Non-invasive and accurate readout of superoxide anion in biological systems by near-infrared light.

Infectious and inflammatory diseases involve superoxide anion (O2•-) production. Real-time and non-invasive evaluation of O2•- in intact biological systems has been a significant challenge in biology and medicine. Here, I report that an advanced near-infrared chemiluminescent probe, MCLA-800, enables reliable non-invasive optical readout of O2•-ex vivo and in vivo. MCLA-800 allowed highly selective and sensitive monitoring of O2•- in undiluted human whole blood ex vivo. For the first time, the use of MCLA-800 revealed two reproducible types of O2•- production in response to stimulation by unop

Effect of IP6 on human neutrophil cytokine production and cell morphology.

Inositol hexaphosphate (IP6) has anti-cancer properties, but recently other extracellular functions have been observed for IP6, including enhancing superoxide production and phagocytosis by neutrophils in the presence of microbial stimuli. This study investigated other inflammatory functions of IP6 on adherent neutrophils. The effect of IP6 on the release of IL-8, tumour necrosis factor (TNF-alpha) and IL-6 by neutrophils attached to either plastic or laminin for up to 6 hours in response to stimulation with lipopolysaccharide or N-formyl-Met-Leu-Phe (fMLP) was investigated. An increase in IL-

Association of mental disorders with sepsis: a bidirectional Mendelian randomization study.

BACKGROUND: Substantial research evidence supports the correlation between mental disorders and sepsis. Nevertheless, the causal connection between a particular psychological disorder and sepsis remains unclear. METHODS: For investigating the causal relationships between mental disorders and sepsis, genetic variants correlated with mental disorders, including anorexia nervosa (AN), attention-deficit hyperactivity disorder (ADHD), autism spectrum disorder (ASD), bipolar disorder (BD), major depressive disorder (MDD), obsessive-compulsive disorder (OCD), panic disorder (PD), posttraumatic stres

[The use of sustained-release preparations for eye-platelet diagnosis for noninvasive determination of leukocyte migration in the eye].

An in vivo method for measuring leukocyte migration in man involves the insertion of membrane filter platelets into the lower conjunctival sac. Before insertion, the filter platelets were soaked in solutions of chemotactic test substances and dried. The chemotactic gradient formed by a dissolving test substance stimulates leukocytes from the conjunctival mucosa to immigrate into a filter platelet. The number and the distribution of immigrant cells within the filter platelet yield information on pathological processes. To obtain reliable results, the chemotactic substance used has to show a def

Movement of polymorphonuclear leukocytes: an elastimetry analysis.

By utilizing the technique of cell elastimetry, which measures the negative pressure required to aspirate human polymorphonuclear leukocytes (PMNs) into a micropipette, a correlation between PMN deformability and cellular movement is confirmed. The effects of the synthetic chemotactic peptide, N-formyl-methionyl-leucyl-phenylalanine (f-met-leu-phe) on the deformability and chemotaxis of PMNs have been investigated, and a relationship between peptide concentrations and PMN deformability, which parallels the effects of the peptide on filter (chemotactic) studies, has been established. Experiment

Concentration-dependent regulatory effects of prostaglandin E1 on human neutrophil function in vitro.

Prostaglandin E1 (PGE1) has recently been used clinically as a purported modulator of activated neutrophils in certain forms of the adult respiratory distress syndrome (ARDS). We now report that PGE1 does not have a uniform inhibitory effect upon human neutrophil functions in vitro. Cells were first pretreated with PGE1 followed by incubation with either N-formyl-methionyl-leucyl-phenylalanine (FMLP), phorbol myristate acetate (PMA), or C5a. Lysosomal enzyme release (myeloperoxidase, lysozyme), superoxide anion generation, and chemotaxis were then quantitated. PGE1 alone lacked any appreciable

Increased sensitivity of the chemoattractant-induced chemiluminescence in eosinophils isolated from atopic individuals.

