Ischemia and the metabolic disorder it entails would seem to be the pathogenic mechanism behind acute cochlear deafness, irrespective of the triggering process. The prognosis is entirely dependent on the rapid initiation of an effective treatment. At the end of a double-blind therapeutic trial comparing Ginkgo biloba extract and a standard alpha blocker (nicergoline), a significant recovery was observed in both therapeutic groups, but improvement was distinctly better in the Ginkgo biloba group.

A double-blind crossover study of the alpha-blocker Nicergoline was carried out in sixteen patients affected by benign prostatic bladder outflow obstruction. The "irritative" symptoms of prostatic hypertrophy, including nocturnal frequency and dysuria, were improved after Nicergoline significantly more than after placebo. In 10 further patients with prostatic hypertrophy, peak and mean flow rates increased by 50% and 77% respectively after the acute administration of Nicergoline. No side effects were detected. Unilateral cerebral ischaemia was induced in 18-month-old Long-Evans rats by injection of 2,000 labelled microspheres (phi 50 microns) into the carotid blood stream. This results in an ipsilateral decrease in cerebral blood flow, development of severe oedema and modifications of glucose uptake and consumption. Furthermore, this ischaemia led to a deterioration of the avoidance response in conditioned animals. All these disturbances, including the cerebral oedema, diminished with nicergoline pretreatment.

It has been found that the drug sermion (nicergoline)--a synthetic ergot alkaloid with alpha-adrenoblocking action--produces a pronounced vasoactive (spasmolytic) effect in the elderly patients suffering of initial atherosclerotic encephalopathy. A course of sermion treatment improves, via its effect on the brain and lower extremity vessels, the flow in most involved vascular regions. High concentrations of the drug after intravenous administration of 4 mg of sermion (nicergoline) result in a rise of the sympathetic vascular tone, high adrenoblocking action.

OBJECTIVE: To study reflexes coming from the chemoreceptors in the humoral-isolated vertebral artery zone on systemic arterial pressure, cerebral and intra-ear hemodynamics in separate stimulation with angioprotectors.
RESULTS: A more pronounced reflex depression of the above-described effectors was caused by Nicergoline, it was less pronounced when using Vinpocetine and Pentoxifylline.
