[Ocular hypotensive action of l-stepholidine].

Ocular hypotension was made by iv 20% NaCl 2.5 ml/kg in rabbits. On this model, l-stepholidine (l-SPD) lowered intraocular pressure when administered topically to the eyes. The ocular hypotensive action of 0.5% l-SPD was weaker than that of 0.5% timolol, but stronger than that of 0.5% pilocarpine. The maximal actions of the 3 drugs were similar. There were no significant changes of pupil diameter and carbonic anhydrase activity after l-SPD administered, but which suppressed aqueous humor formation. As the ciliary process of the eye behaves much like the renal glomerulus, dopamine increases the…

Fadolmidine-induced ocular hypotension in normotensive rabbits.

Fadolmidine, a novel selective alpha2-adrenoceptor agonist, was evaluated for its efficacy to lower intraocular pressure in normotensive rabbits (n=5-6). The dose-response profile between 0.004 microg and 12.5 microg of fadolmidine was determined. The effect of pH on the partition of fadolmidine was studied in order to select an optimal pH for topical fadolmidine administration. After topical administration, fadolmidine significantly lowered the intraocular pressure in normotensive rabbits. The onset of action was immediate, with no initial increase in intraocular pressure. A significant decre

Mature secondary cataracts and reversible ocular hypotension.

Three eyes of three patients are reported with ocular hypotension and mature, white cataracts secondary to other ocular disease. Ocular pressures rapidly rose to and remained at normal levels with recovery of excellent vision in all three eyes following cataract extraction, with follow-up periods of 16 to 28 months. Thus a hypotensive eye with a mature, white cataract secondary to other eye disease is not necessarily hopeless, and hypotony is not a contraindication to cataract extraction in such patients.

The effect of ocular hypotensive agents on macula.

We evaluated the effect of long-term topical antiglaucoma drugs on the macula using the noninvasive macular visual field threshold test in 100 eyes of 75 patients with ocular hypertension or primary open-angle glaucoma. We found that topical antiglaucoma agents can alter macular sensitivity after long-term therapy and advise evaluation of the macula in glaucoma patients receiving antiglaucomatous agents.

Prostaglandin derivates as ocular hypotensive agents.

Low doses of naturally occurring prostaglandins reduce the intraocular pressure (IOP) in many species. Species differences do occur both in terms of efficiency and mechanism of action, and also among the different prostaglandins. Among the prostaglandins mainly PGF2 alpha has been tested in human eyes. Although it is an effective ocular hypotensive drug it is not clinically useful due to pronounced ocular side-effects, mainly conjunctival hyperemia and irritation, at doses that produce a maximal effect on IOP. Modification of the drug has resulted in two analogues that are now in clinical use,

Correlates of acute exercise-induced ocular hypotension.

PURPOSE: To understand those factors that determine the decrease in intraocular pressure (IOP) that occurs during acute dynamic exercise. METHODS: Three aspects of the exercise-IOP relationship were studied. These included graded exercise, with and without CO2 addition for isocapnia; comparison of the IOP response of trained and sedentary subjects to a fixed external work load; and exercise after ocular beta-adrenoceptor blockade. Graded exercise consisted of 7 minutes each at 30 and 90 watts on a cycle ergometer, then progressive work to exhaustion. Trained and sedentary subjects were define

Ocular hypotension, ocular toxicity, and neurotoxicity in response to marihuana extract and cannabidiol.

Marihuana extract containing 21.3% delta-9-tetrahydrocannabinol (100 micrograms/hr), delta-9-tetrahydrocannabinol (20 micrograms/hr), cannabidiol (20 micrograms/hr), or the polyethylene glycol vehicle (1 microliter/hr) was delivered topically to cat eyes via osmotic minipumps over a 9-day period. Intraocular pressure differences between treated and untreated eyes of cats receiving marihuana extract remained 3-4 mmHg lower than those for vehicle controls, while differential values for the delta 9-THC-treated group remained reduced by 3-5 mmHg; data for these two groups did not differ statistica

Effect of vehicle on pilocarpine-induced ocular hypotension.

The effect of 1.4% polyvinyl alcohol and castor oil vehicles on pilocarpine-induced hypotension was studied in 50 patients with open-angle glaucoma. Diurnal tension curves showed that the mean intraocular pressure was lower and the maximum diurnal variation smaller during the treatment with oily drops of 2% and 4% pilocarpine instilled 2-3 times daily than when 2% and 4% pilocarpine in polyvinyl alcohol was instilled 3-4 times daily. The differences were statistically significant. In long-term treatment no marked pilocarpine tolerance developed in cases treated with oily drops of pilocarpine.

Topical ocular hypotensive activity and ocular penetration of dichlorphenamide sodium in rabbits.

A single topical instillation (50 microliter) of the water soluble sodium salt of dichlorphenamide (10%) produced a pronounced and prolonged lowering of intraocular pressure (IOP), compared to suspensions of its free acid, in rabbits with alpha-chymotrypsin-induced ocular hypertension. In normal rabbits, instillation of dichlorphenamide sodium also produced a markedly elevated drug aqueous humor level relative to instillation of its free acid, indicating enhanced penetration into the eye. The IOP response after ocular instillation of dichlorphenamide sodium was equivalent to that produced by o

Interventions for improving adherence to ocular hypotensive therapy.

BACKGROUND: Poor adherence to therapy is a significant healthcare issue, particularly in patients with chronic disease such as open-angle glaucoma. Treatment failure may necessitate unwarranted changes of medications, increased healthcare expenditure and risk to the patient if surgical intervention is required. Simplifying eye drop regimes, providing adequate information, teaching drop instillation technique and ongoing support according to the patient need may have a positive effect on improving adherence. OBJECTIVES: To summarise the effects of interventions for improving adherence to ocula

[The ocular hypotensive effect and safety of 0.2% brimonidine].

PURPOSE: To study the ocular hypotensive efficacy and safety of 0.2% Brimonidine when used in chinese people. METHODS: Using randominzed parallel group control study, 0.2% Brimonidine twice a day or 1% Carteolol twice a day was applied in patients with primary open angle glaucoma or ocular hypertension for three months. RESULTS: The results showed that 0.2% Brimonidine and 1% Carteolol both have good ocular hypotensive efficacy. There was no significant difference between them. 0.2% Brimonidine did not produce significant changes in pupil size and heart rate. Change in blood pressure was not

Ocular hypotensive effects of medetomidine and its analogs.

The ocular hypotensive effects of medetomidine, a relatively selective alpha 2-agonist, and its analogs were tested in rabbits and cats and their inhibition of adenylate cyclase in the isolated bovine ciliary process was also studied. It was found that topical unilateral administration of medetomidine (0.5-2.0%) to the normotensive rabbits produced a dose-dependent bilateral decrease in IOP with peak reduction in IOP at 2 hr in the treated eye and 1 hr in the untreated eye. A dose-dependent mydriasis was also observed in the treated eye. The dose-response curves of medetomidine and its analogs
