"Frizzy" aggregation genes of the gliding bacterium Myxococcus xanthus show sequence similarities to the chemotaxis genes of enteric bacteria.

The frz genes of Myxococcus xanthus are necessary for proper aggregation of cells to form fruiting bodies. Mutations in the frz genes affect the frequency with which individual cells reverse their direction of movement. We have subcloned and determined the nucleotide sequence of three of the frz genes. From the sequence we predict three open reading frames corresponding to frzA, frzB, and frzCD. The putative FrzA protein (17,094 Da) exhibits 28.1% amino acid identity with the CheW protein of Salmonella typhimurium. The putative FrzCD protein (43,571 Da) contains a region of about 250 amino aci

Assessment of fed-batch, semicontinuous, and continuous epothilone D production processes.

Epothilone D is a member of a class of potent antineoplastic natural products produced by myxobacteria. Previously, we have described a fed-batch epothilone D production process in which an adsorber resin is incorporated into the bioreactor setup to capture and stabilize the product in situ, preventing its degradation within the bioreactor. The capture of epothilone D by these relatively large resin beads enables the development of continuous and semicontinuous culturing systems incorporating bead retention mechanisms to completely retain the product within the bioreactor, increasing the epoth

A predator-prey interaction between myxobacteria and fungi mediated by outer membrane vesicles.

INTRODUCTION: Outer membrane vesicles (OMVs) of Gram-negative bacteria mediate diverse functions in natural ecosystems. As a keystone taxon in soil, myxobacteria produce OMVs for cargo packing and microbial predation. However, the roles of OMVs in the interactions of myxobacteria with fungi remain poorly understood. OBJECTIVES: This work aims to clarify the role of outer membrane vesicles in the interaction between myxobacteria and fungi and the regulatory mechanism during the interaction process. METHODS: We found that OMVs play a significant role in the interaction between Myxococcus sp. M

Hidden paths to endless forms most wonderful: ecology latently shapes evolution of multicellular development in predatory bacteria.

Ecological causes of developmental evolution, for example from predation, remain much investigated, but the potential importance of latent phenotypes in eco-evo-devo has received little attention. Using the predatory bacterium Myxococcus xanthus, which undergoes aggregative fruiting body development upon starvation, we tested whether adaptation to distinct growth environments that do not induce development latently alters developmental phenotypes under starvation conditions that do induce development. In an evolution experiment named MyxoEE-3, growing M. xanthus populations swarmed across agar

Aggregative chimeric multicellularity in the absence of lethal kin discrimination.

Aggregative multicellularity is a cooperative strategy used by some microbes. Unlike plant and animal development, which proceeds through clonal expansion from a single progenitor cell, aggregation is susceptible to genetic conflict and cheating, threatening multicellular stability. Myxococcus xanthus and Dictyostelium spp. are soil-dwelling models that form spore-producing fruiting bodies by aggregation upon starvation. Studies of natural M. xanthus fruiting bodies show that this process is confined to clonemates or close relatives, whereas Dictyostelium spp. can form polyclonal fruiting bodi

Laboratory biases hinder Eco-Evo-Devo integration: Hints from the microbial world.

How specific environmental contexts contribute to the robustness and variation of developmental trajectories and evolutionary transitions is a central point in Ecological Evolutionary Developmental Biology ("Eco-Evo-Devo"). However, the articulation of ecological, evolutionary and developmental processes into integrative frameworks has been elusive, partly because standard experimental designs neglect or oversimplify ecologically meaningful contexts. Microbial models are useful to expose and discuss two possible sources of bias associated with conventional gene-centered experimental designs: t

Production optimization and biosynthesis revision of corallopyronin A, a potent anti-filarial antibiotic.

Corallopyronins (COR) are α-pyrone antibiotics from myxobacteria representing highly promising lead structures for the development of antibacterial therapeutic agents. Their ability to inhibit RNA polymerase through interaction with the "switch region", a novel target, distant from binding sites of previously characterized RNA polymerase inhibitors (e.g. rifampicin), makes them particularly promising as antibiotic candidates. Corallopyronin A is currently also investigated as a lead compound for the treatment of lymphatic filariasis because of its superb activity against the nematode symbiont

The Chemical Ecology of Predatory Soil Bacteria.