Eosinophilia in peripheral blood is a common phenomenon associated with atopic condition. The present study illustrates the enhanced sensitivity of these eosinophils for formyl-methionyl-leucylphenylalanine (FMLP) and platelet-activating-factor (PAF) (measured as luminol-enhanced chemiluminescence) compared with eosinophils isolated from the peripheral blood of normal individuals. The PAF-induced chemiluminescence, which is peroxidase dependent and of extracellular origin, is inhibited by the PAF antagonist BN52021. The enhanced sensitivity of eosinophils isolated from atopic patients for FMLP

Polymorphonuclear leukocyte chemotaxis in generalized pustular psoriasis.

We studied polymorphonuclear leukocyte (PMNL) chemotaxis in 3 patients with generalized pustular psoriasis using 3 chemoattractants: Zymosan-activated serum from both the patients and normal individuals, and n-formyl-methionyl-leucyl-phenylalanine (FMLP). These attractants were assayed against both patient-derived and normal PMNLs. All patients had quiescent skin disease at the time of assay. We found that all patients had decreased PMNL chemotaxis to autologous Zymosan-activated serum, but not to FMLP. PMNLs from normal individuals responded normally to serum from the patients. We conclude th

Optimization of the Transwell® assay for the analysis of neutrophil chemotaxis using flow cytometry to refine the clinical investigation of immunodeficient patients.

Chemotaxis is the directed movement of neutrophils towards an infected site. This physiological process can be reproduced using a modified Boyden chamber, such as the Transwell® support. Different techniques can be used to count neutrophils after migration to the lower chamber of the holder. The present study supports the use of an optimized Transwell® assay coupled with a flow cytometry-based method (Sysmex XN-9000) to detect chemotaxis abnormalities. A reference interval of neutrophil's chemotaxis was determined as part of this work. A first step involves the extraction of neutrophils from w

Altered diacylglycerol level and metabolism in neutrophils from patients with localized juvenile periodontitis.

Diacylglycerol, a physiological activator of protein kinase C, was elevated nearly twofold in unstimulated peripheral blood neutrophils from patients with localized juvenile periodontitis compared with cells from normal individuals. These cells also showed an enhanced and prolonged elevation of diglyceride in response to N-formylmethionylleucylphenylalanine. The metabolism of a cell-permeant diacylglycerol by diglyceride kinase was significantly decreased, because of a fivefold or higher elevation in the apparent Km of cellular diglyceride kinase.

Retinoic acid induces hypersegmentation and enhances cytotoxicity of neutrophils against cancer cells.

Hypersegmentation of nuclei is considered a distinct characteristic of the antitumoral phenotype of neutrophils. Retinoic acid, a metabolite of retinol, reorganizes and induces segmentation of the nucleus during the differentiation of neutrophils. However, the role of retinoic acid in the phenotype polarization of neutrophils has not been fully established. Here, we investigated the effect of retinoic acid on phenotype polarization of neutrophils. Retinoic acid-induced the hypersegmentation of human neutrophils via retinoic acid receptors and mTOR pathways. Retinoic acid-induced hypersegmented

Recurrent infection in glycogenosis type Ib: abnormal neutrophil motility related to impaired redistribution of adhesion sites.

Neutrophil function was investigated in a male child with glycogenosis type Ib who demonstrated susceptibility to staphylococcal infections and neutropenia. Random motility and directed migration of the patient's neutrophils in vitro were profoundly diminished. The patient's neutrophils stimulated in suspension with chemotactic factors (CFs) generated chemiluminescence that was comparable to or greater than that generated by neutrophils from controls, but the patient's neutrophils failed to assume a normal bipolar configuration in response to chemotactic stimuli. They also failed to demonstrat

Circulating N-lactoyl-amino acids and N-formyl-methionine reflect mitochondrial dysfunction and predict mortality in septic shock.

INTRODUCTION: Sepsis is a highly morbid condition characterized by multi-organ dysfunction resulting from dysregulated inflammation in response to acute infection. Mitochondrial dysfunction may contribute to sepsis pathogenesis, but quantifying mitochondrial dysfunction remains challenging. OBJECTIVE: To assess the extent to which circulating markers of mitochondrial dysfunction are increased in septic shock, and their relationship to severity and mortality. METHODS: We performed both full-scan and targeted (known markers of genetic mitochondrial disease) metabolomics on plasma to determine

Diosgenin inhibits superoxide generation in FMLP-activated mouse neutrophils via multiple pathways.

