The leg depressor and levator muscles in the squat lobster Munida quadrispina (Galatheidae) and the crayfish Procambarus clarkii (Astacidae) have multiple heads with potentially different functions.

The proximal leg muscles of decapod crustaceans, controlling movements at the first two joints, are anatomically more complex than the better-studied distal leg muscles. Despite extensive research on their involvement in diverse behaviors, no complete descriptions of the anatomy and innervation of these muscles for any species have been published. We describe the anatomy and innervation of the depressor muscle in the second leg of the squat lobster Munida quadrispina and compare its anatomy with that of its homologue in the crayfish Procambarus clarkii and its antagonist, the levator, in both

The lethality of Salmosan (Azamethiphos) to American lobster (Homarus americanus) larvae, postlarvae, and adults.

The pesticide formulation Salmosan (47.5% w/w azamethiphos) is currently registered for use, in Canada, to treat salmonids for infestations of the copepod parasites, Lepeophtheirus salmonis and Caligus elongatus (sea lice). Determination was made of the acute lethality of this product to the three larval stages, the first postlarval stage, and the adult of the American lobster (Homarus americanus), a species of significant economic importance in Eastern Canada. The 48-h LC50 (as azamethiphos) is 3.57 microg/liter for Stage I, 1.03 microg/liter for Stage II, 2.29 microg/liter for Stage III, 2.1

Sodium flux through the sodium channels of axon membrane fragments isolated from lobster nerves.

The efflux of 22Na from vesicles formed by axolemma fragments isolated from lobster nerves was studied in the presence and in the absence of drugs having well-known action on the sodium channels. The vesicles were equilibrated 12-14 h at 4 degrees C with 22Na in lobster solution containing 1 mM ouabain. Afterwards the suspension was divided: one portion was used as control and the others were treated with veratrine (0.025-0.50 mg/ml), tetrodotoxin (1-2,000 nM) in the presence of veratrine, or tetrodotoxin alone. After 3 h at 20-22 degrees C, the suspensions were diluted into nonradioactive sol

Effects of analgesia on the response to a noxious stimulus in Norway lobsters (Nephrops norvegicus).

There is evidence that decapod crustaceans have the capacity to experience aversive states associated with nociception, highlighting the need for humane treatment. Behavioural and physiological responses to nociceptive stimuli may be reduced with analgesic drugs, yet little is known for decapods about effective options. In this study, the impact of electric shock on behaviour and physiology was investigated in Norway lobsters. Behaviour was monitored from before the shock to two hours after, with and without drugs with analgesic properties. Haemolymph and nervous tissues were analysed for stre

Dead Matter, Living Machines: Repurposing Crustaceans' Abdomen Exoskeleton for Bio-Hybrid Robots.

Bio-hybrid robots utilize living organisms for robot design, however, their use of living bodies makes maintenance, control, and fabrication of robot challenging. As an alternative, exoskeletons stand out for retaining mobility after the organism's death, making them an accessible candidate. In particular, crustacean exoskeletons, often discarded as food waste, provide both structural strength and flexibility from their segmented rigid shell. By repurposing dead animals' part from bio-waste, a sustainable cyclic design process is proposed in which materials can be recycled and adapted for new

Interaction of dopamine and cardiac sac modulatory inputs on the pyloric network in the lobster stomatogastric ganglion.

Dopamine and cardiac sac network activity each have strong, and different, modulatory actions on the pyloric rhythm in the stomatogastric ganglion of the spiny lobster. When combined, the two modulatory inputs have a complex effect. Dopamine and cardiac sac activity cancel one another's effects to restore normal pyloric activity to 4 of the 6 classes of pyloric neurons. In the remaining two pyloric neurons, dopamine's strong modulatory effects are completely overruled during cardiac sac network related activity. Possible cellular mechanisms underlying these interactions are discussed.

The modulus of elasticity of lobster aorta microfibrils.

The presence of elastic fibres in the extracellular matrix (ECM) provides physiologically important elastic properties for many tissues. Until recently, microfibrils, one component of the ECM, were thought primarily to serve as a scaffolding on which elastin is deposited during development to form elaunin fibres [1]. The most prominent protein that forms mammalian microfibrils is fibrillin. It is known that mutations in the fibrillin gene cause a heterogenous connective tissue disease called Marfan syndrome [2], so information on mechanical properties of microfibrils or their role in tissue fu

The presence of inorganic and organic mercury and selenium in Nephrops norvegicus from the Ligurian Sea.

Samples of Nephrops norvegicus from the Ligurian Sea were analysed for contents of organic mercury, total mercury and selenium. Results for 37 specimens are reported. Total mercury concentrations increase with size of the specimens. Percentage of organic mercury ranges from 31 to 88% with an average of 58%. Selenium does not correlate with size and the average content is 0.88 micrograms g-1 fresh weight (range 0.30-1.74 micrograms g-1 fresh weight).

Topology selection by chaotic neurons of a pyloric central pattern generator.

The pyloric Central Pattern Generator (CPG) in the lobster has an architecture in which every neuron receives at least one connection from another member of the CPG. We call this a "non-open" network topology. An "open" topology, where at least one neuron does not receive synapses from any other CPG member, is found neither in the pyloric nor in the gastric mill CPG. Here we investigate a possible reason for this topological structure using the ability to perform a biologically functional task as a measure of the efficacy of the network. When the CPG is composed of model neurons that exhibit r

Suppressive control of a rhythmic central pattern generator by an identified modulatory neuron in crustacea.

