Olfactory Mucosa Mesenchymal Stem Cells (OM-MSCs)-Derived Exosomes Mitigate Neuroinflammation and Nerve Injury After Intracranial Hemorrhage (ICH) Through miR-30-5p and Its Associated Mechanisms.

Exosomes derived from olfactory mucosa mesenchymal stem cells (OM-MSCs-EXOs) offer a novel therapeutic strategy for various diseases due to their role in regulating intercellular signaling. However, the potential of OM-MSCs-EXOs to improve intracranial hemorrhage (ICH) and its underlying mechanisms is unclear. In this study, we found that OM-MSCs-EXOs can perfect neurofunctional recovery after ICH. The level of miR-30b-5p declined in thrombin-triggered OM-MSCs. Suppression of miR-30b-5p in OM-MSCs-EXOs inhibited mitophagy and promoted reactive oxygen species (ROS) and inflammasome-related prot

The potential therapeutic applications of olfactory ensheathing cells in regenerative medicine.

Olfactory ensheathing cells (OECs) are unique glia cells restricted to the primary olfactory system including the olfactory mucosa, olfactory nerve, and the outer nerve layer of the olfactory bulb. OECs guide growing olfactory axons from the neurons of the nasal cavity olfactory mucosa to the olfactory bulb to connect both the peripheral nervous system (PNS) and central nervous system (CNS). Based on these specialized abilities of OECs, transplantation of OECs to injury sites has been widely investigated for their potential therapeutic applications in neural repair in different injuries. Odor detection ability and thallium-201 transport in the olfactory nerve of traumatic olfactory-impaired mice.

Although olfactory nerve damage is a contributing factor in the diagnosis of posttraumatic olfactory loss, at present, there are no methods to directly assess injury to these nerves. We have shown that following olfactory nerve injury in mice, thallium-201 (201 Tl) transport from the nasal cavity to the olfactory bulb decreases. To determine if olfactory function after nerve injury could be assessed with nasal administration of 201 Tl, we measured the correlation between odor detection ability (ODA) and the rate of transport of 201 Tl in olfactory nerves. Both ODA and 201 Tl transport were mea

Blockade of interleukin-6 receptor suppresses inflammatory reaction and facilitates functional recovery following olfactory system injury.

We previously reported that anti-inflammatory treatment with steroids improves recovery outcome in an olfactory nerve injury model. Clinically, however, steroid administration is not recommended in the acute phase of head injury because of concerns regarding side effects and no evidence of its efficacy. Recently, it has been reported that interleukin-6 (IL-6) plays an important role in the inflammatory reaction. The present study investigates if anti-IL-6 receptor (IL-6R) antibody can facilitate functional recovery in the olfactory system following injury. Rat anti-mouse IL-6R antibody (MR16-1

Thallium transport and the evaluation of olfactory nerve connectivity between the nasal cavity and olfactory bulb.

Little is known regarding how alkali metal ions are transported in the olfactory nerve following their intranasal administration. In this study, we show that an alkali metal ion, thallium is transported in the olfactory nerve fibers to the olfactory bulb in mice. The olfactory nerve fibers of mice were transected on both sides of the body under anesthesia. A double tracer solution (thallium-201, (201)Tl; manganese-54, (54)Mn) was administered into the nasal cavity the following day. Radioactivity in the olfactory bulb and nasal turbinate was analyzed with gamma spectrometry. Auto radiographic

The risk of olfactory disturbance from conchal plate injury during ethmoidectomy.

BACKGROUND: Even though the olfactory function usually is improved or unchanged after ethmoidectomy, some patients complain of olfactory disturbance, which may be caused by direct injury to the olfactory nerves. However, the detailed anatomic pathway of the olfactory nerves that leave the olfactory mucosa and after going through the cribriform plate insert into the olfactory bulb is uncertain. In the studies of lateral nasal wall, we referred to the thin bone that connects the middle, superior, and supreme turbinates to the skull base as the "conchal plate." We undertook this study to determin

High-dose IgG suppresses local inflammation and facilitates functional recovery after olfactory system injury.

OBJECTIVE: Head trauma can be a cause of refractory olfactory dysfunction due to olfactory nervous system injury. Anti-inflammatory treatment using steroids or anti-cytokine agents is known to contribute to functional recovery of the central and peripheral nervous systems in injury models, while there is a concern that they can induce adverse reactions. The present study examines if high-dose immunoglobulin G (IgG) can facilitate olfactory functional recovery following injury. METHODS: Olfactory nerve transection (NTx) was performed in OMP-tau-lacZ mice to establish injury models. High-dose I

Aging inhibits olfactory recovery from traumatic olfactory system injury.

Although recent advances in treatment of traumatic olfactory dysfunction, which had a low rate of improvement through treatment, have increased the improvement rate in younger patients, it remains low in middle-aged and older patients. Although olfactory function declines with age, its impact on traumatic dysfunction recovery remains unclear. This study was conducted to investigate the effects of aging on olfactory nerve regeneration and olfactory function recovery in an animal model of traumatic olfactory injury. Behavioral experiment using olfactory preference and morphological measurements

Chapter 22: Transplantation of olfactory ensheathing cells for peripheral nerve regeneration.

