Ornithodoros moubata, a soft tick vector for Rickettsia in east Africa?

Omithodoros moubata complex (Argasidae) ticks collected from human dwellings in central Tanzania were found to carry a novel rickettsial species that clustered among the spotted fever group. Although no evidence of human infection was evident, these ticks feed primarily on man, thus providing opportunity for zoonotic infection.

The diversity of neurosecretory cell types in the cave tick Ornithodoros tholozani.

Neurosecretory cells are arranged in 16 NSC centers in the different regions of the brain of O. tholozani. The component cells belong to 13 different types, of which five types are paraldehyde fuchsin positive and the remaining types are negative. Three of these cell types are here described for the first time. Some of these NSC show an affinity to certain localities inside the brain. Cells comprising a neurohaemal organ were found near the exit of the esophagus.

Differential Expression of Putative Ornithodoros turicata Defensins Mediated by Tick Feeding.

Additional research on soft ticks in the family Argasidae is needed to bridge the knowledge gap relative to hard ticks of the family Ixodidae; especially, the molecular mechanisms of Ornithodoros biology. Ornithodoros species are vectors of human and animal pathogens that include tick-borne relapsing fever spirochetes and African swine fever virus. Soft tick vector-pathogen interactions involving components of the tick immune response are not understood. Ticks utilize a basic innate immune system consisting of recognition factors and cellular and humoral responses to produce antimicrobial pept

Ornithodoros cf. mimon infected with a spotted fever group Rickettsia in Brazil.

Ornithodoros mimon is an argasid tick primarily associated with bats that also infest other animals including birds, opossums and humans. In this paper, we report the finding of an argasid species resembling O. mimon, which similarly may be found in human dwellings and parasitize humans in Brazil. We also provide molecular evidence that this argasid tick species may carry a rickettsial organism, whose pathogenicity remains unknown. A total of 16 ticks (two females, two males and 12 nymphs) were collected in the bedroom and in the attic of a human house, where cases of "insect" bites have been

Acquisition and subsequent transmission of Borrelia hermsii by the soft tick Ornithodoros hermsi.

Tick-borne relapsing fever is caused by spirochetes within the genus Borrelia. The hallmark of this disease is recurrent febrile episodes and high spirochete densities in mammalian blood resulting from immune evasion. Between episodes of spirochetemia when bacterial densities are low, it is unknown whether ticks can acquire the spirochetes, become colonized by the bacteria, and subsequently transmit the bacteria once they feed again. We addressed these questions by feeding ticks, Omnithodoros hermsi Wheeler (Acari: Argasidae),daily on an infected mouse during low andhigh levels of spirochete i

Bloodmeal size and spirochete acquisition of Ornithodoros hermsi (Acari: Argasidae) during feeding.

Ornithodoros hermsi Wheeler (Acari: Argasidae) is the vector of Borrelia hermsii, the primary cause of tick-borne relapsing fever in North America. This tick is one of the smallest Ornithodoros species involved with the biological transmission of spirochetes; yet, the amount of blood ingested while feeding is unknown. Therefore, we determined the amount of blood O. hermsi ingested during a bloodmeal to establish its potential for spirochete acquisition while feeding on an infected host. Ticks at different developmental stages were weighed before and after feeding and the volume of blood ingest

Development and field validation of an indirect ELISA for serosurveillance of Ornithodoros lahorensis exposure.

BACKGROUND: Ornithodoros lahorensis is the primary vector of numerous tick-borne diseases. Effective tick surveillance could facilitate the prevention of tick-borne diseases. Direct surveillance methods for tick bites are impractical, the development of an indirect approach to detect specific antibodies against tick salivary proteins in animal samples would provide a more convenient surveillance and diagnostic tool. Previous studies have confirmed that some salivary gland proteins can be used as detection markers for tick bites. METHODS: The recombinant BTSP protein of O. lahorensis (rOl-BTSP

Evaluation of mechanical transmission of HIV by the African soft tick, Ornithodoros moubata.