The activity of the 14 neuron network which organizes the pyloric motor rhythm in the stomatogastric ganglion of the lobster, Homarus gammarus, is controlled by neuromodulatory inputs which have been described as having mainly 'permissive' effects. By contrast, here we identify a neuron, the pyloric suppressor (PS) neuron which exerts a 'suppressive' effect on the pyloric activity. We show that PS neuron discharge can terminate in a long-lasting manner, spontaneous pyloric rhythmic activity. Its effect results from a direct suppression of the endogenous ability of the pyloric pacemaker neurons

Intercellular junctions in the hepatopancreas of the lobster Nephrops norvegicus.

The hepatopancreas of the lobster has recently been found to be a rich source of material from which to isolate arthopod gap junctions biochemically (Finbow et al., 1983a; 1984). It has therefore been studied here to assess the features of these intercellular junctions and any others that may be present, in vivo. The tissue consists of columnar epithelial cells which possess apical microvilli and basal infoldings. In thin sections the lateral borders of these cells are characterized by desmosomes and smooth septate junctions as well as by gap junctions. The desmosomes exhibit no apparent freez

Identification and Semi-quantification of Protozoa from the Digestive System Microbiota of the Lobster Cockroach Nauphoeta cinerea Oliver, 1789 (Insecta:Blattaria).

INTRODUCTION: The lobster cockroach Nauphoeta cinerea (N. cinerea) is indicated as a promising non-mammalian model, because it presents behavioral and biochemical alterations also observed in conventional models. In this research, we identified and characterized the distribution of protozoa that inhabit the digestive system (DS) of N. cinerea cockroaches. METHODS: The adult specimens of N. cinerea used in this study (n = 32) were obtained at the Federal University of Santa Maria, dissected and had their visceral contents observed in bright-field microscopy without staining and after applicati

The actions of crustacean cardioactive peptide on adult and developing stomatogastric ganglion motor patterns.

The motor patterns produced by the stomatogastric ganglion (STG) are strongly influenced by descending modulatory inputs from anterior ganglia. With these inputs intact, in control saline, the motor patterns produced by the stomatogastric nervous system of embryonic and larval lobsters are slower and less regular than those of adult lobsters. We studied the effects of the hormonal modulator, crustacean cardioactive peptide (CCAP) on the discharge patterns of STG motor patterns in embryos, larvae, and adult Maine lobsters, Homarus americanus, with the anterior inputs present and absent. In adul

Differential expression and targeting of K+ channel genes in the lobster pyloric central pattern generator.

A molecular analysis of motor pattern generation is an essential complement to electrophysiological and computational investigations. In arthropods, A-channels are posttranslationally modified multimeric proteins containing Shaker family alpha-subunits that may interact with beta-subunits, gamma-subunits, and other auxiliary proteins. One consequence of A-channel structure is that several mechanisms could underlie the cell-specific differences in pyloric IAs including differential gene expression, alternate splicing, and posttranslational modifications. Transduction of temporal patterns by single neurons.

As our ability to communicate by Morse code illustrates, nervous systems can produce motor outputs, and identify sensory inputs, based on temporal patterning alone. Although this ability is central to a wide range of sensory and motor tasks, the ways in which nervous systems represent temporal patterns are not well understood. I show here that individual neurons of the lobster pyloric network can integrate rhythmic patterned input over the long times (hundreds of milliseconds) characteristic of many behaviorally relevant patterns, and that their firing delays vary as a graded function of the p

Gastric mill activity in the lobster. I. Spontaneous modes of chewing.

1. The gastric central pattern generator (CPG) driving the three teeth of the gastric mill inside the lobster stomach has often been used as a model for the study of central nervous systems, but the actual functioning of the mill has never been observed directly. By using a small endoscope inserted through the esophagus a video analysis of the tooth movements was performed with restrained, but otherwise intact lobsters. 2. The teeth show spontaneous periodic chewing (cycle duration from 4 to 70 s) in two different basic modes. In the squeeze mode only the cusps of the three teeth move together

Vortex fluidics-mediated DNA rescue from formalin-fixed museum specimens.

DNA from formalin-preserved tissue could unlock a vast repository of genetic information stored in museums worldwide. However, formaldehyde crosslinks proteins and DNA, and prevents ready amplification and DNA sequencing. Formaldehyde acylation also fragments the DNA. Treatment with proteinase K proteolyzes crosslinked proteins to rescue the DNA, though the process is quite slow. To reduce processing time and improve rescue efficiency, we applied the mechanical energy of a vortex fluidic device (VFD) to drive the catalytic activity of proteinase K and recover DNA from American lobster tissue (

Understanding circuit dynamics using the stomatogastric nervous system of lobsters and crabs.

Studies of the stomatogastric nervous systems of lobsters and crabs have led to numerous insights into the cellular and circuit mechanisms that generate rhythmic motor patterns. The small number of easily identifiable neurons allowed the establishment of connectivity diagrams among the neurons of the stomatogastric ganglion. We now know that (a) neuromodulatory substances reconfigure circuit dynamics by altering synaptic strength and voltage-dependent conductances and (b) individual neurons can switch among different functional circuits. Computational and experimental studies of single-neuron

An autoradiographic study of binding of iodinated spider toxin to lobster muscle.

Distribution of the binding sites of Joro spider toxin (JSTX), a specific inhibitor of the glutamate receptors in the crustacean neuromuscular synapse, was studied by using autoradiography. JSTX was synthesized and made radioactive by conjugation with iodine-125. 125I-JSTX irreversibly blocked the excitatory postsynaptic potentials of the lobster neuromuscular synapse in a similar manner as the natural spider toxin. Light microscopic autoradiography of 125I-JSTX treated muscle showed sporadic aggregates of reduced silver grains on the surface of muscles. Electron microscopy of adjoining ultrat
