A case of human infection with Newcastle disease virus.

A case of Newcastle disease virus infection in a female laboratory technician is reported for the first time in Malaysia. Infection was acquired by droplet infection of the eye while grinding infected chicken in the laboratory. The case was confirmed by isolation of Newcastle disease virus from an eye swab taken from the subject on the first day of clinical signs. A four-fold rise of haemagglutination-inhibition titre was shown when sera on the third day of infection and 15 days later were compared.

In-ovo Newcastle disease virus vaccine strain TS09-C protects commercial chickens against Newcastle disease in the presence of maternally derived antibodies.

Maternally derived antibodies (MDA) substantially interfere with active immunity in post-hatch vaccination, although they provide early protection against disease through passive immunity in young chickens. Previously, Newcastle disease virus (NDV) strain TS09-C was demonstrated to be safe and immunogenic as in-ovo vaccine in specific-pathogen-free chickens. Here, we evaluated the safety, protective efficacy, and duration of clinical protection of the TS09-C virus as an in-ovo vaccine for commercial chickens in the presence of Maternally derived antibodies against NDV. This vaccine was safe in

Efficacy of Thermostable Newcastle Disease Virus Strain I-2 in Broiler Chickens Challenged with Highly Virulent Newcastle Virus.

Newcastle disease (ND) is a major threat to poultry industry production throughout developing countries. The Newcastle disease viruses (NDVs) infecting industrialized and indigenous poultry in Iran are velogenic strains and responsible for the frequent outbreaks of ND in poultry farms even in vaccinated flocks causing serious economic losses in the commercial and indigenous poultry. However, vaccination is the only way to protect against endemic ND, and the conventional vaccines are not heat stable and consequently require complex cold-chains to be transferred to users leading to not much resi

Recombinant Newcastle disease virus expressing the infectious bronchitis virus S1 gene protects chickens against Newcastle disease virus and infectious bronchitis virus challenge.

The recombinant LaSota strain expressing a chimeric IBV S1 gene (rLaSota-S1) was constructed with the S1 gene of the LX4 type IBV ck/CH/LDL/091022. The expression of the S1 protein was detected by an indirect immunofluorescence assay and Western blotting. The rLaSota-S1 strain was slightly attenuated, and its growth dynamics were similar to that of the parental LaSota strain. Vaccination of specific pathogen-free chickens with the rLaSota-S1 strain induced NDV hemagglutination inhibition antibodies, and it protected chickens from challenge with virulent NDV. In addition, vaccination with the r

Heterologous prime-boost immunization of Newcastle disease virus vectored vaccines protected broiler chickens against highly pathogenic avian influenza and Newcastle disease viruses.

Avian Influenza virus (AIV) is an important pathogen for both human and animal health. There is a great need to develop a safe and effective vaccine for AI infections in the field. Live-attenuated Newcastle disease virus (NDV) vectored AI vaccines have shown to be effective, but preexisting antibodies to the vaccine vector can affect the protective efficacy of the vaccine in the field. To improve the efficacy of AI vaccine, we generated a novel vectored vaccine by using a chimeric NDV vector that is serologically distant from NDV. In this study, the protective efficacy of our vaccines was eval

Rescue of Recombinant Newcastle Disease Virus Expressing Heterologous Genes.

Reverse genetics allows for the generation of recombinant infectious viruses from viral sequences or complete viral genomes cloned into plasmids. Using reverse genetics, it is then possible to introduce changes in the genome of infectious viruses for multiple applications.Newcastle disease virus (NDV) is a non-segmented, negative-sense RNA virus that has been amenable to manipulation by reverse genetics for more than two decades. Since then, recombinant NDVs have been extensively used as viral vectors to express heterologous proteins. We describe the key steps required to design and introduce

Newcastle disease virus: is an updated attenuated vaccine needed?

Newcastle disease virus (NDV) is a major cause of infectious mortality and morbidity in poultry worldwide. It is an enveloped virus with two outer-membrane proteins-haemagglutinin-neuraminidase (HN) and fusion protein (F)-that induce neutralizing antibodies. All NDV strains belong to one serotype. Yet, NDV vaccines, derived from genotype II, do not fully prevent infection or shedding of viruses from other genotypes. The aim of this study was to test if an updated vaccine is required. For this purpose, NDVs isolated from infected, albeit heavily vaccinated, flocks were genetically and immunolog

Characterization of emerging Newcastle disease virus isolates in China.

BACKGROUND: Newcastle disease (ND) is a devastating worldwide disease of poultry characterized by increased respiration, circulatory disturbances, hemorrhagic enteritis, and nervous signs. Sequence analysis shows several amino acid residue substitutions at neutralizing epitopes on the F and HN proteins of recent Shaanxi strains. Both Cross protection and cross serum neutralization tests revealed that the traditional vaccine strains were unable to provide full protection for the flocks. METHODS: To better understand the epidemiology of Newcastle disease outbreak, a portion of the F gene and th

Adaptive immune response of poultry to Newcastle disease vaccines.

