In-situ aberration correction for Laguerre-Gaussian optical tweezers via optimization of orbit shape.

We introduce a concept of aberration correction under microscopy that is based on observation of circular Brownian motion of an object driven by orbital angular momentum of a Laguerre-Gaussian (LG) beam. Following the concept, we establish an aberration-correction scheme by using a holographic optical tweezers setup equipped with a spatial light modulator that produces the LG beam as well as corrects the light wavefront. The light wavefront is modified adaptively to improve circular symmetry and uniformity of the orbit of a colloidal dielectric sphere revolving in mid-water under the irradiati

Characterizing light shift of an open optical Zeeman transition with a single cesium atom in an optical tweezer.

Characterizing the trap-light-induced frequency shifts of optical Zeeman transitions is essential for the precise control of the interacting dynamics between optically trapped single atoms and external light fields. The closed hyperfine transition, for example, 6S1/2 | F = 4〉 → 6P3/2 | F' = 5〉 of the cesium (Cs) atom, is commonly used because of its high transition strength. However, within this hyperfine transition line, different Zeeman transitions experience distinct light shifts due to diverse susceptibilities to the light field, which are usually sorted as dynamic scalar, vector, and tens

Multifunctional chip-scale plasmonic optical tweezers enabled by a polarization- and wavelength-dependent metasurface.

The ability of surface plasmons to confine light at a subwavelength scale enables new perspectives and opportunities in the field of nanotechnology. Making use of this unique advantage, plasmonic trapping techniques have been developed to tackle new challenges in a wide range of areas from physics to biology. However, it is still a challenge to achieve multifunctional chip-scale manipulation of small particles in a single plasmonic device. Here, we proposed a polarization- and wavelength-dependent plasmonic metasurface to realize multifunctional plasmonic optical tweezers. By fully utilizing t

Robot aided optical tweezers and microneedle assisted minimally invasive single cell mitochondria biopsy system.

Single cell surgery, which involves removing or manipulating subcellular organelles from single cells, is increasingly being utilized in precision medicine to investigate illnesses and their causes. This article describes an optical tweezers (OTs)-assisted mitochondria biopsy method for performing minimally invasive automated organelle biopsy of single cells. A microfluidic chip device is utilized to hold a single cell, and OTs is used to trap and move the mitochondria to the edge of the cell membrane automatically, followed by automatic mitochondria biopsy with a bevelled microneedle. An imag

Dipolar spin-exchange and entanglement between molecules in an optical tweezer array.

Ultracold polar molecules are promising candidate qubits for quantum computing and quantum simulations. Their long-lived molecular rotational states form robust qubits, and the long-range dipolar interaction between molecules provides quantum entanglement. In this work, we demonstrate dipolar spin-exchange interactions between single calcium monofluoride (CaF) molecules trapped in an optical tweezer array. We realized the spin-[Formula: see text] quantum XY model by encoding an effective spin-[Formula: see text] system into the rotational states of the molecules and used it to generate a Bell

Comparative mechanical profiling of MCF-7 cells in 2D and 3D cultures via optical tweezers.

Breast cancer cells experience distinct mechanical forces within the 3D tumor microenvironment, and these forces strongly influence cell behavior, invasion potential, and treatment response. While conventional two-dimensional culture systems offer experimental simplicity, they fail to reproduce the complex mechanical interactions present in vivo, which play an important role in the development of therapeutic resistance. In this work, we investigate the mechanical properties of breast cancer cells grown under both 2D and 3D conditions using dual-beam optical tweezers. Our measurements show that

Activation dynamics study of trapped platelets using a lab-built optical tweezers micro-raman spectrometer.

It is well documented that platelet disorders can result from various causes and can lead to different disease conditions such as cardiovascular diseases (CVDs), thrombocythemia, thrombocytopenia, Autoimmune diseases, Alzheimer's disease (AD), and even cancer, to name a few. The diagnosis of many of these diseases mainly depends on imaging examinations, clinical analysis and neuropsychological tests, these may be time taking procedures and, have a high chances of false positive/false negative results. The Raman tweezers spectroscopy can provide trust worthy results without much time delay. In

Optimal calibration of optical tweezers with arbitrary integration time and sampling frequencies: a general framework [Invited].

