Thimerosal positivities: the rôle of SH groups and divalent ions.

For a better understanding of the mechanistic details of the interactions of organomercury compounds inside the skin, 32 subjects who previously had given positive patch-test reactions to thimerosal (TH) and negative reactions to thiosalicylic acid, were divided into 2 groups. 16 subjects were repatch tested to ethylmercury chloride (EtHgCl) and to solutions containing EtHgCl mixed with L-cysteine and glutathione, respectively. The remaining 16 were repatch tested to EtHgCl and to solutions containing EtHgCl mixed with chlorides of Zn, Mg, and Mn, respectively. The results showed that whilst L

Factors affecting blood mercury concentrations in practicing dentists.

It has been suggested that mercury vapor may be transformed into highly toxic organomercury compounds by micro-organisms in the oral cavity and gastrointestinal tract. If this hypothesis is correct, practicing dentists might be expected to have concentrations of organic mercury in their blood higher than that found in non-dentists. Blood mercury concentrations of practicing dentists and non-dentists were determined by means of cold-vapor atomic absorption spectrophotometry. Potential sources of mercury exposure were identified in both dentists and non-dentists through a questionnaire completed

Synthesis and application of organomercury haptens for enzyme-linked immunoassay of inorganic and organic mercury.

Two organomercury haptens were synthesized via the classical oxymercuration reaction. An intramolecular oxymercuration reaction was the strategy employed to prepare a structurally simple, but chemically robust, organomercury hapten that was conjugated to chicken immunoglobulin G (IgG). The resulting immunogen afforded mouse anti-mercury antibodies that were evaluated in an enzyme-linked immunosorbent assay (ELISA). Antibodies demonstrating high titers were obtained, and various immunoassay parameters were investigated. The sensitivity and selectivity of the resulting antibodies were evaluated

Experimental and theoretical evidence of the existence of gold(I)···mercury(II) interactions in solution through fluorescence-quenching measurements.

Heteronuclear complexes {[Hg(R)2][Au(R')(PMe3)]2}n (R=R'=C6Cl2F3 (3); R=R'=C6F5 (4); R=C6Cl2F3, R'=C6F5 (5); R=C6F5, R'=C6Cl2F3 (6)) were prepared by the treatment of the corresponding organomercury compounds, [Hg(C6X5)2], with two equivalents of [Au(C6X5)(PMe3)]. Their crystal structures, as determined by using X-ray diffraction methods, display Au···Hg interactions. Although only compound 4 and 5 show luminescence in the solid state, all of these compounds quench the fluorescence of naphthalene in solution. Solution studies of these derivatives suggest a cooperative effect of the gold(I) cen

Mercury binding to proteins disclosed by ESI MS experiments: The case of three organomercurials.

Solution interactions of three organomercury compounds, i.e., methylmercury chloride, thimerosal and phenylmercury acetate, with a group of biochemically relevant proteins, namely cytochrome c (Cyt c), ribonuclease A (RNase A), carbonic anhydrase I (hCA I), superoxide dismutase (SOD), and serum albumin (HSA), were investigated using an established ESI MS approach. Temporal analysis of sample aliquots provided insight into the binding kinetics, while comparative analysis of the obtained mass spectra disclosed adduct formation of each mercurial with the tested proteins and the relative abundance

Speciation of organomercurials in marine samples using capillary electrophoresis.

A rapid method for speciation and determination of organomercury compounds in biological samples of marine origin using Capillary Electrophoresis (CE) is reported. Organomercurials were extracted from the samples by means of the classical Westöö procedure thus giving organomercury-cysteine complexes which can be separated from each other by means of CE resulting in effective speciation. Electrophoretic separation was achieved in an open silica capillary tube at 15-18 kV using a 100mM sodium borate buffer (pH 8.35). All mercury species were distinctively separated within 12 min. Results are pre

Organic derivatives of mercury and tin as promoters of membrane lipid peroxidation.

The toxicity mechanisms of mercury and tin organic derivatives are still under debate. Generally the presence of organic moieties in their molecules makes these compounds lipophilic and membrane active species. The recent results suggest that Hg and Sn compounds deplete HS-groups in proteins, glutathione and glutathione-dependent enzymatic systems; this process also results in the production of reactive oxygen species (ROS), the enhancement of membrane lipids peroxidation and damage of the antioxidative defence system. The goal of this review is to present recent results in the studies oriente

Coupling two mercury resistance genes in Eastern cottonwood enhances the processing of organomercury.

Eastern cottonwood (Populus deltoides Bartr. ex Marsh.) trees were engineered to express merA (mercuric ion reductase) and merB (organomercury lyase) transgenes in order to be used for the phytoremediation of mercury-contaminated soils. Earlier studies with Arabidopsis thaliana and Nicotiana tabacum showed that this gene combination resulted in more efficient detoxification of organomercurial compounds than did merB alone, but neither species is optimal for long-term field applications. Leaf discs from in vitro-grown merA, nptII (neomycin phosphotransferase) transgenic cottonwood plantlets wer

Possible alkylation of inorganic Hg(II) by photochemical processes in the environment.

The methylation of inorganic Hg by anaerobic bacteria in aquatic environments is considered to be the major pathway for methylmercury (MeHg) production. However, recent research has suggested that abiotic or chemical methylation by humic substances and other low-molecular-weight organic compounds in natural environments is also possible. Here, the aqueous photo-transformation of Hg(2+) to organomercurials was investigated in the presence of ketones, aldehydes and low molecular weight organic acids under UV irradiation. MeHg and/or ethylmercury (EtHg) were identified as the main organomercurial

In situ ethylation of organolead, organotin and organomercury species by bromomagnesium tetraethylborate prior to GC-ICP-MS analysis.

