To investigate if viruses are involved in the pathogenesis of vestibular schwannomas (VS), we have screened biopsies from VS patients using different molecular techniques. Screening for the presence of known viruses using a pan-viral microarray assay (ViroChip) indicated the presence of several viruses including human endogenous retrovirus K (HERV-K) and human herpes virus 2 (HHV2). But with the exception of HERV-K, none of the findings could be verified by other methods. Whole transcriptome sequencing showed only the presence of HERV-K transcripts and whole genome sequencing showed only the p

The innate immune system is important as the first line of defense to sense invading pathogens. Nucleic acids represent critical pathogen signatures that trigger a host proinflammatory immune response. Much progress has been made in understanding how DNA and RNA trigger host defense countermeasures, however, several aspects of how cytosolic nucleic acids are sensed remain unclear. This special issue reviews how the host innate immune system senses nucleic acids from Brucella abortus, Mycobacterium sp and Legionella pneumophila, viral DNA, the role of STING in DNA sensing and inflammatory disea

DNA interaction with artificial binders is of great interest, especially in light of the broad range of possible biomedical applications. The growing understanding of replication, transcription and translation opened the path for new approaches to target pathological effects at a very early stage. Meanwhile, the competitive binding to nucleic acids by designed molecules, which, for example, block certain sequences for natural binders, such as transcription factors, has become a promising concept in the context of gene therapy. On the other extreme, the transport of nucleic acids over the cell

The discovery of circulating nucleic acids in the 1940s opened up new possibilities for the non-invasive detection, monitoring and screening of various human disorders. Several tumour markers that enable early cancer detection or tumour behaviour prediction have been detected in the plasma of cancer patients. Maternal plasma analysis can be used to detect certain fetal abnormalities, with the quantification of cell-free nucleic acids used to screen for several pregnancy-associated disorders. Some other applications are in transplant monitoring and graft rejection assessment, and in certain med

BACKGROUND: Research into cell-free fetal (cff) nucleic acids has primarily focused on maternal plasma; however, cff DNA and RNA are also detectable in other body fluids such as amniotic fluid (AF). In AF, cff DNA is present in much greater concentrations than in maternal plasma and represents a pure fetal sample uncontaminated by maternal- and trophoblast-derived nucleic acids. The aim of this review was to summarize the current knowledge on cff nucleic acids in AF and to outline future research directions.
METHODS: MEDLINE and PREMEDLINE were searched up to August 2010 for original investig

The existence of circulating nucleic acids in plasma and serum (CNAPS) was first described almost six decades ago. However, the prognostic and diagnostic utility of this circulating DNA/RNA has only really begun to be appreciated in the last decade. Earlier studies concentrated mainly on investigations concerned with fetal medicine and oncology, and significant progress was made in both specialities. More recently the field of enquiry has extended further, and attention has turned to other pathologic states, including trauma, sepsis, myocardial infarction, stroke, transplantation, diabetes mel

Treatment of cell smears (chicken blood, Trypanosoma cruzi epimastigotes, Ehrlich ascites tumor cells and mouse spleen) with simple beta-carboline alkaloids induced a strong bluish white fluorescence emission of condensed chromatin and basophilic cytoplasm under ultraviolet excitation. The compounds used were harmine, harmol, harman, norharman, harmalol and harmaline (25-50 micrograms/ml aqueous solutions for 2-3 min). Spectrofluorometric studies on harmine solutions in vitro (emission peak at 420 nm) showed fluorescence quenching at high concentrations as well as in the presence of DNA. Micro

To test the relationship between tumor malignancy and content and distribution of polyamines and nucleic acids, 2 forms of human gestational trophoblastic tumors were examined: the hydatidiform mole (self-limited form) and the human chorio-carcinoma (invasive form) xenografted into nude mice. The results indicate that there are 2 significant differences between the choriocarcinoma and the mole: 1) the choriocarcinoma is characterized by increased polyamine and nucleic acid levels, 2) tissues differ in their putrescine:spermidine and spermidine:spermine ratios. There is an increase in polyamine

Viral nucleic acids have been found to be infectious for tissues and animals, yet are nonantigenic and resistant to antibodies against whole virus. Other unique properties, such as their host range and their susceptibility to nuclease action, render them a wholly new dimension to be reckoned with in virology. These properties may explain a number of conditions which at present are considered anomalous. The release from infected tissues of even a small proportion of total virus as free nucleic acid could, in an otherwise immune individual, lead to a low level of infection which would, perhaps,

