Blood donor screening for parvovirus B19.

A programme of blood donor screening for parvovirus B19 was conducted from January to May 1990. The main aim of the study was to identify a B19 positive donation that could be used as a source of viral antigen for diagnostic serology. Out of 24,000 donors tested one was positive for B19 antigen by counter current immunoelectrophoresis and over 100 ml of undiluted B19 containing material was obtained. However, much of the positive donation was incorporated in a plasma pool of 28 donations. An acid dissociation technique was used to recover B19 antigen from immune complexes formed in the plasma

Respiratory tract infection of fatal severe human bocavirus 1 in a 13-month-old child: A case report and literature review.

Human bocavirus 1 (HBoV1) belongs to the family Parvoviridae and it is acknowledged that HBoV1 is a respiratory pathogen. We report the case of a 13-month-old boy who presented with a cough, shortness of breath, and wheezing, and who eventually died of severe pneumonia and acute respiratory distress syndrome (ARDS). Metagenomics next-generation sequencing (mNGS) showed that HBoV1 was the only detected pathogen. The nasopharyngeal aspirate viral load was 2.08 × 1010 copies/ml and the serum viral load was 2.37 × 105 copies/ml. The child was still oxygen deficient under mechanical ventilation. Ch

Parvovirus B19 in Rheumatic Diseases.

Parvovirus B19 (B19V) is a human pathogen belonging to the Parvoviridae family. It is widely diffused in the population and responsible for a wide range of diseases, diverse in pathogenetic mechanisms, clinical course, and severity. B19V infects and replicates in erythroid progenitor cells (EPCs) in the bone marrow leading to their apoptosis. Moreover, it can also infect, in an abortive manner, a wide set of different cell types, normally non-permissive, and modify their normal physiology. Differences in the characteristics of virus-cell interaction may translate into different pathogenetic me

Human bocavirus-the first 5 years.

Four species of human bocavirus (HBoV) have been recently discovered and classified in the Bocavirus genus (family Parvoviridae, subfamily Parvovirinae). Although detected both in respiratory and stool samples worldwide, HBoV1 is predominantly a respiratory pathogen, whereas HBoV2, HBoV3, and HBoV4 have been found mainly in stool. A variety of signs and symptoms have been described in patients with HBoV infection including rhinitis, pharyngitis, cough, dyspnea, wheezing, pneumonia, acute otitis media, fever, nausea, vomiting, and diarrhea. Many of these potential manifestations have not been s

A novel Chaphamaparvovirus detected in breeding Muscovy Ducks with hepatitis.

Since 2022, a decrease in egg production and hatchability, along with hepatitis-like symptoms, has been frequently reported in Muscovy duck farms in southern China. Using metagenomic sequencing, a novel Chaphamaparvovirus (ChPV), designated MuChPV-GD2022, was detected in the livers of the diseased ducks. Phylogenetic analysis revealed that the MuChPV-GD2022 strain shares 61.8-77.4% genome identity with duck-origin Chaphamaparvoviruses, and 44.3-77.4% with avian-origin Chaphamaparvoviruses. The NS1 protein amino acid sequence showed a 29.3-71.5% similarity to those of other known Chaphamaparvov

Acute diarrhea in West African children: diverse enteric viruses and a novel parvovirus genus.

Parvoviruses cause a variety of mild to severe symptoms or asymptomatic infections in humans and animals. During a viral metagenomic analysis of feces from children with acute diarrhea in Burkina Faso, we identified in decreasing prevalence nucleic acids from anelloviruses, dependoviruses, sapoviruses, enteroviruses, bocaviruses, noroviruses, adenoviruses, parechoviruses, rotaviruses, cosavirus, astroviruses, and hepatitis B virus. Sequences from a highly divergent parvovirus, provisionally called bufavirus, were also detected whose NS1 and VP1 proteins showed <39% and <31% identities to those

[Parvovirus B19 infection].

In 1975, during blood screening for hepatitis B, Cossart et al. discovered the human parvovirus B19 (B19). It is a small, single strand DNA virus of the Parvoviridae family. This virus is widespread with 40-80% of adults showing evidence of infection. It is found in the respiratory secretions of viraemic patients and direct contact has been suggested as the most likely mode of transmission. Parenteral transmission is common during treatment with clotting-factor concentrates, but rarely occurs during transfusion with single donor products. Although B19 usually causes a self-limited illness, com

In search of human protoparvovirus acute infections.

Three protoparvoviruses (PPV)-bufavirus, cutavirus, and tusavirus-are the most recent members of the Parvoviridae virus family discovered in humans. All were initially found in stool but have since then been associated with gastroenteritis or cutaneous T-cell lymphoma, or found to be of zoonotic origin, respectively. In the current study, we developed novel PPV IgM enzyme immunoassays (EIA) and aimed to search for and characterize human protoparvovirus acute infections. We also provide a more comprehensive analysis of PPV seroprevalences. We screened, with in-house IgG, IgM, and PCR assays, a

Application of quantitative PCR for quantization of densovirus genome.

Densovirus is classified as invertebrate virus belonging to the subfamily Densovirinae of Parvoviridae family. This group of viruses infects only insects and several densoviruses have been isolated from indigenous mosquitoes and mosquito cell lines. A number of mosquitoes, especially Aedes aegypti and Ae. albopictus are important vectors of viruses, which are the major causes of dengue hemorrhagic fever and yellow fever in humans. As densoviruses do not cause any pathology in humans, these viruses have been proposed to be a potential vector for use in biological control of mosquitoes and insec

Large expansion of plasma commensal viruses is associated with SIV pathogenesis in Macaca leonina.