The study of natural products is entering a renaissance, driven by the discovery that the majority of bacterial secondary metabolites are not produced under standard laboratory conditions. Understanding the ecological role of natural products is key to efficiently directing our screening efforts, and to ensuring that each screen efficiently captures the full biosynthetic repertoire of the producing organisms. Myxobacteria represent one of the most common and diverse groups of bacteria, with roughly 2500 strains publically available. Fed largely through predation, the myxobacteria have develope

Architecture of the Vibrio cholerae toxin-coregulated pilus machine revealed by electron cryotomography.

Type IV pili (T4P) are filamentous appendages found on many Bacteria and Archaea. They are helical fibres of pilin proteins assembled by a multi-component macromolecular machine we call the basal body. Based on pilin features, T4P are classified into type IVa pili (T4aP) and type IVb pili (T4bP)1,2. T4aP are more widespread and are involved in cell motility3, DNA transfer4, host predation5 and electron transfer6. T4bP are less prevalent and are mainly found in enteropathogenic bacteria, where they play key roles in host colonization7. Following similar work on T4aP machines8,9, here we use ele

LtpA, a CdnL-type CarD regulator, is important for the enzootic cycle of the Lyme disease pathogen.

Little is known about how Borrelia burgdorferi, the Lyme disease pathogen, adapts and survives in the tick vector. We previously identified a bacterial CarD N-terminal-like (CdnL) protein, LtpA (BB0355), in B. burgdorferi that is preferably expressed at lower temperatures, which is a surrogate condition mimicking the tick portion of the enzootic cycle of B. burgdorferi. CdnL-family proteins, an emerging class of bacterial RNAP-interacting transcription factors, are essential for the viability of Mycobacterium tuberculosis and Myxococcus xanthus. Previous attempts to inactivate ltpA in B. burgd

Encapsulin-Protected Immunotherapeutic Complexes: Bacteria-Derived Nanoparticles for mRNA Delivery to Eukaryotic Cells.

Encapsulins are protein nanocompartments that self-assemble into icosahedral structures that capture cargo proteins through a cargo-loading peptide (CLP). Here, we aimed to develop an mRNA delivery vector for eukaryotic cells by fusing the CLP from Myxococcus xanthus encapsulin cargoes with a short peptide that binds to a complementary RNA aptamer, creating an encapsulin-RNA adaptor peptide (ERAP). Additionally, we designed a chimeric mRNA (CmRNA) molecule for translation in eukaryotic cells, featuring an internal ribosome entry site (IRES) in the 5'-UTR to enable cap-independent translation.

Understanding the mechanisms of biological struvite biomineralisation.

The mechanisms of struvite production through biomineralisation were investigated for five microorganisms (Bacillus pumilus, Brevibacterium antiquum, Myxococcus xanthus, Halobacterium salinarum and Idiomarina loihiensis). After 72-96 h of incubation, the microbial strains tested increased the solution pH from 7.5 to 7.7 to 8.4-8.7, and removed ortho-phosphate (63-71%) and magnesium (94-99%) by biomineralisation. The minerals formed were identified as struvite (i.e. bio-struvite). Within the initial 24 h of incubation, microbial growth rates of 0.16-0.28 1/h were measured, and bio-struvite prod

Aggregicyclins Shed Light on Type II Polyketide Biosynthesis in Myxococcota.

Polycyclic bacterial specialized metabolites such as anthraquinones, angucyclines, and tetracyclines are predominantly produced by type II polyketide synthase (PKS) systems and represent an important source of antibiotics and anticancer agents. While type II PKS pathways are well characterized in Gram-positive bacteria, their biosynthetic potential in Gram-negative bacteria remains largely unexplored. Here, we report the discovery and activation of a cryptic type II PKS biosynthetic gene cluster from the myxobacterium Aggregicoccus edonensis MCy10622. Using a rapid PCR-based cloning and promot

Unraveling the allosteric mechanisms of prolyl endopeptidases for celiac disease therapy: Insights from molecular dynamics simulations.

