Bacteriocin activity against various pathogens produced by Pediococcus pentosaceus VJ13 isolated from Idly batter.

Bacteriocins, an antimicrobial peptide, is known to have wide spectrum antimicrobial activity against various pathogens. Because they are easily digested in the intestine, they are considered as safe and are widely used as food preservatives. Hence their purification and characterization have attracted considerable attraction, especially for those having activity against human pathogens. In this study, the bacteriocin produced by Pediococcus pentosaceus VJ13 was precipitated with cold acetone and purified by gel permeation chromatography and hydrophobic interaction chromatography. The bacterio

Effects of a Bacterial Hay Preservative (Pediococcus pentosaceus) on Hay under Experimental Storage Conditions.

The abundant growth of molds and thermophilic actinomycetes in stored hay decreases its quality and can be hazardous for the producer who inhales these contaminants when the moldy hay is fed in closed barns. These microbes are responsible for a respiratory disease called farmer's lung. Products, including bacterial cultures that can be inoculated in hay, are available to prevent hay deterioration by molds and bacteria. The aim of this study was to verify the effectiveness of Pediococcus pentosaceus (a bacterial inoculant) in preventing hay deterioration at different humidity levels in a labora

Influence of Lactobacillus plantarum and Pediococcus pentosaceus addition on the nutritional quality of Sourdough-based bread.

This study explored the nutritional benefits of using a mixed culture of Lactobacillus plantarum and Pediococcus pentosaceus in sourdough bread fermentation. Over a 120-h fermentation period, spontaneous microbial activity resulted in peak lactic acid bacteria populations of 516.29 × 106 cfu/g at 72 h in acha-based dough and 1082.78 × 106 cfu/g at 24 h in rice-based dough (P < .001). The fermentation process led to a steady reduction in pH alongside a rise in titratable acidity. Bread produced with the mixed LAB cultures showed a significant decrease in phytic acid content and a notable enhanc

Impact of encapsulating a probiotic (Pediococcus pentosaceus Li05) within gastro-responsive microgels on Clostridium difficile infections.

Antibiotic treatment is often followed by Clostridium difficile infection (CDI), which causes severe diarrhea and other health issues. Oral administration of Pediococcus pentosaceus Li05 (Li05) has been shown to have great potential in preventing CDI. However, the viability of Li05 is greatly reduced during storage and passage through the gastrointestinal (GI) tract, which limits its biological activity. In this study, a gastro-responsive microgel was designed to encapsulate and protect Li05 to enhance its efficacy against CDI. The viability of Li05 encapsulated within the microgels was signif

Novel bacteriocinogenic Enterococcus hirae and Pediococcus pentosaceus strains with antilisterial activity isolated from Brazilian artisanal cheese.

We isolated and characterized bacteriocin producers Enterococcus hirae ST57ACC and Pediococcus pentosaceus ST65ACC from raw milk artisanal cheeses. Their bacteriocins were tolerant to temperatures from 4°C to 100°C and under sterilization conditions (121°C for 15 min). Additionally, the tested bacteriocins remained active after being exposed to pH 2.0 to 10.0 for 2 h. The activity of the bacteriocins was affected by proteolytic enzymes but remained stable after treatment with EDTA, sodium dodecyl sulfate, NaCl, skim milk, and Tween 80. Cell-free supernatants were capable of inhibiting Listeria

Pediococcus pentosaceus DF29-derived extracellular vesicles ameliorate EPEC-induced intestinal injury via M2 macrophage polarization.

Enteropathogenic Escherichia coli (EPEC) infection remains a major cause of intestinal barrier dysfunction and diarrhea, necessitating the development of novel non-antibiotic therapies. While Pediococcus pentosaceus (P. pentosaceus) exhibits probiotic potential, the specific role of its postbiotic components, particularly extracellular vesicles (EVs), in host-pathogen interactions remains obscure. In this study, we systematically evaluated the protective efficacy and underlying mechanisms of P. pentosaceus DF29 (DF29) and its derived EVs (PpEVs) against EPEC-induced enteritis. Characterization

Trehalose improves cold tolerance of Pediococcus pentosaceus OL77 and enhances low-temperature oat silage fermentation.

