Microsporidia Nosema bombycis CQ1 can be vertically transmitted in silkworm Bombyx mori but Vairimorpha necatrix BM cannot. Therefore, the pathological differences in silkworm infected with these two microsporidia required clarification. Here, we compared the virulence of N. bombycis CQ1 and V. necatrix BM against silkworm. The pathological characteristics in intestine, testis and ovary were surveyed using paraffin sections, scanning electron microscopy and transmission electron microscopy. Our data firstly showed that the virulence of V. necatrix BM was weaker than that of N. bombycis CQ1. Se

Honey bees provide essential pollination services in natural and agricultural ecosystems, yet little is known about honey bee-associated pathogens in Cambodia and Laos. Here, we screened 53 colonies from 10 sites, including Apis cerana (n = 28), A. mellifera (n = 18), A. dorsata (n = 4), and A. florea (n = 3), for common honey bee viruses and the microsporidian Nosema ceranae using PCR and RT-PCR assays. Six viruses-acute bee paralysis virus, black queen cell virus, deformed wing virus type A, Israeli acute paralysis virus, Kashmir bee virus, and Lake Sinai virus-and N. ceranae were detected i

The native honey bee of the Arabian Peninsula, Apis mellifera jemenitica, is well adapted to live in arid environments, but could potentially remain vulnerable to pathogens and parasites. We present a molecular report of Nosema and viral infections from eleven commercial apiaries across Southwestern Saudi Arabia. We confirmed that all Nosema-positive samples belonged to N. ceranae, with infection levels varying by location. The average spore count via microscopy was 3.43 × 107 spores per bee across all locations, with the highest being 3.39 × 108 spores per bee. No spores were detected in thre

Nosema ceranae is an intracellular parasite that infects honeybees' gut altering the digestive functions; therefore, it has the potential of affecting the composition of the gut microbiome. In this work, individual bees of known age were sampled both in spring and autumn, and their digestive tracts were assessed for N. ceranae infection. Intestinal microbiome was assessed by sequencing the bacterial 16S rRNA gene in two different gut sections, the anterior section (AS; midgut and a half of ileum) and the posterior section (PS; second half of ileum and rectum). A preliminary analysis with a fir

Few areas of the world have western honey bee (Apis mellifera) colonies that are free of invasive parasites Nosema ceranae (fungi) and Varroa destructor (mites). Particularly detrimental is V. destructor; in addition to feeding on host haemolymph, these mites are important vectors of several viruses that are further implicated as contributors to honey bee mortality around the world. Thus, the biogeography and attendant consequences of viral communities in the absence of V. destructor are of significant interest. The island of Newfoundland, Province of Newfoundland and Labrador, Canada, is free

The intestinal immune system is essential for insects to defend against pathogenic infections, while the dual oxidase-reactive oxygen species (DUOX-ROS) pathway is a key component. However, the role of the DUOX-ROS pathway in Bombyx mori against Nosema bombycis, a significant pathogen of this species, remains poorly characterized. In this study, four core components of the DUOX-ROS pathway in B. mori (BmGαq, BmPLCβ1, BmPLCβ4, and BmDUOX) were identified using bioinformatics. Pathogen induction experiments showed significant upregulation of BmDUOX and increased ROS production following N. bomby

Honey bees health is compromised by many factors such as the use of agrochemicals in agriculture and the various diseases that can affect them. Multiple studies have shown that these factors can interact, producing a synergistic effect that can compromise the viability of honey bees. This study analyses the interactions between different pesticides and the microsporidium Nosema ceranae and their effect on immune and detoxification gene expression, sugar consumption and mortality in the Iberian western honey bee (Apis mellifera iberiensis). For this purpose, workers were infected with N. cerana

Vairimorpha (Nosema) ceranae, a microsporidian parasite that primarily affects honeybees, has caused concern due to its role in compromising bee health. Although the DNA of this microsporidium has been detected in Bombus terrestris and other bumblebees, evidence of active parasite development in these hosts remains limited. Here, we experimentally inoculated B. terrestris workers with V. ceranae spores to investigate whether the parasite can establish intracellular development in this host. We combined molecular and histopathological approaches, including conventional PCR, parasite-specific PT

Microsporidia represent a group of eukaryotic, obligate intracellular parasites that form environmentally resistant spores. Spore wall proteins (SWPs) can mediate pathogen-host interactions for specific parasitism among different species. However, limited research has been conducted on SWPs inNosema pernyi that causes microsporidiosis in the economically-important Chinese oak silkworm, Antheraea pernyi. Here, we identified a total of 636 proteins fromN. pernyiusing LC-MS/MS, among which 8 SWPs were listed. A hydrophilic SWP13, whichhas identity with other homologous SWP13 among the Nosema genu

The development of beekeeping in Ecuador has generated the need to strengthen the bee health program. Research on the main pathogens responsible for diseases like nosemosis, which can severely impact bee health, is of special interest. This study aims to identify the Nosema apis and/or Nosema ceranae species infecting honey bee colonies located in the northern Andean region of Ecuador using multiplex PCR targeting the RNA polymerase II gene (RPB1), and the phylogenetic analysis of N. ceranae based on the 16 S rRNA gene sequences. Among the 164 honey bee samples collected from colonies in the p

