Enhancing wine fermentation through concurrent utilization of Lachancea thermotolerans and lactic acid bacteria (Oenococcus oeni and Lactiplantibacillus plantarum) or Schizosaccharomyces pombe.

Most commercially available red wines undergo alcoholic fermentation by Saccharomyces yeasts, followed by a second fermentation with the lactic acid bacteria Oenococcus oeni once the initial process is complete. However, this traditional approach can encounter complications in specific scenarios. These situations pose risks such as stalled alcoholic fermentation or the growth of undesirable bacteria while the process remains incomplete, leaving residual sugars in the wine. To address these challenges and the issue of low acidity prevalent in warmer viticultural regions, several novel alternati

Indigenous Lactiplantibacillus plantarum and Oenococcus oeni in malolactic fermentation: effects of co-inoculation strategy and inoculation timing on fermentation kinetics and aroma profiles.

Malolactic fermentation (MLF) plays a key role in regulating acidity, microbial stability, and aroma development in red wine. Indigenous Lactiplantibacillus plantarum isolates were assessed for their potential as malolactic starter cultures, and their role in mixed fermentation with Oenococcus oeni was investigated under different inoculation strategies during Cabernet Sauvignon winemaking. Sixty isolates were screened for ethanol tolerance, β-glucosidase activity, and malolactic performance, and three strains (C10, B16, and D30) were selected for microvinification trials. Mixed MLF was associ

Rapid assessment of Oenococcus oeni activity by measuring intracellular pH and membrane potential by flow cytometry, and its application to the more effective control of malolactic fermentation.

The aim of this study is to highlight the changes in the physiological cellular state of Oenococcus oeni during malolactic fermentation (MLF), and to use its cellular parameters to improve existing knowledge of O. oeni behaviour and to more effectively control the performance of the bacteria during MLF in wine. To do this, measurements of intracellular pH, transmembrane potential and vitality were performed using flow cytometry with different fluorescent probes: CFDA-SE and CDCF, DiBAC and CFDA, respectively. The kinetics of the cellular changes in these parameters were determined during MLF i

Microbial Decontamination of Red Wine by Pulsed Electric Fields (PEF) after Alcoholic and Malolactic Fermentation: Effect on Saccharomyces cerevisiae, Oenococcus oeni, and Oenological Parameters during Storage.

New techniques are required to replace the use of sulfur dioxide (SO2) or of sterilizing filtration in wineries, due to those methods' drawbacks. Pulsed electric fields (PEF) is a technology capable of inactivating microorganisms at low temperatures in a continuous flow with no detrimental effect on food properties. In the present study, PEF technology was evaluated for purposes of microbial decontamination of red wines after alcoholic and malolactic fermentation, respectively. PEF combined with SO2 was evaluated in terms of microbial stability and physicochemical parameters over a period of f

Acetaldehyde metabolism by wine lactic acid bacteria.

Acetaldehyde is a volatile flavor compound present in many fermented foods and is important in the production of red and white wines. Nine strains of the genera Lactobacillus and Oenococcus were able to metabolize acetaldehyde in a resting cell system, whereas two Pediococcus strains were not. Acetic acid and ethanol were produced from its degradation. A Lactobacillus and an Oenococcus were able to degrade SO(2)-bound acetaldehyde, as well. A coincubation of resting cells of Saccharomyces bayanus Première Cuvée and Oenococcus oeni Lo111 showed that strain Lo111 metabolized acetaldehyde produce

Effects of the addition of fumaric acid on the spontaneous alcoholic fermentation of white grape must.

Consumer demand is driving the production of wines without added sulfur dioxide or yeast inoculation. However, this approach can lead to organoleptic defects or health risks due to unwanted microbial activity during alcoholic fermentation (AF). Lactic acid bacteria (LAB), responsible for malolactic fermentation (MLF), may also affect wine quality. Fumaric acid (FUM) inhibits LAB growth and could serve as an alternative to SO₂ in controlling MLF. The objective of this work was to study the effects of adding FUM (0.6 g/L) to natural white grape must without SO2 on the growth of LAB during sponta

Immobilized lysozyme for the continuous lysis of lactic bacteria in wine: Bench-scale fluidized-bed reactor study.

