Functional Metagenomics Reveals Gut Microbial Biosynthetic Potential for Essential Nutrients in Juvenile Chum Salmon.

The fish gut microbiota contributes to host nutrition, immunity and homeostasis, yet how its metabolic capacity relates to gaps in the host genome remains poorly characterized. Here we used shotgun metagenomic sequencing and KEGG module completion analysis to compare the biosynthetic potential of the gut microbiome with that of the host genome in juvenile chum salmon (Oncorhynchus keta) at 35 days posthatching, when exogenous feeding begins. Because small juvenile guts yield low microbial biomass and a high proportion of host DNA, we first optimized a protocol combining enzymatic lysis with NE

Behavioral responses by migratory chum salmon to amino acids in natal stream water.

We propose that amino acids in natal stream water have Important roles in Pacific salmon homing. This study hypothesized that amino acids found in natal stream water have a role in the ability of mature male chum salmon (Oncorhynchus keta) to home to the Osaru River (OR), Hokkaido, Japan. Behavioral experiments were conducted in a two-choice test tank using various combinations of control water (natural Toya Lake water; NLW and three artificial stream waters using amino acids: 1) artificial OR water (AOR); 2) AOR without L-glutamic acid, the major amino acid in OR water (AOR-E); and 3) artific

Lack of circadian regulation of in vitro melatonin release from the pineal organ of salmonid teleosts.

In many teleost species, the photoreceptive pineal organ harbors the circadian clock that regulates melatonin release in the pineal organ itself. However, the pineal organ of three salmonids (rainbow trout Oncorhynchus mykiss, masu salmon Oncorhynchus masou, and sockeye salmon Oncorhynchus nerka) did not exhibit circadian rhythms in melatonin release when maintained under constant darkness (DD) in vitro, suggesting that the pineal organs of all salmonids lack the circadian regulation of melatonin production. To test this hypothesis, the pineal organ of seven salmonids (common whitefish Coregon

Trends in sea lice infestations on chum and pink salmon in the Broughton Archipelago remain unchanged despite removal of finfish aquaculture.

To better understand the relative contributions of sea lice Lepeophtheirus salmonis and Caligus clemensi from farmed and non-farmed sources, infestations with sea lice are described on juvenile chum salmon Oncorhynchus keta and pink salmon O. gorbuscha from the Broughton Archipelago (BA) in coastal British Columbia, Canada, during a period of declining salmon aquaculture presence. A total of 2868 salmon were collected by beach seine from 14 sites between 2016 and 2024 and examined for sea lice infestation by microscope. During this time, production of Atlantic salmon in open netpens in the BA

Transmission dynamics of parasitic sea lice from farm to wild salmon.

Marine salmon farming has been correlated with parasitic sea lice infestations and concurrent declines of wild salmonids. Here, we report a quantitative analysis of how a single salmon farm altered the natural transmission dynamics of sea lice to juvenile Pacific salmon. We studied infections of sea lice (Lepeophtheirus salmonis and Caligus clemensi) on juvenile pink salmon (Oncorhynchus gorbuscha) and chum salmon (Oncorhynchus keta) as they passed an isolated salmon farm during their seaward migration down two long and narrow corridors. Our calculations suggest the infection pressure imposed

Pharmacological Activity and Clinical Use of PDRN.

PDRN is a proprietary and registered drug that possesses several activities: tissue repairing, anti-ischemic, and anti-inflammatory. These therapeutic properties suggest its use in regenerative medicine and in diabetic foot ulcers. PDRN holds a mixture of deoxyribonucleotides with molecular weights ranging between 50 and 1,500 KDa, it is derived from a controlled purification and sterilization process of Oncorhynchus mykiss (Salmon Trout) or Oncorhynchus keta (Chum Salmon) sperm DNA. The procedure guarantees the absence of active protein and peptides that may cause immune reactions. In vitro a

[Peter-the-Great Gulf fishes are a source of human invasion of the nematode Anisakis simplex (the family Anisakidae)].

