Octopamine plasma levels and hepatic encephalopathy: a re-appraisal of the problem.

An investigation on the blood levels of octopamine was carried out on 70 adult individuals. There was a statistically significant correlation between the levels of octopamine and hepatic encephalopathy. Normal subjects had values below 1 ng/ml, while patients with grade 3 or grade 4 encephalopathy constantly showed values above 3.2 ng/ml. In these two groups the distribution was fairly homogeneous. Through the differences between cirrhotics without neurologic involvement and those with grade 1 or 2 hepatic encephalopathy displayed statistical significance, distribution of values in these group

Pharmacology of the octopamine receptor from locust central nervous tissue (OAR3).

1. The present study characterized highly effective agonists from different classes of compounds for the neuronal octopamine receptor (OAR3) of the migratory locust (Locusta migratoria L.). Biogenic amines and phenyliminoimidazolidines (PIIs) were employed for the study of structure-activity relationships. 2. The highest affinity PIIs were predominantly those with substitutions at the positions 2 and 4 of the phenolic ring (e.g. NC 7, KI = 0.3 nM, NC 8, KI = 0.81 nM). Substitutions at these positions always had positive effects on the affinity of the respective agonists. 3. Substitutions at th

Octopamine immunoreactive cell populations in the locust thoracic-abdominal nervous system.

We describe octopamine-immunoreactive somata and their projections in the pro- meso-, meta- and pregenital abdominal-ganglia of locusts. Immunoreactive midline somata were identified as dorsal- and ventral- unpaired median (DUM- and VUM-, respectively) neurones due to their: characteristic large size and positions of somata, primary neurites in DUM-tracts giving rise to T-junctions, and bilaterally projecting axons. In the prothoracic ganglion there are most likely 8 such cells; in the meso- and metathoracic, some 20 each; and in each individual pregenital abdominal ganglion, typically 3. All

AmOctα2R: Functional Characterization of a Honeybee Octopamine Receptor Inhibiting Adenylyl Cyclase Activity.

The catecholamines norepinephrine and epinephrine are important regulators of vertebrate physiology. Insects such as honeybees do not synthesize these neuroactive substances. Instead, they use the phenolamines tyramine and octopamine for similar physiological functions. These biogenic amines activate specific members of the large protein family of G protein-coupled receptors (GPCRs). Based on molecular and pharmacological data, insect octopamine receptors were classified as either α- or β-adrenergic-like octopamine receptors. Currently, one α- and four β-receptors have been molecularly and pha

Octopamine-mediated neuronal plasticity in honeybees: implications for olfactory dysfunction in humans.

Biogenic amines, such as norepinephrine (in vertebrates) and octopamine (in invertebrates), have structural and functional similarities. These amines play crucial roles in animal behavior by modifying the synaptic output of relevant neurons. Increased levels of norepinephrine in the olfactory bulb preferentially increase mitral cell excitatory responses to olfactory nerve inputs, suggesting its critical role in modulating olfactory function including memory formation and/or recall of specific olfactory memories. Increased levels of octopamine in the antennal lobe play an important role in a re

Sustainable and efficient electrochemical determination of octopamine in Apis mellifera for stress monitoring.

This paper presents the design and validation of an electrochemical sensor for the detection of octopamine (OA) in vitro-reared larvae of honey bees (Apis mellifera) using glassy carbon electrodes modified with multi-walled carbon nanotubes and Kolliphor (p188). Octopamine, a key neurohormone involved in the stress response in insects, is studied as an indicator of the physiological and health status of bees under various stress factors. To validate the electrochemical method, a chromatographic approach based on ultra-high-performance liquid chromatography with fast-scanning fluorescence detec

Cloning, expression and functional analysis of an octopamine receptor from Periplaneta americana.

