Morphological and phylogenetic analyses of Gonatobotryum (Dothideaceae, Dothideales), and the description of a new species from a mangrove sediment in Brazil.

Gonatobotryum is an anamorphic fungus reported mainly as a saprophyte, parasite, or endophyte. The genus has traditionally been identified based on morphological features, with its phylogenetic placement remaining uncertain due to the lack of molecular data. During the analysis of fungi obtained from mangrove sediments in Brazil, nine isolates were morphologically identified as Gonatobotryum and subsequently analysed using the internal transcribed spacer (ITS) region, including the intervening 5.8S nRNA gene and the nuclear ribosomal RNA large subunit (LSU) gene, leading to the description of

Antifungal activity of a Pinus monticola antimicrobial peptide 1 (Pm-AMP1) and its accumulation in western white pine infected with Cronartium ribicola.

Pinus monticola antimicrobial peptide 1 (Pm-AMP1) was expressed and purified from bacterial cell lysate and its identity and purity confirmed by Western blot analysis using the Pm-AMP1 antibody. Application of Pm-AMP1 resulted in visible hyphal growth inhibition of Cronartium ribicola , Phellinus sulphurascens , Ophiostoma montium , and Ophiostoma clavigerum 3-12 days post-treatment. Pm-AMP1 also inhibited spore germination of several other phytopathogenic fungi by 32%-84% 5 days post-treatment. Microscopic examination of C. ribicola hyphae in contact with Pm-AMP1 showed distinct morphological

The MAT1-1:MAT1-2 ratio of Sporothrix globosa isolates in Japan.

In order to understand the reproductive biology of pathogenic species in the Sporothrix schenckii complex, we characterized the partial mating type (MAT1-1) loci of Sporothrix schenckii, as well as the S. globosa MAT1-1-1 gene, which encoded 262 amino acid sequences. The data confirmed that the MAT1-1 locus of S. globosa was divergent from the MAT1-2 locus of the opposite mating type, suggesting that the fungus is heterothallic. To determine the mating type ratio of 20 isolates from Japanese patients, we analyzed the MAT loci by specific PCR amplification of MAT1-1-1 and MAT1-2-1 genes. The MA

Isolation and characterization of Sporothrix schenckii from clinical and environmental sources associated with the largest U.S. epidemic of sporotrichosis.

The largest recorded epidemic of sporotrichosis in the United States occurred in 1988 and involved a total of 84 cases in 15 states. All cases were associated with Wisconsin-grown sphagnum moss. Twenty-one clinical isolates of Sporothrix schenckii and 69 environmental isolates of Sporothrix spp. from the epidemic were characterized and compared. The environmental isolates were recovered from 102 samples of sphagnum moss and other material by using direct plating techniques. Characteristics examined included macroscopic and microscopic morphology, conversion to a yeast phase, exoantigen reactio

Ecology of root-feeding beetles and their associated fungi on longleaf pine in Georgia.

Root-feeding beetles, particularly of the curculionid subfamilies Scolytinae and Molytinae, are known to be effective vectors of Ophiostomatoid fungi. Infestation by these insects and subsequent infection by the Ophiostomatoid fungi may play an important role in accelerating symptom progression in pine declines. To examine the relationship between beetles and fungi in longleaf pine stands, root-feeding curculionids were collected in pitfall traps baited with ethanol and turpentine for 62 wk, and Ophiostomatoid fungi were isolated from their body surfaces. The most abundant root-feeding beetles

Ophiostomatalean fungi associated with bark beetles infesting Pinus armandii Franch., with characterizations of seven new species from Shaanxi, China.

Pinus armandii is widely distributed in western China and is of considerable ecological significance. However, recent bark beetle outbreaks have caused severe damage to forests in Shaanxi Province, and these outbreaks are closely associated with ophiostomatalean fungi (Ascomycota, Ophiostomatales). These fungi not only incite plant diseases but also facilitate the successful establishment of the beetles. At present, knowledge of the ophiostomatalean fungal communities associated with bark beetles infesting Pinus armandii in Shaanxi Province remains limited. This study aimed to identify ophiost

Symbiont-Induced HSP83 Establishes Two-Tiered Control of Antifungal Immunity in an Invasive Beetle-Fungus Complex.

