An adenosine triphosphate-phosphate exchange catalyzed by a soluble enzyme couple inhibited by uncouplers of oxidative phosphorylation.

The sulfenic acid form of glyceraldehyde-3-phosphate dehydrogenase (EC 1.2.1.12), which is an acyl phosphatase, will catalyze an acetyl phosphate-Pi exchange reaction. This exchange reaction is reversibly inhibited by the uncouplers of oxidative phosphorylation, 2,4-dinitrophenol, m-Cl carbonylcyanide-phenylhydrazone, pentachlorophenol, and 5-chloro-3-tert-butyl-2'-chloro-4'-nitrosalicylanalide, and is irreversibly inhibited by cyanide and dicumarol. An ATP-Pi exchange reaction similar to that catalyzed by mitochondria can be simulated by a system composed of oxidized glyceraldehyde-3-phosphat

Mitochondrial fission factor senses and governs ferroptosis.

Ferroptosis is a form of regulated cell death driven by iron-dependent lipid peroxidation and is implicated in diverse pathological states1-4. Although mitochondria and other organelles are increasingly being recognized as important modulators of ferroptosis5-7, a unifying mechanism that couples organelle dynamics to ferroptotic execution has remained elusive. Here using quantitative phosphoproteomics, we identified mitochondrial fission factor (MFF) as a key ferroptosis-selective regulator. Mechanistically, the lipid mediator 17-HETE promotes phosphorylation of MFF at Ser155, which triggers t

KIF18A knockdown enhances oxidative phosphorylation, suggesting a metabolic basis for antiviral activity against influenza A virus.

BACKGROUND: Influenza A virus (IAV) infection remains a major global health burden, underscoring the need to identify novel host-directed antiviral mechanisms. KIF18A, a kinesin-8 family motor protein, has emerged as a proviral host factor that supports IAV replication, and its pharmacological inhibition suppresses viral infection. However, the transcriptional and metabolic processes underlying this antiviral effect remain incompletely understood. OBJECTIVE: We aimed to characterize the transcriptional consequences of KIF18A knockdown in IAV-infected cells and identify metabolic pathways pote

Revising NHE-1: From Cardiac Homeostasis to Heart Failure and Future Drug Development.

NHE-1 is a Na+/H+ exchanger that receives phosphorylation signals, binds calmodulin and responds to neurohormonal input from angiotensin II, endothelin-1, and adrenergic pathways. In cardiac myocytes, NHE-1 maintains pH homeostasis and couples to Na+/Ca2+ exchange and mitochondrial ion handling. During heart disease sustained activation drives intracellular Na+ accumulation, promoting Ca2+ overload and mitochondrial dysfunction. Oxidative stress then creates amplifying cycles that activate signaling pathways resulting to arrhythmias and fibrosis. Clinical trials failed despite preclinical prom

From lipid switch to tissue repair: how resolvins reprogram macrophage polarization and function.

Specialized pro-resolving mediators (SPMs) derived from docosahexaenoic acid (DHA), particularly D-series resolvins (RvD1, RvD2, RvD3, and RvD5), function to terminate inflammation while preserving host defense. They are synthesized from DHA by lipoxygenases and act through G‑protein‑coupled receptors and lipid‑sensing transcription factors (TFs). These mediators reprogram macrophage metabolism towards fatty‑acid oxidation and oxidative phosphorylation, accelerate efferocytosis, and promote tissue repair. Here, we synthesize current knowledge on their biosynthesis, receptor signaling, and immu

Hydrogen sulfide protects retinal pigment epithelium cells against ferroptosis through the AMPK- and p62-dependent non-canonical NRF2-KEAP1 pathway.

Oxidative stress-induced ferroptosis of retinal pigment epithelium (RPE) cells contributes to retinal degenerative diseases. The antioxidant molecule hydrogen sulfide (H2S) regulates oxidative stress response, but its effect on the ferroptosis of RPE cells is unclear. In this study, sodium hydrosulfide (NaHS) was used as an exogenous H2S donor to intervene tert-butyl hydroperoxide (t-BHP)-induced ferroptosis of APRE-19 cells. We found that NaHS pretreatment attenuates t-BHP-induced oxidative stress and ferroptosis. Analysis of mRNA-sequencing coupled with FerrDb database identified nuclear fac

Lactate induces oxidative phosphorylation in osteoblasts via Gpr81-Stat3 signaling.

