[Decrease in the Activity of Striatal-Enriched Protein-Tyrosine-Phosphatase (STEP) in the Brain of Danio rerio Treated with p-Chlorophenylalanine and Pargyline].

Fundamental neurophysiological processes are often studied using Danio rerio fish as a model. A selective inhibitor of striatal-enriched protein tyrosine phosphatase (STEP) reduces serotonin metabolism in the D. rerio brain. Both STEP and serotonin are involved in the development of neurodegenerative behavioral disorders. Reduction or elevation of the serotonin level in the brain of mice caused by the administration of p-chlorophenylalanine or pargyline, respectively, results in a decrease in the level of ptpn5 mRNA in the striatum, ptpn5 being the gene encoding STEP. However, it has not been

Monoaminergic mediation of masculine and feminine copulatory behavior in female rats.

Ovariectomized rats treated with testosterone propionate (TP; 100 mug/kg X 6 days) and para-chlorophenylalanine (pCPA; 100 mg/kg X 3 days), a serotonin synthesis inhibitor, showed more masculine copulatory behavior (including the ejaculatory pattern) than did females receiving either TP or pCPA alone. The facilitatory effect of pCPA on the masculine copulatory behavior in females was not potentiated by pargyline (50 or 100 mg/kg), a monoamine oxidase inhibitor; instead, pargyline antagonized the effect of pCPA. Apomorphine (100 mug/kg), a dopamine receptor stimulant, did not increase masculine

The effects of some neuroleptics and d-amphetamine on striatal 2-phenylethylamine in the mouse.

1. Mouse striatal 2-phenylethylamine was not changed at 2 hr following the administration of chlorpromazine, fluphenazine or spiperone. 2. In contrast, when the mice were first given pargyline (2 mg kg-1), treated with chlorpromazine, fluphenazine or spiperone 2 hr later and killed at 4 hr, a significant increase (to 130-170%) in the accumulation of 2-phenylethylamine was observed with respect to the pargyline controls. 3. The effect of chlorpromazine was consistently observed after pretreatment with either deprenyl (2 mg kg -1) or high doses (200 mg kg-1) of pargyline that produced different

Effects of 6-hydroxydopamine on dopamine and noradrenaline content in dog blood vessels and heart. Evidence for a noradrenaline-independent dopamine pool.

The noradrenaline and dopamine depletion induced by 6-hydroxydopamine (6-OHDA) and pargyline plus 6-OHDA was investigated in the heart, mesenteric, renal, splenic and femoral arteries and saphenous vein of the dog. Catecholamine concentrations in plasma were also analyzed in these two experimental conditions. 6-OHDA and pargyline plus 6-OHDA induced a parallel decrease of the noradrenaline and dopamine content in the main trunk of the mesenteric artery, femoral artery and heart. In the proximal branches of the mesenteric artery, renal and splenic arteries 6-OHDA selectively reduced noradrenali

A systematic microdialysis study of dopamine transmission in the accumbens shell/core and prefrontal cortex after acute antipsychotics.

RATIONALE: The only systematic in vivo studies comparing antipsychotic (AP) effects on nucleus accumbens (NAc) shell and core dopamine (DA) transmission are voltammetric studies performed in pargyline-pretreated, halothane-anaesthetized rats. Studies in freely moving rats not pretreated with pargyline are not available. This study was intended to fill this gap by the use of in vivo microdialysis in freely moving rats. METHODS: Male Sprague-Dawley rats were implanted with microdialysis probes in the NAc shell and core and medial prefrontal cortex (PFCX). The next day, rats were administered in

Cardiac microdialysis of salicylic acid .OH generation on nonenzymatic oxidation by norepinephrine in rat heart.

The effect of pargyline, a monoamine oxidase inhibitor, on the generation of hydroxyl free radicals (.OH) was investigated using cardiac microdialysis. Salicylic acid in Ringer's solution (0.5 nmol x microL(-1) x min(-1)) was infused directly through a microdialysis probe to detect the generation of .OH as reflected by the formation of dihydroxybenzoic acid (DHBA) in the myocardium of anesthetized rats. When pargyline (100 nmol x microL(-1) x min(-1)) was infused in rat heart, the level of norepinephrine (NE) gradually increased in a time-dependent manner and an increase of DHBA was also obser

Effect of endogenous catecholamines in lymphocytes on lymphocyte function.

Our previous work has showed that lymphocytes can synthesize catecholamines (CAs). However, role and mechanism of the endogenous CAs in lymphocytes in modulation of immune function are less known. In the present study, we used alpha-methyl-p-tyrosine (alpha-MT), an inhibitor of tyrosine hydroxylase (TH), and pargyline, an inhibitor of monoamine oxydase, to block the synthesis and degradation of CAs in lymphocytes and then observed changes of lymphocyte proliferation induced by concanavalin A (Con A). Phentolamine and propranolol, antagonists respectively to alpha- and beta-adrenoreceptors, wer

Monoamine oxidase inhibitor-induced blockade of locomotor sensitization to quinpirole: role of striatal dopamine uptake inhibition.

Previous studies have shown that the monoamine oxidase inhibitor (MAOI) clorgyline, blocks locomotor sensitization to the D(2)/D(3) dopamine agonist quinpirole and sensitizes self-directed mouthing behavior in rats by a mechanism independent of MAO inhibition. However, clorgyline is also an inhibitor of striatal dopamine uptake, and this mechanism could account for the effect of clorgyline on quinpirole sensitization. To investigate this possibility, the effects of clorgyline and pargyline were examined. Of these two MAOIs, only clorgyline inhibits dopamine uptake in the striatum. Rats receive

Genome-scale in vivo CRISPR screen identifies RNLS as a target for beta cell protection in type 1 diabetes.