Hosts must distinguish mutualistic symbionts from antagonists while avoiding harmful immune overactivation, yet the mechanisms maintaining this balance remain unclear. This challenge is acute during biological invasions, where exposure to novel microbial communities can outpace host genetic adaptation. Dendroctonus valens (RTB) associates with its mutualistic fungus Leptographium procerum (Lp), forming an invasive beetle-fungus complex during attacks on Chinese pines. Lp fails to trigger antimicrobial peptide expression but induces the heat shock protein 83 (HSP83). The native antagonist Ophio

Effects of available water on growth and competition of southern pine beetle associated fungi.

Competitive, interactions among bark beetle associated fungi are potentially influenced by abiotic factors. Water potential, in particular, undergoes marked changes over the course of beetle colonization of tree hosts. To investigate the impact of water potential on competition among three southern pine beetle associated fungi, Ophiostoma minus, Entomocorticium sp. A and Ceratocystiopsis ranaculosus, we utilized artificial media with water potentials of 0, -5, -10, and -20 MPa. Growth of all three fungi, when grown alone, decreased on media with lower water potentials. Growth rates of all thre

Interactions between the yeast Ogataea pini and filamentous fungi associated with the western pine beetle.

Ecologically important microbes other than filamentous fungi can be housed within the fungal-transport structures (mycangia) of Dendroctonus bark beetles. The yeast Ogataea pini (Saccharomycetales: Saccharomycetaceae) was isolated from the mycangia of western pine beetle (Dendroctonus brevicomis) populations in northern Arizona (USA) with a frequency of 56%. We performed a series of in vitro assays to test whether volatile organic compounds produced by O. pini affected radial growth rates of mutualistic and antagonistic species of filamentous fungi that are commonly found in association with t

Use of a mouse model to evaluate clinical and environmental isolates of Sporothrix spp. from the largest U.S. epidemic of sporotrichosis.

Five clinical and 69 environmental isolates from the largest U.S. epidemic of sporotrichosis were evaluated in NYLAR male mice following intravenous injection of 5 x 10(6) to 2 x 10(8) conidia per mouse. The clinical isolates and eight environmental isolates produced 100% mortality in groups of three mice each between 12 and 24 days after injection. These virulent isolates grew at 37 degrees C, were dematiaceous by virtue of melanin (melanized) on permissive media (e.g., potato dextrose agar), produced ovoid conidia borne sympodially on lateral conidiophores and pleurogenously about the main h

Simultaneous induction of jasmonic acid and disease-responsive genes signifies tolerance of American elm to Dutch elm disease.

Dutch elm disease (DED), caused by three fungal species in the genus Ophiostoma, is the most devastating disease of both native European and North American elm trees. Although many tolerant cultivars have been identified and released, the tolerance mechanisms are not well understood and true resistance has not yet been achieved. Here we show that the expression of disease-responsive genes in reactions leading to tolerance or susceptibility is significantly differentiated within the first 144 hours post-inoculation (hpi). Analysis of the levels of endogenous plant defense molecules such as jasm

Chemical Signals of Vector Beetle Facilitate the Prevalence of a Native Fungus and the Invasive Pinewood Nematode.

In China, the invasive Bursaphelenchus xylophilus, the vector Monochamus alternatus beetle, and associated fungi exhibit a symbiotic relationship causing serious losses to pine forests. Although this complex system has been intensively investigated, the role of vector beetles on the development of associated fungi and their indirect contribution to the prevalence of pinewood nematode (PWN) is yet unknown. Here, three of the highly prevalent fungal species, viz., Sporothrix sp. 1, Ophiostoma ips, and Sporothrix sp. 2 were isolated from beetle chambers in diseased trees in Guangdong province, so

Macrorestriction analysis of clinical and environmental isolates of Sporothrix schenckii.

Sporothrix schenckii causes sporotrichosis, a disease that most commonly presents as a subacute or chronic skin infection. An unusually high incidence of clinical cases of sporotrichosis occurred in the southwest of Western Australia over the last 5 years. Anecdotal accounts from patients implicated contact with hay prior to infection. Isolates of S. schenckii from hay and clinical cases were investigated by traditional phenotypic methods and pulsed-field gel electrophoresis (PFGE). The phenotypic evaluation separated S. schenckii from Ophiostoma spp. A DNA macrorestriction method using SfiI a

Leptographium tereforme sp. nov. and other Ophiostomatales isolated from the root-feeding bark beetle Hylurgus ligniperda in California.