Lactate has long been regarded as an end product of glycolysis and a metabolic "waste product" under hypoxic conditions, but recent studies have revealed that lactate plays a central role in energy metabolism reprogramming and intercellular communication. However, it remains unknown whether lactate promotes osteogenic differentiation through metabolic reprogramming. Here, we showed that lactate significantly increased the cellular ATP content, activated succinate dehydrogenase activity, and enhanced oxygen consumption rate in pre-osteoblast MC3T3-E1 cells. Moreover, lactate treatment increased

Far-Infrared Therapy: Mechanisms of Action.

Far-infrared therapy, with wavelengths ranging from 3 to 25 μm, has emerged as a noninvasive photothermal and photobiomodulatory intervention with promising therapeutic applications in cardiovascular medicine, wound healing, and chronic inflammatory diseases. Far-infrared penetrates superficial and subcutaneous tissues, inducing mild, sustained hyperthermia and wavelength-specific molecular signaling. These effects activate mechanosensitive and thermosensitive pathways, including the transient receptor potential channels, phosphoinositide 3-kinase-Akt pathway, and adenosine monophosphate-activ

Banting Lecture 1995. A lesson in metabolic regulation inspired by the glucokinase glucose sensor paradigm.

Special features of glucose metabolism in pancreatic beta-cells are central to an understanding of the physiological role of these cells in glucose homeostasis. Several of these characteristics are emphasized: a high-capacity system for glucose transport; glucose phosphorylation by the high-Km glucokinase (GK), which is rate-limiting for glucose metabolism and determines physiologically the glucose dependency curves of many processes in beta-cell intermediary and energy metabolism and of insulin release and is therefore viewed as glucose sensor; remarkably low activity of lactate dehydrogenase

Mitochondrial dysfunction in heart failure.

Heart failure (HF) is a complex chronic clinical syndrome. Energy deficit is considered to be a key contributor to the development of both cardiac and skeletal myopathy. In HF, several components of cardiac and skeletal muscle bioenergetics are altered, such as oxygen availability, substrate oxidation, mitochondrial ATP production, and ATP transfer to the contractile apparatus via the creatine kinase shuttle. This review focuses on alterations in mitochondrial biogenesis and respirasome organization, substrate oxidation coupled with ATP synthesis in the context of their contribution to the chr

Understanding and Sensitizing Density-Dependent Persistence to Quinolone Antibiotics.

Physiologic and environmental factors can modulate antibiotic activity and thus pose a significant challenge to antibiotic treatment. The quinolone class of antibiotics, which targets bacterial topoisomerases, fails to kill bacteria that have grown to high density; however, the mechanistic basis for this persistence is unclear. Here, we show that exhaustion of the metabolic inputs that couple carbon catabolism to oxidative phosphorylation is a primary cause of growth phase-dependent persistence to quinolone antibiotics. Supplementation of stationary-phase cultures with glucose and a suitable t

Mitochondrial functions in five cases of human neuromuscular disorders.

We determined the respiration, respiratory control, and Pi:O ratios with different substrates in mitochondria isolated from five cases of human neuromuscular disorders (two cases of central core disease, two cases of neuropathy of Dejerine-Sottas, and one case of Kugelberg-Welander's disease) and compared them with normal human muscle. In all the myopathies studied, a severe derangement of the respiratory control with variable derangement of oxidative phosphorylation was found. This supports the idea that a group of neuromyopathies shares the same biochemical lesion as the so-called mitochondr

CTRP13 Preserves Endothelial Function by Targeting GTP Cyclohydrolase 1 in Diabetes.