Diosgenin possesses anti-inflammatory and anticancer properties. Activated neutrophils produce high concentrations of the superoxide anion which is involved in the pathophysiology of inflammation-related diseases and cancer. In the present study, the inhibitory effect and possible mechanisms of diosgenin on superoxide generation were investigated in mouse bone marrow neutrophils. Diosgenin potently and concentration-dependently inhibited the extracellular and intracellular superoxide anion generation in Formyl-Met-Leu-Phe (FMLP)- activated neutrophils, with IC50 values of 0.50 ± 0.08 μM and 0.

Effects of tenoxicam on superoxide anion formation, beta-glucuronidase release and fMLP binding in human neutrophils: comparison with other NSAIDs.

Non-steroidal anti-inflammatory drugs (NSAIDs) are considered to exert their activity by interfering with the generation of arachidonate metabolites in various cells, mainly in neutrophils and monocytes. The inhibition of cellular cyclooxygenase enzyme, however, does not always correlate with the in vivo activity of these drugs. Recent evidence indicates that several NSAIDs may interfere with the stimulus-response coupling of inflammatory cells. In this study, the effects of tenoxicam, an oxicam derivative with a thienothiazine structure, on neutrophil activation were evaluated by the assessme

The role of G protein-coupled receptor in neutrophil dysfunction during sepsis-induced acute respiratory distress syndrome.

Sepsis is defined as a life-threatening dysfunction due to a dysregulated host response to infection. It is a common and complex syndrome and is the leading cause of death in intensive care units. The lungs are most vulnerable to the challenge of sepsis, and the incidence of respiratory dysfunction has been reported to be up to 70%, in which neutrophils play a major role. Neutrophils are the first line of defense against infection, and they are regarded as the most responsive cells in sepsis. Normally, neutrophils recognize chemokines including the bacterial product N-formyl-methionyl-leucyl-p

Antimicrobial peptides from scorpion venom induce Ca(2+) signaling in HL-60 cells.

Parabutoporin (PP) and opistoporin 1 (OP1) are amphipathic alpha-helical antimicrobial peptides that were recently isolated from scorpion venom. In assays in which single granulocyte-like HL-60 cells as well as cells in suspension were used, both peptides were able to induce a reversible Ca(2+) release from intracellular stores and to increase Ca(2+) influx. Both effects could be clearly differentiated for OP1, inducing Ca(2+) release at lower concentrations. The Ca(2+) release was pertussis toxin-sensitive indicating the involvement of G-proteins. Ca(2+) release depended on the stage of diffe

Diacylglycerol accumulation and superoxide anion production in stimulated human neutrophils.

Exogenous diacylglycerols stimulate neutrophil superoxide anion production, suggesting that endogenous diacylglycerols may function as second messengers for this biological response. We have measured the diacylglycerol mass in human neutrophils stimulated by fMet-Leu-Phe, ionomycin, and concanavalin A and have correlated the kinetics and magnitude of the diacylglycerol response with those for superoxide anion production. For each stimulus, no increase in diacylglycerol mass was detected prior to the onset of superoxide anion generation. However, large sustained increases in diacylglycerol conc

The effect of six prostaglandins, prostacyclin and iloprost on generation of superoxide anions by human polymorphonuclear leukocytes stimulated by zymosan or formyl-methionyl-leucyl-phenylalanine.

Prostaglandins (PG) E2,E1,6-keto-E1 and D2 at concentrations of 0.15-0.80 microM inhibited by 25% the generation of superoxide anions (O2-) in human polymorphonuclear leukocytes (PMNs) stimulated with formyl-methionyl-leucyl-phenylalanine (FMLP). The potency of that inhibition by either PGD2 or PGE1 was the same when zymosan was used as a stimulator whereas PGE2 and 6-keto-PGE1 were by 13 and 21 times less potent inhibitors of O2-) in zymosan-stimulated as compared to FMLP-activated PMNs. PGF2 alpha inhibited the generation of O2- by activated PMNs only when used at the highest concentration s