Lysozyme from hen egg white (HEWL) was covalently immobilized on spherical supports based on microbial chitosan in order to develop a system for the continuous, efficient and food-grade enzymatic lysis of lactic bacteria (Oenococcus oeni) in white and red wine. The objective is to limit the sulfur dioxide dosage required to control malolactic fermentation, via a cell concentration typical during this process. The immobilization procedure was optimized in batch mode, evaluating the enzyme loading, the specific activity, and the kinetic parameters in model wine. Subsequently, a bench-scale fluid

Cytofluorometric detection of wine lactic acid bacteria: application of malolactic fermentation to the monitoring.

In this study we report for the first time a rapid, efficient and cost-effective method for the enumeration of lactic acid bacteria (LAB) in wine. Indeed, up to now, detection of LAB in wine, especially red wine, was not possible. Wines contain debris that cannot be separated from bacteria using flow cytometry (FCM). Furthermore, the dyes tested in previous reports did not allow an efficient staining of bacteria. Using FCM and a combination of BOX/PI dyes, we were able to count bacteria in wines. The study was performed in wine inoculated with Oenococcus oeni (10(6) CFU ml(-1)) stained with ei

Interaction effects of fumaric acid, pH and ethanol on the growth of lactic and acetic acid bacteria in planktonic and biofilm states.

The microbial stability of wine can be compromised by the presence of lactic acid bacteria (LAB) and acetic acid bacteria (AAB), which can cause spoilage via off flavour production, increased acetic acid production, or biofilm formation. To manage the growth of LAB in winemaking, fumaric acid (FA) has been proposed as an alternative to traditional antimicrobial agents, such as sulfur dioxide (SO2). This study aimed to evaluate the inhibitory effects of FA on the growth of LAB and AAB based on the influence of pH and ethanol in a synthetic wine-like medium. The research involved the determinati

Bacterial spoilage of wine and approaches to minimize it.

Bacteria are part of the natural microbial ecosystem of wine and play an important role in winemaking by reducing wine acidity and contributing to aroma and flavour. Conversely, they can cause numerous unwelcome wine spoilage problems, which reduce wine quality and value. Lactic acid bacteria, especially Oenococcus oeni, contribute positively to wine sensory characters, but other species, such as Lactobacillus sp. and Pediococcus sp can produce undesirable volatile compounds. Consequences of bacterial wine spoilage include mousy taint, bitterness, geranium notes, volatile acidity, oily and sli

Molecular characterization of lactic acid populations associated with wine spoilage.

We have investigated the prevalence of spoilage lactic acid bacteria (LAB) in table wines produced in the Apulia region. The occurrence of LAB was evaluated in wines produced with low sulphur dioxide doses and not supplemented with selected malolactic starters such as Oenococcus oeni. About 150 strains were isolated from wine must and a molecular characterization was performed using PCR-based techniques. Most of the strains analysed belonged to Lactobacillus plantarum species. However, some of the strains were identified as Pediococcus damnosus and Leuconostoc sp. The amplified fragments of Pe

Encapsulated lactic acid bacteria for control of malolactic fermentation in wine.

The kinetics of both malolactic fermentation in Chardonnay wine by encapsulating Lactobacillus casei cells in pectate gel and lyophilized Oenococcus oeni culture has been carried out. The influence of acidity, sulfur dioxide content, and organic acid content on the malolactic activity of the bacteria has been controlled. Encapsulated bacteria degraded 30%, of malic acid in white wine, deacidifying it from pH 3.15 to 3.40, whereas the lyophilized culture degraded 48% of malic acid, deacidifying from pH 3.15 to 3.60. The degree of conversion of malic acid in wine by the encapsulated cells was tw

Updated insights into esterified Flavan-3-ols in grapevine and wine: Origin, transformation, and taste response of epicatechin-3-O-vanillate and -3-O-gallate.

