Virome Analysis of Normal and Growth Retardation Disease-Affected Macrobrachium rosenbergii.

The giant freshwater prawn, Macrobrachium rosenbergii, is an important aquaculture species in China. Growth retardation disease (GRD) is a common contagious disease in M. rosenbergii, resulting in slow growth and precocious puberty in prawns, and has caused growing economic losses in the M. rosenbergii industry. To investigate the viral diversity of M. rosenbergii and identify potentially high-risk viruses linked to GRD, virome analysis of the GRD-affected and normal M. rosenbergii was carried out using next-generation sequencing (NGS). A total of 327 contigs (>500 bp) were related to viral se

Pyrethroid insecticides pose greater risk than organophosphate insecticides to biocontrol agents for human schistosomiasis.

Use of agrochemicals, including insecticides, is vital to food production and predicted to increase 2-5 fold by 2050. Previous studies have shown a positive association between agriculture and the human infectious disease schistosomiasis, which is problematic as this parasitic disease infects approximately 250 million people worldwide. Certain insecticides might runoff fields and be highly toxic to invertebrates, such as prawns in the genus Macrobrachium, that are biocontrol agents for snails that transmit the parasites causing schistosomiasis. We used a laboratory dose-response experiment and

Repellency and mortality effects of sunscreens on the shrimp Palaemon varians: Toxicity dependent on exposure method.

Contamination by sunscreens has become a serious environmental problem due to the increasing use of these products in coastal regions. Their complex chemical composition supposes an input of different chemical compounds capable of producing toxic effects and repelling organisms. The aim of the current study was to experimentally check the repellency of three commercial sunscreens [A (lotion), B (gel) and C (milk spray)] by assessing the escape (displacement towards areas with lower sunscreen levels) of the estuarine shrimp Palaemon varians exposed (4 h) to a gradient (0-300 mg/L) of the sunscr

Effects of Chemical Compounds on the Activity of the N-acetyl-β-D-Glucosaminidase of the Marine Prawn, Palaemon serratus: Screening In Vitro.

N-acetyl-β-D-glucosaminidase (NAGase) is important for crustaceans because the enzyme activity is necessary for the molting process. The present study aimed to assess the sensitivity of Palaemon serratus NAGase activity to a set of compounds of diverse chemical families in the context of in vitro exposures. Compounds representing different chemical families were selected according to their abundance, impact in the environment, and relevance as disruptors of the molting process. In a first step, four solvents (dimethylsulfoxide [DMSO], methanol, acetone, and ethanol) were tested to determine th

First case of Coxiella burnetii infection in Palaemonetes sinensis cultured in Liaoning, China.

The Palaemonetes sinensis aquaculture industry in Panjin City, Liaoning Province, China, experienced heavy losses in October 2018. Morbidity of cultured shrimp reached 50% and was characterized by cloudiness of muscle and the gradual spread of disease within the population. When the infection was mild, histopathological examinations revealed that the muscle cells contained a considerable number of microorganisms. In extreme cases, the structure of the hepatopancreatic glandular and muscle fiber was obscured or even vanished. Electron microscope observations revealed the presence of granular cy

Resistance to the crayfish plague pathogen, Aphanomyces astaci, in two freshwater shrimps.

Aphanomyces astaci, the causal agent of the crayfish plague, has recently been confirmed to infect also freshwater-inhabiting crabs. We experimentally tested the resistance of freshwater shrimps, another important decapod group inhabiting freshwaters, to this pathogen. We exposed individuals of two Asian shrimp species, Macrobrachium dayanum and Neocaridina davidi, to zoospores of the pathogen strain isolated from Procambarus clarkii, a known A. astaci carrier likely to get into contact with shrimps. The shrimps were kept in separate vessels up to seven weeks; exuviae and randomly chosen indiv

Functional Characterization of Tret1-like Gene and Its Critical Role in Regulating Growth in Macrobrachium nipponense by RNA Interference.

Macrobrachium nipponense is a commercially significant freshwater prawn species in China, with larger individuals commanding higher market prices than smaller ones. Previous transcriptomic analyses have shown that a facilitated trehalose transporter gene Tret1-like (Tret1) exhibits significantly elevated expression levels in fast-growing individuals compared to their slow-growing counterparts, suggesting a potential involvement of this gene in modulating growth performance. In the present study, we explored the functional roles of Mn-Tret1 in growth regulation using reverse transcription quant

Sex-Specific Screening Mitigates Basal Metabolic Dimorphism Bias and Reveals DIV1 Survival-Associated Metabolic Signatures in Macrobrachium rosenbergii.