The redhaired pine bark beetle Hylurgus ligniperda (F.) is native to Europe but was discovered in Los Angeles, California, in 2003. This root-and stump-feeding beetle is a common vector of Ophiostomatales, which are potential tree pathogens or causes of blue stain of conifer sapwood. In this study Ophiostomatales were isolated on a cycloheximide-amended medium from 118 adult H. ligniperda collected from infested logs of Pinus halepensis and P. pinea at two sites in California. In total eight species of Ophiostomatales were identified and seven species that occasionally were isolated were unide

The mutualistic fungi of the bark beetle Pityokteines vorontzowi are nutrient-rich and efficiently deplete their medium of fir chemical defenses.

Conifers are a challenging host for herbivores since their tissues are very low in essential nutrients but high in chemical defenses. For herbivorous insects, such as phloem-colonizing bark beetles, mutualistic fungi may improve their diet by providing a nutritious mycelium. A recent study revealed that two filamentous fungi are mutualists of the European fir engraver beetle Pityokteines vorontzowi, but a potential nutritional contribution of the fungi, as well as their capability to degrade plant antiherbivore defenses remains unknown. We analyzed the nutrient content of the fungal mutualists

Quorum-driven microbial consortium for Bioplastic production from agro-waste.

Microbial consortia have high relevance in natural environments. Here we present the production of polyhydroxyalkanoates (PHA) from agro-industrial residues by a synthetic interkingdom consortium formed by the saprotrophic fungus Ophiostoma piceae CECT 20146, which encodes a wide range of lignocellulolytic enzymes, and a natural PHA producer, Pseudomonas putida KT2440. Two agro-industrial residues were utilized: Brewer's Spent Grain (BSG) as a carbon/nitrogen source and biofilm scaffold and waste cooking oil (WCO) as a carbon source for PHA synthesis. Through biochemistry, microscopy, and omic

Phylogeny and ecological roles shaping yeast-hyphae morphogenesis in Ophiostomataceae.

The Ophiostomataceae family encompasses numerous species that exhibit yeast-hyphae dimorphism, including well-known pathogens like the causal agents of Dutch elm disease, blue-staining fungi, and sporotrichosis. This dimorphism is not only observed in pathogenic interactions but also plays a crucial role in symbiotic relationships, such as ambrosia fungi. The ability to switch between these morphological forms may contribute to their adaptability and ecological functions within different environments and host interactions. This study seeks to elucidate the intricate interplay between dimorphis

Semiochemicals from fungal associates of bark beetles may mediate host location behavior of parasitoids.

In laboratory olfactometer bioassays, females of two hymenopteran parasitoid species, Roptrocerus xylophagorum (Ratzeburg) (Hymenoptera: Pteromalidae) and Spathius pallidus (Ashmead) (Hymenoptera: Braconidae), were attracted to odors from bark or bolts of loblolly pine, Pinus taeda L., colonized by bluestain fungi (genus Ophiostoma) associated with the parasitoids' bark beetle hosts. Mock-inoculated bolts and bark were less attractive or unattractive in these bioassays. Bark infested with host larvae that lacked their fungal and other normal microbial associates was attractive to R. xylophagor

Gene expression differences among three Neurospora species reveal genes required for sexual reproduction in Neurospora crassa.

Many fungi form complex three-dimensional fruiting bodies, within which the meiotic machinery for sexual spore production has been considered to be largely conserved over evolutionary time. Indeed, much of what we know about meiosis in plant and animal taxa has been deeply informed by studies of meiosis in Saccharomyces and Neurospora. Nevertheless, the genetic basis of fruiting body development and its regulation in relation to meiosis in fungi is barely known, even within the best studied multicellular fungal model Neurospora crassa. We characterized morphological development and genome-wide

Molecular Diagnosis of Pathogenic Sporothrix Species.

BACKGROUND: Sporotrichosis is a chronic (sub)cutaneous infection caused by thermodimorphic fungi in the order, Ophiostomatales. These fungi are characterized by major differences in routes of transmission, host predilections, species virulence, and susceptibilities to antifungals. Sporothrix species emerge in the form of outbreaks. Large zoonoses and sapronoses are ongoing in Brazil and China, respectively. Current diagnostic methods based on morphology and physiology are inaccurate due to closely related phenotypes with overlapping components between pathogenic and non-pathogenic Sporothrix.
