Cd detoxification varied in response to mycorrhiza development or Cd application. Expression of a metallothionein-encoding gene increased strongly in roots of Cd-treated non-mycorrhizal plants. Genes encoding gamma-glutamylcys

Arbuscular mycorrhizal fungi (AMF) can enhance drought tolerance of host plants; however, the underlying molecular mechanisms remain unclear. Here, PtPIP1;2, a plasma membrane intrinsic protein gene strongly induced by AMF (Funneliformis mosseae (T.H. Nicolson & Gerd.) C. Walker & A. Schüßler) colonization under drought stress, was identified. Subcellular localization confirmed that PtPIP1;2 targets the plasma membrane. To investigate its function, PtPIP1;2-overexpressing (OE) hairy root lines were generated in trifoliate orange (Poncirus trifoliata L. Raf.). Under drought stress, OE plants ex

BACKGROUND: Studies on mycorrhiza associated bacteria suggest that bacterial-fungal interactions play important roles during mycorrhiza formation and affect plant health. We surveyed Streptomyces Actinobacteria, known as antibiotic producers and antagonists of fungi, from Norway spruce mycorrhizas with predominantly Piloderma species as the fungal partner.
RESULTS: Fifteen Streptomyces isolates exhibited substantial variation in inhibition of tested mycorrhizal and plant pathogenic fungi (Amanita muscaria, Fusarium oxysporum, Hebeloma cylindrosporum, Heterobasidion abietinum, Heterobasidion a

The arbuscular mycorrhizal (AM) association between terrestrial plants and soil fungi of the phylum Glomeromycota is the most widespread beneficial plant-microbe interaction on earth. In the course of the symbiosis, fungal hyphae colonise plant roots and supply limiting nutrients, in particular phosphorus, in exchange for carbon compounds. Owing to the obligate biotrophy of mycorrhizal fungi and the lack of genetic systems to study them, targeted molecular studies on AM symbioses proved to be difficult. With the emergence of plant genomics and the selection of suitable models, an application o

The Matanza-Riachuelo river basin (MRB) is considered one of the most polluted sites in the world, with very high levels of heavy metal(loid)s (HMs). However, as in almost all urban basins, studies on biological indicators of soil health are limited. In this work, we sampled soil and roots of mycorrhizal plants in three regions of the MRB (upper, middle, and lower basins) with the aim of exploring the potential of arbuscular mycorrhizal fungi (AMF) richness, based on spore morphology, plant mycorrhizal status, and glomalin-related soil proteins (GRSP) concentration as bioindicators of soil hea

Microscope slide preparations made for the observation of the mycorrhiza of the mycoheterotrophic orchid Gastrodia elata were rediscovered and confirmed to be the original materials used by Shunsuke Kusano for his 1911 publication. In this study, we re-examined these historical slides to assess their preservation and scientific significance. Among the 115 slides, 61 clearly displayed the mycorrhiza of G. elata. Intracellular organelles closely resembling the 'vesicle', 'globule', 'tubular sheath', 'globular bodies', and 'lignified papillae' described by Kusano were observed, and staining react

Transport of phosphorus (P) into host plants and its release to root cells is an important function of arbuscular mycorrhizal fungi (AMF). However, relatively little is known about the forms and water solubilities of P compounds in specific locations in the intraradical fungal structures. We determined concentrations and solubility of P components in these structures in white clover (Trifolium repens L.). Plants were grown in the field (colonised by indigenous AMF) or in the glasshouse (inoculated with Glomus intraradices). Mycorrhizas were cryo-fixed in liquid nitrogen immediately (control) o

The main challenge for plants growing in nutrient poor, contaminated soil is biomass reduction, nutrient deficiency and presence of heavy metals. Our aim is to overcome these challenges using different microbial combinations in mining-impacted soil and focus on their physiological and biochemical impacts on a model plant system, which has multiple applications. In the current study, sorghum BTx623 seedlings grown in mining-impacted soil in greenhouse were subjected to plant growth promoting bacteria (PGPB or B) alone, PGPB with arbuscular mycorrhizal fungi (My), My alone and control group with

Arbuscular mycorrhizas (AM) can increase plant acquisition of P and N. No published studies have investigated the impact of P and AM on the allocation of N to the plant defence, cyanogenic glucosides. We investigated the effects of soil P and AM on cyanogenic glucoside (dhurrin) concentration in roots and shoots of two forage sorghum lines differing in cyanogenic potential (HCNp). Two harvest times allowed plants grown at high and low P to be compared at the same age and the same size, to take account of known ontogenetic changes in shoot HCNp. P responses were dependent on ontogeny and tissue

Arbuscular mycorrhiza (AM), a symbiosis between plants and Glomeromycotan fungi is based on the exchange of nutrients, which majorly occurs between fungal tree-shaped arbuscules and root cortex cells. Two homologous plant GRAS transcription factors, RAM1 and RAD1, were previously reported to interact, and both were implicated in regulating arbuscule branching in Lotus japonicus. We revisited the role of L. japonicus RAM1 and RAD1 and asked whether they act together in mediating arbuscule development and gene expression, or whether they act additively. We performed comprehensive genetic, phenot

