MzmL, a novel marine derived N-acyl homoserine lactonase from Mesoflavibacter zeaxanthinifaciens that attenuates Pectobacterium carotovorum subsp. carotovorum virulence.

Quorum sensing (QS) is a conserved cell-cell communication mechanism widely distributed in bacteria, and is oftentimes tightly correlated with pathogen virulence. Quorum quenching enzymes, which interfere with QS through degrading the QS signaling molecules, could attenuate virulence instead of killing the pathogens, and thus are less likely to induce drug resistance. Many Gram-negative bacteria produce N-acyl homoserine lactones (AHLs) for interspecies communication. In this study, we isolated and identified a bacterial strain, Mesoflavibacter zeaxanthinifaciens XY-85, from an Onchidium sp. c

Two Novel AHL-Degrading Enzymes, PoAcy1 and PoLac, Attenuate Pectobacterium carotovorum and Pseudomonas aeruginosa Virulence through Quorum-Sensing Disruption.

Ensuring food safety remains a significant challenge due to the persistent contamination and spoilage caused by biofilm-forming and resistant pathogenic bacteria. Quorum quenching presents an ecofriendly control strategy. We identified a novel acylase (PoAcy1) and a broad-spectrum lactonase (PoLac) from marine-derived bacterium Pseudooceanicola onchidii XY-99. Both the XY-99 strain and purified PoLac effectively suppressed Pectobacterium carotovorum subsp. carotovorum (Pcc)-induced soft rot in potato tubers. Heterologous expression of either enzyme in Pseudomonas aeruginosa PAO1 significantly

Development of a multiplex TaqMan qPCR targeting unique genomic regions for the specific and sensitive detection of Pectobacterium species and P. parmentieri.

AIM: The newly defined species Pectobacterium parmentieri has emerged as an aggressive pathogen that causes soft rot and blackleg diseases on potato and has been widely disseminated across the globe, jeopardizing the productivity and potato food safety. The implementation of a fast and accurate detection tool is imperative to control, monitor and prevent further spread of these pathogens. The objective of this work was to develop a specific and sensitive multiplex TaqMan qPCR to detect P. parmentieri and distinguish it from all known Pectobacterium species. A universal internal control was inc

Genomic characteristics of vB_PpaP_PP74, a T7-like Autographivirinae bacteriophage infecting a potato pathogen of the newly proposed species Pectobacterium parmentieri.

Bacteriophage vB_PpaP_PP74 (PP74) is a novel virulent phage that infects members of the species Pectobacterium parmentieri, a newly established species of soft-rot-causing bacteria in the family Pectobacteriaceae, derived from potato-specific Pectobacterium wasabiae. vB_PpaP_PP74 was identified as a member of the family Podoviridae by transmission electron microscopy. The phage has a 39,790-bp dsDNA genome containing 50 open reading frames (ORFs). Because of the absence of genes encoding toxins or lysogeny factors, PP74 may be considered a candidate phage for pathogen biocontrol applications.

Elicitation of induced resistance against Pectobacterium carotovorum and Pseudomonas syringae by specific individual compounds derived from native Korean plant species.

Plants have developed general and specific defense mechanisms for protection against various enemies. Among the general defenses, induced resistance has distinct characteristics, such as broad-spectrum resistance and long-lasting effectiveness. This study evaluated over 500 specific chemical compounds derived from native Korean plant species to determine whether they triggered induced resistance against Pectobacterium carotovorum supsp. carotovorum (Pcc) in tobacco (Nicotiana tabacum) and Pseudomonas syringae pv. tomato (Pst) in Arabidopsis thaliana. Salicylic acid influences the resistance of colored calla lily to soft rot (Pectobacterium carotovorum) via the TGA2-LNC88940-miR528-SOD module.

Soft rot is a major disease affecting the production of colored calla lily, significantly compromising its ornamental, commercial, and market value. In this study, we identified potential long non-coding RNAs (lncRNAs) in colored calla lily (Zantedeschia elliottiana) leaves infected with soft rot via transcriptome sequencing. Among them, long non-coding RNA 88490 (LNC88940) was identified as a differentially expressed lncRNA that was significantly upregulated following soft rot infection. The results revealed that LNC88940 functions as an endogenous target mimic (eTM) for miR528, reducing the

Protein Language Model-Driven Optimisation of Antimicrobial Peptide Pth-Ca1 Against Pectobacterium brasiliense Using ESMFold-Predicted Structures and the ESM-3 Model.