OBJECTIVES: To assess the ability of the African Hut Tampan, Ornithodoros moubata, to mechanically transmit HIV-1 and to re-appraise HIV-1 infectivity in an arthropod cell line at 28 and 35 degrees C. DESIGN: To evaluate HIV-1 transmission by O. moubata, as determined by HIV-1 survival, 'blood-meal' size and interval between feeds, various tick developmental stages were allowed to feed on a heavily infected lymphoblast-rich blood-meal containing HIV-1BRU in an in vitro feeding chamber. METHODS: Blood-meal regurgitation was evaluated using 51Cr-labelled human erythrocytes, and human lymphobla

African swine fever virus infection in the argasid host, Ornithodoros porcinus porcinus.

The pathogenesis of African swine fever virus (ASFV) infection in Ornithodoros porcinus porcinus was examined in nymphal ticks infected with the ASFV isolate Chiredzi/83/1. At times postinfection (p.i.) ranging from 6 h to 290 days, ticks or dissected tick tissues were titrated for virus and examined ultrastructurally for evidence of virus replication. The ASFV infection rate in ticks was 100% in these experiments, and virus infection was not associated with a significant increase in tick mortality. Initial ASFV replication occurred in phagocytic digestive cells of the midgut epithelium. Subse

New World relapsing fever Borrelia found in Ornithodoros porcinus ticks in central Tanzania.

Ticks were collected from 8 houses in Mvumi Mission village, near Dodoma, Tanzania. All ticks were examined for Borrelia infestation by flagellin gene-based nested polymerase chain reaction. All houses were highly infested with ticks, and all ticks collected were of the Ornithodoros porcinus species. Fifty-one out of 120 ticks were infected with spirochetes, and a flagellin gene sequence comparison showed that most of the spirochetes belonged to Borrelia duttonii, which is the causative agent of tick-borne relapsing fever in East Africa. The rest of the spirochetes were quite different from B.

Detection and quantification of Ornithodoros-specific anti-tick antibody by competitive inhibition ELISA.

The objective of this study was to develop a highly specific enzyme-linked immunosorbent assay (ELISA) for the serological detection of anti-Ornithodoros tick antibodies in animals. Affinity-purified rabbit anti-Ornithodoros IgG antibodies were employed in indirect competitive inhibition ELISA assays designed to measure the anti-Ornithodoros antibody titers in other animal species using the domestic goat (Capra hircus) as a large animal model. Repeated infestation of goats with Ornithodoros coriaceus was found to elicit the formation of antibodies capable of inhibiting the binding of the Ornit

A proteomics informed by transcriptomics insight into the proteome of Ornithodoros erraticus adult tick saliva.

BACKGROUND: The argasid tick Ornithodoros erraticus is the main vector of tick-borne human relapsing fever (TBRF) and African swine fever (ASF) in the Mediterranean Basin. The prevention and control of these diseases would greatly benefit from the elimination of O. erraticus populations, and anti-tick vaccines are envisaged as an effective and sustainable alternative to chemical acaricide usage for tick control. Ornithodoros erraticus saliva contains bioactive proteins that play essential functions in tick feeding and host defence modulation, which may contribute to host infection by tick-born

Discovery of Ornithodoros tholozani, the Main Vector of Iranian Tick-born Relapsing Fever.

Doctor Joseph Désiré Tholozan was a French physician who became the special physician of Nassereddin Shah, King of Persia (Iran) in the 19th century. He studied lots of topics in the field of epidemiology of infectious diseases. His efforts led to the discovery of the main vector of Iranian tick-born relapsing fever, Ornithodoros tholozani. He was also one of the pioneers of the Iranian Sanitary Council, whose efforts led to the foundation of the Ministry of Health of Iran a few decades later. This paper is a brief review of his biography and his roles in promoting health in Iran in the 19th c

Morphological and molecular confirmation of Ornithodoros hasei (Schulze, 1935) (Acari: Argasidae) in Colombia.

A large number of tick species are proven vectors for the transmission of bacteria, protozoa, and viruses. Soft ticks (Acari: Argasidae) in South America have been found to be the most frequent carriers of borreliae of the relapsing fever group (RFG); however, there are several information gaps specially on the taxonomy and distribution of some tick species. Here, we used light microscopy, scanning electron microscopy, and PCR amplification of a fragment of the mitochondrial 16S rRNA gene to evaluate 174 larvae of Ornithodoros (Argasidae) collected from three bat species (Eptesicus orinocensis
