Sudden-onset Blindness from a Spontaneous Carotid-cavernous Fistula with Secondary Central Retinal Artery Occlusion and Posterior Ischemic Optic Neuropathy.

Our case describes a patient diagnosed with a carotid-cavernous fistula (CCF) secondary to a spontaneously ruptured cavernous carotid aneurysm, presenting with sudden vision loss, and a concomitant central retinal artery occlusion as visualized by a cherry-red spot in the macula and posterior ischemic optic neuropathy. Computed tomography of the brain and orbits showed mild hydrocephalus, orbital fat haziness, and proptosis with concern for fluid in the basal cisterns. Cerebral angiography confirmed the suspected diagnosis of CCF. After angiography, a Magnetic resonance imaging of the brain de

New near-infrared optical probes of cardiac electrical activity.

Styryl voltage-sensitive dyes (e.g., di-4-ANEPPS) have been widely and successfully used as probes for mapping membrane potential changes in cardiac cells and tissues. However, their utility has been somewhat limited because their excitation wavelengths have been restricted to the 450- to 550-nm range. Longer excitation/emission wavelength probes can minimize interference from endogenous chromophores and, because of decreased light scattering and lower absorption by endogenous chromophores, improve recording from deeper tissue layers. In this article, we report efforts to develop new potentiom

Retroviral misexpression of cVax disturbs retinal ganglion cell axon fasciculation and intraretinal pathfinding in vivo and guidance of nasal ganglion cell axons in vivo.

The transcription factor cVax (Vax2) is expressed in the ventral neural retina and restricted expression is a prerequisite for at least three prominent aspects of retinal dorsal-ventral patterning: polarized expression of EphB/B-ephrin molecules, the retinotectal projection and the distribution of rod photoreceptors across the retina. In the chick retina, the fasciculation pattern of ganglion cell axons also differs between the dorsal and ventral eye. To investigate the molecular mechanisms involved, the nerve fiber layer was analyzed after retroviral misexpression of several factors known to

Incremental training increases the plasticity of the auditory space map in adult barn owls.

The plasticity in the central nervous system that underlies learning is generally more restricted in adults than in young animals. In one well-studied example, the auditory localization pathway has been shown to be far more limited in its capacity to adjust to abnormal experience in adult than in juvenile barn owls. Plasticity in this pathway has been induced by exposing owls to prismatic spectacles that cause a large, horizontal shift of the visual field. With prisms, juveniles learn new associations between auditory cues, such as interaural time difference (ITD), and locations in visual spac

Deciphering Genomic Complexity of Multiple Myeloma Using Optimized Optical Genome Mapping.

The genomic evaluation of multiple myeloma in routine diagnostics involves isolating plasma cells expressing CD138, usually followed by fluorescence in situ hybridization analyses. However, cell sorting often yields a limited number of cells, restricting the number of probes that can be used and limiting the analysis to a few markers required for minimal prognostic classification. Optical genome mapping is a high-resolution technology capable of identifying structural variants and copy number variations across the entire genome; however, it currently requires 1 million cells. To overcome this

Self-Training Strategy Based on Finite Element Method for Adaptive Bioluminescence Tomography Reconstruction.

Bioluminescence tomography (BLT) is a promising pre-clinical imaging technique for a wide variety of biomedical applications, which can non-invasively reveal functional activities inside living animal bodies through the detection of visible or near-infrared light produced by bioluminescent reactions. Recently, reconstruction approaches based on deep learning have shown great potential in optical tomography modalities. However, these reports only generate data with stationary patterns of constant target number, shape, and size. The neural networks trained by these data sets are difficult to rec

A novel Asp380Ala mutation in the GLC1A/myocilin gene in a family with juvenile onset primary open angle glaucoma.

Glaucoma describes a clinically and genetically heterogeneous group of diseases that result in optic neuropathy and progressive loss of visual fields. A gene for juvenile onset primary open angle glaucoma JOAG) has recently been mapped to 1q21-31. Mutations in the trabecular meshwork induced glucocorticoid response gene (TIGR, also known as myocilin or the GLC1A locus) have been found to cause both juvenile and later onset primary open angle glaucoma. Family TCD-POAG1 is a Spanish kindred, which segregates JOAG in an autosomal dominant fashion. This family was found to be linked to the previou

Nutrient restriction preserves calcium cycling and mitochondrial function in cardiac myocytes during ischemia and reperfusion.

Nutrient restriction (NR) prolongs longevity via enhanced mitochondrial function. We tested the hypothesis that NR enhances resistance to ischemia/reperfusion (IR) arrhythmias via preserved calcium (Ca) cycling and mitochondrial function. We examined the protective effects of NR on regional IR in cultured neonatal rat ventricular myocyte monolayers. Optical mapping of intracellular Ca and mitochondrial membrane potential Δψ(m) was performed using Rhod 2-AM and TMRE, respectively. Regional ischemia was mimicked by covering a portion of monolayer with a glass coverslip until loss of Ca propagati

Visual defects in a mouse model of fetal alcohol spectrum disorder.

Alcohol consumption during pregnancy can lead to a multitude of neurological problems in offspring, varying from subtle behavioral changes to severe mental retardation. These alterations are collectively referred to as Fetal Alcohol Spectrum Disorders (FASD). Early alcohol exposure can strongly affect the visual system and children with FASD can exhibit an amblyopia-like pattern of visual acuity deficits even in the absence of optical and oculomotor disruption. Here, we test whether early alcohol exposure can lead to a disruption in visual acuity, using a model of FASD to mimic alcohol consump

Rapid sonication-assisted whole tissue clearing and immunostaining.

