Persistent bovine pestivirus infection localized in the testes of an immuno-competent, non-viraemic bull.

A post-pubertal bull on an artificial insemination station was found to be persistently shedding bovine viral diarrhoea virus (BVDV) in semen over a period of eleven months, while demonstrating no viraemia. Circulating antibodies to BVDV were consistently high, suggesting that the immune system was challenged repeatedly. Post-mortem findings confirmed that the virus was sequestered in the testes of the bull. It is hypothesized that the BVDV in this immuno-competent bull was protected from the bull's immune response by the blood-testes barrier. The barrier becomes functional only at puberty whe

A multiplex RT-PCR assay for the rapid and differential diagnosis of classical swine fever and other pestivirus infections.

Classical swine fever is a highly contagious viral disease causing severe economic losses in pig production almost worldwide. All pestivirus species can infect pigs, therefore accurate and rapid pestivirus detection and differentiation is of great importance to assure control measures in swine farming. Here we describe the development and evaluation of a novel multiplex, highly sensitive and specific RT-PCR for the simultaneous detection and rapid differentiation between CSFV and other pestivirus infections in swine. The universal and differential detection was based on primers designed to amp

Dynamics of Infection of Atypical Porcine Pestivirus in Commercial Pigs from Birth to Market: A Longitudinal Study.

Atypical porcine pestivirus (APPV) was found to be associated with pigs demonstrating congenital tremors (CT), and clinical signs in pigs have been reproduced after experimental challenge. Subsequently, APPV has been identified in both symptomatic and asymptomatic swine of all ages globally. The objective of this research was to perform a longitudinal study following two cohorts of pigs, those born in litters with pigs exhibiting CT and those born in litters without CT, to analyze the virus and antibody dynamics of APPV infection in serum from birth to market. There was a wide range in the per

Viral load and histological distribution of atypical porcine pestivirus in different tissues of naturally infected piglets.

A newly identified atypical porcine pestivirus (APPV) associated with congenital tremors in newborn piglets has been shown to have a worldwide geographic distribution. In view of the function of Erns in pestivirus infection and replication, the viral load and histological distribution of APPV in different tissues of naturally infected piglets were analyzed by quantitative RT-PCR and immunohistochemical detection using Erns as the target. The results showed that the viral copy number was higher in the cerebellum, submandibular lymph nodes, and thymus than in other tissues, indicating that these

Detection and localization of atypical porcine pestivirus in the testicles of naturally infected, congenital tremor affected piglets.

Atypical porcine pestivirus (APPV) belongs to the genus Pestivirus within the family Flaviviridae. Recently, APPV has been identified as the causative agent of congenital tremor (CT) type AII. The disease is a neurological disorder that affects newborn piglets and is characterized by generalized trembling of the animals and often splay legs. CT is well known worldwide, and the virus seems to be highly prevalent in major swine producing areas. However, little is known about the epidemiology of the infection, transmission and spread of the virus between herds. Here, we show the high prevalence o

Viremia and antibody dynamics following atypical porcine pestivirus infection: a cohort study of pigs with congenital tremor.

Atypical porcine pestivirus (APPV) induces congenital tremor (CT) in piglets. Limited understanding of the relationship between viremia, antibody responses and clinical signs has impeded the development of effective preventive measures, as it is unclear which individuals are infectious and for how long. To characterize these aspects, a cohort field study with 43 piglets from herds experiencing CT and 21 external control piglets was conducted. CT-pigs and their healthy littermates were viremic at birth, with APPV RNA detected in serum by RT-qPCR, and viremia persisted in eight of the 15 pigs fr

Protective Efficacy of a Chimeric Pestivirus KD26_E2LOM Vaccine Against Classical Swine Fever Virus Infection of Pigs.

A chimeric pestivirus KD26_E2LOM strain can induce antibodies that can be partially distinguished from antibodies from classical swine fever virus (CSFV) infection. The chimeric pestivirus vaccine strain was created using bovine viral diarrhea virus as the backbone; however, the entire BVDV E2 gene region was replaced with the E2 gene, which encodes the major target for neutralizing antibodies against CSFV. Pigs were vaccinated once or twice with the chimeric pestivirus KD26_E2LOM strain, and protective efficacy was evaluated after subsequent challenge with virulent CSFV. Pigs inoculated with

Congenital Neurological Disease Associated With HoBi-like Pestivirus Infection in a Newborn Dairy Calf From Brazil.