Prolyl endopeptidases (PEP) from Sphingomonas capsulata (sc) and Myxococcus xanthus (mx) selectively degrade gluten peptides in vitro, offering a potential therapeutic strategy for celiac disease. However, the mechanisms governing the interaction of these enzymes with their substrates remain unclear. In this study, conventional molecular dynamics simulations with a microsecond timescale and targeted molecular dynamics simulations were performed to investigate the native states of mxPEP and scPEP enzymes, as well as their allosteric binding with a representative substrate, namely, Z-Ala-Pro-p-n

Metamorphic change in EP37 expression: members of the βγ-crystallin superfamily in newt.

EP37 is an epidermis-specific protein found in the developing embryo of the Japanese newt, Cynops pyrrhogaster. Our previous study predicted the presence of genes homologous to EP37, which show temporary shared expression at the turn of metamorphosis. In this study, we isolated and characterized three cDNAs encoding novel EP37 homologues; two from the skin of an adult newt and the other from swimming larva. Conceptual translation of the open reading frames of these cDNAs predicted proteins carrying βγ-crystallin motifs and putative calcium-binding sites, both of which are features shared by th

Engineering an Artificial Myxopyronin Derivative with Enhanced Metabolic Stability via Mutasynthesis.

The rise of multidrug-resistant pathogens, such as Staphylococcus aureus and Mycobacterium tuberculosis, underscores an urgent need for therapeutic innovation. The antibiotic development pipeline targeting these bacteria is critically limited, with most discovered candidates exhibiting structurally similar features of prominent chemical entities and with well-established molecular targets or binding modes. The myxobacterial α-pyrone antibiotics, myxopyronins, represent a highly promising compound class due to their ability to inhibit RNA polymerase by binding to the "switch region", a distinct

Speed switching of gonococcal surface motility correlates with proton motive force.

Bacterial type IV pili are essential for adhesion to surfaces, motility, microcolony formation, and horizontal gene transfer in many bacterial species. These polymers are strong molecular motors that can retract at two different speeds. In the human pathogen Neisseria gonorrhoeae speed switching of single pili from 2 µm/s to 1 µm/s can be triggered by oxygen depletion. Here, we address the question how proton motive force (PMF) influences motor speed. Using pHluorin expression in combination with dyes that are sensitive to transmembrane ΔpH gradient or transmembrane potential ΔΨ, we measured b

Mycoplasma pneumoniae, an underutilized model for bacterial cell biology.

In recent decades, bacterial cell biology has seen great advances, and numerous model systems have been developed to study a wide variety of cellular processes, including cell division, motility, assembly of macromolecular structures, and biogenesis of cell polarity. Considerable attention has been given to these model organisms, which include Escherichia coli, Bacillus subtilis, Caulobacter crescentus, and Myxococcus xanthus. Studies of these processes in the pathogenic bacterium Mycoplasma pneumoniae and its close relatives have also been carried out on a smaller scale, but this work is ofte

Immunization with Virus-Like Particles Displaying Ara h 2‑Derived Peptides to Mitigate Peanut Allergy.

Peanut allergy is caused by aberrant IgE-driven immune responses to a well-defined set of peptide sequences within allergenic proteins. Oral and sublingual desensitization therapies are associated with the elicitation of IgG antibodies against allergenic proteins that interfere with IgE-induced effects. We tested whether inducing peptide-specific IgG responses by displaying key epitopes on immunogenic protein nanoparticle platforms protected against peanut allergy. A panel of 10 IgE epitopes from Ara h 2, the major degranulation-inducing peanut allergen, were displayed on two virus-like partic

In vivo fluorescence imaging of exogenous enzyme activity in the gastrointestinal tract.

Exogenous enzymes are administered orally to treat several diseases, such as pancreatic insufficiency and lactose intolerance. Due to the proteinaceous nature of enzymes, they are subject to inactivation and/or digestion in the gastrointestinal (GI) tract. Here we describe a convenient fluorescence-based assay to monitor the activity of therapeutic enzymes in real time in vivo in the GI tract. To establish the proof of principle, the assay was applied to proline-specific endopeptidases (PEPs), a group of enzymes recently proposed as adjuvant therapy for celiac disease (a highly prevalent immun
