The polyacrylamide-gel disc electrophoresis has been applied to the fractionation of 27 samples of unconcentrated urine obtained from the patients with monoclonal gammopathy. It was shown that 44% of the whole Bence Jones protein bands examined were detected in the area between 0.70 and 0.90 expressed in terms of relative migration based on transferrin. The method provided a satisfactory separation of native urine proteins with good reproducibility and few technical difficulties. It may become an efficient tool in clinical chemistry.

In this study we tested the hypothesis that reduced myofibrillar ATPase activities in end-stage heart failure are associated with a redistribution of myosin isozymes. Lysates of barley chloroplasts release more radioactivity into acid soluble form from UV-irradiated and alkylated-depurinated E. coli [3H] DNA than from intact DNA. By means of affinity chromatography on depurinated DNA-cellulose and/or UV irradiated DNA-cellulose and by electrophoresis in polyacrylamide gels, four activities on depurinated DNA were separated. A major 50-kDa soybean agglutinin (SBA)-reactive component present in extracts of Schistosoma mansoni eggs was isolated by SBA lectin affinity chromatography. In polyacrylamide gel electrophoresis (PAGE), the SBA-reactive component was seen as a 100-kDa polypeptide band that after reduction and alcylation was substituted by a 50-kDa component. This suggests that it occurs in native form as a dimer. Monoclonal antibodies produced against gp50 reacted with miracidial and cercarial secretions and with adult worm components including tegumental structures suggestive of a secretory function.

An autoantibody to liver cytosol was previously described in childhood autoimmune chronic active hepatitis type 2. The antigen, liver cytosol antigen type 1, was for the first time partially purified using gel filtration and ion exchange chromatography, and it was characterized using immunodiffusion, immunoblot and sodium dodecyl sulfate-polyacrylamide gel electrophoresis analysis of the immunoprecipitate. Acetate kinase (ATP:phosphotransferase, EC 2.7.2.1) has been purified 294-fold from acid-producing cells of Clostridium acetobutylicum DSM 1731 to a specific activity of 1087 U mg-1 (ADP-forming direction).
