Previous studies have shown that perirhinal cortex lesions in monkeys impair visual discriminations with a high degree of "feature ambiguity," a property of visual discriminations that can emerge when features are a part of both rewarded and unrewarded stimuli. The effects of damage to the hippocampus on these perirhinal-dependent feature-ambiguous tasks are, however, unknown. Prominent theories of medial temporal lobe function predict similar effects of perirhinal cortex and hippocampal lesions on cognitive tasks. In contrast, our hypothesis is that perirhinal cortex, and not the hippocampus,

The formation of associations, which involves binding disparate pieces of information, is fundamental to constructing episodic memory. This process primarily relies on the neural circuitry within the medial temporal lobe, specifically the hippocampal-parahippocampal network. Within this network, the perirhinal cortex (PER) and the hippocampus (HPC) are recognized as essential components for associative processing. While the traditional dual-pathway model depicts a hierarchically organized, sequential transmission of information along the medial temporal lobe, recent anatomical and functional s

Episodic memory involves the representation of memories shared across a global context as well as that of unique events that occurred within it. To examine how parahippocampal subregions support this balance, we had participants encounter multiple objects in each building of a virtual environment and later asked them to recall the building associated with each cued object during fMRI scanning. Across the MTL, objects from the same building showed greater representational similarity than those from different buildings, indicating the integration of contextually associated memories. This effect

Recent studies have suggested that the perirhinal cortex (Prh) supports representations of feature conjunctions in the visual modality during the acquisition/encoding of complex discriminations. To extend this idea to other sensory modalities and to another stage of the discrimination process, we studied the effect of Prh lesions on the expression of a series of tactual discrimination tasks learned preoperatively. These tasks differed from one another in the degree of feature overlap of the stimuli and in the difficulty of the task. During pre- and post-operative testing phases, rats had to di

The perirhinal cortex, a small strip of the anterior medial temporal cortex, first came into prominence through studies of memory. While examining patients with damage to the medial temporal lobe as well as animals with similar regional damage, findings showed that combined damage to the hippocampus, amygdala, and adjacent cortical areas, including the perirhinal cortex, were responsible for the profound memory loss observed. Later, however, the evidence demonstrated that the accompanying damage to the underlying medial temporal cortical areas were largely responsible for the memory deficit th

Rats were tested in a series of two-choice visual discrimination tasks in a computer-controlled testing apparatus. The discriminations used a range of discriminanda, which varied in complexity. The discriminations included relatively simple form discriminations, more complex form discriminations and discriminations between compound stimuli that shared many features. It was found that rats with perirhinal cortex lesions were unimpaired in all discriminations except those that involved the compound stimuli with overlapping features. Using these stimuli, rats with perirhinal cortex lesions were u

Several lines of evidence in humans and experimental animals suggest that the hippocampus is critical for the formation and retrieval of spatial memory. However, although the hippocampus is reciprocally connected to adjacent cortices within the medial temporal lobe and they, in turn, are connected to the neocortex, little is known regarding the function of these cortices in memory. Here, using a reference spatial memory task in the radial maze, we show that neurotoxic perirhinal cortex lesions produce a profound retrograde amnesia when learning-surgery intervals of 1 or 50 d are used (Experime

Temporal lobe epilepsy (TLE) is frequently accompanied by memory impairments and, although their bases are unknown, most research has focused on the hippocampus. The present study investigated the importance of another medial temporal lobe structure, the perirhinal cortex (Prh), in changes in memory in TLE using kindling as a model. Rats were kindled twice daily with anterior Prh stimulation until three fully generalized seizures were evoked. Beginning 7 days later and on successive days, rats were tested in an elevated plus maze, a large circular open field, an open field object exploration t

Damage to perirhinal cortex (PRh) impairs object recognition memory in humans, monkeys, and rats when tested in tasks such as delayed nonmatching to sample, visual paired comparison, and its rodent analog, the spontaneous object recognition task. In the present study, we have capitalized on the discrete one-trial nature of the spontaneous object recognition task to investigate the role of PRh in several distinct stages of object recognition memory. In a series of experiments, transient inactivation of PRh was accomplished with bilateral infusions of lidocaine directly into PRh immediately befo

Methamphetamine (meth) use disorder is part of an overarching use disorder that encompasses continued drug seeking and an increased risk of returning to drug use following periods of abstaining. Chronic meth use results in drug-induced cortical plasticity in the perirhinal cortex (PRC) that mediates responses to novelty. PRH projection targets are numerous and include the nucleus accumbens core (NAc). Whereas the PRH-prefrontal cortex is involved in object recognition; we propose that the PRH-NAc is involved in novelty salience. Rats underwent short-access (ShA, 1 hr) or long-access (LgA, 6 hr

