Permissiveness of different TMEM154 genotype cell lines to different SRLV genotypes/subtypes.

Small ruminant lentiviruses (SRLVs) are a heterogeneous group of viruses that infect goats and sheep worldwide, for which no cures or effective vaccines are available. Control measures have been based on early diagnosis and accurate management with limited success due to re-emergence of seropositivity and disease in controlled flocks. Association between the genetic variation in the ovine transmembrane 154 (TMEM154) gene and susceptibility to certain viral subtypes was previously demonstrated, representing a possible control strategy to reduce infection prevalence in sheep. Thus, the identific

Antimicrobial resistance level and conjugation permissiveness shape plasmid distribution in clinical enterobacteria.

Conjugative plasmids play a key role in the dissemination of antimicrobial resistance (AMR) genes across bacterial pathogens. AMR plasmids are widespread in clinical settings, but their distribution is not random, and certain associations between plasmids and bacterial clones are particularly successful. For example, the globally spread carbapenem resistance plasmid pOXA-48 can use a wide range of enterobacterial species as hosts, but it is usually associated with a small number of specific Klebsiella pneumoniae clones. These successful associations represent an important threat for hospitaliz

PI3-kinase activation is critical for host barrier permissiveness to Listeria monocytogenes.

Invasion of nonphagocytic cells, a critical property of Listeria monocytogenes (Lm) that enables it to cross host barriers, is mediated by the interaction of two bacterial surface proteins, InlA and InlB, with their respective receptors E-cadherin and c-Met. Although InlA-E-cadherin interaction is necessary and sufficient for Lm crossing of the intestinal barrier, both InlA and InlB are required for Lm crossing of the placental barrier. The mechanisms underlying these differences are unknown. Phosphoinositide 3-kinase (PI3-K) is involved in both InlA- and InlB-dependent pathways. Indeed, InlA-

A distinct interphase microtubule state marks host cell permissiveness to Chlamydia pneumoniae entry.

Entry of intracellular pathogenic bacteria is widely considered an actin-driven process, potentially overlooking contributions of the microtubule cytoskeleton. Interphase microtubules exist in distinct states, but whether this heterogeneity influences bacterial entry is unknown. Here, using the obligate intracellular bacterium Chlamydia pneumoniae, we show that microtubules not only contribute to bacterial entry but that their pre-existing state influences host-cell permissiveness. Cells enriched in acetylated microtubules are preferentially infected, whereas detyrosinated microtubules show no

Endothelial-to-mesenchymal transition enhances permissiveness to AAV vectors in cardiac endothelial cells.

A major obstacle in inducing therapeutic angiogenesis in the heart is inefficient gene transfer to endothelial cells (ECs). Here, we identify compounds able to enhance the permissiveness of cardiac ECs to adeno-associated virus (AAV) vectors, which stand as ideal tools for in vivo gene delivery. We screened a library of >1,500 US Food and Drug Administration (FDA)-approved drugs, in combination with AAV vectors, in cardiac ECs. Among the top drugs increasing AAV-mediated transduction, we found vatalanib, an inhibitor of multiple tyrosine kinase receptors. The increased AAV transduction efficie

Defending the Ethical Permissibility of Laryngeal Transplantation.

Transplantation of the larynx is a rare and controversial procedure. Unlike solid organ transplantation, in which the primary goal is preservation of life, laryngeal transplantation aims to improve a patient's quality-of-life by restoring function. Several successful laryngeal transplants have been reported in the scientific literature: restoring patient speech, improving swallowing and breathing, and providing substantial quality-of-life improvements. Nonetheless, laryngeal transplantation poses both scientific and ethical challenges. The novelty of the procedure, uncertainty about appropriat

"Permissive AKI" with treatment of heart failure.

Renin-angiotensin-aldosterone system inhibitors have proven clinical benefit in management of patients with heart failure with reduced ejection fraction. However, there are no guidelines to manage decline in kidney function that occurs with initiation and titration of renin-angiotensin-aldosterone system agents during management of heart failure. We discuss the complex interplay of kidney function and heart failure in the presence of renin-angiotensin-aldosterone system agents and suggest a clinical algorithm for management of acute decline in kidney function.

Permissive and protective roles for neutrophils in leishmaniasis.