Lactobacillus salivarius A3iob was administered to productive colonies belonging to commercial apiaries of small beekeepers (around 30-50 hives each one), from four departments of the province of Jujuy (Argentina): Yala, Tilquiza, El Carmen, and Los Alisos. The incidence of Varroa destructor and Nosema spp., before and after winter, was monitored during 2 years of study (2014-2015). Depending on the geographical location of each apiary and the application time, a monthly dose of the bacteria (105 CFU/mL) reduced the levels of varroasis between 50 and 80%. Interestingly, L. salivarius A3iob cel

Microsporidians (M) are supposed to be ancient eukaryotic parasites with a broad range of animal hosts, being especially abundant in Arthropoda. They are supposed to pass a long way of adaptation to parasitism, that usually means inhibiting or avoiding host immune reactions alongside with the reduction of pathogenicity. However M, unlike other eukaryotic obligate parasites, preserved a high pathogenicity, comparable with one of viruses, and thus they could be expected to possess a unique mode of interactions with their hosts. The goal of the present work is to assess how M influence the cellul

Honey bees (Apis mellifera) have a vital role as pollinators of various crops in the global food supply. Honeybee colonies in Egypt have recently experienced an unexplained rise in annual loss due to a phenomenon known as Colony Collapse Disorder (CCD). In the current study, honey bees were collected from 14 sites from eight governorates in Egypt. The genetic diversity among the collected honey bee populations was investigated using the mitochondrial DNA cytochrome oxidase subunit I (COI). The amplified COI regions were sequenced, analyzed and aligned with other GenBank entries. The nucleotide

Nosema ceranae infections in honey bees (Apis mellifera) pose a severe threat to colony health. Beekeepers have used dicyclohexylammonium fumagillin to control Nosema apis, although it may be ineffective against N. ceranae. We investigated the ability of various propolis extracts collected from Upstate New York (United States) to decrease in vivo N. ceranae infection levels when fed ad libitum to N. ceranae-infected honey bees. Propolis extracts, most notably a dichloromethane extract, significantly lowered spore levels in a dose-dependent fashion 4 days post inoculation. When testing the in v

Early on-site detection of microscopic pathogens is essential for the effective management and health of honeybee colonies. Currently, the gold standard for molecular detection of bee pathogens is qPCR or RT-qPCR. Here, we present a rapid, sensitive, and cost-effective alternative for field-deployable DNA pathogen detection. This method combines direct, amplification-ready cell lysis of worker bee abdomens with recombinase polymerase amplification (RPA), CRISPR/Cas12a-mediated trans-cleavage of reporter probes, and detection via lateral flow assays. We successfully validated this protocol in h

Nosemosis C, a Nosema disease caused by microsporidia parasite Nosema ceranae, is a significant disease burden of the European honey bee Apis mellifera which is one of the most economically important insect pollinators. Nevertheless, there is no effective treatment currently available for Nosema disease and the disease mechanisms underlying the pathological effects of N. ceranae infection in honey bees are poorly understood. Iron is an essential nutrient for growth and survival of hosts and pathogens alike. The iron tug-of-war between host and pathogen is a central battlefield at the host-path

USDA-ARS Bee Research Laboratory received symptomatic honey bee (Apis mellifera L.) samples across the United States for disease diagnosis. Here, we present a retrospective study and cartography of ectoparasite Varroa destructor and intracellular microsporidia parasite Nosema spp. These two major parasites were identified in the diseased honey bee samples between 2015 and 2022. Varroa infestation level (VIL) was examined by a wash technique (Mites/100 bees) and calculated as a percentage, while Nosema infection was quantified by microscopical spore count (Million Spores/Bee). Data were analyze

Nosema podocotyloidis n. sp. (Microsporidia, Nosematidae) is described from Podocotyloides magnatestis (Trematoda: Opecoelidae), a parasite of the fish Parapristipoma octolineatum (Teleostei) in the Atlantic Ocean. Electron microscopy reveals that all the stages of the cycle (merogony and sporogony) are diplokaryotic and in direct contact with the cytoplasm of host cells. There is no sporophorous vesicle (pansporoblast). The earliest stages observed are meronts, which have a simple plasmic membrane. Their cytoplasm is granular, rich in ribosomes and contains some sacculi of endoplasmic reticul

Nosema ceranae is a microsporidium parasite that silently affects honey bees, causing a disease called nosemosis. This parasite produces resistant spores and germinates in the midgut of honey bees, extrudes a polar tubule that injects an infective sporoplasm in the host cell epithelium, proliferates, and produces intestinal disorders that shorten honey bee lifespan. The rapid extension of this disease has been reported to be widespread among adult bees, and treatments are less effective and counterproductive weakening colonies. This work aimed to evaluate the antifungal activity of a prototype
