BACKGROUND: Nefopam is a non-opioid, non-steroidal analgesic drug with fewer adverse effects than narcotic analgesics and nonsteroidal anti-inflammatory drugs, and is widely used for postoperative pain control. Because nefopam sometimes causes side effects such as nausea, vomiting, somnolence, hyperhidrosis and injection-related pain, manufacturers are advised to infuse it slowly, over a duration of 15 minutes. Nevertheless, pain at the injection site is very common. Therefore, we investigated the effect of warmed carrier fluid on nefopam injection-induced pain.
METHODS: A total of 48 patient

BACKGROUND: Nefopam is a centrally-acting but non-opioid analgesic drug of the benzoxazocine chemical class, developed in the early 1970s. It is widely used, mainly in European countries, for the relief of moderate to severe pain as an alternative to opioid analgesic drugs, and used in rheumatic disease and other musculoskeletal disorders in the UK. This review sought to evaluate the efficacy and safety of oral nefopam in acute postoperative pain, using clinical studies of patients with established pain, and with outcomes measured primarily over 6 hours using standard methods. Nefopam is widely used for the relief of moderate acute pain. Its safety profile remains to be specified. The objective of the study was to review adverse reactions to nefopam spontaneously reported to the French Pharmacovigilance system. All cases of adverse drug reactions (ADRs) associated with nefopam, registered in the French Pharmacovigilance database from January 1, 1995 to December 31, 2004, were reviewed. For each reported ADR, information about patient (age, gender, medical history), drug exposure (suspected and concomitantly used drugs), characteristics of ADRs (imputability score, t

One hundred and ninety-six hospitalized patients were randomly assigned to one of eight treatment groups consisting of oral or intramuscular analgesics and placebo. Patients were monitored for 6 hours for pain relief, vital signs, and side effects. No statistically significant differences in pain intensity or relief were seen for nefopam (Acupan), diphenhydramine (Benadryl), aspirin, or placebo despite a numerically higher score for nefopam. This is attributed to an unexpectantly high placebo response. Significant pain relief and reduction of pain intensity were seen with both parenteral nefop

BACKGROUND: The recently known analgesic action mechanisms of nefopam (NFP) are similar to those of anticonvulsants and antidepressants in neuropathic pain treatment. It is difficult to prescribe high doses of oral neuropathic drugs without titration due to adverse effects. Unfortunately, there are few available intravenous analgesics for the immediate management of acute flare-ups of the chronic neuropathic pain. The aim of this study was to determine the additional analgesic effects for neuropathic pain of NFP and its adverse effects during the titration of oral medications for neuropathic p

A new analgesic compound, neopam HCl, was studied for possible gastrointestinal blood loss liabilities by Cr-51 red cell tagging. It was compared with aspirin in usual therapeutic doses of 1.8 Gm per day. Dosage of nefopam HCl was 180 mg per day (two tablets t.i.d). Twenty healthy male volunteers had fecal blood loss determined after one-week crossover periods of drug investigation, each preceded by one-week no-drug control periods. Results show a significant (P is less than 0.01) increase in occult bleeding in the aspirin-treated subjects from a mean of 0.5 ml to 1.63 ml per 24 hours, the mea

INTRODUCTION: Nefopam is a non-opioid analgesic used for postoperative pain control. For intravenous home use, a portable elastomeric device is often preferred for administration. The nefopam in the device must be stable at 32°C because this device is positioned close to the patient.
OBJECTIVES: This study aimed to evaluate the physicochemical stability of nefopam solutions at 0.2 and 3.33 mg/mL diluted in 0.9% NaCl in silicone portable elastomeric devices at 32°C in the dark.
METHODS: Three portable elastomeric devices were prepared for each condition. Chemical stability was assessed by pH

BACKGROUND: Nefopam is a safe analgesic with mild side effects including drowsiness, nausea, vomiting, and sweating. Nevertheless, research is scarce on the impact of nefopam in managing postoperative pain following laparoscopic cholecystectomy (LC), and the advantageous effects of nefopam remain unclear. In this research, the effectiveness of nefopam was compared to placebo for the treatment of postoperative pain following LC.
METHODS: A literature search for randomized controlled trials (RCTs) was performed through June 2024 using online databases including Cochrane Library, PubMed/Medline,

