The effect of naftidrofuryl on red blood cell aggregation detected in vitro with ultrasound.

Red blood cell aggregates are mainly responsible for the echogenicity of flowing blood. Thus, ultrasound can be used to observe the degree of red blood cell aggregation in slow flow conditions. We quantified blood echogenicity to study aggregation tendency of red blood cells in blood of patients with claudication, patients with suspected venous thrombosis, and normal volunteers without and with in vitro addition of naftidrofuryl (10(-6) M). Normal volunteers showed lower original echogenicity than any group of patients, and claudication patients showed the highest echogenicity. Naftidrofuryl c

Efficacy of naftidrofuryl in patients with vascular or mixed dementia: results of a multicenter, double-blind trial.

BACKGROUND: Dementia is a cerebral disorder resulting in a progressive deterioration of intellectual function that compromises the patient's ability to function. The diagnostic criteria for dementia are primarily clinical and are based on the Diagnostic and Statistical Manual of Mental Disorders, Fourth Edition. The Hachinski score and computed tomography of the brain help distinguish between degenerative and vascular dementias. OBJECTIVE: This study examined the efficacy of naftidrofuryl in patients with vascular or mixed dementia. METHODS: This multicenter, randomized, double-blind study c

Drugs affecting the inner ear. A review of their clinical efficacy, mechanisms of action, toxicity, and place in therapy.

Many drugs have a site of action within the inner ear. The list includes therapeutic, diagnostic and ototoxic agents. Therapeutic agents are most useful in cases of infections, endolymphatic hydrops, vascular insufficiency, vertigo of peripheral origin, autoimmune disease, otosclerosis (otospongiosis), sudden hearing loss and tinnitus. For infections, the most widely used anti-microbial agents are the penicillins and cephalosporins. There are no antiviral agents that have been proven useful for inner ear viral infections. However, steroids have been of some value for controlling some of the se

[Consequences of obliterating arteriopathy of the legs on the professional activity of patients and external assistance].

Peripheral arterial occlusive disease of the lower limbs results in increased risks of cardiovascular and cerebrovascular morbidity and mortality. However, peripheral arterial disease also has an incapacitating effect upon patients' daily lives, and indeed can have considerable consequences upon their quality of life as well as upon their professional activities. The purpose of this study was to describe patients in terms of their professional activities, the consequences of the disease upon such activities, need for external assistance, and the effects of treatment over a six-month treatment

Naftidrofuryl oxalate improves impaired brain glucose metabolism after microsphere-induced cerebral embolism in rats.

The present study was designed to elucidate possible therapeutic effects of naftidrofuryl on the brain glucose metabolism after cerebral ischemia. Cerebral ischemia was induced by injecting 680 microspheres with a diameter of 48 microns into the right internal carotid artery of the rat. After ensuring the onset of symptoms of stroke on the first day after the operation, the rats were treated with intraperitoneal injections of 15 mg/kg naftidrofuryl oxalate twice a day. The behavioral and metabolic changes of operated rats were monitored up to the 5th day after surgery. The symptoms gradually f

[Allergic reaction in therapy with naftidrofuryl (Dusodril). A case report].

There are many drugs marketed for the purpose of altering vascular blood flow in various regions, especially of the central nervous system and in peripheral arterial insufficiency. More than 50 different methods are described for the treatment of sudden deafness. Considerations of the therapy of sudden deafness are influenced by the fact that the cause of the disease is unknown. The dysfunction of the hair-cells of the organ of CORTI is thought to be caused by a deficit of oxygen due to disorders of micro-circulation in the inner ear. The infusion of vaso-active drugs in the early state of dis

Intravenous naftidrofuryl for critical limb ischaemia.

BACKGROUND: Peripheral arterial disease affects five per cent of men and women by late middle age. Approximately 25% of those affected will develop critical limb ischaemia (rest pain, ulceration and gangrene) within five years. Naftidrofuryl is a vasoactive drug which may be beneficial in the treatment of critical limb ischaemia. OBJECTIVES: To determine whether naftidrofuryl, when administered intravenously, is effective in alleviating symptoms and reducing progression of disease in patients with critical limb ischaemia. SEARCH METHODS: The Cochrane Peripheral Vascular Diseases Group Trials

The effect of oxygen inhalation and intravenous naftidrofuryl on the transcutaneous partial oxygen pressure in ischaemic lower limbs.

The effect of oxygen inhalation at atmospheric pressure and Naftidrofuryl infusion (N) on the TCpO2 is shown. At the central control site--5 cm below the midclavicular line--oxygen inhalation produced a significant increase in TCpO2, whereas there was no change after Naftidrofuryl infusion. At the 10 cm below-knee site, there were significant rises after oxygen inhalation alone, Naftidrofuryl alone and both combined. The study was conducted on 20 patients (23 legs). It is suggested that this study can form the basis for a regimen to improve the viability of ischaemic limbs showing borderline T

Meniere's disease: the selection and assessment of patients for surgery using electrocochleography.