The emergence of antimicrobial resistance (AMR) poses a significant threat to global health and food security. Antimicrobial peptides (AMPs), particularly those characterised by α-helical structures, represent a promising alternative due to their broad-spectrum activity and unique mechanisms of action. Pectobacterium brasiliense is a destructive bacterial pathogen affecting solanaceous crops but effective control measures remain insufficient. This study aims to optimise pseudothionin AMPs using AI-based protein language models, ESM-3 and ESMFold, to enhance their antibacterial efficacy. Using

First Report of Bacterial Soft Rot Disease on Pak Choi (Brassica rapa subsp. chinensis) caused by Pectobacterium brasiliense in the United States.

Pak choi (Brassica rapa subsp. chinensis) is an important vegetable crop native to China, known for high water content and low caloric value, containing high quality of protein, carbohydrates, fiber, vitamins, minerals, and secondary plant metabolites (Acikgoz, 2016). A pak choi field (8,000 sq. ft.) on Oahu, Hawaii, was visited in May 2020. About 10% plants were infected and showed characteristic symptoms of soft rot, wet lesions, macerated infected stem and necrotic leaves (Figure1A-D); leading to the suspect of one of the most devastating bacterial pathogens within genus Pectobacterium (Bol

The Effect of Some Wild Grown Plant Extracts and Essential Oils on Pectobacterium betavasculorum: The Causative Agent of Bacterial Soft Rot and Vascular Wilt of Sugar Beet.

The bacterial soft rot and vascular wilt of sugar beet are the major diseases of sugar crops globally induced by Pectobacterium betavasculorum and P. carotovorum subsp. carotovorum (Pcc). The control of this bacterial disease is a severe problem, and only a few copper-based chemical bactericides are available for this disease. Because of the limitations of chemicals to control plant bacterial pathogens, the essential oils and extracts have been considered one of the best alternative strategies for their control. In this study, twenty-seven essential oils and twenty-nine plant extracts were ext

The type III secreted effector DspE is required early in solanum tuberosum leaf infection by Pectobacterium carotovorum to cause cell death, and requires Wx(3-6)D/E motifs.

Pectobacterium species are enterobacterial plant-pathogens that cause soft rot disease in diverse plant species. Unlike hemi-biotrophic plant pathogenic bacteria, the type III secretion system (T3SS) of Pectobacterium carotovorum subsp. carotovorum (P. carotovorum) appears to secrete only one effector protein, DspE. Previously, we found that the T3SS regulator HrpL and the effector DspE are required for P. carotovorum pathogenesis on leaves. Here, we identified genes up-regulated by HrpL, visualized expression of dspE in leaves, and established that DspE causes host cell death. DspE required i

Combining Asymmetric PCR with PAM-Independent Cas12a Analysis of Single-Stranded Amplicons for On-Site Nucleic Acid Testing.

A biosensing system relying on asymmetric PCR (aPCR) and PAM-independent recognition of single-stranded DNA amplicons by a Cas12a/gRNA complex is adapted for on-site nucleic acid testing (NAT). aPCR was performed on a low-cost, portable, smartphone-controlled thermocycler operating in endpoint mode. Subsequently, a selective visual readout of the aPCR outcome was achieved using a Cas12a-based assay. A mechanism to prevent false positives was incorporated into the aPCR/Cas12a biosensing system by introducing uracil into the amplicons produced during amplification. Pectobacterium species-harmful

Functional study of PtPTA and PtDLO1 in disease resistance of Pinellia ternata to soft rot by an efficient Agrobacterium tumefaciens-mediated transformation system.

Pinellia ternata (Thunb.) Breit, belonging to the genus Pinellia in the Araceae family, is globally distributed. The dried tuber of P. ternata has significant medicinal value in traditional Chinese medicine, with a history of usage spanning approximately 2000 years, particularly for alleviating cough and vomiting. However, soft rot disease caused by Pectobacterium carotovorum poses a severe threat to P. ternata plants. This study screened 44 germplasms and identified the disease-resistant Pinellia ternata P-9 and disease-susceptible Pinellia ternata P-15. A de novo transcriptome analysis was c

Differences in the constituents of bacterial microbiota of soils collected from two fields of diverse potato blackleg and soft rot diseases incidences, a case study.

