Loss of Zeb2 in mesenchyme-derived nephrons causes primary glomerulocystic disease.

Primary glomerulocystic kidney disease is a special form of renal cystic disorder characterized by Bowman's space dilatation in the absence of tubular cysts. ZEB2 is a SMAD-interacting transcription factor involved in Mowat-Wilson syndrome, a congenital disorder with an increased risk for kidney anomalies. Here we show that deletion of Zeb2 in mesenchyme-derived nephrons with either Pax2-cre or Six2-cre causes primary glomerulocystic kidney disease without tubular cysts in mice. Glomerulotubular junction analysis revealed many atubular glomeruli in the kidneys of Zeb2 knockout mice, which expl

Effects of systemic hypertension, antidiuretic hormone, and prostaglandins on remnant nephrons.

Renal function was evaluated in normal and after 30 days of 5/6 renal mass reduction (CRF) in Munich-Wistar (MW) rats, spontaneously hypertensive rats with superficial glomeruli (EPM), and in Brattleboro rats with congenital diabetes insipidus (DI). Mean arterial pressure was higher in EPM-Control and EPM-CRF rats as compared with MW and DI rats. MW and EPM rats with CRF showed increases of 120% and 196%, respectively, in single nephron glomerular filtration rate as compared with their controls. However, DI rats with CRF did not show any increase in single nephron glomerular filtration rate as

Pathways to recovery and loss of nephrons in anti-Thy-1 nephritis.

The present histopathologic study of anti-Thy-1.1 models of mesangioproliferative glomerulonephritis in rats provides a structural analysis of damage development and of pathways to recovery and to nephron loss. As long as the disease remains confined to the endocapillary compartment, the damage may be resolved or recover with a mesangial scar. Irreversible lesions with loss of nephrons emerge from extracapillary processes with crucial involvement of podocytes, leading to tuft adhesions to Bowman's capsule (BC) and subsequent crescent formation. Two mechanisms appeared to be responsible: (1) Ep

Sensitivity of residual nephrons to high dose furosemide described by diuretic efficiency.

Ten haemodialysis (HD) patients with a median residual creatinine clearance (CLCR) of 1.9 ml.min-1 x 1.73 m-2 (range 0.6-5.3) were treated with oral furosemide (F) 2.0 g. Overall-efficiency (O-E, daily sodium excretion/total urinary F) and total-efficiency (delta-E, increase in daily sodium excretion/total urinary F) were measured on the last 24 hours of each interdialysis interval. In addition, O-E was measured during the complete interdialysis interval in 10 HD patients with a median CLCR of 5.6 ml.min-1 x 1.73 m-2 (range 0.7-6.8) treated for 1 year with a fixed oral dose of F between 250-10

Disorders of distal nephron function.

In this review, the distal nephron is considered to be that portion of the renal tubule commencing with the thick ascending limb of the loop of Henle and ending with the papillary collecting duct. The collecting duct, including its subdivisions in the cortex and medulla, originates from a different embryologic anlage than more proximal nephron segments, which may explain its morphologic and functional dissimilarities from the thick ascending limb and the distal convoluted tubule. This review summarizes selected aspects of the physiology of the distal nephron, with particular emphasis on the ph

Effects of experimental obstructive hydronephrosis on the immature nephrons in newborn rats.

Partial obstruction of the left ureter was created in rats less than 36 h of age and its effects were studied at the age of 2-6 days. A considerable hydronephrosis with distension of the kidney developed within 1 day after obstruction, but there was no parenchymal weight reduction. The thickness of the mature, inner cortical layer was significantly decreased, suggesting a delay in the differentiation of the cortex. The parenchymal weight reduction, previously documented in our laboratory after long-standing ureteric obstruction, may thus be caused by a retardation of renal growth, which is nev

Alpha 1-antitrypsin and antithrombin III in atrophic nephrons of chronic pyelonephritis.

In a series of investigation to elucidate the roles of chemical substances in the atrophic process of nephron in chronic pyelonephritis (CPN), two protease inhibitors (PI) and enterobacterial antigen were examined by immunofluorescence (IF). Alpha-antitrypsin (alpha 1AT) was always found in the sclerotic glomerular tuft (GT) and along the thickened Bowman's capsule (BC) as well as the thickened tubular basement membrane (TBM) in every atrophic nephron of CPN. Antithrombin III (AT III) was always found along the inside of thickened TBM in every atrophic nephron, but not found in the sclerotic G

Different functional characteristics of residual nephrons in infantile vs adult diffuse cortical necrosis.

In this report we study the functional characteristics of residual nephrons in a 37 year-old woman, 7 months after diffuse bilateral cortical necrosis (CN) of unknown etiology, and in two infants, aged 13 and 15 months, who suffered CN in early infancy after surgical shock and acute dehydration, respectively. In the three cases CN was proven histologically by renal biopsy but undamaged nephrons were only present in the juxtamedullary area in the adult patient whereas in the two infants they were located in the outer part of the cortex. At the time of the study all patients presented a similar

Mechanism of cystogenesis by Cd79a-driven, conditional mTOR activation in developing mouse nephrons.