Peripheral nerve injury is a common clinical problem, and the development of novel strategies to enhance peripheral nerve regeneration is important. Traumatic events, including motor vehicle accidents, sports-related injuries, violence, and falls, lead to significant numbers of peripheral nerve lesions. Traumatic nerve injuries are often associated with life-threatening injuries, which must be treated first. During the delay in nerve repair, the transected nerves undergo Wallerian degeneration. Therefore, delay before surgical treatment is critical, but care must also be taken to ensure that n

CNPase expression in olfactory ensheathing cells.

A large body of work supports the proposal that transplantation of olfactory ensheathing cells (OECs) into nerve or spinal cord injuries can promote axonal regeneration and remyelination. Yet, some investigators have questioned whether the transplanted OECs associate with axons and form peripheral myelin, or if they recruit endogenous Schwann cells that form myelin. Olfactory bulbs from transgenic mice expressing the enhanced green fluorescent protein (eGFP) under the control of the 2-3-cyclic nucleotide 3-phosphodiesterase (CNPase) promoter were studied. CNPase is expressed in myelin-forming

A Time Limit for Initiating Anti-Inflammatory Treatment for Improved Olfactory Function after Head Injury.

We previously reported that treatment with an anti-inflammatory drug, specifically a steroid, is effective in improving recovery during the acute phase of head injury. Clinically, however, patients with head injury usually become aware of their olfactory loss several weeks or months after the injury, which may be a critical factor in poor recovery from olfactory dysfunction. This raises an important question: When should steroid administration begin in order to achieve optimum improvement of olfactory dysfunction? The present study was designed to reveal the time limit for starting anti-inflam

Retinoic acid enhances the rate of olfactory recovery after olfactory nerve transection.

In the olfactory system, retinoic acid (RA) plays an important role in development and may affect growth in the adult animal. To explore the potential effects of RA on recovery after injuries, adult mice were trained in a buried food paradigm and were given a single oral supplement of RA after olfactory nerve transection. Results demonstrate that RA accelerates the recovery of olfactory functions after injury.

Ultrastructural analysis of olfactory ensheathing cells derived from olfactory bulb and nerve of neonatal and juvenile rats.

Olfactory nerve derived and olfactory bulb derived olfactory ensheathing cells (OECs) have the ability to promote axonal regeneration and remyelination, both of which are essential in a successful cell transplant. Thus, morphological identification of OECs is a key aspect to develop an applicable cell therapy for injuries to the nervous system. However, there is no clear definition regarding which developmental stage or anatomical origin of OECs is more adequate for neural repair. In the present study, an ultrastructural comparison was made between OECs recovered from primary cultures of olfac

Recovery of olfactory behavior. I. Recovery after a complete olfactory bulb lesion correlates with patterns of olfactory nerve penetration.

The olfactory system is an excellent system in which to study issues related to potential functional recovery after a debilitating brain injury. The olfactory system is well-characterized, easily accessible and there are a vast number of studies available from a variety of perspectives. The experimental aim of this research is to examine the anatomical correlates associated with potential behavioral recovery in rats that receive complete olfactory bulb lesions as neonates or as adults. The results show that behavioral recovery occurs only when olfactory nerve penetration of the central nervous

Subjective and Objective Assessments of Post-traumatic Olfactory Dysfunction.

Introduction: Traumatic brain injuries are the most common cause of olfactory dysfunction. Deficits in olfaction may be conductive or neurosensory in nature, with varying degrees of impairment resulting in a diminished quality of life and an increased risk for personal injury among patients. The aim of this research is to evaluate the results of the subjective and objective quantitative examinations of olfactory function in a group of patients with post-traumatic anosmia in order to predict its value in identifying olfactory deficits in clinical practice. Materials and Methods: The present stu

Techniques to prevent olfactory nerve reconstitution in the pigeon.

Transectioned olfactory nerves in birds can reestablish structural and functional connections with the olfactory bulbs in comparatively short time spans, thereby hampering long-range studies of avian olfaction and behavior. Accordingly, techniques are described that are suitable for impeding the reconstitution of the olfactory nerve after its transection in the pigeon. These involve the use of inexpensive and easily obtainable materials including cotton pellets, glass beads, and polyethylene tubing.

Neural regeneration and the peripheral olfactory system.

The peripheral olfactory system is able to recover after injury, i.e., the olfactory epithelium reconstitutes, the olfactory nerve regenerates, and the olfactory bulb is reinnervated, with a facility that is unique within the mammalian nervous system. Cell renewal in the epithelium is directed to replace neurons when they die in normal animals and does so at an accelerated pace after damage to the olfactory nerve. Neurogenesis persists because neuron-competent progenitor cells, including transit amplifying and immediate neuronal precursors, are maintained within the population of globose basal

The olfactory network of larval Xenopus laevis regenerates accurately after olfactory nerve transection.

Across vertebrate species, the olfactory epithelium (OE) exhibits the uncommon feature of lifelong neuronal turnover. Epithelial stem cells give rise to new neurons that can adequately replace dying olfactory receptor neurons (ORNs) during developmental and adult phases and after lesions. To relay olfactory information from the environment to the brain, the axons of the renewed ORNs must reconnect with the olfactory bulb (OB). In Xenopus laevis larvae, we have previously shown that this process occurs between 3 and 7 weeks after olfactory nerve (ON) transection. In the present study, we show t