Functional nucleic acids (FNAs), including aptamers and DNAzymes, offer programmable recognition and catalysis for biosensing, but their performance in real samples is often compromised by nuclease degradation, matrix effects, and interface-associated losses. For diagnostic translation, stability must, therefore, be defined not only as strand persistence, but as retained folding, binding, catalysis, and signal generation under assay-relevant conditions. This review examines strategies for stabilizing FNAs in complex biological matrices and organizes the field around four design levers: Chemica

INTRODUCTION: Cell-free fetal nucleic acids in maternal plasma or serum have become important tools in the pursuance of new methods for non-invasive prenatal diagnosis, such as the determination of fetal blood groups and fetal gender. During these pioneering explorations, elevations in the concentration of these new-found biological analytes were noted in several pregnancy-related disorders, including preterm labor, pre-eclampsia and malimplantation. As these elevations appeared to occur before onset of clinical symptoms, it was proposed that such analyses might assist in screening for at-risk

Novel amino-reactive derivatives of lanthanide-based luminescent labels of enhanced brightness and metal retention were synthesized and used for the detection of cDNA oligonucleotides by molecular beacons. Time-resolved acquisition of the luminescent signal that occurs upon hybridization of the probe to the target enabled the avoidance of short-lived background fluorescence, markedly enhancing the sensitivity of detection, which was less than 1 pM. This value is about 50 to 100 times more sensitive than the level achieved with conventional fluorescence-based molecular beacons, and is 10 to 60

It has been known for decades that it is possible to detect small amounts of extracellular nucleic acids in plasma and serum of healthy and diseased human beings. The unequivocal proof that part of these circulating nucleic acids (CNAs) is of tumor origin, initiated a surge of studies which confirmed and extended the original observations. In the past few years many experiments showed that tumor-associated alterations can be detected at the DNA and RNA level. At the DNA level the detection of point mutations, microsatellite alterations, chromosomal alterations, i.e. inversion and deletion, and

Sodium bisulfite is a food and beverage additive. It is also a salt of the urban air pollutant, sulfur dioxide. Bisulfite catalyzes, at neutral pH and physiological temperatures, the transamination reactions of cytosine derivatives with amines. The products of the reactions are N4-substituted cytosines. Both the alpha and epsilon-amino groups of L-lysine react with cytosine, and its nucleosides, in the presence of bisulfite. Bisulfite catalyzes the binding of cytosine to polylysine, lysine to polycytidylic acid, and polysine to polycytidylic acid. Polylysine crosslinks with heat-denatured, but

As the world has been facing several deadly virus crises, including Zika virus disease, Ebola virus disease, severe acute respiratory syndrome (SARS), Middle East respiratory syndrome (MERS), and Coronavirus disease 2019 (COVID-19), lateral flow assays (LFAs), which require minimal equipment for point-of-care of viral infectious diseases, are garnering much attention. Accordingly, there is an increasing demand to reduce the time and cost required for manufacturing LFAs. The current study introduces an equipment-free method of salt-mediated immobilization of nucleic acids (SAIoNs) for LFAs. Com

Medicinal plants, as a core constituent of traditional medicine, provide an essential foundation for preventing and treating human diseases. Research on their pharmacologically active constituents has long focused on secondary metabolites, including alkaloids, terpenoids, flavonoids, and other compounds. However, with the rapid advancement of molecular biology techniques, nucleic acids have gradually become a research focus as a novel class of active components in medicinal plants that possess both nutritional and regulatory functions. This paper systematically reviews the types, characteristi

Therapeutic oligonucleotides targeting GATA3 have emerged as promising immune-modulating drugs for treating ulcerative colitis (UC). However, achieving effective in vivo delivery of therapeutic oligonucleotides to selectively modulate target cells is challenging due to the intestinal barrier and cellular heterogeneity. Here, we demonstrated the intravenous delivery of GATA3-targeted oligonucleotide (hgd40 DNAzyme) to the intestine utilizing tetrahedral framework nucleic acids (tFNAs). The hgd40 DNAzyme was assembled into tFNAs (hgd40-tFNA) via an inflammation-responsive DNA duplex, thereby ena
