Permafrost thaw lakes including thermokarst lakes and ponds are ubiquitous features of Subarctic and Arctic landscapes and are hotspots of microbial activity. Input of terrestrial organic matter into the planktonic microbial loop of these lakes may greatly amplify global greenhouse gas emissions. This microbial loop, dominated in the summer by aerobic microorganisms including phototrophs, is radically different in the winter, when metabolic processes shift to the anaerobic degradation of organic matter. Little is known about the viruses that infect these microbes, despite evidence that viruses

Microbial dynamics in thawing permafrost induce a climate feedback of uncertain magnitude. Upon thaw, active layer microorganisms may establish in formerly frozen layers and introduce missing functions. Here we test the prevalence of functional limitations and how active layer microorganisms can mitigate these across four widespread permafrost types over 389 days. By restoring functions with a multifunctional microbial community, we demonstrate widespread functional limitations, as increased CO2 emissions across four permafrost types, and onset N2O emissions in two of those. However, realistic

The loss of ecosystem carbon (the sum of vegetation, litter, and soil carbon) may occur in a permafrost region under mitigation pathways, which could reduce the efficiency of carbon dioxide removal. Here, we investigate changes in permafrost under net-zero and negative emissions, based on idealized emission-driven simulations using a state-of-the-art Earth system model. While acting as a net ecosystem carbon sink during most of the positive emission phase, permafrost becomes a net ecosystem carbon source just before reaching net-zero and negative emissions. Permafrost slowly recovers, especial

While wetlands are major sources of biogenic methane (CH4), our understanding of resident microbial metabolism is incomplete, which compromises the prediction of CH4 emissions under ongoing climate change. Here, we employed genome-resolved multi-omics to expand our understanding of methanogenesis in the thawing permafrost peatland of Stordalen Mire in Arctic Sweden. In quadrupling the genomic representation of the site's methanogens and examining their encoded metabolism, we revealed that nearly 20% of the metagenome-assembled genomes (MAGs) encoded the potential for methylotrophic methanogene

Nitrogen oxides (NOx) play an important role for atmospheric chemistry and radiative forcing. However, NOx emissions from the vast northern circumpolar permafrost regions have not been studied in situ due to limitations of measurement techniques. Our goals were to validate the offline analytical technique, and based on this, to widely quantify in situ NOx emissions from peatlands in the southern Eurasian permafrost region. To this end, we conducted a comparison of online and offline flux measurements in 2018 and 2019 using the synthetic air flushing, steady-state opaque chamber method. With di

Climate warming in the Arctic may increase the risk of zoonoses due to expansion of vector habitats, improved chances of vector survival during winter, and permafrost degradation. Monitoring of soil temperatures at Siberian cryology control stations since 1970 showed correlations between air temperatures and the depth of permafrost layer that thawed during summer season. Between 1900s and 1980s, the temperature of surface layer of permafrost increased by 2-4°C; and a further increase of 3°C is expected. Frequent outbreaks of anthrax caused death of 1.5 million deer in Russian North between 189

Permafrost soils store vast amounts of organic carbon, and their thawing due to climate warming accelerates the release of carbon as methane and carbon dioxide, exacerbating global climate change. Understanding the distribution of greenhouse gases trapped in these soils and predicting their behavior upon thawing is essential for accurately modeling climate feedbacks. This study presents an integrated biogeochemical and microbial dataset from ~1.8 m deep soil cores collected across a 970 km latitudinal gradient in Alaskan permafrost regions, spanning boreal forest and Arctic tundra biomes. This

Permafrost thaw in peatlands risks increasing the production and mobilization of methylmercury (MeHg), a bioaccumulative neurotoxin that poses a health hazard to humans. We studied 12 peatlands on a trophic gradient in northwestern Canada, including permafrost peat plateaus and thawed bogs and fens, to determine the effects of thaw on MeHg production from measures of soil and porewater MeHg and in situ methylation assays. The production of MeHg was greater in thawed peatlands, especially rich fens, as indicated by higher potential rates of microbial methylation of inorganic mercury (Hg) to MeH

With Arctic amplification, hydrological conditions in Arctic permafrost regions are expected to change substantially, which can have a strong impact on carbon budgets. To date, detailed mechanisms remain highly uncertain due to the lack of continuous observational data. Considering the large carbon storage in these regions, understanding these processes becomes crucial for estimating the future trajectory of global climate change. This study presents findings from 8 years of continuous eddy-covariance measurements of carbon dioxide ( CO 2 $$ {\mathrm{CO}}_2 $$ ) and methane ( CH 4 $$ {\

Recent climate change has increased arctic soil temperatures and thawed large areas of permafrost, allowing for microbial respiration of previously frozen C. Furthermore, soil destabilization from melting ice has caused an increase in thermokarst failures that expose buried C and release dissolved organic C (DOC) to surface waters. Once exposed, the fate of this C is unknown but will depend on its reactivity to sunlight and microbial attack, and the light available at the surface. In this study we manipulated water released from areas of thermokarst activity to show that newly exposed DOC is >