Endothelial dysfunction plays a crucial role in the progress of diabetic vasculopathy. C1q/tumor necrosis factor-related protein 13 (CTRP13) is a secreted adipokine that can ameliorate atherosclerosis and vascular calcification. However, the role of CTRP13 in regulating endothelial function in diabetes has yet to be explored. In this study, CTRP13 treatment improved endothelium-dependent relaxation in the aortae and mesenteric arteries of both db/db mice and streptozotocin-injected mice. CTRP13 supplement also rescued the impaired endothelium-dependent relaxation ex vivo in the db/db mouse aor

The role of mitochondrial DNA in bone/cartilage diseases.

Skeletons, as a human sports scaffold, provide attachment points for muscle and ligament attachment, and store significant quantities of calcium and phosphate. The aetiology of bone-related diseases is multifactorial, involving a complex interplay between osteoblasts and osteoclasts, as well as immune system abnormalities. These factors can result in impaired motor function and physical discomfort. Recent studies have indicated an association between mitochondrial DNA (mtDNA) abnormalities and the development of bone and cartilage diseases. The level of mtDNA in body fluid is expected to becom

Cognitive impairment caused by hypoxia: from clinical evidences to molecular mechanisms.

Hypoxia is a state of reduced oxygen supply and excessive oxygen consumption. According to the duration of hypoxic period, it can be classified as acute and chronic hypoxia. Both acute and chronic hypoxia could induce abundant neurological deficits. Although there have been significant advances in the pathophysiological injuries, few studies have focused on the cognitive dysfunction. In this review, we focused on the clinical evidences and molecular mechanisms of cognitive impairment under acute and chronic hypoxia. Hypoxia can impair several cognitive domains such as attention, learning and m

Time-Resolved Oxidative Stress and Apoptosis in Murine Retina under Acute Hypobaric Hypoxia with Parallel Activation of the LIF-JAK-STAT3 Axis.

High-altitude retinopathy (HAR) arises under acute hypobaric hypoxia (AHH), yet the temporal coupling of oxidative stress, apoptosis and cytokine signaling in the retina remains unclear. We exposed healthy male C57BL/6 mice to a hypobaric chamber (≈5,000 m) for 2-72 h, with normoxic controls at 1,500 m (n = 3 eyes/group). Haematoxylin-eosin sections quantified total and laminar thickness; at 24 h, bulk RNA-seq profiled transcripts. Reactive oxygen species (ROS) were assayed by dihydroethidium, and Western blotting/immunofluorescence evaluated Bax, Bcl-2/Bcl-xL, cleaved caspase-3, cytochrome c,

New aspects of pathogenesis of atrial fibrillation: remodelling of intercalated discs.

This review deals with the understanding of the role of key factors of ageing, oxidative stress and inflammation, in relation to development of age-related cardiovascular disease, atrial fibrillation. Increased production of reactive oxygen species and systemic inflammation promote cardiac structural and electrophysiologic remodeling that is crucial with respect to development and sustaining of atrial fibrillation. Data suggest that alterations in atrial connexin-43 and/or connexin-40 expression, phosphorylation and distribution affect cell-to-cell electrical coupling and molecular signalling

Increased protein phosphatase 5 expression in inflammation-induced left ventricular dysfunction in rats.

BACKGROUND: Titin phosphorylation contributes to left ventricular (LV) diastolic dysfunction. The independent effects of inflammation on the molecular pathways that regulate titin phosphorylation are unclear. METHODS: We investigated the effects of collagen-induced inflammation and subsequent tumor necrosis factor-α (TNF-α) inhibition on mRNA expression of genes involved in regulating titin phosphorylation in 70 Sprague-Dawley rats. LV diastolic function was assessed with echocardiography. Circulating inflammatory markers were quantified by enzyme-linked immunosorbent assay and relative LV ge

Inflammation, oxidative stress, and obesity.

Obesity is a chronic disease of multifactorial origin and can be defined as an increase in the accumulation of body fat. Adipose tissue is not only a triglyceride storage organ, but studies have shown the role of white adipose tissue as a producer of certain bioactive substances called adipokines. Among adipokines, we find some inflammatory functions, such as Interleukin-6 (IL-6); other adipokines entail the functions of regulating food intake, therefore exerting a direct effect on weight control. This is the case of leptin, which acts on the limbic system by stimulating dopamine uptake, creat
