Addressing MRSA infection and antibacterial resistance with peptoid polymers.

Methicillin-Resistant Staphylococcus aureus (MRSA) induced infection calls for antibacterial agents that are not prone to antimicrobial resistance. We prepare protease-resistant peptoid polymers with variable C-terminal functional groups using a ring-opening polymerization of N-substituted N-carboxyanhydrides (NNCA), which can provide peptoid polymers easily from the one-pot synthesis. We study the optimal polymer that displays effective activity against MRSA planktonic and persister cells, effective eradication of highly antibiotic-resistant MRSA biofilms, and potent anti-infectious performan

Toward a clinical antifungal peptoid: Investigations into the therapeutic potential of AEC5.

Cryptococcus neoformans is a fungal pathogen that causes cryptococcal meningitis in immunocompromised individuals. Existing antifungal treatment plans have high mammalian toxicity and increasing drug resistance, demonstrating the dire need for new, nontoxic therapeutics. Antimicrobial peptoids are one alternative to combat this issue. Our lab has recently identified a tripeptoid, AEC5, with promising efficacy and selectivity against C. neoformans. Here, we report studies into the broad-spectrum efficacy, killing kinetics, mechanism of action, in vivo half-life, and subchronic toxicity of this

Modification of receptor selectivity and functional activity in cholecystokinin peptoid ligands.

Hybrid analogs of the cholecystokinin A (CCK-A) receptor selective tetrapeptide agonist Boc-Trp-Lys(Tac)-Asp-MePhe-NH2 (1,A-71623) and the CCK-B receptor selective antagonists PD-135118 (2) and CI-988 (3) were prepared. Incorporation of the Lys(Tac) side chain into 2 produced a moderately potent antagonist of CCK-8 in the isolated guinea pig gallbladder (GPGB). Incorporation of the Lys(Tac) side chain into 3 produced the novel agonist analog 7 (EC50 = 28 nM in the GPGB) with excellent affinity for both human CCK-A (IC50 = 12 nM) and CCK-B (IC50 = 17 nM) receptors. Analog 7 was a full agonist (

Nicotinamide-Loaded Peptoid Nanotubes for Energy Regeneration in Acute Brain Injury.

Acute brain injuries such as perinatal asphyxia, stroke, and traumatic brain injury result in ischemia, oxidative stress, excitotoxicity, and inflammation, leading to a depletion of ATP. Nicotinamide adenine dinucleotide (NAD+) is crucial for ATP regeneration and DNA repair during postinjury recovery. However, the therapeutic benefits of NAD+ and its precursors, such as nicotinamide (NAM), are limited by challenges in achieving effective cell-specific intracellular delivery. In this study, we use a nanopeptoid delivery strategy to replenish the cellular redox state and increase energy producti

NMR-Guided Studies to Establish the Binding Interaction between a Peptoid and Protein.

Ligand discovery of nonenzymatic proteins can be accomplished through screening methods utilizing libraries comprising small molecules, peptides, and peptidomimetics. Incorporating peptoids, which are oligomers of N-substituted glycine monomers, into high-throughput screens can produce libraries of large structural diversity. Due to their malleable structures, peptoids can occupy unique protein binding sites, but determination of the peptoid binding pose is challenging. For example, the peptoid KDT-11 is reported to bind with low micromolar binding affinity to the proteasome subunit Rpn-13. Po

Selective chromium(VI) ligands identified using combinatorial peptoid libraries.

Hexavalent chromium [Cr(VI)] is a worldwide water contaminant that is currently without cost-effective and efficient remediation strategies. This is in part due to a lack of ligands that can bind it amid an excess of innocuous ions in aqueous solution. We present herein the design and application of a peptoid-based library of ligand candidates for toxic metal ions. A selective screening process was used to identify members of the library that can bind to Cr(VI) species at neutral pH and in the presence of a large excess of spectator ions. There were 11 sequences identified, and their affinitie

New Treatment Approach Using Peptoid-Based Vaccines in Parkinson's Disease.

As the population ages, the prevalence of neurodegenerative disease, such as Parkinson's disease (PD) continues to grow and new and alternative treatment modalities will be required to deal with these issues. Peptoids are oligomers of N-substituted glycine. These peptidomimetics molecules provide many advantages over peptide-based vaccines. Unlike peptides, peptoids are resistant to proteolytic degradation, and allow peptoids to retain therapeutic potential in some difficult physiological environments. Such molecules can also be engineered for structural stability and biomarker detection. All

Data-Driven Engineering of Thermostable Collagen-Mimetic Peptoid Triple Helices.