The presence of bacteria from the Dickeya spp. and Pectobacterium spp. in farmlands leads to global crop losses of over $420 million annually. Since 1982, the scientists have started to suspect that the development of disease symptoms in crops might be inhibited by bacteria present in the soil. Here, we characterized in terms of physicochemical properties and the composition of bacterial soil microbiota two fields differing, on the basis of long-term studies, in the occurrence of Dickeya spp.- and Pectobacterium spp.-triggered infections. Majority, i.e. 17 of the investigated physicochemical f

Purification, Characterization, and Assessment of Antimicrobial Activity and Toxicity of Portulaca elatior Leaf Lectin (PeLL).

Lectins are carbohydrate-binding proteins with several bioactivities, including antimicrobial properties. Portulaca elatior is a species found at Brazilian Caatinga and data on the biochemical composition of this plant are scarce. The present work describes the purification of P. elatior leaf lectin (PeLL) as well as the assessment of its antimicrobial activity and toxicity. PeLL, isolated by chromatography on a chitin column, had native liquid charge and subunit composition evaluated by electrophoresis. Hemagglutinating activity (HA) of PeLL was determined in the presence of carbohydrates or

Pathogenicity of and plant immunity to soft rot pectobacteria.

Soft rot pectobacteria are broad host range enterobacterial pathogens that cause disease on a variety of plant species including the major crop potato. Pectobacteria are aggressive necrotrophs that harbor a large arsenal of plant cell wall-degrading enzymes as their primary virulence determinants. These enzymes together with additional virulence factors are employed to macerate the host tissue and promote host cell death to provide nutrients for the pathogens. In contrast to (hemi)biotrophs such as Pseudomonas, type III secretion systems (T3SS) and T3 effectors do not appear central to pathoge

The rsmS (ybaM) mutation causes bypass suppression of the RsmAB post-transcriptional virulence regulation system in enterobacterial phytopathogens.

Plant cell wall degrading enzymes (PCWDEs) are the primary virulence determinants of soft rotting bacteria such as the potato pathogen, Pectobacterium atrosepticum. The regulation of secondary metabolite (Rsm) system controls production of PCWDEs in response to changing nutrient conditions. This work identified a new suppressor of an rsmB mutation - ECA1172 or rsmS (rsmB suppressor). Mutants defective in rsmB (encoding a small regulatory RNA), show reduced elaboration of the quorum sensing molecule (N-3-oxohexanoyl-homoserine lactone; OHHL) and PCWDEs. However, OHHL and PCWDE production were p

Comparative Analyses of the Order Enterobacterales Support the Importance of Methyl-Accepting Chemotaxis Proteins and ATP-Binding Cassette Transporters in the Pathogenicity of Soft Rot Pectobacteriaceae.

Methyl-accepting chemotaxis proteins (MCPs) are bacterial chemoreceptors that sense environmental and intracellular signals and regulate flagellar rotation through interactions with Che proteins. Some MCPs interact with ATP-binding cassette (ABC) transporters, which import and export various compounds across cell membranes. Using annotated prokaryotic genome assemblies, we assessed the abundance and organization of MCPs and ABC transporters in plant pathogenic Pectobacteriaceae and compared these features with those in other members of the Enterobacterales order, including Escherichia, Yersini

Comparative essentialome analysis of six Pectobacteriaceae strains using the TNSEEK pipeline identifies conserved and strain-specific fitness determinants.

Transposon sequencing (Tn-seq) is a powerful technique for defining the essential genes required for bacterial survival. However, gene essentiality can vary significantly across taxonomic levels, and comparing large Tn-seq datasets from multiple strains presents considerable analytical challenges. To address this, we developed TNSEEK, a fully automated bioinformatics pipeline for the systematic and comparative analysis of Tn-seq experiments. We applied TNSEEK to analyse newly generated data for six soft rot Pectobacteriaceae strains, encompassing species from the Dickeya and Pectobacterium gen