Esterified flavan-3-ols, prevalent in grapes and wine as monomers or subunits within condensed tannins, significantly influence the mouthfeel of wine. However, comprehensive knowledge of their molecular structures and transformations during vinification is still limited. This study investigated tannin derivatives, specifically vanillate and gallate forms, using UHPLC-Q-TOF to elucidate their distribution across four Vitis vinifera L. cultivars, their transformations during vinification, and their metabolic fate mediated by Oenococcus oeni and β-glucosidase activity. The taste responses of epic

Inhibition of malolactic fermentation by a peptide produced by Saccharomyces cerevisiae during alcoholic fermentation.

The ability of Saccharomyces to inhibit Oenococcus oeni during the alcoholic fermentation by mechanisms other than SO(2) production was investigated. During fermentation in synthetic grape juice, S. cerevisiae strain RUBY.ferm inhibited the malolactic fermentation by O. oeni while strain EC1118 did not despite both strains producing similar amounts of SO(2). The bacterial inhibition exerted by RUBY.ferm was diminished when the wine was treated with proteases but not through the addition of nutrients. Wine fermented by RUBY.ferm was fractionated based on molecular weight and each fraction teste

The 'buttery' attribute of wine--diacetyl--desirability, spoilage and beyond.

The diketone, diacetyl, is a major flavour metabolite produced by lactic acid bacteria (LAB). Of the LAB associated with wine, Oenococcus oeni is encouraged during the malolactic (ML) fermentation, a biodeacidification of wine during which the metabolism of diacetyl occurs. Diacetyl, which imparts a buttery aroma and flavour to many fermented foods and beverages, is a key flavour compound of most fermented dairy products. In wine, diacetyl has important stylistic implications. The biosynthesis of diacetyl is dependent upon citric acid metabolism and diacetyl is an intermediate metabolite which

Wild Australian niches as a source of novel lactic acid bacteria and malolactic fermentation candidates.

Malolactic fermentation (MLF) by lactic acid bacteria (LAB) is essential for most red wine production, yet commercial starter cultures are largely limited to Oenococcus oeni, a species constrained by slow growth, fastidious nutritional requirements, and poor co-inoculation competitiveness. To identify novel MLF candidates from diverse ecological niches, we assembled 714 wild LAB strains from seven Australian geographic regions and eight ecological sources, including insects, flowers, honey, and fermented foods, spanning 21 genera. Four hundred strains were screened in a microplate assay quanti

Evaluation of Specific Torulaspora delbrueckii Fractions to Stimulate Malolactic Fermentation in Limiting Conditions.

Malolactic fermentation (MLF) is a bioprocess driven by lactic acid bacteria (LAB), which is desired in red and highly acidic white wines. Among all LAB, Oenococcus oeni is the main species in wine, followed by Lactiplantibacillus plantarum. The harsh conditions found in wine-not only due to the low nutrient concentration but also the presence of antimicrobial compounds such as ethanol, high acidity, SO2, and polyphenols-can compromise MLF performance. In recent years, the use of certain non-Saccharomyces yeasts, such as Torulaspora delbrueckii or Metschnikowia pulcherrima, as starter cultures

Pesticides' influence on wine fermentation.

Wine quality strongly depends on the grape quality. To obtain high-quality wines, it is necessary to process healthy grapes at the correct ripeness stage and for this reason the farmer has to be especially careful in the prevention of parasite attacks on the grapevine. The most common fungal diseases affecting grape quality are downy and powdery mildew (Plasmopara viticola and Uncinula necator), and gray mold (Botrytis cinerea). On the other hand, the most dangerous insects are the grape moth (Lobesia botrana), vine mealybug (Planococcus ficus), and the citrus mealybug (Planococcus citri). Far