High-resolution mapping of three-dimensional structures in biological tissues is essential for understanding various biological processes. However, the optical heterogeneity of these tissues, marked by varying optical properties, causes light scattering and absorption, complicating imaging. Current tissue clearing methods can take between 48 h and 32 days, and the limited diffusion depth of fluorescent probes restricts whole-tissue imaging. This study introduces an innovative Sonication-Assisted Tissue Clearing and Immunofluorescent Staining method (SoniC/S), which combines low-frequency ultra

Pseudo-depth-based deep neural network model for object detection.

Current machine learning methods only utilize the three-channel color features of optical images for computer visual tasks. However, the optical images only explicitly present information of RGB color and two-dimensional planar shape, where the third-dimensional spatial features are not fully exploited. This limitation restricts the potential improvement in recognition performance. To address this issue, we propose a detection scheme to enhance model's detection capabilities based on four independent features by combining the pseudo-depth and the RGB features without adding any additional hard

Localized Refractive Changes Induced by Symmetric and Progressive Asymmetric Intracorneal Ring Segments Assessed with a 3D Finite-Element Model.

To build a representative 3D finite element model (FEM) for intracorneal ring segment (ICRS) implantation and to investigate localized optical changes induced by different ICRS geometries, a hyperelastic shell FEM was developed to compare the effect of symmetric and progressive asymmetric ICRS designs in a generic healthy and asymmetric keratoconic (KC) cornea. The resulting deformed geometry was assessed in terms of average curvature via a biconic fit, sagittal curvature (K), and optical aberrations via Zernike polynomials. The sagittal curvature map showed a locally restricted flattening int

OptiGAN for crystal arrays: physics-informed generative modeling of optical photon transport in PET detector arrays.

Objective.Monte Carlo simulations of optical photon transport are computationally prohibitive for large-scale optical systems including detector arrays and positron emission tomography systems, restricting their practical use to single-crystal studies. This work presents an enhanced conditional generative adversarial network capable of replacing optical simulations at the crystal array level, extending our previous single-crystal approach to a 3  3 bismuth germanate detector array.Approach.We introduce Fourier feature encoding and a learnable latent mapping network as the modifications enablin

Training on a visual task improves the outcome of optic nerve regeneration.

Optic nerve regeneration in a lizard, Ctenophorus ornatus, is dysfunctional despite survival of most retinal ganglion cells and axon regeneration to the optic tectum. The regenerated retino-tectal projection at 6 months has crude topography but by 1 year is disordered; visually-elicited behavior is absent via the experimental eye. Here, we assess the influence of training on the outcome of optic nerve regeneration. Lizards were trained to catch prey presented within the monocular field of either eye. One optic nerve was then severed and visual stimulation resumed throughout regeneration. In th

Secondary Imaging Architecture for Fast and Ultra-Wide LWIR Optics with Low Rectilinear Distortion.

Wide-swath longwave infrared (LWIR) imaging from Low Earth Orbit (LEO) demands fast optics and rectilinear (F-tan) mapping for thermal mapping and multi-frame registration. Achieving an F/1.2 aperture with a 112° diagonal field of view (FOV) and distortion within ±5% is challenging, as mapping constraints and field-dominant off-axis aberrations become strongly coupled at large chief-ray angles. The low-distortion target is not only a geometric specification, but also a practical requirement that reduces peripheral compression, helps maintain edge-detail consistency, and lowers digital de-warpi

Signaling - transcription interactions in mouse retinal ganglion cells early axon pathfinding -a literature review.

Sending an axon out of the eye and into the target brain nuclei is the defining feature of retinal ganglion cells (RGCs). The literature on RGC axon pathfinding is vast, but it focuses mostly on decision making events such as midline crossing at the optic chiasm or retinotopic mapping at the target nuclei. In comparison, the exit of RGC axons out of the eye is much less explored. The first checkpoint on the RGC axons' path is the optic cup - optic stalk junction (OC-OS). OC-OS development and the exit of the RGC pioneer axons out of the eye are coordinated spatially and temporally. By the time

Multiplexed Nanoscopy via Buffer Exchange.

Understanding cellular functions, particularly in their intricate complexity, can greatly benefit from the spatial mapping of diverse molecules through multitarget single-molecule localization microscopy (SMLM). Existing methodologies, primarily restricting the encoding dimensions to color and lifetime or requiring cyclic staining, often involve broad chromatic detection, specialized optical configurations, or sophisticated labeling techniques. Here, we propose a simple approach called buffer-exchange stochastic optical reconstruction microscopy (beSTORM), which introduces an additional dimens

Layer-Specific Developmentally Precise Axon Targeting of Transient Suppressed-by-Contrast Retinal Ganglion Cells.

The mouse retina encodes diverse visual features in the spike trains of >40 retinal ganglion cell (RGC) types. Each RGC type innervates a specific subset of the >50 retinorecipient brain areas. Our catalog of RGC types and feature representations is nearing completion. Yet, we know little about where specific RGC types send their information. Furthermore, the developmental strategies by which RGC axons choose their targets and pattern their terminal arbors remain obscure. Here, we identify a genetic intersection (Cck-Cre and Brn3cCKOAP ) that selectively labels transient Suppressed-by-Contrast
