Nanogels for bone tissue engineering - from synthesis to application.

Nanogels are cross-linked hydrogel nanoparticles with a three-dimensional, tunable porous structure that merges the best features of hydrogels and nanoparticles, including the ability to retain their hydrated nature and to swell and shrink in response to environmental changes. Nanogels have attracted increasing attention for use in bone tissue engineering as scaffolds for growth factor transport and cell adhesion. Their three-dimensional structures allow the encapsulation of a wide range of hydrophobic and hydrophilic drugs, enhance their half-life, and impede their enzymatic breakdown in vivo

Ferrocene/ β-cyclodextrin based supramolecular nanogels as theranostic systems.

A supramolecular redox responsive nanogel (NG) with the ability to sense cancer cells and loaded with a releasing therapeutic agent was synthesized using hostguest interactions between polyethylene glycol-grafted-β-cyclodextrin and ferrocene boronic acid. Cyclic voltammetry matched with other spectroscopy and microscopy methods provided strong indications regarding host-guest interactions and formation of the NG. Moreover, the biological properties of the NG were evaluated using fluorescence silencing, confocal laser scanning microscopy, and cell toxicity assays. Nanogel with spherical core-sh

Investigating the Effects of Nanogels in Promoting Protein Crystallization.

X-ray crystallography is still the most widely used and versatile method for structural studies of biological macromolecules. This study concerns the application of nanogels to facilitate protein crystallization, a prerequisite for X-ray crystallography. Nanogels (NGs) are nano-sized, highly crosslinked polymeric particles that have been extensively studied for chemical catalysis and drug delivery but not for protein crystal nucleation. The efficacy of six types of nanogels (three N-isopropylacrylamide-based and three acrylamide-based) was tested, with promising results. They were subsequently

Hydrophobic interactions enhance doxorubicin delivery from hyaluronic acid nanogels.

This study introduces a novel targeted chemotherapy approach employing biocompatible nanogels engineered from hyaluronic acid (HA) and pitaya-derived DNA. Both polymers were functionalized with aldehyde groups, enabling crosslinking and subsequent encapsulation of doxorubicin (DOX). The incorporation of DNA significantly enhanced DOX loading, achieving a remarkable encapsulation efficiency of 93.3%. Notably, the integration of a hydrophobic 4,4'-dithiodianiline (DTD) linker facilitated controlled drug release within the reductive tumor microenvironment, with 36% of DOX released within 24 h in

Surfactant-Free Emulsion-Based Preparation of Redox-Responsive Nanogels.

A surfactant-free emulsion-based approach is developed for preparation of nanogels. A water-in-oil emulsion is generated feasibly from a mixture of water and a solution of disulfide-containing hyperbranched PEGylated poly(amido amine)s, poly(BAC2-AMPD1)-PEG, in chloroform. The water droplets in the emulsion are stabilized and filled with poly(BAC2-AMPD1)-PEG, and the crosslinked poly(amido amine)s nanogels are formed via the intermolecular disulfide exchange reaction. FITC-dextran is loaded within the nanogels by dissolving the compound in water before emulsification. Transmission electron mic

Nanogels as pharmaceutical carriers: finite networks of infinite capabilities.

Nanogels are swollen nanosized networks composed of hydrophilic or amphiphilic polymer chains. They are developed as carriers for the transport of drugs, and can be designed to spontaneously incorporate biologically active molecules through formation of salt bonds, hydrogen bonds, or hydrophobic interactions. Polyelectrolyte nanogels can readily incorporate oppositely charged low-molecular-mass drugs and biomacromolecules such as oligo- and polynucleotides (siRNA, DNA) as well as proteins. The guest molecules interact electrostatically with the ionic polymer chains of the gel and become bound

Cyclodextrin-Based Nanogels with Chemistry-Tunable Intracellular Stability and Distribution.

INTRODUCTION: Cyclodextrin-based nanogels (CD-nGels) uniquely enable encapsulation of hydrophobic pharmaceuticals within hydrophilic networks through host-guest interactions, thereby improving drug solubility, stability, and therapeutic efficacy. METHODS: Here, we report the surfactant-free synthesis of Nile Blue-labeled fluorescent CD-nGels with tunable hydrolytic properties by integrating amide or ester linkages in both β-cyclodextrin (βCD) moieties and crosslinking sites. Three formulations were prepared with progressively increasing hydrolytic sensitivity: NG1 (fully amide-linked), NG2 (e

Celastrol-Loaded Zwitterionic Nanogels Trigger Pyroptosis for Potent Antitumor Effects.

Pyroptosis has recently emerged as a promising therapeutic strategy owing to its unique advantages in inducing lytic cell death and eliciting immunogenic responses. Moderate induction of pyroptosis not only directly eliminates cancer cells but also activates antitumor immunity through the release of tumor-associated antigens and proinflammatory cytokines. However, most available pyroptosis inducers suffer from poor solubility, instability, and nonspecific biodistribution, which significantly hinder their translational potential. To address these challenges, we developed a pyroptosis-inducing d

Multifunctional chitin nanogels for simultaneous drug delivery, bioimaging, and biosensing.

In this work, we developed biodegradable chitin nanogels (CNGs) by controlled regeneration method. For multifunctionalization, we have conjugated CNGs with MPA-capped-CdTe-QDs (QD-CNGs) for the in vitro cellular localization studies. In addition, the Bovine Serum Albumin (BSA) was loaded on to QD-CNGs (BSA-QD-CNGs). The CNGs, QD-CNGs, and BSA-QD-CNGs were well-characterized by SEM and AFM, which shows that the nanogels are in the range of <100 nm. These were further characterized by FT-IR and Cyclic Voltametry. The cytocompatibility assay showed that the nanogels are nontoxic to L929, NIH-3T3,

Carbonization of quercetin into nanogels: a leap in anticoagulant development.

Quercetin, a flavonoid abundantly found in onions, fruits, and vegetables, is recognized for its pharmacological potential, especially for its anticoagulant properties that work by inhibiting thrombin and coagulation factor Xa. However, its clinical application is limited due to poor water solubility and bioavailability. To address these limitations, we engineered carbonized nanogels derived from quercetin (CNGsQur) using controlled pyrolysis and polymerization techniques. This led to substantial improvements in its anticoagulation efficacy, water solubility, and biocompatibility. We generated

Extracellular vesicle-mimetic nanogels: overcoming barriers in advanced breast cancer.

Breast cancer remains one of the most common malignancies among women worldwide. Conventional treatment strategies including surgery, adjuvant/neoadjuvant chemotherapy, and targeted therapies, are often associated with severe side effects, poor selectivity, low drug solubility, and high recurrence rates. Nanogel-based drug delivery systems have emerged as promising platforms in precision oncology due to their ability to enhance drug solubility, improve bioavailability, and facilitate targeted delivery through enhanced permeability and retention (EPR) effects. This review focuses on stimuli-res

Mitochondria-Targeted Zwitterionic Nanogels Trigger Photopyroptosis for Enhanced Cancer Therapy.

Precise induction of pyroptosis remains a formidable challenge in cancer therapy due to the stringent requirements for spatiotemporal and subcellular control. Herein, we report a zwitterionic nanogels platform (poly(N-oxide dimethylaminoethyl methacrylate), PODMMA) that enables precise induction of photopyroptosis via intrinsic mitochondrial targeting. Owing to its unique N-oxide zwitterionic structure, the PODMMA nanogels exhibit prolonged blood circulation, selective mitochondrial localization without the need for additional targeting ligands, and enhanced tumor accumulation.