Type 1 diabetes (T1D) is caused by the autoimmune destruction of pancreatic beta cells. Pluripotent stem cells can now be differentiated into beta cells, thus raising the prospect of a cell replacement therapy for T1D. However, autoimmunity would rapidly destroy newly transplanted beta cells. Using a genome-scale CRISPR screen in a mouse model for T1D, we show that deleting RNLS, a genome-wide association study candidate gene for T1D, made beta cells resistant to autoimmune killing. Structure-based modelling identified the U.S. Food and Drug Administration-approved drug pargyline as a potentia

Blockade of corticosterone synthesis reduces serotonin turnover in the dorsal hippocampus of the rat as measured by microdialysis.

The influence of plasma corticosterone concentration on serotonin (5-HT) turnover in the dorsal hippocampus was investigated. The experiments were performed in freely moving male Wistar rats in their home cage. Blood samples were taken via a permanent jugular vein catheter to determine plasma corticosterone levels. Extracellular levels of 5-HT and its metabolite 5-hydroxy-indole acetic acid (5-HIAA) were measured using in vivo microdialysis. The rats received an intravenous (i.v.) infusion of the steroid synthesis-inhibitor metyrapone (150 mg/kg/ml) in order to manipulate circulating corticost

On the use of selective neurotoxic amine analogs to measure the blockade of norepinephrine and 5-hydroxytryptamine uptake systems by antidepressants.

In pargyline or carbidopa pretreated mice, 6-hydroxydopa causes a depletion of brain norepinephrine. Several antidepressant compounds block the depletion after pargyline but not after carbidopa pretreatment. Similarly, p-chloromethamphetamine-induced depletion of brain 5-hydroxytryptamine is blocked by several antidepressants. Diazepam antagonizes the ability to imipramine to block the norepinephrine depletion induced by 6-hydroxydopa but not the 5-hydroxytryptamine depletion induced by p-chloromethamphetamine. Blockade of selective neurotoxic induced depletions of biogenic amines is a useful

Protection and potentiation of MPTP-induced toxicity by cytochrome P-450 inhibitors and inducer: in vitro studies with brain slices.

Exposure to 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP) causes loss of dopaminergic neurons in humans, primates and mice. Exposure of sagittal slices of mouse brain to MPTP (100 pM) caused inhibition of mitochondrial NADH-dehydrogenase activity. Leakage of lactate dehydrogenase from the slice into the medium was observed following incubation of slices with 1 nM MPTP. Neurotoxicity induced by MPTP was prevented by prior exposure of the slices to the dopamine uptake inhibitor GBR 12935. Deprenyl and pargyline (inhibitors of monoamine oxidase), also protected the slices from MPTP-induced

Pharmacological and chemical screening of Byrsonima crassifolia, a medicinal tree from Mexico. Part I.

Leaf and bark extracts of Byrsonima crassifolia displayed concentration-dependent, spasmogenic effects on rat fundus in vitro and biphasic effects on rat jejunum and ileum in vitro. Dose-related in vivo effects in intact rats using hippocratic screening were: decrease in motor activity, mild analgesia, back tonus, enophthalmos, reversible palpebral ptosis, ear blanching, Robichaud positive, catalepsy (awake) and strong hypothermia. Rat fundus in vitro was used as the bioassay to carry out an activity-directed separation. Bioactive material was concentrated in a 2% acetic acid leaf extract (HOA

Harman (1-methyl-beta-carboline) is a natural inhibitor of monoamine oxidase type A in rats.

Harman (1-methyl-beta-carboline) displaces [3H]pargyline in vitro from high affinity binding sites on membranes from cerebral cortex, provided that experimental conditions are chosen under which [3H]pargyline labels selectively monoamine oxidase type A. Norharman (beta-carboline) is a much weaker displacing compound. It is well known that the type A enzyme can be blocked irreversibly in vivo by treatment of rats with clorgyline. Under these conditions no specific binding of [3H]harman and [3H]pargyline to monoamine oxidase type A was detected in brain, whereas the specific binding was reduced

Effect of different photoperiods on the diurnal rhythm of 5-methoxytryptamine in the pineal gland of golden hamsters (Mesocricetus auratus).

This study tested the photo-dependency of the rhythmic synthesis of 5-methoxytryptamine (5-MT) in the pineal gland of golden hamsters. After pargyline administration, pineal 5-methoxytryptamine and melatonin were measured by HPLC in male golden hamsters kept under short and long photoperiod. In both photoperiodic regimes, a clear 5-MT rhythm was observed which fitted a sinusoidal function with high values occurring during the daytime and low values occurring during the night-time. The duration of the low night-time levels was clearly proportional to the length of the dark phase. A marked rhyth

Effects of opiate antagonists on serotonin turnover and on luteinizing hormone and prolactin secretion in estrogen- or morphine-treated rats.

Previous findings indicate that estradiol benzoate (EB) acutely activates serotonergic projections in the medial preoptic area that are stimulatory to prolactin (PRL) secretion. Because opioid agonists also stimulate both PRL release and serotonin (5-HT) turnover, the present experiments tested whether endogenous opioid neurons may mediate estrogen feedback effects. In experiment 1, ovariectomized rats received either 50 micrograms EB or oil vehicle, and either saline or a long-acting opiate receptor blocker, nalmetrene (10 mg/kg), simultaneously, 3 h prior to decapitation.