Transtympanic electrocochleography has been used to measure the action potential (AP), summating potential (SP), and cochlear microphonic (CM) in patients with Menière's disease. In this condition the widening of the AP/SP complex is thought to be due to an enhanced negative SP which shows no adaptation and this may be the result of an increased asymmetry produced by endolymphatic hydrops. Measurements have also been made using two groups of agents -- those designed to increase the cochlear blood flow (the metabolic activator naftidrofuryl and inhalations of carbon dioxide) and those designed

Praxilene (naftidrofuryl oxalate) as an alternative for the augmentation of femoro-distal bypass blood flow.

In 30 patients undergoing femoro-distal bypass the effect of papaverine and praxilene on blood flow in the graft was measured. The mean resting flow was 129 ml min-1 (range: 91-167) and after papaverine was 202 ml min-1 (142-262) and after praxilene was 205 ml min-1 (143-267). Praxilene has a similar effect to papaverine in the augmentation of blood flow, and further investigation is needed to see if long-term praxilene administration might improve graft survival.

Study on the anti-hypoxic effect of cinnarizine and its interaction with prostacyclin.

The anti-hypoxic effect of cinnarizine was studied using the following experimental methods: hypobaric and anoxic hypoxia in mice, complete ischemia by decapitation in mice and hemic hypoxia in rats. Papaverine, xanthinol nicotinate and naftidrofuryl were used as reference drugs. In hypobaric and anoxic hypoxia the interaction of cinnarizine with the effect of prostacyclin (PGI2) was investigated. Cinnarizine showed an anti-hypoxic effect in all the methods used. It was more effective in hypobaric and anoxic hypoxia, in incomplete ischemia by decapitation, and less effective in hemic hypoxia.

Prostanoids for critical limb ischaemia.

BACKGROUND: Peripheral arterial occlusive disease (PAOD) is a common cause of morbidity and mortality due to cardiovascular disease in the general population. Although numerous treatments have been adopted for patients at different disease stages, no option other than amputation is available for patients presenting with critical limb ischaemia (CLI) unsuitable for rescue or reconstructive intervention. In this regard, prostanoids have been proposed as a therapeutic alternative, with the aim of increasing blood supply to the limb with occluded arteries through their vasodilatory, antithrombotic

[Effect of naftidrofuryl (Praxilene) in primary open angle glaucoma: prospective double-blind study].

The positive effect of Naftidrofuryl (Praxilène), a specific serotonine-S2 receptor antagonist, has already been described in the treatment of normal pressure glaucoma. Despite the fact that in POAG intraocular pressure (IOP) is the primary ethiopathogenic mechanism, vascular factors could also influence the progression of the disease, especially in old patients with cardiovascular risk factors. We have studied the effect of Naftidrofuryl versus placebo in a randomised double-blind study on 42 POAG patients. Experimental and clinical study of the effect of naftidrofuryl on the recovery from peripheral nerve lesions.

After primary repair of lesions of nerves and vessels, patients who were administered a vasoactive drug, naftidrofuryl, postoperatively, had a better functional recovery than untreated patients. Evidence for the favourable effect of the drug on nerve regeneration was first obtained from experiments in the rat. The sciatic nerve was transected and repaired with standard microsurgical techniques. Results were evaluated by electromyography and histology. Data indicated that treated animals had better motor responses and distal latency than untreated rats. Fibre counts showed a greater number of n

Incidentally diagnosed melorheostosis of upper limb: case report.

BACKGROUND: Melorheostosis is quite a rare bone disease with still unclear ethiology. Although multifocal affection is highly debilitating with unfavorable prognosis, there is no clear consensus about therapeutical approach. There is still insufficient evidence in the literature for almost a century after the first description. Affected bone has a typical appearance of melting wax. Diagnosis is usually incidental with pain as a leading symptom. Diagnosis itself is relatively easy, routine X-ray examination is sufficient. Even though it could be easily overlooked and mistaken with other disease

[Prognostic criteria in sudden deafness].

The present study was devised to determine whether the pattern of hearing loss has any meaning for prognosis in patients with sudden hearing loss. The audiograms and clinical records of 145 patients with sudden deafness between 1991 and 1993 were reviewed. Forty-eight of the patients were females and 97 were males. All patients had been hospitalized and treated with intravenous infusions of naftidrofuryl. The best recovery rate was seen in patients with low-tone hearing losses (68.8% complete and 25% partial recoveries), whereas in patients with high-tone and flat losses the recovery rates wer

Fatal intoxication with naftidrofuryl.

A 52-year-old man was found dead in his bed. He had financial and psychosocial problems like separation from his wife and children or unemployment due to alcoholism. Under treatment of disulfiram he was presently abstinent from alcohol. As he had suffered from epileptic seizures and dizziness, he received valproic acid and the vasodilator naftidrofuryl, respectively. Autopsy showed no morphologic cause of death. Chemical analysis of blood revealed concentrations for valproic acid and disulfiram in the therapeutic and above the therapeutic range but far below the lethal level, respectively. No