Octopamine regulates multiple physiological functions in invertebrates. The biological effects of octopamine and the pharmacology of octopamine receptors have been extensively studied in the American cockroach, Periplaneta americana. This paper reports the cloning of the first octopamine receptor from Periplaneta americana. A cDNA encoding a putative 7 transmembrane receptor was isolated from the head of Periplaneta americana. The encoded protein contains 628 amino acids and has sequence similarity to other biogenic amine receptors. This protein was expressed in COS-7 cells for radioligand bin

The cardiorespiratory impairment in cirrhosis and sepsis. An experimental interpretation using octopamine infusion.

The effects of octopamine on systemic and pulmonary circulation and on respiratory parameters have been studied in ten O2 100% breathing pigs. A rise in cardiac index (CI) due to an increase in the dynamics of the left ventricle and a progressive hypoxemia, notwithstanding the hyperventilation, was found. The decrease in arterial O2 tension was due to a rise in the pulmonary shunt fraction (QS/QT). The statistical analysis of cardiorespiratory parameters demonstrated that the high flow state linked with a non-Starling mechanism was the causing factor of a ventilation: perfusion ratio (VA/QT) d

CREB promotes starvation-induced sleep loss by regulating octopamine production in Drosophila.

Starvation induces robust food-seeking behavior in animals as a fundamental survival response. In Drosophila, biogenic amine octopamine (OA) plays a critical role in starvation-induced locomotor hyperactivity and awakeness, while the underlying signaling cascade remains not fully understood. Here, we demonstrated that the conserved transcription factor cAMP-Responsible Element Binding Protein (CREB), activated during starvation through the SIK2-CRTC pathway, contributes to starvation-induced sleep reduction. Mechanistically, we found that tyramine β-hydroxylase, the rate-limiting enzyme for OA

Octopamine signaling from clock neurons plays dual roles in Drosophila long-term memory.

Circadian clock genes are best known for regulating circadian rhythms, but they also play crucial roles in memory processes. This suggests that memory is modulated by neural networks containing clock neurons, although the underlying mechanisms remain unclear. In Drosophila melanogaster, approximately 240 clock neurons are grouped into at least eight distinct clusters. Among them, the dorsal-lateral neurons (LNds) are required for maintaining long-term memory (LTM). In contrast, the neuropeptide Pigment-dispersing factor (Pdf), expressed in both small and large ventral-lateral neurons (s-LNvs a

Selective activation of beta3-adrenoceptors by octopamine: comparative studies in mammalian fat cells.

Numerous synthetic agonists selectively stimulate beta3-adrenoceptors (ARs). The endogenous catecholamines, noradrenaline and adrenaline, however, stimulate all the beta-AR subtypes, and no selective physiological agonist for beta3-ARs has been described so far. The aim of this study was to investigate whether any naturally occurring amine can stimulate selectively beta3-ARs. Since activation of lipolysis is a well-known beta-adrenergic function, the efficacy and potency of various biogenic amines were compared with those of noradrenaline, isoprenaline, and beta3-AR agonists 4-(-{[2-hydroxy-(3

Octopamine relaxes rabbit jejunal smooth muscle by selective activation of dopamine D1 receptors.

The effect of octopamine on intestinal smooth muscle of rabbit isolated jejunum has been studied. Octopamine induced a dose-dependent decrease of muscle tone and this reproducible relaxation was not modified by tetrodotoxin or by agents that acted on adrenergic nerve terminals. Adrenoceptor antagonists, at concentrations sufficient to block each adrenoceptor type, did not reduce the actions of octopamine. On the other hand, octopamine-induced relaxations were affected by agents that have the ability to change cyclic AMP (cAMP) content; such as alloxan (an adenylate cyclase inhibitor), imidazol

Dopamine and octopamine influence avoidance learning of honey bees in a place preference assay.

Biogenic amines are widely characterized in pathways evaluating reward and punishment, resulting in appropriate aversive or appetitive responses of vertebrates and invertebrates. We utilized the honey bee model and a newly developed spatial avoidance conditioning assay to probe effects of biogenic amines octopamine (OA) and dopamine (DA) on avoidance learning. In this new protocol non-harnessed bees associate a spatial color cue with mild electric shock punishment. After a number of experiences with color and shock the bees no longer enter the compartment associated with punishment. Intrinsic

An octopamine-glutamate cotransmitting neuron gates aggressive escalation through layered inhibition in Drosophila.

