Mechanistic insights into ochratoxin A biodetoxification by Pediococcus acidilactici NX8 involving a multi-hydrolase toolbox.

This study aimed to develop efficient and environmentally friendly detoxification strategies for ochratoxin A (OTA). Pediococcus acidilactici NX8, isolated from traditional fermented dairy products, completely degraded 1 μg/mL OTA within 24 h. By integrating degradation pathway analysis and multi-omics approaches, the molecular mechanism underlying OTA degradation was elucidated, and three novel OTA-hydrolyzing enzymes (XlyAB, AmiE, and Pbp4b) were identified. The recombinant variants completely degraded 2 μg/mL OTA within 5 min and remained stable at pH 4-7 and 4-50 °C. Molecular simulations

Comprehensive safety assessment and therapeutic potential of Pediococcus acidilactici NMCC-B in attenuating arthritis progression.

Dysbiosis of gut microbiota and loss of gut-barrier integrity contribute to the development and severity of rheumatoid arthritis (RA). The available treatments pose a burden of major adverse effects and new treatment strategies are therefore the need of time. In this study, Pediococcus acidilactici NMCC-B (Probiotic) was evaluated for its safety and efficacy in complete Freund's adjuvant (CFA)-induced mice model of RA. Mice were treated with either Escherichia coli (1 × 109 CFU/ml) or P. acidilactici NMCC-B (1 × 109 CFU/ml, 2 × 109 CFU/ml) to assess acute, sub-acute, and chronic toxicities. In

Aggregation, adhesion and efficacy studies of probiotic candidate Pediococcus acidilactici NCDC 252: a strain of dairy origin.

Aggregation and adhesion capability and survival efficacy of candidate probiotic strain Pediococcus acidilactici NCDC 252 under simulated gastric, intestinal and vaginal conditions was studied. The strain exhibited strong autoaggregation phenotype and coaggregation with other Lactic acid bacteria (LAB) and E. coli. The adhesion studies of NCDC 252 to pig's intestinal epithelial cells showed its adhesive ability. Aggregation and adhesiveness were related through cell surface proteins as removal/extraction of surface proteins resulted in altered aggregation and no adhesiveness. Cell surface prot

In vitro and in vivo anti-clostridial activity of newly isolated Pediococcus acidilactici SPM138 against Clostridium difficile.

Clostridium difficile infection (CDI) has become a growing health concern, as evident from the increase in the mortality rate among elderly or hospitalized patients. Treatment of CDI is usually based on antibiotics (metronidazole and vancomycin), but it has some limitations, including cost and antibiotic resistance. Probiotics could offer an effective remedy to prevent CDI and could be an auxiliary agent in treatment CDI. Here, the anti-clostridial activity of a newly isolated probiotic strain, Pediococcus acidilactici SPM138 (SPM138) was evaluated. The cultivation of C. difficile (CD) with SP

Investigation of the Effects of Postbiotics Obtained from Pediococcus acidilactici on Specific Biomarker Expressions in Intestinal Tissue.

The intestinal mucosal barrier is a layered structure comprising fundamental components that play important roles in regulating paracellular permeability. Disruption of intestinal barrier homeostasis predisposes to infections, mucosal damage, and metabolic and allergic diseases. To provide protection against potential damage to the intestinal mucosa, agents such as prebiotics and probiotics are recommended due to their ability to secrete components and metabolites (e.g., bacteriocins, organic acids, enzymes) that can exert beneficial biological effects. The aim of this study is to comprehensiv

In vivo study on the effectiveness of pediocin PA-1 and Pediococcus acidilactici UL5 at inhibiting Listeria monocytogenes.

The anti-listerial effect of pediocin PA-1 and its producing strain, Pediococcus acidilactici UL5, was investigated in vivo using an ICR mouse model. The effect of intra-gastric administration of a single dose of P. acidilactici UL5 (4 x 10(10) CFU/animal) on the propagation of Listeria monocytogenes LSD348 in intestine, liver and spleen was negligible. P. acidilactici UL5 did not appear competitive with the mouse intestinal flora and was not detectable in fecal samples collected two days after administration. However, double-agar-layer activity assay showed the ability of P. acidilactici UL5

Selenium-Enriched Pediococcus acidilactici MRS-7 Alleviates Patulin-Induced Jejunum Injuries in Mice and Its Possible Mechanisms.

Patulin (PAT) is a common mycotoxin. Oral ingestion of PAT could damage the intestinal mucosa. Both selenium and probiotics can alleviate intestinal damage, but there are few reports on selenium-enriched probiotics. Here, we studied the protective effects of a new selenium-enriched Pediococcus acidilactici MRS-7 (SeP) on PAT-induced jejunum injuries in mice. Results show that PAT induced jejunum injuries such as loss of crypts, ulceration of the mucosa, and intestinal epithelial barrier function impairment. However, SeP could protect against PAT-induced jejunum injuries and significantly inhib
