Towards the Establishment of a Sea Urchin Sperm Biobank: Key Challenges and Opportunities for the Conservation and Sustainable Aquaculture of Paracentrotus lividus.

The Mediterranean Sea urchin Paracentrotus lividus is a key benthic herbivore and a valuable fishery resource because of the high commercial and gastronomic value of its edible gonads. Increasing market demand, overharvesting, climate change, habitat degradation, and pollution have contributed to the decline of wild populations in several Mediterranean areas. This narrative review, including a preliminary case study, provides an integrated perspective on the role of P. lividus in conservation-oriented aquaculture, with particular attention to Integrated Multi-Trophic Aquaculture (IMTA), microb

Preliminary evaluation of ferrous sulphate toxicity on larvae of the sea urchin Paracentrotus lividus: implications for ocean iron fertilization.

Ocean iron fertilization (OIF) has emerged as a potential marine carbon dioxide removal strategy, involving the addition of ferrous sulphate (FeSO₄) to iron-limited waters to stimulate phytoplankton blooms and enhance carbon sequestration. However, the ecotoxicological impacts on non-target organisms remain poorly characterised. We evaluated FeSO₄ toxicity using embryo-larval bioassays with the sea urchin Paracentrotus lividus, a standard model for marine ecotoxicology. FeSO₄ demonstrated an EC₅₀ of 60.81 μg/L (95% CI: 53.29-68.33 μg/L), compared to 17.90 μg/L for copper used as positive contr

Toxicity of engineered micro- and nanomaterials with antifouling properties to the brine shrimp Artemia salina and embryonic stages of the sea urchin Paracentrotus lividus.

Antifouling booster biocides are chemicals used in protective paints to tackle the adhesion of fouling organisms to maritime artificial structures. However, they are also known to exert toxic effects on non-target organisms. Recent research developments have highlighted the potential use of engineered micro/nanomaterials (EMNMs) as carriers of antifouling booster biocides in order to control their release and to reduce the harmful effects on living biota. In the present study, we sought to assess the toxicity of two commercially-available booster biocides: (zinc pyrithione (ZnPT) and copper py

The Effect of Photobiomodulation on the Sea Urchin Paracentrotus lividus (Echinodermata) Using Higher-Fluence on Fertilization, Embryogenesis, and Larval Development: An In Vitro Study.

OBJECTIVE: The aim of this study was to investigate the photobiomodulation (PBM) effect of the 808 nm diode laser irradiation on spermatozoa, eggs, fertilized eggs, embryos, and larvae of Paracentrotus lividus, using two different power settings. BACKGROUND DATA: Studies have shown the possible use of PBM in artificial insemination. These have shown the potential effect of low-power laser irradiation on spermatozoa, while there are few studies on the effect of laser photonic energy on oocytes and almost no reports on the influence of lasers in embryogenesis. METHODS: P. lividus gametes, zygo

Antioxidant and immune response of the sea urchin Paracentrotus lividus to different re-suspension patterns of highly polluted marine sediments.

Marine pollution due to disused industrial activities is a major threat to ecosystems and human health, for example through the effects of re-suspension of toxic substances that are present in contaminated sediments. Here, we examined the effects of different re-suspension patterns of polluted sediments from the site of national interest Bagnoli-Coroglio, on the immune system of the sea urchin Paracentrotus lividus. An indoor experiment was set up exposing sea urchins for 34 days to such sediments and evaluating the effects of two patterns of water turbulence, mimicking natural storms at sea.

Ecotoxicological evaluation of four UV filters using marine organisms from different trophic levels Isochrysis galbana, Mytilus galloprovincialis, Paracentrotus lividus, and Siriella armata.

Due to the concern about the negative effects of exposure to sunlight, combinations of UV filters like 4-Methylbenzylidene-camphor (4-MBC), Benzophenone-3 (BP-3), Benzophenone-4 (BP-4) and 2-Ethylhexyl-4-methoxycinnamate (EHMC) are being introduced in all kind of cosmetic formulas. These chemicals are acquiring a concerning status due to their increasingly common use and the potential risk for the environment. The aim of this study is to assess the behaviour of these compounds in seawater, the toxicity to marine organisms from three trophic levels including autotrophs (Isochrysis galbana), her

Responses to environmentally relevant microplastics are species-specific with dietary habit as a potential sensitivity indicator.