The paper presents the results of the rate of infection with the nematode Anisakis simplex in Peter-the-Great gulf fishes that are a source of human invasion. Five of 19 fish types were found to be nematode-free, and the extensiveness and intensity of invasion were observed in Siberian salmons (Oncorhynchus keta) (61% and 2-700 worms, respectively), Alaska pollacks (Theragra chalcogramma) (75.7% and 1-31 worms), and brown rock trouts (Hexagrammos octogrammus) (100% and 1-60 worms). Autopsy showed that the distribution of larvae was ambiguous in different fish species. Data on the survival of A

Space-time cluster analysis of sea lice infestation (Caligus clemensi and Lepeophtheirus salmonis) on wild juvenile Pacific salmon in the Broughton Archipelago of Canada.

Sea lice infestation levels on wild chum and pink salmon in the Broughton Archipelago region are known to vary spatially and temporally; however, the locations of areas associated with a high infestation level had not been investigated yet. In the present study, the multivariate spatial scan statistic based on a Poisson model was used to assess spatial clustering of elevated sea lice (Caligus clemensi and Lepeophtheirus salmonis) infestation levels on wild chum and pink salmon sampled between March and July of 2004 to 2012 in the Broughton Archipelago and Knight Inlet regions of British Columb

Short communication: Divergent life histories link thermal tolerance variation across Pacific salmon species and populations.

Divergent life histories among organisms are linked to variations in thermal response. Although there have been extensive studies on interpopulation variations in thermal tolerance, the ecological relevance of the tolerance to life history remains largely unexplored. We compared critical thermal maximum (CTmax) among two Pacific salmon species with different freshwater life histories, masu salmon (Oncorhynchus masou) and chum salmon (O. keta), across two local Japanese populations. CTmax was higher for masu salmon fry (26.9 °C) than for chum salmon fry (26.1 °C). Although the mean CTmax differ

Marine-Derived N-Terminal Mitochondrial-Targeting Sequences Exhibit Antimicrobial and Anticancer Activities.

The potential of N-terminal mitochondrial-targeting sequences (MTSs) as potent antimicrobial peptides (AMPs) has been previously reported. Building on this, 3923 mitochondrial proteins were identified from various marine organisms, among which 470 MTSs were predicted using MitoFates. Of these, 25 MTSs were synthesized and assessed for antimicrobial activity. All MTSs exhibited antifungal activity against Candida albicans, while 22 and 20 MTSs demonstrated activity against Escherichia coli and Staphylococcus aureus, respectively. Notably, the MTS of methylcrotonyl-CoA carboxylase subunit 1 (MCC

The diversity of microsporidia in parasitic copepods (Caligidae: Siphonostomatoida) in the Northeast Pacific Ocean with description of Facilispora margolisi n. g., n. sp. and a new Family Facilisporidae n. fam.

Three distinct microsporidia were identified from parasitic copepods in the northeast Pacific Ocean. Sequencing and phylogenetic analysis of a partial small subunit ribosomal RNA gene (SSU rDNA) sequence identified a genetically distinct variety of Desmozoon lepeophtherii from Lepeophtheirus salmonis on cultured Atlantic salmon Salmo salar, and this was confirmed by transmission electron microscopy. Phylogenetic analysis resolved the SSU rDNA sequence of the second organism in a unique lineage that was most similar to microsporidia from marine and brackish water crustaceans. The second occurre

Salmon nasal cartilage proteoglycan up-regulates Listeria monocytogenes-mediated immune response in mice.

Proteoglycan (PG) is a complex glycohydrate that is widely distributed in the extracellular matrix. We have reported that daily oral administration of PG extracted from salmon (Oncorhynchus keta) nasal cartilage modulates the severity and proinflammatory cytokine responses in Escherichia coli-stimulated macrophages, and attenuates mouse models of various inflammatory diseases. Thus, PG has shown anti-inflammatory activities. In this study, we investigated the effect of salmon nasal cartilage PG on a bacterial infection by using a mouse model of infection with Listeria monocytogenes, which is a

Ontogenetic changes in neuropeptide Y-like-immunoreactivity in the terminal nerve of the chum salmon and the cloudy dogfish, with special reference to colocalization with gonadotropin-releasing hormone-immunoreactivity.

We observed ontogenetic changes of neuropeptide Y (NPY)-like-immunoreactivity in the terminal nerve (TN) of the chum salmon, a teleost, and the cloudy dogfish, an elasmobranch. In the chum salmon, NPY-like-immunoreactive (NPY-IR) cells first appeared in the olfactory placode of embryos at 16 days after fertilization, and then extended sequentially and centrally in the olfactory system. Colocalization of NPY- and gonadotropin-releasing hormone (GnRH)-like-immunoreactivities was also observed in TN ganglion cells. In the cloudy dogfish, NPY-IR cells first appeared in the rudimental TN ganglion o

Wild juvenile salmonids in Muchalat Inlet, British Columbia, Canada: factors associated with sea lice prevalence.