Newcastle disease (ND), caused by virulent Newcastle disease virus (NDV), poses a major global threat to the poultry industry. ND vaccines have been used for nearly 70 years for disease prevention and control, and much is known about adaptive immune responses to infection and vaccination. However, our current knowledge remains incomplete and biased. While humoral immunity, especially regarding the induction of hemagglutination-inhibiting and virus-neutralizing antibodies, has been studied extensively, research on mucosal and cellular immunity has been more limited, despite their critical roles

Current situation and future direction of Newcastle disease vaccines.

Newcastle disease (ND) is one of the most economically devastating infectious diseases affecting the poultry industry. Virulent Newcastle disease virus (NDV) can cause high mortality and severe tissue lesions in the respiratory, gastrointestinal, neurological, reproductive and immune systems of poultry. Tremendous progress has been made in preventing morbidity and mortality caused by ND based on strict biosecurity and wide vaccine application. In recent decades, the continual evolution of NDV has resulted in a total of twenty genotypes, and genetic variation may be associated with disease outb

Immunity Elicited by a Turkey Herpesvirus-Vectored Newcastle Disease Vaccine in Turkey Against Challenge With a Recent Genotype IV Newcastle Disease Virus Field Strain.

Newcastle disease (ND) is still a major poultry disease worldwide. Vaccination remains the principal method of controlling ND in endemic countries. Various vaccination strategies, including the use of recently developed recombinant vaccines, have been used to control it. Recombinant vaccines that use the herpesvirus of turkey (HVT) as a vector to express one of the key antigens of Newcastle disease virus (NDV) have been developed to overcome some of the drawbacks related to the use of conventional vaccines. HVT as a vector appears to have unique beneficial characteristics: it is extremely safe

Reverse Genetics for Newcastle Disease Virus as a Vaccine Vector.

Newcastle disease virus (NDV) is an economically important pathogen in the poultry industry worldwide. Recovery of infectious NDV from cDNA using reverse genetics has made it possible to manipulate the genome of NDV. This has greatly contributed to our understanding of the molecular biology and pathogenesis of NDV. Furthermore, NDV has modular genome and accommodates insertion of a foreign gene as a transcriptional unit, thus enabling NDV as a vaccine vector against diseases of humans and animals. Avirulent NDV strains (e.g., LaSota and B1) have been commonly used as vaccine vectors. In this p

Newcastle disease vaccines-A solved problem or a continuous challenge?

Newcastle disease (ND) has been defined by the World Organisation for Animal Health as infection of poultry with virulent strains of Newcastle disease virus (NDV). Lesions affecting the neurological, gastrointestinal, respiratory, and reproductive systems are most often observed. The control of ND must include strict biosecurity that prevents virulent NDV from contacting poultry, and also proper administration of efficacious vaccines. When administered correctly to healthy birds, ND vaccines formulated with NDV of low virulence or viral-vectored vaccines that express the NDV fusion protein are

The isolation of lentogenic strains of Newcastle disease virus in Australia.

Twelve isolations of Newcastle disease virus were made from 77 clinical samples from chickens from conjunctivitis, respiratory disease, proventriculitis and bursal atrophy. Nine of the isolations were made from chickens with conjunctivitis. The viruses were identified as Newcastle disease virus by inhibition of their haemagglutinins with specific antiserum to Newcastle disease virus. The viruses failed to kill chicken embryos after inoculation into the allantoic cavity and they were judged to be lentogenic strains. There was no evidence that the Newcastle disease viruses were responsible for a

Advances in the Study of Antitumour Immunotherapy for Newcastle Disease Virus.

This article reviews the preclinical research, clinical application and development of Newcastle disease virus (NDV) in the field of cancer therapy. Based on the distinctive antitumour properties of NDV and its positive interaction with the patient's immune system, this biologic could be considered a major breakthrough in cancer treatment. On one hand, NDV infection creates an inflammatory environment in the tumour microenvironment, which can directly activate NK cells, monocytes, macrophages and dendritic cells and promote the recruitment of immune cells. On the other hand, NDV can induce the

Ocular pathogenesis of Newcastle disease virus in rabbits and monkeys.

Studies on the ocular pathogenesis of Newcastle disease virus (NDV) in rabbits and monkeys revealed that conjunctivitis can only be produced by the virus when conjunctival epithelium is traumatized; multiple instillations of large doses of virus over the intact conjunctiva failed to do so. The presence of serum HI antibodies showed no correlation with the clinical form of the disease in rabbits. However, in monkeys, the disease showed a uniform pattern of immune response irrespective of the strain of the virus. Monkeys may be useful for further studies.
