SIGNIFICANCE: The molecular mechanisms driving the progression from nonalcoholic fatty liver (NAFL) to fibrosing steatohepatitis (NASH) are insufficiently understood. Techniques enabling the characterization of different lipid species with both chemical and spatial information can provide valuable insights into their contributions to the disease progression.
AIM: We extend the utility of stimulated Raman scattering (SRS) microscopy to characterize and quantify lipid species in liver tissue sections from patients with NAFL and NASH.
APPROACH: We applied a dual-band hyperspectral SRS microscop

Accelerating the measurement for discrimination of samples, such as classification of cell phenotype, is crucial when faced with significant time and cost constraints. Spontaneous Raman microscopy offers label-free, rich chemical information but suffers from long acquisition time due to extremely small scattering cross-sections. One possible approach to accelerate the measurement is by measuring necessary parts with a suitable number of illumination points. However, how to design these points during measurement remains a challenge. To address this, we developed an imaging technique based on a

Anisotropic materials with intrinsic one-dimensional architectures, where chains or tubes align along a crystallographic axis, exhibit direction-dependent optical responses and serve as ideal building blocks for polarization-sensitive optoelectronics. While progress exists in engineered compounds, discovering elemental crystals with naturally ordered one-dimensional building blocks exhibiting giant optical anisotropy remains challenging. Here, we report the synthesis of a direct-bandgap semiconducting one-dimensional phosphorus single crystal composed of unique wavy polygonal tubes. The monocl

Magnetic order coupled second harmonic generation (SHG) provides a unique nonlinear probe of symmetry breaking in low dimensional magnets. However, its capability to resolve nonuniform magnetic domain structures and identify domain boundaries remains largely unexplored. Here, we demonstrate optical SHG imaging of labyrinthine magnetic domains in room temperature Fe3GaTe2, showing high sensitivity to magnetic configurations consistent with magnetic force microscopy. Magnetic field dependent SHG measurements reveal coherent interference between structural and magnetization coupled contributions,

In this study, we present a semi-supervised inspection framework that integrates second harmonic generation (SHG) microscopy with pixel-level anomaly detection (PLAD) for high-sensitivity defect mapping in AlGaN/GaN heterostructures grown on Si (111). SHG is intrinsically sensitive to local inversion symmetry breaking and modulation of the effective second-order nonlinear susceptibility. As a result, spatial variations in the nonlinear optical response serve as a physical contrast mechanism for detecting structural inhomogeneities. The SHG-derived defect features are further supported by corre

Background and Objectives: Extracellular matrix (ECM) and collagen remodeling contribute to chronic kidney disease (CKD) progression and vascular access dysfunction. Conventional histological techniques rely on staining and provide limited sensitivity for detecting early or subtle ECM alterations. Nonlinear optical imaging modalities, including second-harmonic generation (SHG), third-harmonic generation (THG), and multiphoton fluorescence (MPF) microscopy, enable label-free, high-resolution visualization of fibrillar collagen and may offer additional structural information. This study aimed to

Thermally evaporated pyrano[3,2-c]quinoline semicarbazone (PQMHC) and its Cu(II) complex thin films were systematically investigated to elucidate the effect of metal coordination on their structural, morphological, photoluminescence, and optical properties. X-ray diffraction (XRD) revealed broad halos, confirming the amorphous nature of both films. Field-emission scanning electron microscopy (FE-SEM) showed that the PQMHC film possesses a relatively uniform surface composed of closely packed granular features, whereas Cu(II) incorporation induces a heterogeneous morphology with irregular clust

Raman spectroscopy has long been known to provide sufficient information to discriminate distinct cell phenotypes. Underlying this discriminating capability is that Raman spectra provide an overall readout of the metabolic profiles that change with transcriptomic activity. Robustly associating Raman spectral changes with the regulation of specific signaling pathways may be possible, but the spectral signals of interest may be weak and vary somewhat among individuals. Establishing a Raman-to-transcriptome mapping will thus require tightly controlled and easily manipulated biological systems and

Antibiotics resistance developed by biofilms has posed a clinical challenge in the effective treatment of bacterial infections. However, the resistance mechanisms have not been well understood due to a lack of suitable tools for dynamic observation of the interplay between antibiotics and biofilm. In this work, with the use of rapid hyperspectral stimulated Raman scattering microscopy associated with an aryl-alkyne-based Raman tag synthesized, we investigate dynamic interactions between vancomycin and Staphylococcus aureus (S. aureus) biofilm to gain new insights into the resistance mechanisms

Vibrational sum frequency generation (VSFG) spectroscopy and surface pressure measurements are used to investigate the adsorption of a globular protein, bovine serum albumin (BSA), at the air/water interface with and without the presence of salts. We find at low (2 to 5 ppm) protein concentrations, which is relevant to environmental conditions, both VSFG and surface pressure measurements of BSA behave drastically different from at higher concentrations. Instead of emerging to the surface immediately, as observed at 1000 ppm, protein adsorption kinetics is on the order of tens of minutes at low

