In vitro activity of novel antifungals, natamycin, and terbinafine against Fusarium.

The genus Fusarium includes several trans-kingdom pathogens, infecting plants, animals, and humans, with widespread occurrence. Invasive Fusarium infections are often destructive due to their prevalence in critically ill patients. Furthermore, severe superficial infections, like keratitis or endophthalmitis with potential loss of vision, are rising in incidence globally. Infections are difficult to treat due to the intrinsic resistance of Fusarium against echinocandins and often high minimal inhibitory concentrations (MICs) for azoles. In this article, we assessed the in vitro activity of the

Improved Topical Ophthalmic Natamycin Suspension for the Treatment of Fungal Keratitis.

Purpose: Natamycin (NT) is used as a first-line antifungal prescription in the treatment of fungal keratitis (FK) and is commercially available as a 5% w/v ophthalmic suspension. NT shows poor water solubility and light sensitivity. Thus, the present investigation is aimed to enhance the fraction of NT in solution in the commercial formulation by adding cyclodextrins (CDs), thereby improving the delivery of the drug into deeper ocular tissues. Methods: The solubility of NT in different CDs, the impact of ultraviolet (UV) light exposure, stability at 4°C and 25°C, in vitro release, and ex vivo

The combined utilization of Chlorhexidine and Voriconazole or Natamycin to combat Fusarium infections.

BACKGROUND: Fusarium species are the fungal pathogens most commonly responsible for the mycotic keratitis, which are resistant to the majority of currently available antifungal agents. The present study was designed to assess the efficacy of a combination of low doses chlorhexidine with two other commonly used drugs (voriconazole and natamycin) to treat Fusarium infections. RESULTS: We utilized combinations of chlorhexidine and natamycin or voriconazole against 20 clinical Fusarium strains in vitro using a checkerboard-based microdilution strategy. In order to more fully understand the synerg

Multiple transporters are involved in natamycin efflux in Streptomyces chattanoogensis L10.

Antibiotic-producing microorganisms have evolved several self-resistance mechanisms to prevent auto-toxicity. Overexpression of specific transporters to improve the efflux of toxic antibiotics has been found one of the most important and intrinsic resistance strategies used by many Streptomyces strains. In this work, two ATP-binding cassette (ABC) transporter-encoding genes located in the natamycin biosynthetic gene cluster, scnA and scnB, were identified as the primary exporter genes for natamycin efflux in Streptomyces chattanoogensis L10. Two other transporters located outside the cluster,

Utilizing confocal microscopy and topical natamycin in metarhizium keratitis of Caribbean origin.

A 32-year-old contact lens-wearing man with recent travel history to the Caribbean was referred for a corneal infiltrate in the left eye that worsened following 1-week of steroid-antibiotic therapy. Corneal cultures were obtained and sent to our facility's clinical microbiology laboratory for analysis. Same-day in vivo confocal microscopy revealed fungal elements. Nucleic acid sequencing performed on the isolated determined it to be a member of the entomopathogenic genus Metarhizium. Over the course of 3 months, the patient's corneal infiltrate ultimately resolved following topical natamycin 5

Efficacy and toxic action of the natural product natamycin against Sclerotinia sclerotiorum.

BACKGROUND: Sclerotinia stem rot caused by Sclerotinia sclerotiorum seriously endangers oilseed rape production worldwide, and the occurrence of fungicide-resistant mutants of S. sclerotiorum leads to control decline. Thus, it is critical to explore new green substitutes with different action mechanisms and high antifungal activity. Herein, the activity and the action mechanism of natamycin against S. sclerotiorum were evaluated. RESULTS: Natamycin showed potent inhibition on the mycelial growth of S. sclerotiorum, and half-maximal effective concentration (EC50 ) values against 103 S. sclerot

Platelet membrane-camouflaged PLGA loaded natamycin improve the prognosis of fungal keratitis.

