An Investigation of a Cluster of Parapoxvirus Cases in Missouri, Feb-May 2006: Epidemiologic, Clinical and Molecular Aspects.

In the spring of 2006, four human cases of parapoxvirus infections in Missouri residents were reported to the Centers for Disease Control and Prevention (CDC), two of which were initially diagnosed as cutaneous anthrax. This investigation was conducted to determine the level of recognition of zoonotic parapoxvirus infections and prevention measures, the degree to which veterinarians may be consulted on human infections and what forces were behind this perceived increase in reported infections. Interviews were conducted and clinical and environmental sampling was performed. Swab and scab specim

Effects of coadministration of foot and mouth disease vaccine and inactivated parapoxvirus ovis on humoral immunity in cattle.

Foot and mouth disease (FMD) is an acute viral disease in animals. Inactive parapoxvirus ovis (IPPVO) strengthens humoral immunity. This study aimed to determine the effect of IPPVO application together with FMD vaccine on cattle immunity. It included 30 Holstein cattle randomly divided into two groups: one was administered only the FMD vaccine, and the other was administered the FMD vaccine and IPPVO simultaneously. Control blood was collected from all animals at 0 hours. Serum TNF-α, IL-1β, and IL-6 levels were measured in blood samples collected at 4, 8, 12, 16, and 24 hours post-vaccinatio

Novel immune-modulator identified by a rapid, functional screen of the parapoxvirus ovis (Orf virus) genome.

BACKGROUND: The success of new sequencing technologies and informatic methods for identifying genes has made establishing gene product function a critical rate limiting step in progressing the molecular sciences. We present a method to functionally mine genomes for useful activities in vivo, using an unusual property of a member of the poxvirus family to demonstrate this screening approach. RESULTS: The genome of Parapoxvirus ovis (Orf virus) was sequenced, annotated, and then used to PCR-amplify its open-reading-frames. Employing a cloning-independent protocol, a viral expression-library was

A new rabies vaccine based on a recombinant ORF virus (parapoxvirus) expressing the rabies virus glycoprotein.

The present study describes the generation of a new Orf virus (ORFV) recombinant, D1701-V-RabG, expressing the rabies virus (RABV) glycoprotein that is correctly presented on the surface of infected cells without the need of replication or production of infectious recombinant virus. One single immunization with recombinant ORFV can stimulate high RABV-specific virus-neutralizing antibody (VNA) titers in mice, cats, and dogs, representing all nonpermissive hosts for the ORFV vector. The protective immune response against severe lethal challenge infection was analyzed in detail in mice using dif

Comparison of the Efficacy of Filgrastim and an Inactivated Parapoxvirus ovis Paraimmune Activator in Naturally Infected Cats with Feline Panleukopenia.

Feline panleukopenia (FPL) is a serious viral disease caused by Feline panleukopenia virus (FPV) that causes leukopenia, lymphopenia, and neutropenia, particularly in young or unvaccinated cats. There is no specific antiviral treatment available for FPL, and treatment protocols generally consist of fluid therapy and supportive care. This study evaluated the clinical and hematological efficacy of filgrastim, a granulocyte colony-stimulating factor (G-CSF) that has shown successful results in treating FPL in various studies, and the paraimmune activator-inactivated Parapoxvirus ovis (iPPVO) in 4

First report of a parapoxvirus red deer infection in reindeer (Rangifer tarandus tarandus): clinical presentation and full-genome characterization.

In September 2023, five semi-domesticated reindeer calves (Rangifer tarandus tarandus) from Norrbotten County, Sweden, displayed clinical signs resembling contagious ecthyma. Samples were collected from the lesion sites, including the skin of the muzzle, eyelids, and oral mucosa. The samples were analysed via real-time PCR (qPCR) for parapoxviruses (PPV) and cervidpoxviruses (CvPV). Both viruses were detected in samples from several calves. Full genome sequencing of the PPV strain 23-MIK191411 revealed close similarity to the parapoxvirus red deer (RDPV) strain HL953 from a red deer (Cervus el

An engineered oncolytic parapoxvirus induces immunogenic tumor cell death and displays antitumor efficacy by enhancing T cell responses.

Oncolytic viruses (OVs) is a class of promising cancer biotherapeutics by selectively lysing tumor cells and stimulating antitumor immunity. However, most OV-based clinical efficacy were often restricted by rapid viral clearance mediated by pre-existing neutralizing antibodies and immunosuppressive tumor microenvironment (TME). These barriers particularly limit the efficient activation of T cell-mediated immune responses, thereby compromising durable systemic antitumor effects. Leveraging the distinctive advantage that oncolytic parapoxvirus ovis (ORFV) does not induce neutralizing antibodies

Putative parapoxvirus-associated foot disease in the endangered huemul deer (Hippocamelus bisulcus) in Bernardo O'Higgins National Park, Chile.