Leishmania parasites are the causative agents of leishmaniasis, a neglected tropical disease that causes substantial morbidity and considerable mortality in many developing areas of the world. Recent estimates suggest that roughly 10 million people suffer from cutaneous leishmaniasis (CL), and approximately 76,000 are afflicted with visceral leishmaniasis (VL), which is universally fatal without treatment. Efforts to develop therapeutics and vaccines have been greatly hampered by an incomplete understanding of the parasite's biology and a lack of clear protective correlates that must be met in

Permissive underfeeding of the critically ill patient.

The rise in the popularity of nutrition support in the 1970s was associated with the concept of "hyperalimentation." This concept was based on the early findings that increased metabolic rates were observed in various disease states such as trauma, sepsis, and burns. The aim was to feed 40% to 100% above the basal metabolic rate to avoid weight loss associated with critical illness. Since that time, several observations have indicated that permissive underfeeding may be beneficial because: (a) the metabolic rate is not markedly increased in most patients with critical illness except burns; (b)

Permissiveness of American bison to infection with severe acute respiratory syndrome coronavirus-2.

The COVID-19 pandemic represents one of the most significant public health events of the last century. As with other coronaviruses (SARS, MERS) the role of animals is of intense interest. Believed to have originated in bats, the role of other animals in the epidemiology of the SARS-CoV-2 pandemic is still unclear, as is the range of susceptible hosts. American bison were intranasally infected with SARS-CoV-2 and monitored for seroconversion and the presence of viral RNA in oronasal secretions and feces. Although clinical signs were not seen, permissiveness of bison to infection with SARS-CoV-2

Evolution of triclosan resistance modulates bacterial permissiveness to multidrug resistance plasmids and phages.

The horizontal transfer of plasmids has been recognized as one of the key drivers for the worldwide spread of antimicrobial resistance (AMR) across bacterial pathogens. However, knowledge remain limited about the contribution made by environmental stress on the evolution of bacterial AMR by modulating horizontal acquisition of AMR plasmids and other mobile genetic elements. Here we combined experimental evolution, whole genome sequencing, reverse genetic engineering, and transcriptomics to examine if the evolution of chromosomal AMR to triclosan (TCS) disinfectant has correlated effects on mod

Clinical effectiveness and safety of permissive hypercapnia.

Experimental and clinical data indicate that ventilator strategies with permissive hypercapnia may reduce lung injury by a variety of mechanisms. Seven randomized controlled trials in preterm neonates suggest that permissive hypercapnia started early, before the initiation of mechanical ventilation (in conjunction with continuous positive airway pressure), followed by prolonged permissive hypercapnia if mechanical ventilation is needed is an alternative to early ventilation and surfactant. Permissive hypercapnia may improve pulmonary outcomes and survival.

Ultrastructural changes in cells induced by temperature-sensitive mutants of fowl plague virus at permissive and non-permissive temperature.

Ultrastructural changes developing in chick embryo fibroblast cultures infected with a wild-type strain of fowl plague virus (FPV) or one of six FPV temperature-sensitive (ts) mutants belonging to different complementation groups were studied. Cells infected with wild-type FPV and incubated at optimal (36 degrees C) or nonpermissive temperature (42 degrees C) displayed changes similar to those described for orthomyxoviruses. The same patterns of changes were observed at 36 degrees C in cells infected with ts mutants belonging to five of the complementation groups. Mutant ts 303, possessing mut

iPSC-based macrophage models reveal interferon-dependent control of RSV permissiveness and syncytia formation.

Respiratory syncytial virus (RSV) is a leading cause of severe respiratory illness worldwide and affects mainly infants, immunocompromised individuals, and older adults. Understanding how immune cells contribute to RSV pathogenesis is crucial for developing interventions. However, the lack of model systems that accurately mimic in vivo immune responses has hampered progress in this field. Here, we present iPSC-derived macrophages (iMacs) as a promising model for the analysis of tissue-resident macrophage responses during RSV infection. Using monocyte-derived macrophages as a comparison, we sho

Neural tube defects induce abnormal astrocyte development by activation and epigenetic permissiveness of STAT3.

The open neural tube defect, myelomeningocele (MMC), is a debilitating congenital defect of the central nervous system, characterized by impaired spinal cord development. The pathological development of MMC spinal cord involves enhanced astrogenesis. However, the mechanisms underlying the abnormal generation of astrocytes in MMC spinal cord are poorly understood. Using a fetal rat model of this defect, we discovered that neural progenitor cells (NPCs) in the developing spinal cord undergo the accelerated acquisition of astrocytic competency and the abnormal induction of GFAP expression, result