Fungal keratitis (FK) is a severe infectious corneal disease and a common cause of blindness. At present, natamycin (NATA) is the most commonly prescribed drug for fungal keratitis. However, these disadvantages, including poor water solubility, poor stability, and significant corneal irritation, limit its effect in clinical application. In this study, we innovatively prepare platelet membranes (PLTm) which are a unique population of cellular fragments that adhere to a variety of pathogens camouflaged with fabulous biocompatibility poly (lactic-co-glycolic acid) (PLGA) loaded NATA. PLTm can hel

Two Cases of Curvularia geniculata Keratitis Successfully Treated with Natamycin-Based Therapy.

This report describes two cases of Curvularia geniculata keratitis, a rare form of fungal keratitis successfully managed with natamycin-based therapy. Both patients presented with characteristic feathery corneal infiltrates following ocular trauma. In vivo confocal microscopy and direct microscopy revealed septate filamentous fungi, and the isolates were definitively identified as C. geniculata through sequence analysis of the translation elongation factor 1-alpha (tef1-α) gene. Antifungal susceptibility testing showed sensitivity to natamycin at 2 μg/mL for both isolates, with variable sensit

Integrated omics analyses reveal natamycin disrupts oxidative phosphorylation pathways in Magnaporthe oryzae.

Magnaporthe oryzae, the causative agent of rice blast, is a devastating fungal pathogen that threatens global rice production. While natamycin is a known antimicrobial compound of microbial origin, its efficacy and precise mode of action against M. oryzae have remained largely unexplored. In this study, treatment with various concentrations of natamycin potently inhibited mycelial growth by 16.43% to 100%, with an EC₅₀ value of 11.77 mg/L. Pathogenicity of M. oryzae was also markedly reduced, declining to 4.1% of the control level under both in vitro and greenhouse conditions. To elucidate the

Therapeutic effect of oxidized bletilla striata polysaccharide-natamycin eye drops on fungal keratitis.

OBJECTIVE: Fungal keratitis (FK) usually develops to a poor clinical prognosis due to the fungal invasion and excessive inflammatory reaction. In order to enhance the therapeutic effect of natamycin (NAT), we used the anti-inflammatory biological polysaccharide bletilla striata polysaccharide (BSP) combined with NAT to prepare a new eye drop -- oxidized bletilla striata polysaccharide-natamycin (OBN). METHODS: UV-vis, FT-IR, and fluorescence spectroscopy were used to identify the synthesis of OBN. Biocompatibility of OBN was determined by CCK-8, scratch assay, and corneal toxicity test. RAW26

Functional properties and antifungal activity of films based on gliadins containing cinnamaldehyde and natamycin.

Gliadin films cross-linked with cinnamaldehyde (1.5, 3, and 5%) and incorporated with natamycin (0.5%) were prepared by casting, and their antifungal activity, water resistance, and barrier properties were characterized. Incorporation of natamycin gave rise to films with greater water uptake, weight loss and diameter gain, and higher water vapor and oxygen permeabilities. These results may be associated to a looser packing of the protein chains as a consequence of the presence of natamycin. The different cross-linking degree of the matrices influenced the natamycin migration to the agar test m

Natamycin sequesters ergosterol and interferes with substrate transport by the lysine transporter Lyp1 from yeast.

Natamycin is a polyene macrolide, widely employed to treat fungal keratitis and other yeast infections as well as to protect food products against fungal molds. In contrast to other polyene macrolides, such as nystatin or amphotericin B, natamycin does not form pores in yeast membranes, and its mode of action is not well understood. Here, we have employed a variety of spectroscopic methods, computational modeling, and membrane reconstitution to study the molecular interactions of natamycin underlying its antifungal activity. We find that natamycin forms aggregates in an aqueous solution with s

Gellan Gum Based Sol-to-Gel Transforming System of Natamycin Transfersomes Improves Topical Ocular Delivery.

Short precorneal residence time and poor transocular membrane permeability are the major challenges associated with topical ocular drug delivery. In the present research, the efficiency of the electrolyte-triggered sol-to-gel-forming system of natamycin (NT) transfersomes was investigated for enhanced and prolonged ophthalmic delivery. Transfersomes were optimized by varying the molar ratios of phospholipid, sorbitan monostearate (Span) and tocopheryl polyethylene glycol succinate (TPGS). NT transfersome formulations (FNs) prepared with a 1:1 molar ratio of phospholipid-to-Span and low levels