There is a lack of information on understanding how marine organisms respond to environmentally relevant microplastics (MP) which hampers decision making for waste management strategies. This study addresses this information gap by determining whether responses to MPs are species specific within a functional group. Benthic residing sea urchins, Psammechinus miliaris and Paracentrotus lividus were used as a case study. Psammechinus miliaris are strong omnivores with dietary intake including hard components (e.g. shell, tubeworms) and therefore likely to cope with the ingestion of MPs, while P.

Locality matters: Variation in the reproductive cycle and population structure of subtropical sea urchins.

The life cycle of many Echinoidea species remains poorly understood despite research conducted in temperate and tropical-subtropical regions. Common species in the Central-Eastern Atlantic's shallow waters include Paracentrotus lividus, Arbacia lixula, and Sphaerechinus granularis. Nevertheless, significant gaps in understanding their life cycles persist. This study discusses the reproductive cycles of three sea urchin species in rocky coastal ecosystems around Gran Canaria Island (Spain) (27º45´N, 15º45´W) from June 2020 to May 2021. Morphological measurements reveal that test length increase

Purification of sea-urchin RNA polymerase II. Characterization by template requirements and sensitivity to inhibitors.

The purification of RNA polymerase II from gastrulae of Paracentrotus lividus is described. The enzyme obtained is homogeneous as judged by electrophoresis under non-denaturing conditions. It is able to transcribe both native high-Mr P. lividus DNA and Psammechinus miliaris h22 histone DNA, although single-stranded and nicked DNAs are better templates. P. lividus RNA polymerase II forms with homologous native DNA stable binary complexes that are able to initiate RNA chains after exposure to heparin. Heparin-resistant complexes do not form on nicks of DNA molecules. Sensitivity of sea-urchin RN

Sea urchin coelomic fluid agglutinin mediates coelomocyte adhesion.

The sea urchin Paracentrotus lividus coelomic fluid was found to contain agglutinin which agglutinates animal erythrocytes and promotes adhesion of autologous coelomocytes. Hemagglutinating activity depended upon the presence of calcium ions and was relatively heat-stable. Through a combination of methods including ammonium sulfate precipitation and both size exclusion and ion exchange chromatographies, we purified the anti-rabbit agglutinating factor. The intact agglutinin migrates as a single band with an apparent M(r) of over 200,000. Three distinct protein bands with a calculated M(r) of 1

Toxicity of tire particle leachates on early life stages of keystone sea urchin species.

Particles from tires are a major fraction of microplastic pollution. They contain a wide range of chemical additives that can leach into the water and be harmful to aquatic organisms. In this study, we investigated the acute toxicity of tire particle leachates in early life stages of three keystone echinoderm species (Paracentrotus lividus, Arbacia lixula, Diadema africanum). Embryos were exposed for 72 h to a range of leachate dilutions, prepared using a concentration of 1 g L-1. Larval growth, abnormal development, and mortality were the measured endpoints. Furthermore, we estimated the acti

Bioaccumulation of organotin compounds in the marbled electric ray (Torpedo marmorata) in the northern Adriatic Sea.

Organotin compounds (OTC), tri-, di- and monobutyl tin, were determined in the tissues of marbled electric ray (Torpedo marmorata) in the Adriatic Sea. Marbled electric ray specimens were provided by local fishermen from three localities in the northern Adriatic: area close to the shipyard in Seča, the natural protected area Strunjan Nature Reserve and along the west Istrian coast. To assess the concentration of OTC in the environment, sediment samples were also analysed. After an adequate extraction of OTC from both matrices, their concentrations were determined by GC-ICP-MS. The results indi

Understanding osmoregulation, cryoprotectant toxicity, and cold tolerance in sea urchin eggs: Implications for cryopreservation.

This study investigates the challenges and potential solutions for cryopreserving sea urchin (Paracentrotus lividus) eggs. It focuses on understanding the thresholds and osmoregulatory mechanisms, cryoprotectant toxicity, and cold tolerance in these eggs, which are especially difficult to preserve due to high sensitivity to cryoprotectants and low-temperature injury. Key findings suggest that stepwise addition of cryoprotectants, particularly dimethyl sulfoxide (Me2SO) combined with other agents like DMF or methanol, reduces toxicity and enhances larval survival. The study also highlights that

Heavy metals in Ras Beirut prawns and sea urchin eggs.