Low temperature remains a major bottleneck in silage fermentation, especially in cold regions and high-altitude areas. This study investigated the regulatory role of compatible solutes in enhancing the cold adaptation of Pediococcus pentosaceus OL77 and their synergistic application in oat silage under suboptimal temperatures (5°C-15°C). RT-qPCR analysis showed that expression of the cold shock protein gene CspP was sharply induced at 5°C, indicating its central role in cold-stress response. Exogenous trehalose and betaine significantly downregulated CspP expression, promoted bacterial growth,

Oral supplementation of Pediococcus pentosaceus SMM847 ameliorates hyperuricemia in rats and alcohol-induced injury in mice.

UNLABELLED: Excessive dietary purine intake can lead to hyperuricemia, facilitating the precipitation and deposition of monosodium urate crystals in joints, which subsequently triggers inflammatory cascades leading to gout. Alcohol consumption exacerbates this condition by accelerating purine metabolism and impairing renal uric acid excretion, thereby further increasing serum uric acid concentrations and aggravating gout severity. In this study, Pediococcus pentosaceus SMM847 was screened based on its inosine degradation efficiency. Notably, the strain SMM847 also exhibited alcohol dehydrogena

Construction of a co-reaction system for ethanol-promoted gamma-aminobutyric acid synthesis by Pediococcus pentosaceus.

The biotransformation of L-glutamic acid (L-Glu) to γ-aminobutyric acid (GABA) using glutamate decarboxylase (GAD) in microbial whole cells represents an ideal approach for biosynthesis of food-grade and pharmaceutical-grade GABA. To overcome the cell membrane barrier, enhance mass transfer efficiency in the whole-cell reaction system, and improve GABA biosynthesis efficiency, we established a novel ethanol-enhanced whole-cell biocatalytic co-reaction system through systematic investigations on the enzymatic characteristics of GAD in Pediococcus pentosaceus whole cells and the regulatory effec

Inoculation With Pediococcus pentosaceus Alters Microbial Interactions, Metabolic Landscapes, and Quality Traits in Spontaneously Fermented Carrot Pickles.

Spontaneous carrot pickling often results in inconsistent product quality, prolonged fermentation, and potential safety risks due to the uncontrolled activity of indigenous microbiota. In this study, Pediococcus pentosaceus CLB16, exhibiting potential for acid and salt tolerance and antibacterial activity, was evaluated as a starter culture for carrot pickle fermentation. Inoculation with CLB16 significantly decreased pH, increased lactic acid production, and reduced nitrite levels compared with natural fermentation. The inoculated group also showed better color retention, moderate and control

Nitrite stress acclimation of Pediococcus pentosaceus improves nitrite degradation in fermented sausages: from phenotype to transcriptomic mechanism.

Nitrite is a widely used additive in processed meat products, but its excessive accumulation raises serious safety concerns due to the formation of carcinogenic N-nitrosamines. Lactic acid bacteria as starter cultures have been explored for nitrite removal, but their efficiency is often limited under nitrite stress. In this study, to improve nitrite degradation performance, stepwise adaptive evolution of Pediococcus pentosaceus (P. pentosaceus) and Leuconostoc mesenteroides (L. mesenteroides) under nitrite stress was conducted followed by nitrite removal test via in vitro and in fermented saus

Pediococcus pentosaceus ZJUAF-4 relieves oxidative stress and restores the gut microbiota in diquat-induced intestinal injury.

Lactic acid bacteria (LAB) play a key role in promoting health and preventing diseases because of their beneficial effects, such as antimicrobial activities, modulating immune responses, maintaining the gut epithelial barrier and antioxidant capacity. However, the mechanisms with which LAB relieve oxidative stress and intestinal injury induced by diquat in vivo are poorly understood. In the present study, Pediococcus pentosaceus ZJUAF-4 (LAB, ZJUAF-4), a selected probiotics strain with strong antioxidant capacities, was appointed to evaluate the efficiency against oxidative stress in diquat-in

Optimized medium via statistical approach enhanced threonine production by Pediococcus pentosaceus TL-3 isolated from Malaysian food.

BACKGROUND: Threonine is an essential amino acid that is extensively used in livestock industry as feed supplement due to its pronounced effect in improving the growth performance of animals. Application of genetically engineered bacteria for amino acid production has its share of controversies after eosinophils myalgia syndrome outbreak in 1980s. This has urged for continuous search for a food grade producer as a safer alternative for industrial amino acid production. Lactic acid bacteria (LAB) appear as an exceptional candidate owing to their non-pathogenic nature and reputation of Generally
