Melanotan II nasal spray: a possible risk factor for oral mucosal malignant melanoma?

The case of a 22-year-old female who developed a mass in the anterior maxilla after using Melanotan II, a nasal spray containing a synthetic analogue of melanocyte-stimulating hormone, is reported. The patient had used the nasal spray for tanning purposes. Melanotan II is unlicensed, and its sale is illegal in the United Kingdom and in many other countries around the world. Histological analysis confirmed the diagnosis of a mucosal malignant melanoma. The patient subsequently underwent surgical resection followed by ongoing immunotherapy. The clinical presentation and initial management of the

Optimizing Mucoadhesive Film-Forming Spray for Efficient Oral Delivery of Fluconazole in Candidiasis Treatment.

Buccal candidiasis has become increasingly prevalent in recent years, with Candida albicans being the primary causative organism. While systemic fluconazole is an effective treatment, its use is associated with adverse effects such as gastric upset, hepatic failure, and potential drug interactions. Therefore, the development of local fluconazole treatment presents a promising solution to these challenges. This study aimed to formulate an efficient local mucoadhesive film-forming spray for the targeted delivery of fluconazole in the treatment of oral candidiasis. The investigation involved the

Spray-dried Solid Lipid Nanoparticles for Enhancing Berberine Bioavailability via Oral Administration.

BACKGROUND: Berberine (BBR), an Eastern traditional medicine, has expressed novel therapeutic activities, especially for chronic diseases like diabetes, hyperlipemia, hypertension, and Alzheimer's disease. However, the low oral bioavailability of BBR has limited the applications of these treatments. Hence, BBRloaded solid lipid nanoparticles (BBR-SLNs) were prepared to improve BBR absorption into systemic circulations via this route. METHODS: BBR-loaded solid lipid nanoparticles (BBR-SLNs) were prepared by ultrasonication and then transformed into solid form via spray drying technique. The si

Eudragit® FS Microparticles Containing Bacteriophages, Prepared by Spray-Drying for Oral Administration.

Phage therapy is recognized to be a promising alternative to fight antibiotic-resistant infections. In the quest for oral dosage forms containing bacteriophages, the utilization of colonic-release Eudragit® derivatives has shown potential in shielding bacteriophages from the challenges encountered within the gastrointestinal tract, such as fluctuating pH levels and the presence of digestive enzymes. Consequently, this study aimed to develop targeted oral delivery systems for bacteriophages, specifically focusing on colon delivery and employing Eudragit® FS30D as the excipient. The bacteriophag

A new oral vaccine candidate based on the microencapsulation by spray-drying of inactivated Vibrio cholerae.

The aim of this work was to evaluate the microencapsulation by spray-drying of inactivated Vibrio cholerae, using methacrylic copolymers Eudragit® L30D-55 and FS30D. The microparticles obtained presented a particle size around 3.0 μm. The preparation temperature affected the morphology and the antigenicity of microparticles, but it did not affect the V. cholerae content. In vitro release studies showed that in acid medium less than 5% of bacteria was released, and in neutral medium, Eudragit® L30D-55 microparticles released 86% after 24 h, whereas FS30D released less than 30%. Rats inoculated

Oral tablets of therapeutic eutectic solvent prepared by spray immobilization into excipient microparticles and direct compression.

Therapeutic eutectic solvents (TES) represent liquid forms of active pharmaceutical ingredients (APIs) that can circumvent the issues of low aqueous solubility and polymorphism associated with conventional solid crystalline APIs. Due to their viscous liquid nature, TES applications beyond transdermal drug delivery have been rarely explored. The present work aimed to develop oral tablets of TES to facilitate their delivery via oral ingestion, which represents the most commonly used route of drug administration. A poorly soluble API ibuprofen (IBU) paired with menthol (MEN) at 1:3 molar ratio wa

Spray drying OZ439 nanoparticles to form stable, water-dispersible powders for oral malaria therapy.

BACKGROUND: OZ439 is a new chemical entity which is active against drug-resistant malaria and shows potential as a single-dose cure. However, development of an oral formulation with desired exposure has proved problematic, as OZ439 is poorly soluble (BCS Class II drug). In order to be feasible for low and middle income countries (LMICs), any process to create or formulate such a therapeutic must be inexpensive at scale, and the resulting formulation must survive without refrigeration even in hot, humid climates. We here demonstrate the scalability and stability of a nanoparticle (NP) formulati