Whole samples of Palaemon elegans Rathke, 1837 and eggs of Paracentrotus lividus Lamarck, 1816 were collected on the coast of Ras Beirut, Lebanon in the summer and fall of 1977 and the spring of 1978. All were analysed for lead, cadmium, copper and other heavy metals. Lead concentrations in both the prawns and sea urchin eggs appeared to be highest in the summer and lowest in the spring, whereas cadmium was low in all seasons with only a few elevated readings. The strongest tendency towards seasonal variation was seen in the copper levels of the prawns, which were high in summer, much lower in

Assessing the toxicity of gadolinium in freshwater and marine ecosystems: Effects across trophic levels.

Rare Earth Elements, particularly gadolinium (Gd), are increasingly used in industrial and medical applications, leading to their dispersion in aquatic environments. This study examines the ecotoxicological effects of Gd on aquatic organisms across different trophic levels in freshwater and marine ecosystems. Using standardized bioassays, the impact of Gd was assessed on Aliivibrio fischeri, Raphidocelis subcapitata, Phaeodactylum tricornutum, Daphnia magna, and Paracentrotus lividus larvae. Results showed that primary producers exhibited the highest sensitivity, with growth inhibition at low

EGTA treatment causes the synthesis of heat shock proteins in sea urchin embryos.

Paracentrotus lividus embryos, at post-blastular stage, when subjected to a rise in temperature from physiologic (20 degrees C) to 31 degrees C, synthesize a large group of heat shock proteins (hsps), and show a severe inhibition of bulk protein synthesis. We show, by mono- and two-dimensional electrophoresis, that also EGTA (ethylene glycol-bis[beta-aminoethyl ether] tetraacetic acid) treatment induces in sea urchin embryos both marked inhibition of bulk protein synthesis and the synthesis of the entire set of hsps. Furthermore, EGTA-treated sea urchin embryos are able to survive at a tempera

Reproductive toxicity assessment of cellulose nanofibers, citric acid, and branched polyethylenimine in sea urchins: Eco-design of nanostructured cellulose sponge framework (Part B).

In the framework of a safe-by-design approach, we previously assessed the eco-safety of nanostructured cellulose sponge (CNS) leachate on sea urchin reproduction. It impaired gamete quality, gamete fertilization competence, and embryo development possibly due to the leaching of chemical additives used during the CNS synthesis process. To extend this observation and identify the component(s) that contribute to CNS ecotoxicity, in the present study, we individually screened the cytotoxic effects on sea urchin Arbacia lixula and Paracentrotus lividus gametes and embryos of the three main constitu

Sea Urchin Extracellular Proteins Design a Complex Protein Corona on Titanium Dioxide Nanoparticle Surface Influencing Immune Cell Behavior.

Extensive exploitation of titanium dioxide nanoparticles (TiO2NPs) augments rapid release into the marine environment. When in contact with the body fluids of marine invertebrates, TiO2NPs undergo a transformation and adhere various organic molecules that shape a complex protein corona prior to contacting cells and tissues. To elucidate the potential extracellular signals that may be involved in the particle recognition by immune cells of the sea urchin Paracentrotus lividus, we investigated the behavior of TiO2NPs in contact with extracellular proteins in vitro. Our findings indicate that TiO

Cytotoxic and antimitotic activities in aqueous extracts of eight cyanobacterial strains isolated from the marine sponge Petrosia ficiformis.

Marine cyanobacteria are photosynthetic prokaryotes of significant ecological interest, living free or in association with invertebrates. They are also considered as excellent sources of antineoplastic, antibacterial, antiviral and antifungal compounds. In this work, aqueous extracts from eight cyanobacterial strains isolated from the Mediterranean sponge Petrosia ficiformis have been investigated for their bioactive properties. Bioassays with human erythrocytes, Artemia salina nauplii, and Paracentrotus lividus gametes and embryos were performed. Some aqueous extracts exhibited citolytic effe