We introduce a plasmonic computing platform that unites multiplexed optical inputs, static weighting, and nonlinear activation within a single nanoscale architecture-marking a critical step toward the realization of a plasmonic neural cell. In this approach, the orbital angular momentum (OAM) of light serves as an orthogonal and inherently parallel encoding scheme, enabling distinct optical channels to be directed into individual plasmonic waveguides. The weighting of input signals is governed by precisely engineered nanoscale gaps, whereas the system's nonlinear activation is revealed through

Ni-doped CdTe nanostructures were successfully synthesized via a facile hydrothermal route, demonstrating the critical role of transition-metal doping in tailoring the functional properties of CdTe-based nanomaterials. Structural analysis using X-ray diffraction (XRD) confirmed the formation of a cubic CdTe phase, with a noticeable reduction in crystallite size from ∼28 to ∼24 nm upon Ni incorporation, indicating lattice modification and enhanced defect density. Field-emission scanning electron microscopy (FESEM) analysis revealed a predominantly nanoparticle-like morphology, suggesting unifor

Nonlinear degradation behavior in semiconductor laser diodes (LDs) is critical for understanding the reliability limitations after long-term operation. Here, we have investigated an observed nonlinear power degradation and a subsequent progressive recovery behavior of GaN-based violet LDs during constant-current aging. The aged devices exhibit a pronounced increase in threshold current and reduced optical output, accompanied by a reduced radiative efficiency at low injection levels. Electrical measurements, below-threshold electroluminescence (EL) spectroscopy, and scanning electron microscopy

BACKGROUND: Prognosis and appropriate treatment of head and neck squamous cell carcinoma (HNSCC) depend on the tumor type routinely derived by invasive histopathology. A promising noninvasive alternative is nonlinear optical imaging, which is capable of in vivo tissue visualization for tumor typing and grading.
METHODS AND RESULTS: Thin tissue sections from 3 patients aged 56 to 60 years presenting advanced carcinoma of the hypopharynx, larynx, and left tonsil were investigated by coherent anti-Stokes Raman scattering (CARS), second-harmonic generation (SHG), and 2 photon excited fluorescence

Innovations in sophisticated optoelectronic devices have increased interest in high-refractive index polymers. Herein, we report innovative nanocomposite films with high linear and nonlinear refractive indices prepared by casting chitosan (Cs) with polyvinyl alcohol (PVA) (50:50 wt%) along with different concentrations (10-50 wt%) of sodium montmorillonite (NaMMT) nanoclay. The refractive indices in addition to other optical parameters of homopolymers and hybrid materials were investigated by UV-Vis. spectroscopy and optical modeling to assess their potential applications in optics. Besides, t

A recently developed nonlinear optical pump-probe microscopy technique uses modulation transfer to sensitively extract excited-state dynamics of endogenous biological pigments, such as eumelanin and pheomelanin. In this work, we use this method to image and characterize several inorganic and organic pigments used in historical art. We show substantial differences in the near-IR pump-probe signatures from nominally similar pigments and suggest extensions to art restoration.

Nonlinear microscopy has proven to be essential for neuroscience investigations of thick tissue preparations. However, the optical recording of fast (approximately 1 msec) cellular electrical activity has never until now been successfully combined with this imaging modality. Through the use of second-harmonic generation microscopy of primary Aplysia neurons in culture labeled with 4-[4-(dihexylamino)phenyl][ethynyl]-1-(4-sulfobutyl)pyridinium (inner salt), we optically recorded action potentials with 0.833 msec temporal and 0.6 microm spatial resolution on soma and neurite membranes. Stimulated Raman scattering (SRS) microscopy has emerged as a vital analytical tool in the fields of biology and medicine. However, the prevalent noise in SRS images has limited its ability for high-sensitivity and high-speed imaging. Here, we introduce DenoiseGAN, a self-supervised network that can reduce random noise while maintaining structural consistency in SRS images. DenoiseGAN was demonstrated in different sample types, including gastric cancer exfoliated cells, bladder cancer tissues, and fungal cells, achieving an up-to 7-fold increase in the signal-to-noise ratio and a structural si

Tellurium (Te) is an elemental semiconductor with a simple chiral crystal structure. Te in a two-dimensional (2D) form synthesized by a solution-based method shows excellent electrical, optical, and thermal properties. In this work, the chirality of hydrothermally grown 2D Te is identified and analyzed by hot sulfuric acid etching and high-angle tilted high-resolution scanning transmission electron microscopy. The gate-tunable nonlinear electrical responses, including the nonreciprocal electrical transport in the longitudinal direction and the nonlinear planar Hall effect in the transverse dir

Being able to image the microstructure of growth cartilage is important for understanding the onset and progression of diseases such as osteochondrosis and osteoarthritis, as well as for developing new treatments and implants. Studies of cartilage using conventional optical brightfield microscopy rely heavily on histological staining, where the added chemicals provide tissue-specific colours. Other microscopy contrast mechanisms include polarization, phase- and scattering contrast, enabling non-stained or 'label-free' imaging that significantly simplifies the sample preparation, thereby also r