To study the local and systemic effects of arbuscular mycorrhizal fungal (AMF) colonization, Nicotiana attenuata plants impaired in their interactions with AMF due to silencing of a calcium- and calmodulin dependent protein kinase (inverted repreat (ir)CCaMK) were grown competitively in pairs with empty vector (EV) plants, with and without two different types of inoculum. When inoculated, EV plants strongly outperformed irCCaMK plants. Foliar transcript profiling revealed that AMF colonization significantly changed gene expression of P-starvation and -transporter genes in irCCaMK plants. The P

It is well known that individual pea (Pisum sativum L.) cultivars differ in their symbiotic responsivity. This trait is typically manifested with an increase in seed weights, due to inoculation with rhizobial bacteria and arbuscular mycorrhizal fungi. The aim of this study was to characterize alterations in the root proteome of highly responsive pea genotype k-8274 plants and low responsive genotype k-3358 ones grown in non-sterile soil, which were associated with root colonization with rhizobial bacteria and arbuscular mycorrhizal fungi (in comparison to proteome shifts caused by soil supplem

The ectoparasitic dagger nematode (Xiphinema index), vector of Grapevine fanleaf virus (GFLV), provokes gall formation and can cause severe damage to the root system of grapevines. Mycorrhiza formation by Glomus (syn. Rhizophagus) intraradices BEG141 reduced both gall formation on roots of the grapevine rootstock SO4 (Vitis berlandieri×V. riparia) and nematode number in the surrounding soil. Suppressive effects increased with time and were greater when the nematode was post-inoculated rather than co-inoculated with the arbuscular mycorrhizal (AM) fungus. Using a split-root system, decreased X.

Interactions between phosphate (Pi) and zinc (Zn) or iron (Fe) nutrition in plants have been widely studied; however, the underlying mechanisms of their cross-talks in arbuscular mycorrhizal (AM) plants remain obscure. Here, we examine the physiological and molecular responses of tomato (Solanum lycopersicum L.) to the combination of Pi, Zn and/or Fe nutrient stresses during symbiosis with Rhizophagus irregularis, revealing the existence of a tripartite Pi-Zn-Fe cross-talk in AM symbiosis. Interestingly, the mycorrhiza-activated SlPT3, a member of the PHOSPHATE TRANSPORTER 1 (PHT1) gene family

Tissue culture and mycorrhiza applications can provide disease-free seedlings and enhanced nutrient absorption, respectively, for organic farming. Ginger (Zingiber officinale Roscoe) is rich in phytochemicals and has various health-protective potentials. This study was aimed at determining effects of tissue culture and mycorrhiza applications alone or in combinations in organic farming on phytochemical contents (total phenolics and flavonoids [TP and TF, respectively], gingerol and shogaol homologues, phenolic acids, and carotenoids) and antioxidant capacities (DPPH [2,2-diphenyl-1-picrylhydra

The sucrose transporter SUT1 functions in phloem loading of photoassimilates in solanaceous plant species. In the present study, wildtype and transgenic potato plants with either constitutive overexpression or antisense inhibition of SUT1 were grown under high or low phosphorus (P) fertilization levels in the presence or absence of the arbuscular mycorrhizal (AM) fungus Glomus intraradices. At a low soil P fertilization level, the extent of AM fungal root colonization was not different among the genotypes. In all plants, the AM symbiosis contributed significantly to P uptake under these condit

Microbial plant growth promoters often perform inconsistently under natural soil conditions most likely due to their poor establishment after inoculation. Here, we conducted a growth chamber pot experiment with maize using pre-activated inoculum of the P-solubilizing fungus Penicillium aculeatum to facilitate its establishment in a natural non-sterile agricultural soil. Hence, P. aculeatum was preincubated for one week in soil amended with cellulose as a C source and with different P sources, i.e. sewage sludge ash, Ca3(PO4)2, and no P fertilization as control. Control treatments of C and P so

The term 'global climate change' encompasses many physical and chemical changes in the atmosphere that have been induced by anthropogenic pollutants. Increases in concentrations of CO2 and CH4 enhance the 'greenhouse effect' of the atmosphere and may contribute to changes in temperature and precipitation patterns at the earth's surface. Nitrogen oxides and SO2 are phytotoxic and also react with other pollutants to produce other phytotoxins in the troposphere such as O3 and acidic substances. However, release of chlorofluorocarbons into the atmosphere may cause depletion of stratospheric O3, in

A collection of 300 isolates of fluorescent pseudomonads was established from Douglas fir-Laccaria bicolor mycorrhizas and mycorrhizosphere and from adjacent bulk soil. These isolates were first phenotypically characterized with the Biolog method. Taxonomic identification assigned 90% of the isolates to the different biovars of Pseudomonas fluorescens, with inverted frequencies of biovars V and I from the bulk soil to the mycorrhizas, suggesting that the mycorrhizas exert a selective stimulation of the P. fluorescens bv. I and a counterselection of the P. fluorescens bv. V present in the soil.

Glomalin is a specific glycoprotein compound synthesized by glomeral fungi and its amount varies according to soil conditions. Due to the symbiosis of these fungi with a multitude of plants, a remarkable amount of glomalin is produced inside the hypha wall of these fungi. Furthermore, while increasing the symbiosis stability, glomalin plays a critical role in reducing the availability of potentially toxic elements (PTEs) through their fixation. In this regard, soil contamination with PTEs such as cadmium (Cd) affects the glomalin production of mycorrhiza fungi. In order to investigate the effe