Optical tweezers (OT) have become an essential technique in several fields of physics, chemistry, and biology as precise micromanipulation tools and microscopic force transducers. Quantitative measurements require the accurate calibration of the trap stiffness of the optical trap and the diffusion constant of the optically trapped particle. This is typically done by statistical estimators constructed from the position signal of the particle, which is recorded by a digital camera or a quadrant photodiode. The finite integration time and sampling frequency of the detector need to be properly tak

Quantification of the interaction forces between dengue virus and dopamine type-2 receptor using optical tweezers.

BACKGROUND: Dengue virus (DENV) causes the most significant mosquito-borne viral disease with a wide spectrum of clinical manifestation, including neurological symptoms associated with lethal dengue diseases. Dopamine receptors are expressed in central nervous system, and dopamine antagonists have been reported to exhibit antiviral activity against DENV infection in vivo and in vitro. Although identification of host-cell receptor is critical to understand dengue neuropathogenesis and neurotropism, the involvement of dopamine receptors in DENV infection remains unclear. RESULTS: We exploited t

Fluorescent II-VI semiconductor quantum dots in living cells: nonlinear microspectroscopy in an optical tweezers system.

In this work we used a setup consisting of an optical tweezers combined with a nonlinear microspectroscopy system to perform scanning microscopy and obtain emission spectra using two photon excited (TPE) luminescence of captured single living cells labeled with core-shell fluorescent semiconductor quantum dots (QDs). The QDs were obtained via colloidal synthesis in aqueous medium with an adequate physiological resulting pH. Sodium polyphosphate was used as the stabilizing agent. The results obtained show the potential presented by this system as well as by these II-VI fluorescent semiconductor

Formation of Lymphoma Hybrid Spheroids and Drug Testing in Real Time with the Use of Fluorescence Optical Tweezers.

Interactions between stromal and lymphoma cells in the bone marrow are closely related to drug resistance and therapy failure. Physiologically relevant pre-clinical three-dimensional (3D) models recapitulating lymphoma microenvironmental complexity do not currently exist. In this study, we proposed a scheme for optically controlled hybrid lymphoma spheroid formation with the use of optical tweezers (OT). Following the preparation of stromal spheroids using agarose hydrogel, two aggressive non-Hodgkin lymphoma B-cell lines, Ri-1 (DLBCL) and Raji (Burkitt lymphoma), were used to conduct multi-ce

Measurement of the binding forces between von Willebrand factor and variants of platelet glycoprotein Ibalpha using optical tweezers.

BACKGROUND AND OBJECTIVE: Thrombus formation is initiated by adhesion of the platelet receptor, glycoprotein (GP) Ib-IX-V complex, to its adhesive ligand, von Willebrand factor (vWf), in the subendothelium or plasma. The vWf-binding domain of GP Ib-IX-V is in the GP Ibalpha subunit of the complex and contains a leucine-rich repeat region. The adhesion of different leucine-rich repeats was studied using optical tweezers in order to determine which ones were critical for the vWf/GP Ibalpha interaction. STUDY DESIGN/MATERIALS AND METHODS: Canine GP Ibalpha does not normally bind to human vWf, an

Sonidegib Inhibits the Adhesion of Acute Myeloid Leukemia to the Bone Marrow in Hypoxia: An Optical Tweezer Study.

Background: Acute myeloid leukemia (AML) is a heterogeneous disease highly resistant to chemotherapeutic agents. Leukemia stem cells (LSCs) can enter a dormant state and avoid apoptosis in the protective niche of the bone marrow (BM) microenvironment. Moreover, bone marrow stromal cells protect leukemia cells by promoting pro-survival signaling pathways and drug resistance. Therefore, attenuating interactions between leukemia cells and BM cells may have a positive therapeutic effect. Objectives: In this work, we hypothesized that sondages may inhibit the adhesion of leukemia cells to the bone

Planar lightsheet optical tweezer pLOT for 2D trapping and imaging of freely-moving live cells.