Decapod Iridescent Virus 1 (DIV1) causes severe economic losses in Macrobrachium rosenbergii farming. Serum metabolic biomarkers are valuable tools for disease-resistant breeding, yet most existing studies employ mixed-sex samples, and the impact of sexual dimorphism on screening reliability remains poorly quantified. In this study, non-targeted serum metabolomics was performed on 28 healthy M. rosenbergii (14 females and 14 males), which were divided into marginally resistant and susceptible groups after DIV1 challenge. We compared results from mixed-sex and sex-specific screening strategies,

The prawn Macrobrachium vollenhovenii in the Senegal River basin: towards sustainable restocking of all-male populations for biological control of schistosomiasis.

Early malacological literature suggests that the outbreak of schistosomiasis, a parasitic disease transmitted by aquatic snails, in the Senegal River basin occurred due to ecological changes resulting from the construction of the Diama dam. The common treatment, the drug praziquantel, does not protect from the high risk of re-infection due to human contact with infested water on a daily basis. The construction of the dam interfered with the life cycle of the prawn Macrobrachium vollenhovenii by blocking its access to breeding grounds in the estuary. These prawns were demonstrated to be potenti

Ovarian microrna transcriptome of Macrobrachium rosenbergii after eyestalk ablation: unveiling molecular switches in reproductive regulation.

BACKGROUND: Macrobrachium rosenbergii is an economically important freshwater prawn species in China. The X-organ–sinus gland complex in crustacean eyestalks regulates reproduction through neurohormones, with ablation significantly promoting ovarian maturation. However, the miRNA mediated molecular mechanisms underlying this eyestalk ovary regulatory axis remain poorly characterized in M. rosenbergii. RESULTS: RNA sequencing of 9 ovarian samples produced 207 million paired-end reads (62.25 Gb total), averaging > 6 Gb per sample with Q30 scores exceeding 94.42%. The reads showed 84.32% alignmen

Experimental evidence of contamination driven shrimp population dynamics: Susceptibility of populations to spatial isolation.

Contamination is likely to affect the composition of an ecological landscape, leading to the rupture of ecological connectivity among habitats (ecological fragmentation), which may impact on the distribution, persistence and abundance of populations. In the current study, different scenarios within a spatially heterogeneous landscape were simulated in the Heterogeneous Multi-Habitat Assay System (HeMHAS) to evaluate the potential effect that contamination (copper at 0.5 and 25 μg/L) might have on habitat selection by the estuarine shrimp Palaemon varians in combination with two other ecologica

Steroids and genes related to steroid biosynthesis in the female giant freshwater prawn, Macrobrachium rosenbergii.

The giant freshwater prawn, Macrobrachium rosenbergii, is important to many Asian countries due to its high economic value as an aquaculture product. With demand increasing, there is requirement for a better understanding of the biosynthetic components that regulate its growth and reproduction, including steroids, in order to help increase production. Vertebrate-type steroids and their receptors were identified in crustaceans and implicated in reproduction. In this study, we presented the sex steroids estradiol and progesterone by LC-MS/MS in female M. rosenbergii, and reveal steroidogenic-rel

Genome-wide identification, phylogeny, expression and eyestalk neuroendocrine regulation of vitellogenin gene family in the freshwater giant prawn Macrobrachium rosenbergii.

Vitellogenin (Vg) is the precursor of vitellin, which is an important female-specific protein stored in oocytes as the major nutrient and energy sources for embryogenesis in oviparous animals. In this study, we performed comprehensive genome-wide analysis of Vg gene family in the prawn Macrobrachium rosenbergii, and eight Vg genes designated as MrVg1a, MrVg1b and MrVg2-7 were identified. MrVg1a clusters with the previously described MrVg1b near the end of chromosome 46 and MrVg2 is on the chromosome 42 while MrVg3-7 cluster on the chromosome 23. All the putative MrVg proteins are characterized

Investigation of the conversion mechanism of endogenous semicarbazide in shrimp on the amino acid level.

Semicarbazide (SEM), the metabolite of antibiotic nitrofurazone, is often used as the biomarker to determine the use of nitrofurazone. Frequent false-positive events of SEM have brought great trouble to the aquatic industry in international trade. In this paper, the situation of endogenous SEM in aquatic products was investigated, and the possible mechanism of amino acid conversion into SEM was studied by establishing a simulated oxidation system and a urea system. The results revealed the presence of endogenous SEM in the muscle tissue of shrimps, and the content of SEM ranged from 0.56 to 5.