Arctic shorelines are vulnerable to climate change impacts as sea level rises, permafrost thaws, storms intensify, and sea ice thins. Seventy-five years of aerial and satellite observations have established coastal erosion as an increasing Arctic hazard. However, other hazards at play-for instance, the cumulative impact that sea-level rise and permafrost thaw subsidence will have on permafrost shorelines-have received less attention, preventing assessments of these processes' impacts compared to and combined with coastal erosion. Alaska's Arctic Coastal Plain (ACP) is ideal for such assessment

This article describes Bacillus anthracis strains isolated during an outbreak of anthrax on the Yamal Peninsula in the summer of 2016 and independently in Yakutia in 2015. A common feature of these strains is their conservation in permafrost, from which they were extracted either due to the thawing of permafrost (Yamal strains) or as the result of paleontological excavations (Yakut strains). All strains isolated on the Yamal share an identical genotype belonging to lineage B.Br.001/002, pointing to a common source of infection in a territory over 250 km in length. In contrast, during the excav

Antibiotic resistance genes (ARGs) and mobile genetic elements (MGEs) can be identified with metagenomic analyses comparing relatively pristine and human-impacted environments. We collected samples from 3 different environments: glacial soil little affected by anthropogenic activity, deep permafrost dated to 5821 BP (before human antibiotics), and sediment from the Pearl River. Sulfonamides, tetracyclines, and fluoroquinolones were common in the sediment samples. Sulfonamides and tetracycline were not found in permafrost; tetracycline was also not found in glacial soil. ARGs from the sediment

In 2007, the baby woolly mammoth (Mammuthus primigenius) named Lyuba was found frozen in the Siberian tundra permafrost along the Yuribey River. She was proclaimed the best-preserved mammoth discovery. As part of the endoscopic examination of Lyuba, tissue samples of hair, muscle, and internal organs were taken. The sectioned biopsies were stained using standard and special histological stains. In general, the microscopic preservation of the tissue was good although no clearly identifiable cell nuclei were found by standard staining methods. Only a few cell nuclei could be identified in some s

Rising temperatures and changing precipitation patterns are causing rapid changes in the Himalayan ecosystem. Changes in climatic conditions affect various aspects of high mountain environments, including glaciers, rock glaciers, and permafrost, and pose significant threats to indigenous mountain communities. This study investigates the first case of permafrost thaw-induced subsidence in the Lindur Village of Lahaul & Spiti district of Himachal Pradesh using MT-InSAR method. We used 15 Sentinel-1 single-look complex images acquired from April 2022 to September 2022 along the ascending orbit tr

Anthropogenic-driven climate change is accelerating permafrost thaw, threatening to release vast carbon stores through increased microbial activity. While microbial roles are increasingly studied, the contributions of viruses remain largely unexplored, in part due to soil-associated technical challenges that have hindered their detection and characterization. Here, we applied an optimized virion enrichment workflow along a permafrost thaw gradient, identifying 9,963 viral populations (vOTUs), including single- and double-stranded DNA viruses, with 99.9% novelty compared to other soils. BACKGROUND: A shallow active layer of soil above the permafrost thaws during the summer months which promotes microbial growth and releases previously confined pathogens which result in bacterial epidemics in circumpolar regions. Furthermore, these permafrost sources harbor several antibiotic resistance genes (ARGs) which may disseminate and pose a challenge for pharmacologists worldwide.
AIMS: The authors examined the potential association between climate change-induced permafrost thawing, and the resulting release of antibiotic-resistant pathogens, as well as the potential impact this can h

We studied the effect of a single intraperitoneal injection of metabolites from Bacillus sp. M3 strain isolated from permafrost (from 5×103 to 50×103 microbial bodies) on differentiation of T cells in the thymus of F1(CBA/Black-6) mice. On day 21 after the injection of metabolites, a dose-dependent decrease in the level of CD34+CD44+ and an increase in the number of CD34+CD44-, CD25-TCR+, CD25+TCR+max, CD4+CD8-, CD4-CD8+, and CD44+TCR+ lymphocytes were observed in the thymus. The increase in thymus level of mature (CD25+TCR+max) and migration-ready (CD44+TCR+) T cells in combination with a mod

Deciphering microbiomes across vertical profiles is critical for understanding biogeochemical cycling and potential biosafety hazards associated with permafrost degradation under climate warming. However, knowledge about microbiome patterns over key profile layers in response to degradation remains limited in the Qinghai-Tibet Plateau. Using metagenomic data obtained from 150 samples of six 15 m-depth alpine permafrost cores along a degradation gradient, we analyzed microbial community structure and functional potential across different main-layers, including the active, frozen fringe, and fro

Dissolved organic matter (DOM) released from permafrost thaw greatly influences the biogeochemical cycles of, among others, downstream carbon, nitrogen and phosphorus cycles; yet, knowledge of the linkages between bacterial communities with permafrost DOM heterogeneity is limited. Here, we aim at unravelling the responses of bacterial diversities and metabolic profiles to DOM quantity and composition across permafrost thawing gradients by coupling an extensive field investigation with bio-incubation experiments. Richness, evenness and dissimilarities of the whole and rare communities decreased
