Pentachlorophenol (PCP) is a possible human carcinogen detected widely in the environment. A quinone metabolite of PCP, tetrachloro-1,4-benzoquinone (Cl4BQ), is a reactive electrophile with the capacity to damage DNA by forming bulky covalent DNA adducts. These quinone adducts may contribute to chlorophenol carcinogenesis, but their structures, occurrence, and biological consequences are not known. Previous studies have indicated that several DNA adducts are formed in vivo in rats exposed to Cl4BQ, but these adducts were not identified structurally. In the present study, we have elucidated the

OBJECTIVE: The objective of this study is to assess the carcinogenicity of pentachlorophenol and tetrachlorophenol using data from the BC sawmill workers cohort study.
METHODS: The cohort consisted of 27,464 men employed by 14 sawmills for 1 year or more between 1950 and 1995. Fatal (1950-1995) and incident (1969-1995) cancers were identified using national registries. Plant records and systematic interviews with senior employees were used to estimate dermal exposure. Comparisons were made with the general BC population and dose-response relationships were assessed using Poisson regression.

Pentachlorophenol (PCP) and its sodium salt are frequently used in wood preservatives. Little is known about the effects on man when being chronically exposed. Only vague skin symptoms, such as rashes, acne and cutaneous infections were described. We present two cases of pemphigus vulgaris with a known non-occupational chronic PCP exposure. The clinical course and the titer of pemphigus antibodies roughly correlate with the PCP levels in serum. In one case of chronic urticaria the exacerbations also run parallel to the PCP serum levels and increased anti-skin antibodies, without any manifestat

A cohort of 2122 US pentachlorophenol (PCP) production workers from four plants in the National Institute for Occupational Safety and Health Dioxin Registry was exposed to PCP and to polychlorinated dibenzo-p-dioxin and dibenzofuran contaminants of PCP production. A subcohort of 720 was also exposed to 2,3,7,8-tetrachlorodibenzodioxin, a contaminant of trichlorophenol (TCP) while using TCP or a TCP derivative. PCP and several production contaminants have been implicated as animal carcinogens. A priori hypotheses were that the cohort would have elevated standardized mortality ratios (SMRs) for

A microbial consortium was developed by continuous enrichment of bacterial population isolated from sediment core of pulp and paper mill effluent in mineral salts medium (MSM) supplemented with pentachlorophenol (PCP) as sole source of carbon and energy in the chemostat. The enriched consortium contained three bacterial strains identified as Escherichia coli (PCP1), Pseudomonas aeruginosa (PCP2) and Acinetobacter sp. (PCP3) by morphological and biochemical tests, further confirmation was done using 16S rDNA sequence analysis. The potency of bacterial isolates in degradation of PCP was monitore

This paper presents a study of the uptake of 2,4,6-tribromophenol (TBP), pentachlorophenol (PCP), and its metabolite pentachloroanisole (PCA) from contaminated sawdust from the forest industry in horticultural products such as apples, raspberries, and fodder maize for cattle feed. The samples were obtained from Bio-Bio Province in South Chile between 2002 and 2006. The analytical parameters of the methodology applied to the different matrices are presented and discussed. The chromatographic method was applied to determine the residues in 413 horticultural product samples. Eleven per cent of fo

A novel approach for the remediation of upland soils contaminated with pentachlorophenol (C6HCl5O; PCP) (1), a fungicide, wood perservative, and herbicide, through the exploitation of plant-endophytic bacteria may overcome the existing issues in bioaugmentaion and phytoremidiation. In this study, we isolated the endophytic Bacillus sp. strain PCP15 and determined its metabolite of PCP (1). This strain degraded 8.03 μmol L-1 PCP (1) within 24 h and generated the novel metabolite PCP phosphate (3). The PCP15 strain showed nearly complete growth inhibition of 20 μmol L-1 PCP (1). In contrast, PCP

Hexachlorobenzene is highly persistent, bioaccumulative, toxic and globally distributed, a model persistent organic pollutant. The major atmospheric removal process for hexachlorobenzene is its oxidation by hydroxyl radicals. Unfortunately, there is no information on the reaction mechanism of this important atmospheric process and the respective degradation rates were measured in a narrow temperature range not of environmental relevance. Thus, the geometries and energies of all stationary points significant for the atmospheric oxidation of hexachlorobenzene are optimized using MP2/6-311G(d,p)

Pentachlorophenol (PCP) was, and still is, one of the most frequently used fungicides and pesticides. The use of PCP has been more restricted during the last few years. PCP's most important industrial application is as a wood preservative. The pentachlorophenol can be absorbed into the body by all the routes of occupational exposure. Some epidemiological observations suggest that exposure to PCP solutions may result in an increased risk for certain diseases in humans, e.g., immunodeficiency, blood disorders, malignancies, congenital anomalies. Chronic poisoning is difficult to detect since sym

Mortality in a cohort of 770 workers with potential pentachlorophenol (PCP) exposure was evaluated from 1940 through 1989. The study cohort is a subset of a larger cohort of workers with potential exposure to higher chlorinated dioxins. Total mortality and cancer mortality in the PCP cohort were slightly lower than expected in comparison to the U.S. white male population. There were 229 total deaths with 242.5 expected (SMR = 94, 95% confidence interval 83-108), and 50 cancer deaths with 52.6 expected (SMR = 95, 95% confidence interval 71-125). In comparison with unexposed employees, the risk

