Efferent and Afferent Connections of Neuropeptide Y Neurons in the Nucleus Accumbens of Mice.

Neuropeptide Y (NPY) is a neural peptide distributed widely in the brain and has various functions in each region. We previously reported that NPY neurons in the nucleus accumbens (NAc) are involved in the regulation of anxiety behavior. Anterograde and retrograde tracing studies suggest that neurons in the NAc project to several areas, such as the lateral hypothalamus (LH) and ventral pallidum (VP), and receive afferent projections from the cortex, thalamus, and amygdala. However, the neural connections between accumbal NPY neurons and other brain areas in mice remain unclear. In this study,

Sympathetic preganglionic efferent and afferent neurons mediated by the greater splanchnic nerve in rabbit.

Motion sickness, a multisymptom disorder characterized by abnormal gastrointestinal motility and emesis, can be induced by vestibular effects on the sympathetic portion of the autonomic nervous system. However, the vestibular-autonomic pathways are unknown. As a first step in the analysis, we identified the locus of preganglionic sympathetic neurons (PSNs) and dorsal root afferent ganglionic neurons (DRGs) which supply sympathetic innervation to major portions of the gastrointestinal tract in the rabbit. Retrograde labeling of neurons was obtained by application of horseradish peroxidase (HRP)

Fast inhibition slows and desynchronizes mouse auditory efferent neuron activity.

The encoding of acoustic stimuli requires precise neuron timing. Auditory neurons in the cochlear nucleus (CN) and brainstem are well-suited for accurate analysis of fast acoustic signals, given their physiological specializations of fast membrane time constants, fast axonal conduction, and reliable synaptic transmission. The medial olivocochlear (MOC) neurons that provide efferent inhibition of the cochlea reside in the ventral brainstem and participate in these fast neural circuits. Effects of medial olivocochlear efferent stimulation on the activity of neurons in the auditory midbrain.

Medial olivocochlear (MOC) efferents are known to suppress spontaneous activity and sound-evoked responses of primary afferents by their actions on outer hair cells in the cochlea. This study investigated the effects of MOC activation on the responses of single neurons in the central nucleus of the inferior colliculus (CNIC) of anaesthetized guinea pigs. Extracellular responses of CNIC neurons to contralateral tones were recorded with and without MOC stimulation in normal animals and in animals acutely treated with gentamicin to eliminate peripheral effects of MOC activation. In normal animals

Effects of early hearing deficits on olivocochlear efferent neuron morphology in mice.

One of the fundamental features of age-related hearing loss is difficulty discriminating signals in the presence of background noise, a functional deficit that may be related to the loss of auditory efferent neurons. We hypothesize that the central degeneration of olivocochlear (OC) efferent neurons results from prolonged lack of afferent input. We investigated the time-course of central degeneration of medial (MOC) and lateral (LOC) OC efferent neurons in genetic mouse models that exhibit hearing loss during development. Tests to verify cochlear function were combined with anatomical and morp

Distinct forms of synaptic plasticity during ascending vs descending control of medial olivocochlear efferent neurons.

Activity in each brain region is shaped by the convergence of ascending and descending axonal pathways, and the balance and characteristics of these determine the neural output. The medial olivocochlear (MOC) efferent system is part of a reflex arc that critically controls auditory sensitivity. Multiple central pathways contact MOC neurons, raising the question of how a reflex arc could be engaged by diverse inputs. We examined functional properties of synapses onto brainstem MOC neurons from ascending (ventral cochlear nucleus, VCN) and descending (inferior colliculus, IC) sources in mice usi

Importance of rostral ventrolateral medulla neurons in determining efferent sympathetic nerve activity and blood pressure.

Accentuated sympathetic nerve activity (SNA) is a risk factor for cardiovascular events. In this review, we investigate our working hypothesis that potentiated activity of neurons in the rostral ventrolateral medulla (RVLM) is the primary cause of experimental and essential hypertension. Over the past decade, we have examined how RVLM neurons regulate peripheral SNA, how the sympathetic and renin-angiotensin systems are correlated and how the sympathetic system can be suppressed to prevent cardiovascular events in patients. Based on results of whole-cell patch-clamp studies, we report that ang

Response properties of cochlear efferent neurons: monaural vs. binaural stimulation and the effects of noise.

1. Discharge properties of olivocochlear efferent neurons were measured in anesthetized cats. Previous studies of these neurons concentrated on monaural stimulation with tones and found sound-evoked discharge rates rarely exceeded 60 spikes/s (16, 20). In the present study, rates as high as 140 spikes/s were achieved by binaural stimulation and/or the addition of noise. Based on studies on the known effects of electrically stimulating the efferents such high rates of sound-evoked efferent activity probably have significant feedback effects on the auditory periphery. 2. Spontaneous discharge ra

Co-release of GABA and ACh from medial olivocochlear neurons fine tunes cochlear efferent inhibition.