The Muchalat Inlet, British Columbia, is among the most westerly points at which aquaculture is practiced in Canada. In this paper, we summarise data from over 18000 wild fish sampled at 16 sites over an 8 yr period, between 2004 and 2011. The most prevalent wild species was chum salmon Oncorhynchus keta (82.4%), followed by Chinook O. tshawytscha (10%) and coho O. kisutch (4.3%). However, inter-annual and seasonal variation was evident, and smaller numbers of other Pacific salmon and stickleback species were sporadically observed. A high percentage of wild salmon (~95%) had no sea lice parasi

Messenger RNA transcription levels of neuronal soluble N-ethylmaleimide-sensitive factor attachment protein receptor complex components in the olfactory nerve system of the anadromous Pacific salmon, masu salmon Oncorhynchus masou.

Anadromous Pacific salmon (genus Oncorhynchus) are known for homing behavior to their natal rivers based on olfactory imprinted memories during seaward migration. The SNARE complex is a regulator of vesicle exocytosis from the presynaptic membrane. Our previous study suggested that its component genes (Snap25, Stx1, and Vamp2) are more highly expressed in the olfactory nervous system (ONS) during the migration stages associated with olfactory imprinting in the evolutionary species of Pacific salmon, such as chum (O. keta) and pink (O. gorbuscha) salmon. Masu salmon (O. masou) has a significant

High sensitivity to ocean acidification in wild out-migrating juvenile Pacific salmon is not impacted by feeding success.

Salmon populations are declining worldwide, with high mortality rates during juvenile marine migration presenting a bottleneck to recruitment. The ocean conditions along the main migratory route of juvenile salmon in British Columbia are characterized by high variability in CO2, with the amplitude, duration, and frequency of ocean acidification events exacerbated by climate change. Similarly, the variability in ocean conditions affects the abundance and diversity of plankton prey, leading to areas of food paucity for juvenile salmon. We investigated the combined effects of ocean acidification

Anisakis spp. in fishery products from Japanese waters: Updated insights on host prevalence and human infection risk factors.

The nematodes of the genus Anisakis are among the most relevant parasitic hazards in fishery products since they are responsible for human infection and allergy cases. In a food safety and epidemiological perspective, several marine hosts from different locations around Japan were examined to characterize the parasitism of Anisakis larvae. Chum salmon (Oncorhynchus keta) and Alaska pollock (Gadus chalcogrammus) showed the highest overall prevalence (100%), followed by blue mackerel (Scomber australasicus) (97.5%), Pacific cod (Gadus macrocephalus) (80%), chub mackerel (Scomber japonicus) (60.1

Carbohydrate recognition by the rhamnose-binding lectin SUL-I with a novel three-domain structure isolated from the venom of globiferous pedicellariae of the flower sea urchin Toxopneustes pileolus.

The globiferous pedicellariae of the venomous sea urchin Toxopneustes pileolus contains several biologically active proteins. We have cloned the cDNA of one of the toxin components, SUL-I, which is a rhamnose-binding lectin (RBL) that acts as a mitogen through binding to carbohydrate chains on target cells. Recombinant SUL-I (rSUL-I) was produced in Escherichia coli cells, and its carbohydrate-binding specificity was examined with the glycoconjugate microarray analysis, which suggested that potential target carbohydrate structures are galactose-terminated N-glycans. rSUL-I exhibited mitogenic

Intracerebral expression of gonadotropin-releasing hormone and growth hormone-releasing hormone is delayed until smoltification in the salmon.

A developmental strategy was employed to investigate the functional assembly of neuropeptidergic systems in the migratory species of chum salmon Oncorhynchus keta. Using immunocytochemistry we have demonstrated that different groups of gonadotropin-releasing hormone- (GnRH)- and growth hormone-releasing hormone- (GHRH)-synthesizing neurons emerged according to very different developmental timetables. From the eye pigmentation stage (23 +/- 2 days after fertilisation (DAF)) through to the pre-smoltification stage (136 DAF), salmon-GnRH neurons originating from the olfactory placodes remained re