Human immunodeficiency virus-1 (HIV-1) infection disrupts the homeostatic equilibrium between the host and commensal microbes. However, the dynamic changes of plasma commensal viruses and their role in HIV/simian immunodeficiency virus (SIV) pathogenesis are rarely reported. Here, we investigated the longitudinal changes of plasma virome, inflammation levels, and disease markers using an SIV-infected Macaca leonina model. Large expansions of plasma Anelloviridae, Parvoviridae, Circoviridae and other commensal viruses, and elevated levels of inflammation and D-dimer were observed since the chro

Acral manifestations of viral infections.

Viruses are considered intracellular obligates with a nucleic acid RNA or DNA. They have the ability to encode proteins involved in viral replication and production of the protective coat within the host cells but require host cell ribosomes and mitochondria for translation. The members of the families Herpesviridae, Poxviridae, Papovaviridae, and Picornaviridae are the most commonly known agents for cutaneous viral diseases, but other virus families, such as Adenoviridae, Togaviridae, Parvoviridae, Paramyxoviridae, Flaviviridae, and Hepadnaviridae, can also infect the skin. Viral Metagenomics Reveals Diverse Viruses in the Feces Samples of Raccoon Dogs.

Raccoon dogs as an ancient species of Canidae are the host of many viruses, including rabies virus, canine distemper virus, severe acute respiratory syndrome coronavirus, and so on. With the development of raccoon dog breeding in recent years, some viruses which infected poultry or pigs were also detected from raccoon dogs. At present, the fecal virome of raccoon dogs has been rarely studied. Using an unbiased viral metagenomic approach, we investigated the fecal virome in raccoon dogs collected from one farm of Jilin Province, China. Many DNA or RNA viruses identified in those fecal samples w

Detection of human bocavirus 1 and 2 from children with acute gastroenteritis in Japan.

This study reports the detection and the genetic characterization of HBoV, a new member in the Parvoviridae family. Among 177 fecal specimens collected from children with diarrhea younger than 5 years in Japan, 11 (6.2%) were positive for HBoV. Co-infection with other enteric viruses (norovirus and adenovirus) was found in nine (81.8%) pediatric patients, while monoinfection by HBoV alone was detected in two (18.2%) cases. Nucleotide sequence analysis of 11 Japanese HBoV strains revealed that seven HBoV sequences were most closely related to other HBoV 1 reference strains, while the other four

Electron microscopic identification of parvovirus virions in erythroid-line cells in fatal hydrops fetalis.

Intrauterine infection with parvovirus B19 may lead to fatal hydrops fetalis. Intranuclear particles, consistent with parvovirus virions, within erythroid cells were readily identified on transmission electron microscopy of formalin-preserved material obtained at necropsy from a neonate and two fetuses in whom clinical and light microscopic criteria for parvovirus B19 infection were met. No such particles were seen in similar material from a neonate and two fetuses with erythroblastosis fetalis due to alpha-thalassemia, maternofetal Rh incompatibility, and an erythrocyte membrane protein defec

Human parvovirus B19: general considerations and impact on patients with sickle-cell disease and thalassemia and on blood transfusions.

Human parvovirus B19 (B19V) is a small (22-24 nm) nonenveloped DNA virus belonging to the genus Erythrovirus (family Parvoviridae). Although it generally causes self-limiting conditions in healthy people, B19V infection may have a different outcome in patients with inherited hemolytic anemias. In such high-risk individuals, the high-titer replication may result in bone marrow suppression, triggering a life-threatening drop of hemoglobin values (profound anemia, aplastic crisis). To date there is no consensus concerning a B19V screening program either for the blood donations used in the hemothe

[Erythrovirus B19 infection].

Erythrovirus B19 has been associated with an expanding range of clinical disorders since its identification as the etiological agent of erythema infectiosum, or fifth disease of childhood: acute arthropathy, dermatologic manifestations, chronic anemia in immunocompromised patients, and transient aplastic crisis in individuals with underlying chronic hemolytic disorders. Furthermore, exposure to and infection by B19 virus can lead to serious complications during pregnancy, which may result in fetal anemia, spontaneous abortion, and hydrops fetalis. Consequently, the B19 immune status of pregnan

Serologic and molecular detection of human Parvovirus B19 infection.

Following its identification by Yvonne Cossart in 1975, human Parvovirus B19 has been recognized as the causative agent of a wide range of diseases. In childhood, the most common disease is a typical exanthema called "fifth disease". In adults, viral infection may be responsible for fetal loss and for aplastic anaemia in immuno-compromised patients. Because persistent viral infection may induce an autoimmune response, Parvovirus B19 is emerging as an environmental factor linked to the pathogenesis of autoimmunity. As a result of its expanding disease spectrum, Parvovirus B19 is the subject of

[A family with hereditary spherocytosis discovered after an infection with human parvovirus B19].

Hereditary spherocytosis was diagnosed in five of six children in one family after a human parvovirus B19 (B19-virus) infection. The diagnosis was made on the basis of severe anaemia, demonstrable haemolysis, decreased osmotic fragility and an increased number of spherocytes. Since in the serum of the patients an increased level of IgG and IgM antibodies against B19 virus was detectable two weeks after the crisis it was concluded that the B19-virus infection caused the severe anaemia.