How verbal and nonverbal visuoperceptual input connects to semantic knowledge is a core question in visual and cognitive neuroscience, with significant clinical ramifications. In an event-related functional magnetic resonance imaging (fMRI) experiment we determined how cosine similarity between fMRI response patterns to concrete words and pictures reflects semantic clustering and semantic distances between the represented entities within a single category. Semantic clustering and semantic distances between 24 animate entities were derived from a concept-feature matrix based on feature generati

The perirhinal cortex (PRh) has extensive interconnections with limbic structures, including the fasciola cinerea (FC), a recently identified node and therapeutic target in temporal lobe epilepsy. However, the neural circuit basis between PRh and FC in seizure progression remains unclear. Here we show that the PRh provides the principal excitatory input to the FC and plays an important role in seizure modulation in male mouse epilepsy models. PRh-FC projections are strongly activated during seizures, and their inhibition delays seizure progression. Moreover, PRh excitatory neurons bidirectiona

RATIONAL: The human perirhinal cortex (PRC) plays critical roles in episodic and semantic memory and visual perception. The PRC consists of Brodmann areas 35 and 36 (BA35, BA36). In Alzheimer's disease (AD), BA35 is the first cortical site affected by neurofibrillary tangle pathology, which is closely linked to neural injury in AD. Large anatomical variability, manifested in the form of different cortical folding and branching patterns, makes it difficult to segment the PRC in MRI scans. Pathology studies have found that in ~97% of specimens, the PRC falls into one of three discrete anatomical

The rodent ventral and primate anterior hippocampus have been implicated in approach-avoidance (AA) conflict processing. It is unclear, however, whether this structure contributes to AA conflict detection and/or resolution, and if its involvement extends to conditions of AA conflict devoid of spatial/contextual information. To investigate this, neurologically healthy human participants first learned to approach or avoid single novel visual objects with the goal of maximizing earned points. Approaching led to point gain and loss for positive and negative objects, respectively, whereas avoidance

The perirhinal cortex is situated on the border between sensory association cortex and the hippocampal formation. It serves an important function as a transition area between the sensory neocortex and the medial temporal lobe. While the perirhinal cortex has traditionally been associated with object coding and the "what" pathway of the temporal lobe, current evidence suggests a broader function of the perirhinal cortex in solving feature ambiguity and processing complex stimuli. Besides fulfilling functions in object coding, recent neurophysiological findings in freely moving rodents indicate

Visual recognition memory is early impaired in Alzheimer's disease. Long-term depression of synaptic transmission in the perirhinal cortex is critically involved in this form of memory. We found that synaptic transmission was impaired in perirhinal cortex slices obtained from 3-month-old Tg2576 mice, and that 3,000 pulses at 5 Hz induced long-term depression in perirhinal cortex slices from age-matched control mice, but not in those from Tg2576 mice. To our knowledge, these data provide the first evidence of synaptic transmission and long-term depression impairment in the perirhinal cortex in

Normal aging causes a decline in object recognition. Importantly, lesions of the perirhinal cortex produce similar deficits and also lead to object discrimination impairments when the test objects share common features, suggesting that the perirhinal cortex participates in perceptual discrimination. The current experiments investigated the ability of young and aged animals to distinguish between objects that shared features with tasks with limited mnemonic demands. In the first experiment, young and old rats performed a variant of the spontaneous object recognition task in which there was a mi

The Coolidge effect is a phenomenon in which males show renewed sexual interest in a novel female following copulation to satiety with another female. In golden hamsters, this phenomenon depends on the ability to recognize conspecifics using chemosensory cues processed through the main olfactory system. Here we tested whether olfactory targets in the hippocampal system support this natural form of recognition memory. Male hamsters received ibotenic acid lesions of the perirhinal-entorhinal cortex (PR-ENT) or hippocampus (H) and were allowed to copulate to satiety with a female conspecific, the

Synaptic transmission is essential for early development of the central nervous system. However, the mechanisms that regulate early synaptic transmission in the cerebral cortex are unclear. PKMζ is a kinase essential for the maintenance of LTP. We show for the first time that inhibition of PKMζ produces a profound depression of basal synaptic transmission in neonatal, but not adult, rat perirhinal cortex. This suggests that synapses in early development are in a constitutive LTP-like state. Furthermore, basal synaptic transmission in immature, but not mature, perirhinal cortex relies on persis