Polycystic kidney disease (PKD) is a common genetic disorder arising from developmental and postnatal processes. Defects in primary cilia and their signaling (eg, mTOR) underlie the pathogenesis. However, how mTOR regulates tubular integrity remains unclear. The paucity of faithful models has limited our understanding of pathogenesis and, therefore, the refinement of therapeutic targets. To understand the role of mTOR in early cystogenesis, we studied an in-house mouse model, Cd79a-Cre;Tsc1ff. (Cd79a-Tsc1 KO hereafter), recapitulating human autosomal-dominant PKD histology. Cre-mediated Tsc1 d

Sex differences in solute transport along the nephrons: effects of Na+ transport inhibition.

Each day, ~1.7 kg of NaCl and 180 liters of water are reabsorbed by nephron segments in humans, with urinary excretion fine tuned to meet homeostatic requirements. These tasks are coordinated by a spectrum of renal Na+ transporters and channels. The goal of the present study was to investigate the extent to which inhibitors of transepithelial Na+ transport (TNa) along the nephron alter urinary solute excretion and how those effects may vary between male and female subjects. To accomplish that goal, we developed sex-specific multinephron models that represent detailed transcellular and paracell

Feedback activation in rat nephrons by sera from patients with acute renal failure.

Tubuloglomerular feedback (TGF) may be triggered by varying the distal electrolyte load. TGF may contribute to acute renal failure (ARF), but maximal TGF stimulation induces only relatively small alterations in single nephron GFR (SNGFR) and stop-flow pressure (SFP), whereas in ARF, GFR may cease, raising the possibility that substances eliciting greater TGF responses than do elelectrolytes occur in ARF. We have sought such substances in serum, urine, and peritoneal fluid from patients with ARF. Sera samples from 1 healthy subject, 4 ARF patients, and 2 other anuric patients were dialyzed agai

Stealing nephrons-a review on how patent ductus arteriosus physiology impacts neonatal kidney health.

Adverse short- and long-term kidney outcomes are increasingly recognized as sequelae of NICU care, oftentimes of multifactorial origin and in the setting of a patent ductus arteriosus (PDA). Many studies have reviewed the interactions between the PDA and the brain, intestine, and lungs, but few have specifically reviewed the potential influence of the PDA on the kidney. This review describes the current breadth of literature as it relates to the pathophysiologic interplay between the PDA and the kidneys, as well as how PDA intervention may influence kidney health while neonates are still in th

Effects of renal pelvic high-pressure perfusion on nephrons in a porcine pyonephrosis model.

The aim of this study was to investigate the effects of various renal pelvic pressure gradients on nephrons with purulent infection. Five miniature test pigs were selected. One side of the kidney was used to prepare the pyonephrosis model and the other side was used as the healthy control. A piezometer and a water fill tube were inserted into the renal pelvis through the ureter. Prior to perfusion, punctures were made on the healthy and purulent sides of the kidneys to obtain tissues (as controls). Subsequently, a puncture biopsy was conducted on the kidneys at five pressure levels: 10, 20, 30

The number of nephrons in the mammalian kidney: environmental influences play a determining role.

Several lines of evidence, mostly derived from animal studies, indicate that changes in the fetal environment may affect the renal development. Fetal growth retardation is associated with a nephron deficit in both humans and animals. Changes in the supply of vitamin A to the fetus may be responsible for the variations in the number of nephrons in the human kidney. In utero exposure to hyperglycemia or drugs may also cause a nephron deficit.

Low Nephron Number and Kidney Disease.

Reduced nephron endowment and acquired nephron loss increase susceptibility to hypertension and chronic kidney disease (CKD). Low birth weight, prematurity, genetic factors, and adverse prenatal exposures can limit nephron development, whereas aging and kidney injury reduce the number of functioning nephrons after birth. Compensatory glomerular hypertrophy and increased single-nephron filtration can initially preserve overall glomerular filtration rate, but sustained glomerular stress may promote podocyte injury, proteinuria, and glomerulosclerosis. These changes can accelerate nephron loss an

From Nephron Number to Global Health.

In 1988, in a seminal paper, Brenner and colleagues proposed that the numbers of nephrons acquired in utero may be a factor that determines an individual's lifelong risk of kidney disease. The hypothesis posited that a kidney with fewer nephrons would have a reduced filtration surface, a reduced capacity to excrete sodium, and a limited capacity to compensate for additional kidney injury and nephron loss. These factors increase the risk of high blood pressure and kidney dysfunction. Potential clinical markers for such risk include preterm birth, small-for-gestational-age birth and low birth we