During development, inner hair cells (IHCs) in the mammalian cochlea are unresponsive to acoustic stimuli but instead exhibit spontaneous activity. During this same period, neurons originating from the medial olivocochlear complex (MOC) transiently innervate IHCs, regulating their firing pattern which is crucial for the correct development of the auditory pathway. Although the MOC-IHC is a cholinergic synapse, previous evidence indicates the widespread presence of gamma-aminobutyric acid (GABA) signaling markers, including presynaptic GABAB receptors (GABABR). In this study, we explore the sou

Whole-brain efferent and afferent connectivity of mouse ventral tegmental area melanocortin-3 receptor neurons.

The mesolimbic dopamine (DA) system is involved in the regulation of multiple behaviors, including feeding, and evidence demonstrates that the melanocortin system can act on the mesolimbic DA system to control feeding and other behaviors. The melanocortin-3 receptor (MC3R) is an important component of the melanocortin system, but its overall role is poorly understood. Because MC3Rs are highly expressed in the ventral tegmental area (VTA) and are likely to be the key interaction point between the melanocortin and mesolimbic DA systems, we set out to identify both the efferent projection pattern

Efferent-like roles of afferent neurons in the gut: Blood flow regulation and tissue protection.

The maintenance of gastrointestinal mucosal integrity depends on the rapid alarm of protective mechanisms in the face of pending injury. To this end, the gastric mucosa is innervated by intrinsic sensory neurons and two populations of extrinsic sensory neurons: vagal and spinal afferents. Extrinsic afferent neurons constitute an emergency system that is called into operation when the gastrointestinal mucosa is endangered by noxious chemicals. The function of these chemoceptive afferents can selectively be manipulated and explored with the use of capsaicin which acts via a cation channel termed

Role of C1 neurons in anti-inflammatory reflex: Mediation between afferents and efferents.

Neuroimmune communication, the connection between the autonomic regulatory pathway and immune cells, has been implicated in the regulation of immune function and inflammation. The role of afferents (vagal afferent and somatic sensory nerves) and efferents (autonomic nervous and hypothalamic-pituitary-adrenal systems) in the inflammatory reflex has been well studied; however, the central pathway remains unknown. C1 neurons include both catecholaminergic and glutamatergic neurons, which are located in the rostral ventrolateral medulla. C1 neurons project to the spinal cord, dorsal motor nucleus

Two types of actions of norepinephrine on identified auditory efferent neurons in rat brain stem slices.

Whole cell voltage-clamp recordings were performed on auditory olivocochlear neurons in the ventral nucleus of the trapezoid body (VNTB) of brain stem slices from neonatal rats. Each neuron was identified by retrograde labeling with Fast Blue injected into the cochlea. Bath application of norepinephrine (NE; 1-10 microM) reversibly induced an inward current in 26 of 38 labeled neurons that were voltage clamped at -75 mV. This was responsible for the membrane depolarization to NE observed in current-clamp mode. The NE-induced inward current appeared to be more prominent at -55 mV than at -75 mV

The cytogenesis and radial migration of cranial nerve efferent neurons in the developing brain stem of the bovine.

The cytogenesis of motor neurons of the immature brain stem of the bovine has been examined by light-and electron microscopy from the periods of origin and migration of the motor neurons until their transformation into relatively mature neurons in their area of destination. Up to 2.7 cm CRL intensive cell migration occurs from the ventromedial cell column in a ventrolateral direction, leading to the formation of the definitive viscero-efferent nucleus of the facial nerve. Electron-optically these cells exhibit features of immature neurons which are extending leading processes in the direction

Mapping of afferent and efferent connections of phenylethanolamine N-methyltransferase-expressing neurons in the nucleus tractus solitarii.

OBJECTIVE: Phenylethanolamine N-methyltransferase (PNMT)-expressing neurons in the nucleus tractus solitarii (NTS) contribute to the regulation of autonomic functions. However, the neural circuits linking these neurons to other brain regions remain unclear. This study aims to investigate the connectivity mechanisms of the PNMT-expressing neurons in the NTS (NTSPNMT neurons). METHODS: The methodologies employed in this study included a modified rabies virus-based retrograde neural tracing technique, conventional viral anterograde tracing, and immunohistochemical staining procedures. Rhombomere-specific origin of the contralateral vestibulo-acoustic efferent neurons and their migration across the embryonic midline.

The bilateral efferent supply to the inner ear receptor fields is located in the hindbrain. In ovo injections of Dil into the common facial/vestibulo-acoustic nerve root at 3 days of chick development (stage 16) followed by analysis at 7 days has revealed the origin of the contralateral efferent neurons of the inner ear and their relation to the transient hindbrain rhombomeres. These neurons have a rhombomere 4-specific origin and form their commissure not by axonal outgrowth but, unusually, by transmedian cell migration into the contralateral rhombomere 4 and rhombomere 5.