An analytical method for simultaneous in situ ethylation, of organolead, organotin and organomercury compounds in aqueous samples was developed using a new derivatisation agent, bromomagnesium tetraethylborate (BrMgEt(4)B). The determination of lead, tin and mercury compounds was done by species-specific isotope dilution, derivatisation and GC-inductively coupled plasma MS (GC-ICP-MS) or by GC-MS. The recovery and accuracy of the derivatisation were evaluated. The effect of pH and the relative quantity of derivatisation agent were studied.

Detection of organomercurials with sensor bacteria.

Mercury and its organic compounds, especially methylmercury, are hazardous compounds that concentrate in biota via biomagnification and cause severe neurological disorders in animals. In this paper, a recombinant whole-cell bacterial sensor for the detection of the organic compounds of mercury was constructed. The sensor carries firefly luciferase gene as a reporter under the control of the mercury-inducible regulatory part of broad spectrum mer operon from pDU1358. In addition, a gene-encoding organomercurial lyase (an enzyme necessary for cleavage of the mercury-carbon bond) was coexpressed

Cleaving mercury-alkyl bonds: a functional model for mercury detoxification by MerB.

The extreme toxicity of organomercury compounds that are found in the environment has focused attention on the mechanisms of action of bacterial remediating enzymes. We describe facile room-temperature protolytic cleavage by a thiol of the Hg-C bond in mercury-alkyl compounds that emulate the structure and function of the organomercurial lyase MerB. Specifically, the tris(2-mercapto-1-t-butylimidazolyl)hydroborato ligand [Tm(Bu(t))], which features three sulfur donors, has been used to synthesize [Tm(Bu(t))]HgR alkyl compounds (R = methyl or ethyl) that react with phenylthiol (PhSH) to yield [

Mercury operon regulation by the merR gene of the organomercurial resistance system of plasmid pDU1358.

The structural basis for induction of the mercury resistance operon with inorganic mercury and with the organomercurial compound phenylmercuric acetate was addressed by DNA sequencing analysis and by lac fusion transcription experiments regulated by merR in trans from broad-spectrum-resistance plasmid pDU1358 (Hg2+ and phenylmercury responding). The lac fusion results were compared with those from a narrow-spectrum-resistance (Hg2+ responding but not phenylmercuric responding) operon and the pDU1358 merR deleted at the 3' end. Rapid separation of elemental species by multicapillary GC.

Multicapillary gas chromatography has been applied to the speciation of organomercury compounds. Basic investigation of the fundamental properties of multicapillary columns, to evaluate their potential and limitations as a rapid separation unit, are presented. For analysis of methylated and ethylated mercury compounds a complete separation can be performed within 45 s under isothermal conditions. The adaptation of the technique for use with a purge and trap system and with an element-selective plasma-emission detector results in a compact and effective system for mercury speciation. Organomercury (II) complexes with anti-carcinogenic agents. I. Synthesis and characterization.

A few organomercury (II) complexes involving anti-carcinogenic ligands have been synthesized. The compounds are of the type p-MeOC6H4HgL1 (I), p-NO2C6H4HgL2 (II), p-MeOC4H4HgL3 (III) and p-MeC6H4HgL4 (IV) (HL1-6-mercaptopurine, HL2-6-thioguanine, HL3-5-fluorouracil, L4-phenyldithiocarbazate). Their composition has been determined from elemental analysis. Thin layer chromatography (TLC) studies demonstrate that the compounds are pure. Conductance measurement reveal that these derivatives are non-electrolytes. From IR and UV spectral studies the bonding modes of the ligands to the mercury (II) i

Bis[N-(4-nitrophenyl)thiobenzamidato]mercury(II).

The molecule of the title compound, [Hg(C(13)H(9)N(2)O(2)S)(2)], has approximate twofold rotation symmetry, with the Hg atom in an essentially linear two-coordinate HgS(2) environment supported by secondary pi interactions with the nitrophenyl rings of both ligands. The ligands are in the imine-thiolate rather than the amine-thione tautomeric form.

Genetically modified Escherichia coli for colorimetric detection of inorganic and organic Hg compounds.

A sensitive colorimetric bacterial system was developed for the detection of Hg(II) and organomercury compounds. The bioactive species, a recombinant Escherichia coli, produces proportionally elevated levels of the enzyme beta-galactosidase with increasing amounts of Hg. This is due to a reporter plasmid which carriers a Hg(II)-inducible promoter (mer promoter) from the Hg resistance transposon Tn501 regulating the transcription of a promoterless lacZ gene. Additionally, a pMB1 origin of replication without the natural RNA polymerase start site is fused downstream of the mer promoter leading t

The Synthesis and Molecular Structure of the First Two-Coordinate, Dinuclear σ-Bonded Mercury(I) RHgHgR Compound.

A linear Si-Hg-Hg-Si arrangement and a Hg-Hg distance of 265.69 pm are exhibited by the first two-coordinate, dinuclear σ-bonded organomercury(I) compound 1. It was formed unexpectedly in the reaction of two equivalents of the silane (Me3 SiMe2 Si)3 SiH with tBu2 Hg. In contrast if the reagents are allowed to react in a 1:1 ratio the expected mercury(II) compound (Me3 SiMe2 Si)3 SiHgtBu is obtained.