We report the realization of the first planar optical trap and imaging system using a sheet of light that allows interrogation of living specimens in a plane. An orthogonal widefield detection is employed to directly visualize the trapping of the target object (dielectric beads/cells) in a plane. The planar trap is realized on an inverted optical stage with illumination from the bottom. The system uses a combination of a cylindrical lens and a high NA objective lens to generate a tightly focused diffraction-limited sheet of light. For trapping objects in a selective XZ-plane, the sample (beads

Plasmonic Optical Tweezers and Surface-Enhanced Raman Spectroscopy: Fundamentals, Single-Entity Applications, and the Evolving Role of Artificial Intelligence.

The ability to manipulate and probe individual nano-particles, viruses, and organelles with high sensitivity and specificity is an essential part of modern nanoscience and molecular biology. Plasmonic optical tweezers (POT), which use localized surface plasmons to create nanoscale-confined optical fields, have emerged as a powerful platform for trapping and manipulating single nano-bio entities at low optical powers. When combined with surface-enhanced Raman spectroscopy (SERS) from the same plasmonic nanostructures, these platforms offer a unique multi-modal capability: simultaneous optical m

Two steps forward, one step back: determining XPD helicase mechanism by single-molecule fluorescence and high-resolution optical tweezers.

XPD-like helicases constitute a prominent DNA helicase family critical for many aspects of genome maintenance. These enzymes share a unique structural feature, an auxiliary domain stabilized by an iron-sulphur (FeS) cluster, and a 5'-3' polarity of DNA translocation and duplex unwinding. Biochemical analyses alongside two single-molecule approaches, total internal reflection fluorescence microscopy and high-resolution optical tweezers, have shown how the unique structural features of XPD helicase and its specific patterns of substrate interactions tune the helicase for its specific cellular fu

Measurements of the binding force between the Helicobacter pylori adhesin BabA and the Lewis b blood group antigen using optical tweezers.

Helicobacter pylori is a world-wide spread bacterium that causes persistent infections and chronic inflammations that can develop into gastritis and peptic ulcer disease. It expresses several adhesin proteins on its surface that bind to specific receptors in the gastric epithelium. The most well-known adhesin is BabA, which has previously been shown to bind specifically to the fucosylated blood group antigen Lewis b (Leb). The adhesion forces between BabA and the Leb antigen are investigated in this work and assessed by means of optical tweezers. A model system for in situ measurements of the

Differentiation of single lymphoma primary cells and normal B-cells based on their adhesion to mesenchymal stromal cells in optical tweezers.

We have adapted a non-invasive method based on optical tweezers technology to differentiate between the normal B-cells and the B-cell non-Hodgkin lymphoma (B-NHL) cells derived from clinical samples. Our approach bases on the nascent adhesion between an individual B-cell and a mesenchymal stromal cell. In this study, a single B-cell was trapped and optically seeded on a mesenchymal stromal cell and kept in a direct contact with it until a stable connection between the cells was formed in time scale. This approach allowed us to avoid the introduction of any exogenous beads or chemicals into the

Optical Tweezer-Driven Mechanotransduction: Probing pN-Scale Forces and Calcium-Mediated Redox Signaling in Single Endothelial Cells.

Endothelial cells (ECs) regulate vascular function by converting mechanical forces into biochemical signals; however, the molecular mechanisms of pN-scale mechanotransduction remain elusive. Here, we develop an optical tweezer-integrated confocal microscopy system that allows precise, noninvasive manipulation of the cell membrane localization with mechanical stimuli within the 0-100 pN range while monitoring Ca2+-mediated NO/ROS redox signaling in situ in single ECs under varying force parameters. We show that pN-scale mechanical stimulation regulates extracellular Ca2+ influx, triggering down

Efficient optical plasmonic tweezer-controlled single-molecule SERS characterization of pH-dependent amylin species in aqueous milieus.

It is challenging to characterize single or a few biomolecules in physiological milieus without excluding the influences of surrounding environment. Here we utilize optical plasmonic trapping to construct a dynamic nanocavity, which reduces the diffraction-limited detection volume and provides reproducible electromagnetic field enhancements to achieve high-throughput single-molecule surface-enhanced Raman spectroscopy (SERS) characterizations in aqueous environments. Specifically, we study human Islet Amyloid Polypeptide (amylin, hIAPP) under different physiological pH conditions by combining