Collagen-mimetic peptides (CMPs) are engineered molecules designed to replicate the triple-helical structure of natural collagen. A repeating x-y-Gly sequence is the defining motif of CMPs and is critical to their triple-helical structure and stability. Substitutions to the residues occupying the x and y positions present a means to modulate the CMP structure and properties. Peptoid residues-N-substituted glycine derivatives-present an attractive potential substitution due to their thermal stability, proteolytic resistance, biocompatibility, and diverse palette of non-natural side chains, but

Noncanonical Folding of Peptoid Oligomers: Formation of a Closed Conformation in Nonpolar Solvent.

Conformational behavior of peptoids in low-dielectric solvents remains poorly understood despite its relevance to membrane environments. Here, conformations of N-(S)-1-phenylethylglycine (Nspe) homo-oligomers were investigated in chloroform using NMR spectroscopy and MD simulations. Nspe7 populated two closed conformations, while Nspe10 adopted a single conformation reminiscent of the Nspe9 threaded-loop structure. End-to-end hydrogen bonding and hydrophobic side-chain shielding stabilize these compact folds, minimizing polar surface area. These findings provide insights into peptoid folding i

Phosphate-Binding Zn(II)-Coordinated Antimicrobial Peptoids: Enhancing Selectivity through Specific Recognition of Bacterial Membranes.

The global spread of multidrug-resistant bacteria underscores the urgent need for antimicrobial agents with enhanced efficacy and selectivity. Here, we developed antimicrobial peptoids conjugated with zinc-dipicolylamine (ZnDPA) and bivalent Zn2BPMP motifs for improved bacterial membrane recognition via phosphate binding. The Zn2BPMP-containing peptoids, 5_Zn2 and 8_Zn4, exhibited the highest bacterial selectivity, achieving an increase in selectivity index of >10-fold. Cytotoxicity assays confirmed reduced toxicity against mammalian cells. Mechanistically, Zn2BPMP conjugation enhanced binding

Structure-function relationships in peptoids: recent advances toward deciphering the structural requirements for biological function.

Oligomers of N-substituted glycine, or peptoids, are versatile tools to probe biological processes and hold promise as therapeutic agents. An underlying theme in the majority of recent peptoid research is the connection between peptoid function and peptoid structure. For certain applications, well-folded peptoids are essential for activity, while unstructured peptoids appear to suffice, or even are superior, for other applications. Currently, these structure-function connections are largely made after the design, synthesis, and characterization process. However, as guidelines for peptoid foldi

Exploring a Ugi-Mediated Polymerization-Induced Self-Assembly Approach to Peptoid Particles.

This work investigates the synthesis of polypeptoid nanoparticles through Ugi-mediated polymerization-induced self-assembly (U-PISA), combining aqueous Ugi four-component polycondensation with PEG-based stabilization. Polypeptoid nano-objects were produced directly from amino acid-derived building blocks via a one-pot process under mild, open-air conditions. The effects of PEG molar mass and feed composition on particle formation and morphology were systematically examined, providing access to tunable spherical and worm-like nanostructures. The in situ formation of amphiphilic PEG-polypeptoid

Expanded polyglutamine-binding peptoid as a novel therapeutic agent for treatment of Huntington's disease.

Polyglutamine(polyQ)-expanded proteins are potential therapeutic targets for the treatment of polyQ expansion disorders such as Huntington's disease (HD) and spinocerebellar ataxia type 3 (SCA3). Here, we used an amino-terminal fragment of a mutant Huntingtin protein (Htt-N-82Q) as bait in an unbiased screen of a 60,000 peptoid library. Peptoid HQP09 was selected from the isolated hits and confirmed as a specific ligand of Htt-N-82Q and Atxn3-77Q mutant proteins in biochemical experiments. We identified three critical residues in the HQP09 sequence that are important for its activity and gener

Parasitic diseases are a worldwide health and socio-economical problem, especially due to increasing parasite resistance and frequent toxicity of commercial drugs. Parasite metallo-aminopeptidases are potential therapeutic targets, as they perform crucial biological functions during parasite development within the human host. Here, we describe the synthesis and biological evaluation of two libraries consisting of a total of 74 peptoid-based compounds. The synthetic approaches use a multicomponent reaction to install various diversity elements, and in library one, this is followed by subsequent