The analyses of low pentachlorophenol (PCP) in soils and earthworms require a sensitive and reliable analytical method. In this paper, several derivatization methods and extraction solvents were compared systematically. The derivatization reagents included acetic anhydride, 2,3,4,5,6-pentafluorobenzyl bromide (PFBBr) and diazomethane. Hexane, acetone, hexane-acetone (1:1), dichloromethane and methanol were used as the extraction solvents. PFBBr derivatization showed the highest sensitivity. The derivatization parameters of PFBBr including the amount of PFBBr, the power and irradiation time of

On incubation of [14C]-hexachlorobenzene with microsomes from livers of rats induced with hexachlorobenzene, the major product (80-90%) was pentachlorophenol. The only other detectable metabolite, tetrachlorohydroquinone (4-15%), was presumably formed from pentachlorophenol. A considerable amount of radioactivity (5-10% of the amount of extracted metabolites) was covalently bound to protein. Microsomes derived from male hexachlorobenzene--induced rats gave by far the highest conversion (approx. 1% of substrate). Microsomes from female hexachlorobenzene--induced rats were 3 times less efficient

Pentachlorophenol (PCP) is a persistent environmental contaminant, but its effect on bladder cancer invasiveness is unclear. Here, we show that environmentally relevant PCP concentrations enhance invasion of T24 and SW780 bladder cancer cells, with limited effects on migration or proliferation. Integrative target prediction, transcriptomics, and machine learning identified EGFR, MAP2K1, and NFE2L2 as candidate contributors. PCP altered the protein stability of these factors, and EGFR or MAP2K1 knockdown or NFE2L2 overexpression partially attenuated PCP-induced invasion. A three-gene signature

A pentachlorophenol (PCP)-degrading bacterium was isolated from possible PCP-contaminated soil from Pusan, Korea and identified as a member of the genus Pseudomonas. It used PCP as its sole source of carbon and energy. This micro-organism was capable of degrading PCP more effectively, certified by the increase in cell density and the decrease in PCP substrate. Pseudomonas sp. Bu34 was able to degrade a much higher concentration of PCP (4000 mg l-1) than any previously reported PCP-degrading bacteria and fungi and to grow in mineral salts solution containing one of a variety of chlorophenols. Incubation of the pentachlorophenol (PCP) metabolites, tetrachloro-p-benzoquinone (chloranil, TCpBQ), tetrachloro-p-hydroquinone (TCpHQ) and tetrachloro-p-benzoquinone (TCoBQ) with V79 Chinese hamster cells led to a significant enhancement of the amount of 8-hydroxydeoxyguanosine (8-OH-dG) in DNA. With PCP itself and the metabolite tetrachloro-o-hydroquinone (TCoHQ) no distinct induction of this lesion could be observed. The average yields of 8-OH-dG were about 2-2.5 times above background levels. In addition, TCpBQ and TCpHQ were able to generate DNA single-strand breaks, while PCP, TCoHQ and

Chromosome analyses were carried out on peripheral lymphocytes from 22 male workers employed at a pentachlorophenol (PCP) producing factory. As compared with a group of 22 matched controls a small, but significant, increase in the frequency of dicentrics and acentrics was observed. There was no significant increase of sister-chromatid exchange (SCEs) in smoking PCP workers, as compared with smoking controls. Within the control group, smokers had a higher incidence of SCEs than non-smokers.

Electrochemical degradation of pentachlorophenol (PCP) in aqueous solution was investigated over Ti/SnO2-Sb electrodes prepared by sol-gel method. X-ray diffraction (XRD), scanning electron microscopy (SEM) and electrochemical measurements were used to characterize the physicochemical properties of the electrodes. The electrochemical degradation of PCP followed pseudo-first-order kinetics. The main influencing factors, including the types of supporting electrolyte (i.e., NaClO4, Na2SO4, Na2SO3, NaNO3, and NaNO2), initial concentrations of PCP (5-1000mgL(-1)), pH values (3.0-11.0), and current

Microbes in contaminated environments often evolve new metabolic pathways for detoxification or degradation of pollutants. In some cases, intermediates in newly evolved pathways are more toxic than the initial compound. The initial step in the degradation of pentachlorophenol by Sphingobium chlorophenolicum generates a particularly reactive intermediate; tetrachlorobenzoquinone (TCBQ) is a potent alkylating agent that reacts with cellular thiols at a diffusion-controlled rate. TCBQ reductase (PcpD), an FMN- and NADH-dependent reductase, catalyzes the reduction of TCBQ to tetrachlorohydroquinon

A collection of 1056 strains in our laboratory were incubated with various xenobiotics among which were two potent fungicides: pentachlorophenol (PCP) and pentachloronitrobenzene (PCNB). The production of extracellular phenoloxidases were examined, using a series of ten different reagents. On the whole, PCNB is less accessible to fungal degradation than PCP. Although no correlation was found between the biodegradative capability of individual fungal strains for PCP or for PCNB, when taxonomic groups were considered as a whole, the same activity profiles were found. Zygomycetes were the most ef