The huemul (Hippocamelus bisulcus) is an endangered cervid endemic to southern Argentina and Chile. Here we report foot lesions in 24 huemul from Bernardo O'Higgins National Park, Chile, between 2005 and 2010. Affected deer displayed variably severe clinical signs, including lameness and soft tissue swelling of the limbs proximal to the hoof or in the interdigital space, ulceration of the swollen tissues, and some developed severe proliferative tissue changes that caused various types of abnormal wear, entrapment, and/or displacement of the hooves and/or dewclaws. Animals showed signs of inten

Milkers' nodules complicated by erythema multiforme and graft-versus-host disease after allogeneic hematopoietic stem cell transplantation for multiple myeloma.

We describe a case of cow-transmitted parapoxvirus infection--also known as milkers' nodules--after a hematopoietic stem cell transplantation for multiple myeloma. The infection was complicated by erythema multiforme and acute exacerbation of graft-versus-host disease. Parapoxvirus was confirmed by electron microscopy. The natural history of milker's nodules in immunocompetent hosts is described and compared to that in our immunocompromised patient.

Milker's nodule: an occupational infection and threat to the immunocompromised.

Milker's nodule virus, also called paravaccinia virus, is a DNA virus of the parapoxvirus genus transmitted from infected cows to humans. It results from contact with cattle, cattle by-products or fomites. Classified as an occupational disorder, those at risk of exposure include farmers, butchers and agricultural tourists. The viral infection begins 5-15 days after inoculation as an erythematous-purple, round nodule with a clear depressed centre and a surrounding erythematous ring. While familiar to those in farming communities, the presence of the nodule may be concerning to others, particula

Nutrition as an etiological factor causing diseases in endangered huemul deer.

OBJECTIVES: Distinct diseases prevent endangered huemul deer (Hippocamelus bisulcus) recovery. Fundamental etiological factors include nutriments, a mayor component of habitat quality. Undernutrition affects growth, skeletal development, osteopathology, reproduction and immunocompetence: this paper amplifies data corroborating micro-nutrient deficiencies among huemul. RESULTS: In Argentina, 57% huemul cadavers exhibited osteopathology, with new cases reported here. Recently, 86% live huemul had osteopathology: cranial lesions involved antemortem tooth loss, reducing feeding efficiency and bod

A Case of Orf Disease Complicated with Erythema Multiforme and Bullous Pemphigoid-Like Eruptions.

Parapoxvirus infection in sheep and goats is usually referred to as contagious pustular dermatitis/ecthyma, or orf, and the corresponding human infection is referred to as orf. In humans, after a brief incubation period of 3 to 5 days, lesions begin as pruritic erythematous macules and then rise to form papules, often with a target appearance. Lesions become nodular or vesicular, and orf lesions often ulcerate after 14 to 21 days. Erythema multiforme and bullous pemphigoid have been associated with parapoxvirus infections and they are rare complications of orf disease. In this case report, we

Recognizing Ocular Milker's Nodule: Diagnostic Clues and Clinical Course in Two Dairy Workers.

Milker's nodule is a zoonotic infection caused by the Parapoxvirus (genus Parapoxvirus , family Poxviridae ), typically affecting dairy workers through direct contact with infected cattle. Ocular involvement is rare and usually results from autoinoculation from hand lesions. We report two cases in dairy workers presenting with vesiculoulcerative eyelid lesions and corneal epithelial defects. One patient developed limbitis and stromal keratitis, which resolved completely with topical lubricants, and corticosteroids after reepithelialization. The second case healed spontaneously with conservativ

Parapoxviral vulvovaginitis in Holstein cows.

A group of Holstein first-calved heifers developed small pustules and ulcers on the vulva and in the vagina during the first 1-4 wk postpartum. The lesions varied from small red pinpoint foci to pustules and ulcers, 3-5 mm diameter. Some ulcers coalesced to form large ulcerated areas up to 15 mm diameter. In some animals, these ulcers progressed to become deep ulceration of the vaginal and vulvar mucosa with >50% of the mucosa involved. Vaginal biopsies from 4 heifers and vaginal individual swabs from 8 heifers for a combined sampling of 9 heifers were taken for clinical assessment. Six of the