INTRODUCTION: The adjuvant-induced arthritis (AIA) model is widely used in research to investigate arthritis pathogenesis. Hind paw inflammation is the main outcome in this model with high loss of mobility function partly related to pain. However, analgesics such as non-steroidal anti-inflammatory drugs or opioid drugs interfere with the inflammation process related to arthritis, thus reducing their beneficial use in this model. Therefore, we investigated the effect of nefopam on arthritis development in order to improve pain management in the AIA model.
METHODS: Female Lewis rats were random

BACKGROUND: Shivering is a frequent complication following surgery and anaesthesia. A large variety of studies have been reported that nefopam may be efficacious for the prevention and treatment of perioperative shivering. Regrettably, there is still no conclusion of the efficacy and safety of nefopam for the prevention of perioperative shivering. The aim of this analysis is to evaluate the efficacy of nefopam for the prevention of perioperative shivering in patients undergoing different types of anaesthesia compared with placebo group and other active interventions.

Intra-operative intramuscular injections of either papaveretum 0.3 mg/kg or nefopam 0.4 mg/kg were given to alternate patients to promote smooth emergence from ENT anaesthesia in a consecutive series of 40 children. Observations over a period of 30 minutes following completion of surgery showed that emergence was satisfactory in 19 out of 20 children given papaveretum, and in 15 out of 20 children given nefopam. The study confirms that this indication for papaveretum is justifiably popular and that nefopam is a useful alternative mainly because it does not cause respiratory depression. There a

Simple and rapid spectrophotometric procedures have been established for quantitation of nefopam hydrochloride (NF) mebevrine hydrochloride (MB) and phenylpropanolamine hydrochloride (PP). The procedures are based on the reaction between the examined drugs (NF, MB and PP) and alizarin (I), alizarin red S (II), alizarin yellow G (III) and quinalizarin (IV) producing ion-pair complexes which can be measured at the optimum wavelength. The optimization of the reaction conditions is investigated. Beer's law is obeyed in the concentration ranges 0.5-30.0 microg ml(-1). The molar absorptivity, Sandel

BACKGROUND: Patients who undergo urinary catheterization may experience postoperative catheter-related bladder discomfort (CRBD). Previous studies have indicated that drugs with antimuscarinic effects could reduce the incidence and severity of CRBD. Accordingly, this study was carried out to investigate whether nefopam, a centrally acting analgesic with concomitant antimuscarinic effect, reduces the incidence and severity of CRBD.
METHODS: Sixty patients with American Society of Anesthesiologists physical status I and II and aged 18-70 years who were scheduled to undergo elective ureteroscopi

The present study investigated the effects of intrathecal nefopam on the pain behavior and on the extracellular levels of serotonin (5-HT), norepinephrine (NE), and glutamate in the spinal cord, in a rat model of pain induced by formalin. Nefopam was intrathecally administered 10 min prior to the formalin test to assess its antinociceptive effects. In another cohorts of animals, dihydroergocristine, yohimbine, or (RS)-α-Methylserine-O-phosphate (MSOP), a serotonergic, α-2 adrenergic receptor, or group III metabotropic glutamate receptor antagonist, respectively, were administered prior to the

INTRODUCTION: The primary goal in treating acute pain is to provide effective analgesia while minimizing the risk of progression to chronic pain. Opioids have long been the mainstay for treating moderate to severe pain. However, their use is increasingly questioned due to challenges such as addiction, tolerance, and adverse effects, including respiratory depression. A multimodal approach (balanced analgesia, multimodal analgesia- MMA), which includes non-opioid analgesic agents (NOAAs), adjuvant medications, and opioids, is recommended. This review was designed to cover the current characteris

BACKGROUND: Nefopam is a centrally acting analgesic which has a theoretical risk of stopping lactation due to its anticholinergic and dopaminergic effects. The aim of this study was to evaluate the effect of nefopam on lactation and to investigate potential adverse effects on newborns.
METHODS: Seventy-two women, scheduled to undergo a caesarean delivery under spinal anaesthesia and wanting to breastfeed, were randomised to one of two groups: nefopam (20mg, six hourly) or paracetamol (1g, six hourly). In both groups, postoperative analgesia was supplemented with ketoprofen (50mg, six hourly)
