In situ detection of the Clostridium botulinum type C1 toxin gene in wetland sediments with a nested PCR assay.

A nested PCR was developed for detection of the Clostridium botulinum type C1 toxin gene in sediments collected from wetlands where avian botulism outbreaks had or had not occurred. The C1 toxin gene was detected in 16 of 18 sites, demonstrating both the ubiquitous distribution of C. botulinum type C in wetland sediments and the sensitivity of the detection assay.

Recognition of 7 genes signature (Cirrhosis Risk Score) in the diagnosed non-responders to DAAs therapy by intra-PBMCs nested HCV RNA PCR.

BACKGROUND AND AIMS: Predictors of chronic HCV response to oral antiviral therapy (OAT) are related to host genetic variations. Single nucleotide polymorphisms (SNP) and alleles variations of host genes in association with hepatic fibro-cirrhotic changes have a distinct role in OAT outcomes. The current research evaluated the association of Cirrhosis-Risk-Scores (CRS) values, based on the correlation of seven genes signature-SNPs, with sonographic liver parenchymal changes in determining OAT outcomes. METHODS: All study subjects (n = 54) were recruited three months after completing OAT and cl

Knocking down Israa, the Zmiz1 intron-nested gene, unveils interrelated T cell activation functions in mouse.

We previously reported Israa (immune-system-released activating agent), a novel gene nested in intron 6 of the mouse Zmiz1 gene. Zmiz1 is involved in several functions such as fertility and T cell development and its knockout leads to non-viable embryos. We also reported ISRAA's expression in lymphoid organs, particularly in the thymus CD3+ T cells during all developmental stages. In addition, we showed that ISRAA is a binding partner of Fyn and Elf-1 and regulates the expression of T cell activation-related genes in vitro. In this paper, we report the generation and characterization of an Isr

Expression of human F8B, a gene nested within the coagulation factor VIII gene, produces multiple eye defects and developmental alterations in chimeric and transgenic mice.

Factor VIII-associated gene B ( F8B ) is a small human gene of unknown function which is nested within the gene encoding coagulation factor VIII ( FVIII ) in chromosome band Xq28. The sequence of F8B includes the C2 cell adhesion motif of factor VIII, which has also been identified in numerous proteins known to play important roles during development. Here we have constructed both chimeric and transgenic mice expressing normal human F8B to investigate its possible developmental effects. The chimeras produced from embryonic stem cells transfected with normal F8B under control of a cytomegalovir

Primary and Nested PCR Amplification of B1 Gene to Confirm Seropositivity of Toxoplasmosis Among Cancer Patients in Sri Lanka.

Toxoplasmosis, caused by the parasite Toxoplasma gondii, affects approximately 30% of the human population worldwide. Reactivation of latent infection in immunocompromised people leads to fatal disease. This study aimed to determine the seropositivity of toxoplasmosis by enzyme-linked immunosorbent assay (ELISA), confirmed by primary polymerase chain reaction (PCR) and nested PCR (nPCR), and validate ELISA against nPCR among a group of cancer patients. Of the 321 participants, six (1.9%) patients were positive for nPCR and both IgG and IgM ELISA tests while 36 (11.2%) and 131 (40.8%) showed ev

Targeting of putative fimbrial gene for detection of S. Typhi in typhoid fever and chronic typhoid carriers by nested PCR.

INTRODUCTION: It is important to identify Salmonella Typhi infection quickly to treat acute fever patients and to prevent transmission by chronic typhoid carriers; therefore, a very specific and sensitive diagnostic technique is highly desirable, especially in endemic areas. The objective of this study was to develop a PCR protocol targeting the putative fimbrial staA gene of S. Typhi. This is a preferred target gene that is specifically amplified in the S. Typhi serotype compared to the commonly targeted fliC gene which may also be amplified from the non-typhoidal Salmonella Munchen serotype.

Development and evaluation of a nested PCR based on spherical body protein 2 gene for the diagnosis of Babesia bovis infection.

We developed and evaluated a nested PCR assay for the diagnosis of Babesia bovis infection in cattle based on the spherical body protein 2 gene (SBP2) from B. bovis. The specificity and sensitivity of the test were compared with the B. bovis RAP-1 gene nPCR. The SBP2 primers have specificities of 100% for B. bovis DNA. The sensitivity of the SBP2 nPCR to B. bovis from the in vitro cultured parasites was higher than that of the B. bovis RAP-1 gene nPCR, and a parasitemia as low as 10(-8)% was detected. The sensitivity of the SBP2 nPCR to B. bovis-diluted genomic DNA was also higher than that of

The association of FKBP5 gene methylation, adolescents' sex, and depressive symptoms among Chinese adolescents: a nested case-control study.

BACKGROUND: Depressive symptoms among adolescents are a serious health concern around the world. Altered DNA methylation in the FK506 binding protein 5 (FKBP5) gene has been reported to regulate stress response, which has been reported to be closely associated with depressive symptoms. However, most of the contributing studies have been conducted among adults and relatively few studies have considered the effect of disparate social influences and sex differences on the DNA methylation of FKBP5 in persons with depressive symptoms. The present study aimed to test the associations of FKBP5 DNA me

Clinical and epidemiological use of nested PCR targeting the repetitive element IS1111 associated with the transposase gene from Coxiella burnetii.

Q fever is a worldwide zoonosis caused by Coxiella burnetii-a small obligate intracellular Gram-negative bacterium found in a variety of animals. It is transmitted to humans by inhalation of contaminated aerosols from urine, feces, milk, amniotic fluid, placenta, abortion products, wool, and rarely by ingestion of raw milk from infected animals. Nested PCR can improve the sensitivity and specificity of testing while offering a suitable amplicon size for sequencing. Serial dilutions were performed tenfold to test the limit of detection, and the result was 10× detection of C. burnetti DNA with i

Effect of Limited Bedding and Nesting in Early Ontogenesis on Gene Expression in the Hippocampus and Frontal Cortex of Adolescent Rats Subjected to Restraint.

INTRODUCTION: According to literature, early stress may lead to a higher susceptibility to the action of various stressors later in life, thus largely contributing to the development of a wide range of affective disorders. Disrupting maternal care is one way to destabilize the environment for pups, which may result in the formation of an altered reaction to acute or moderate stress. METHODS: In this study, we analyzed the effects of limited bedding and nesting material (LBN) in PND2-PND9 on baseline gene expression in the hippocampus and frontal cortex of 1-month-old rats and the expression o
