Effect of the oral intake of probiotic Pediococcus acidilactici BA28 on Helicobacter pylori causing peptic ulcer in C57BL/6 mice models.

Probiotic lactic acid bacteria are being proposed to cure peptic ulcers by reducing colonization of Helicobacter pylori within the stomach mucosa and by eradicating already established infection. In lieu of that, in vitro inhibitory activity of pediocin-producing probiotic Pediococcus acidilactici BA28 was evaluated against H. pylori by growth inhibition assays. Further, chronic gastritis was first induced in two groups of C57BL/6 mice by orogastric inoculation with H. pylori with polyethylene catheter, and probiotic P. acidilactici BA28 was orally administered to study the eradication and cur

Structure-Activity Relationships of the Bacteriocin Bactofencin A and Its Interaction with the Bacterial Membrane.

The antimicrobial peptide bactofencin A is an unmodified non-pediocin-like bacteriocin that inhibits several clinically relevant pathogens, including Listeria monocytogenes and Staphylococcus aureus. Here we report the synthesis and structure-activity relationship studies of bactofencin A and novel analogues thereof. Synthetic bactofencin A was a potent inhibitor of L. monocytogenes (MIC = 8.0 μM) and S. aureus (MIC = 4.0 μM), similar to the bacteriocin produced naturally by Lactobacillus salivarius. Of particular interest is the fact that linear analogues lacking the disulfide bond found in b

Listeria monocytogenes-Can We Reduce or Eliminate It From Food Commodities?

Listeria monocytogenes is a highly virulent foodborne pathogen responsible for listeriosis, a severe infection threatening vulnerable populations such as pregnant women, newborns, the elderly, and immunocompromised individuals. Its resilience, surviving low pH, reduced water activity, and refrigeration, makes it a formidable contaminant in food systems. Traditional control methods include heat treatment, high-pressure processing, irradiation, and acidification, all aimed at reducing bacterial load in ready-to-eat foods. WHO and FAO guidelines emphasize minimizing contamination and growth. Emer

Acidocin A and Acidocin 8912 Belong to a Distinct Subfamily of Class II Bacteriocins with a Broad Spectrum of Antimicrobial Activity.

Within class II bacteriocins, we assume the presence of a separate subfamily of antimicrobial peptides possessing a broad spectrum of antimicrobial activity. Although these peptides are structurally related to the subclass IIa (pediocin-like) bacteriocins, they have significant differences in biological activities and, probably, a mechanism of their antimicrobial action. A representative of this subfamily is acidocin A from Lactobacillus acidophilus TK9201. We discovered the similarity between acidocin A and acidocin 8912 from Lactobacillus acidophilus TK8912 when analyzing plasmids from lacti

Evaluation of antibacterial properties of lactic acid bacteria from traditionally and industrially produced fermented sausages from Germany.

With regards to the frequently reported findings of spoilage bacteria and pathogens in various foods there is a need to explore new ways to control hazards in food production and to improve consumer safety. Fermented sausages from traditional and industrial production in Germany were screened for lactic acid bacteria with antibacterial effects towards important foodborne pathogens (Escherichia coli DSM 1103, Listeria innocua DSM 20649, Listeria monocytogenes DSM 19094, Pseudomonas aeruginosa DSM 939, Staphylococcus aureus DSM 799 and Salmonella Typhimurium DSM 19587). The obtained isolates and

Plant-based fermented foods harbor autochthonous lactic acid bacteria with broad antimicrobial activity, bacteriocin diversity, and probiotic potential.

Plant-based fermented foods are increasingly recognized as an underexplored niche for autochthonous and beneficial lactic acid bacteria (LAB) with multiple biotechnological and functional potential in health and agrifood systems. We isolated LAB from plant-based fermented foods and assessed their antimicrobial, functional, and probiotic potential using agar-well diffusion technique, in vitro colonization resistance and gastric-environment survivability assays, and 16S rRNA sequencing. Thirteen autochthonous LAB identified as Lacticaseibacillus paracasei, Lactiplantibacillus plantarum, Leuconos

Comprehensive Phenotypic and Genomic Study for Understanding the Variability in Susceptibility of Listeria monocytogenes to AMPs and Bacteriocins.

In this study, we investigated the inhibitory activity of six antimicrobial peptides against 40 Listeria monocytogenes isolates from clinical, environmental, or food samples. Antibiotic susceptibility assays revealed high resistance to clindamycin, whereas resistance to ciprofloxacin was less frequent. Additionally, four isolates were resistant to at least one or two antibiotics. Virulome analysis revealed that the prfA-virulence gene cluster, the main pathogenicity determinant marker in Listeria monocytogenes (prfA, plcA, plcB, hly, mpl and actA) was detected in selected strains. The anti-lis

Double-Edged Nanobiotic Platform with Protean Functionality: Leveraging the Synergistic Antibacterial Activity of a Food-Grade Peptide to Mitigate Multidrug-Resistant Bacterial Pathogens.

While persistent efforts are being made to develop a novel arsenal against bacterial pathogens, the development of such materials remains a formidable challenge. One such strategy is to develop a multimodel antibacterial agent which will synergistically combat bacterial pathogens, including multidrug-resistant bacteria. Herein, we used pediocin, a class IIa bacteriocin, to decorate Ag° and developed a double-edged nanoplatform (Pd-SNPs) that inherits intrinsic properties of both antibacterial moieties, which engenders strikingly high antibacterial potency against a broad spectrum of bacterial

Genomic characterization and biological activities of cell-free supernatant from Lactiplantibacillus pentosus CROT01 against clarithromycin-resistant Helicobacter pylori.

Helicobacter pylori is a major gastric pathogen associated with chronic gastritis, peptic ulcers, and gastric cancer, while increasing antibiotic resistance has reduced the effectiveness of standard eradication therapies. This study investigated the antibacterial, antibiofilm, anti-inflammatory, probiotic-related, and genomic properties of cell-free supernatant (LCFS) derived from Lactiplantibacillus pentosus CROT01 against H. pylori. LCFS exhibited inhibitory activity against all tested H. pylori strains, including the clarithromycin-resistant strain ATCC 700684, with inhibition zones ranging

Modulation of Early-Life Calf Gut Microbiota by Bacteriocin-Producing Lactiplantibacillus pentosus AA86 and Pediococcus pentosaceus AA94.

Early-life gut microbiota plays a critical role in livestock health and productivity. Probiotic interventions, particularly those employing bacteriocin-producing lactic acid bacteria (LAB), may contribute to modulation of microbial development during this critical period. This study evaluated Lactiplantibacillus pentosus AA86 (formerly Lactobacillus pentosus) and Pediococcus pentosaceus AA94 for genomic safety, bacteriocin potential, and their effects on calf gut microbiota. Whole-genome sequencing assessed antimicrobial resistance genes, virulence determinants, plasmid-associated elements, an

Unlocking the potential of tumor-targeting peptides in precision oncology.

Targeted cancer therapy has emerged as a promising alternative to conventional chemotherapy, which is often plagued by poor selectivity, off-target effects, and drug resistance. Among the various targeting agents in development, peptides stand out for their unique advantages, including minimal immunogenicity, high tissue penetration, and ease of modification. Their small size, specificity, and flexibility allow them to target cancer cells while minimizing damage to healthy tissue selectively. Peptide-based therapies have shown great potential in enhancing the efficacy of drug delivery, improvi