Aggression is an evolutionarily conserved behavior essential for survival and reproduction, yet escalation to high-intensity forms entails substantial metabolic costs and injury risks, necessitating precise neural control. Using Drosophila melanogaster, we uncovered a multilayered inhibitory circuit that constrains aggressive escalation. This circuit involves a cotransmitting octopamine-glutamate ventral paired medial 4 (VPM4) neuron and its downstream GABAergic target, MBON-11. Neurotransmitter-specific manipulations reveal that octopamine and glutamate release from VPM4 is independently regu

Functional characterization of β-adrenergic-like octopamine receptors in planthopper reproduction and feeding.

As an essential biogenic amine in invertebrates, octopamine (OA) regulates multiple physiological and behavioral processes via binding to octopamine receptors (OARs). The small brown planthopper Laodelphax striatellus is an important agricultural pest. However, little is known about OARs in L. striatellus. Herein, three β-adrenergic-like OARs (LsOA2s) were cloned, including LsOA2B1, LsOA2B2, and LsOA2B3. LsOA2s were expressed at the highest levels in egg stage and brain. Knockdown of LsOA2s significantly decreased the transcript levels of vitellogenin (LsVg) and Vg receptor (LsVgR), and reduce

Octopamine Signaling Regulates Phototactic Behavior via a β-Adrenergic-like Receptor in Diaphorina citri.

The Asian citrus psyllid (Diaphorina citri, ACP) is the primary vector of citrus huanglongbing and exhibits positive phototactic behavior. Octopamine (OA) is a critical neuromodulator of diverse insect behaviors, yet its regulatory mechanism in ACP phototactic behavior remains largely uncharacterized. In this study, three core genes of the OA signaling pathway in ACP were identified and characterized: the biosynthetic enzyme DcTβH and two β-adrenergic-like OA receptors (DcOctβ1R, DcOctβ2R). All three genes exhibited a U-shaped developmental expression pattern and high transcript abundance in t

Characterization of an octopamine receptor, and its involvement in the immune response of cuttlefish Sepiella japonica.

Octopamine (OA) can regulate immune response because immune cells exist their receptors. In the study, the full-length cDNA of octopamine receptor (OAR) from Sepiella japonica (termed as Sj-OAR) was amplified. Sj-OAR was 1633 bp in length encoding 348 amino acid residues. Sj-OAR owned seven conserved transmembrane domains. Sj-OAR shared the highest identity with β2-adrenergic OARs of cephalopods, and clustered together with β2 OAR clade. Sj-OAR had the highest expression level in pancreas, followed in liver, optic lobe and brain; Sj-OAR kept increasing in pancreas during the whole growth perio

Differential effects of β-methylphenylethylamine and octopamine on contractile parameters of the rat gastrointestinal tract.

This study tested the effects of β-methylphenylethylamine (β-MPEA) and octopamine on contractile parameters of the gastrointestinal tract in rats. We hypothesized that some of their effects result from interactions with trace amine (TA)-associated receptors or serotoninergic 5-hydroxytryptamine (5-HT) receptors. β-MPEA-induced contractions in rat gastric fundus strips under resting tonus conditions, but induced relaxation in preparations that were previously contracted with carbachol. Octopamine relaxed gastric fundus strips maintained at resting tonus or contracted with carbachol. The contrac

The effects of p-octopamine on salivary flow rates and protein secretion by rat submandibular glands.

The effects of p-octopamine injected i.v. and i.p on salivary flow rates and proteins secreted by the submandibular glands of rats were studied with and without various types of autonomic blockers at different doses, and with two enzyme inhibitors. The salivary flow rates and the amounts of protein secreted progressively increased with increasing doses injected both i.v. and i.p., whereas they were dramatically reduced with almost all autonomic blockers and disulfiram, a dopamine-beta-hydroxylase inhibitor. Salivation was completely abolished in response to p-octopamine in combination with met