HoBi-like pestivirus (HoBiPeV) has been reported in several biological samples from cattle worldwide, but there are no descriptions of this virus associated with neurological symptoms. This report described the first occurrence of neurological disease associated with HoBiPeV in a newborn dairy calf. A mixed-breed Holstein calf had severe neurological symptoms at birth and died at 21 days old. The tissue fragments (central nervous system (CNS), myocardium, liver, kidney, lung, intestine, and spleen) were submitted to reverse transcription (RT)-PCR assay for the partial 5'-untranslated region (5

Serological evidence of Coxiella burnetii, Leptospira interrogans Hardjo, Neospora caninum and bovine pestivirus infections in a dairy cattle herd from the United Arab Emirates.

The serostatus of five abortigenic agents and the association between abortion history and Coxiella burnetii seropositivity were assessed in 350 dairy cattle from Al Ain, UAE. The bovine sera were ELISA-screened for C. burnetii, Leptospira Hardjo, Neospora caninum, and Brucella abortus antibodies, plus bovine pestivirus (BVDV) antigen. The serology data were collated and the level of significance between the proportions of C. burnetii-seropositive cattle with and without abortion history assessed by the Z test of two proportions. Of the 350 cattle, 41.4%, 1.7%, 1.4%, 0.3%, and 0.0% were seropo

Enhancing the understanding of coinfection outcomes: Impact of natural atypical porcine pestivirus infection on porcine reproductive and respiratory syndrome in pigs.

Atypical porcine pestivirus (APPV) is a novel member of the Pestivirus genus detected in association with congenital tremor (CT) type A-II outbreaks and from apparently healthy pigs, both as singular infection and as part of multi-pathogen infections. 'Classical' pestiviruses are known to cause immunosuppression of their host, which can increase susceptibility to secondary infections, severely impacting health, welfare, and production. To investigate APPV's effect on the host's immune system and characterise disease outcomes, 12 piglets from a natural APPV CT type A-II outbreak were experiment

The new emerging ovine pestivirus can infect pigs and confers strong protection against classical swine fever virus.

Several emerging pestiviruses have been reported lately, some of which have proved to cause disease. Recently, a new ovine pestivirus (OVPV), isolated from aborted lambs, with high genetic identity to classical swine fever virus (CSFV), has proved to induce reproductive disorders in pregnant ewes. OVPV also generated strong serological and molecular cross-reaction with CSFV. To assess the capacity of OVPV to infect swine, twelve piglets were infected either by intranasal or intramuscular route. Daily clinical evaluation and weekly samplings were performed to determine pathogenicity, viral repl

The impact of pestivirus on an artificial breeding program for cattle.

After the introduction of pestivirus into a herd undergoing an embryo transfer and artificial insemination program, substantial post-weaning calf losses occurred. The predominant clinical feature was severe respiratory disease, in contrast to the commonly recognised mucosal disease. Thirty-one of 76 calves were affected, with a case fatality rate of 58%. All calves which were persistently infected with pestivirus died during the study period. There was a significant association in the surviving calves between the occurrence of recent pestivirus infection and respiratory disease. The losses on

Molecular detection and genetic characterization of pestiviruses associated with abortion in domestic small ruminants in Türkiye.

Abortion in small ruminants significantly reduces productivity, with pestiviruses recognized as major viral contributors. Due to their broad host range, sheep can be infected not only by classical pestiviruses, such as bovine viral diarrhea virus (BVDV) and border disease virus, but also by newly identified pestivirus species. This study aimed to molecularly detect and genetically characterize pestiviruses associated with abortion in small ruminants from eastern Türkiye and to evaluate their epidemiological distribution. Samples from 85 sheep and 14 goats were tested for pestivirus RNA using a

Exclusion of Superinfection or Enhancement of Superinfection in Pestiviruses-APPV Infection Is Not Dependent on ADAM17.

Some viruses can suppress superinfections of their host cells by related or different virus species. The phenomenon of superinfection exclusion can be caused by inhibiting virus attachment, receptor binding and entry, by replication interference, or competition for host cell resources. Blocking attachment and entry not only prevents unproductive double infections but also stops newly produced virions from re-entering the cell post-exocytosis. In this study, we investigated the exclusion of superinfections between the different pestivirus species. Bovine and porcine cells pre-infected with non-

Molecular diversity and detection of pestiviruses across ruminant species in Türkiye.

Pestiviruses have historically been described using host-associated disease names, such as Bovine Viral Diarrhoea Virus (BVDV) in cattle and Border Disease Virus (BDV) in small ruminants, and are important pathogens causing abortions, congenital anomalies, reproductive failure, and reduced productivity, resulting in substantial economic losses in livestock. This study investigated pestivirus infections in ruminant samples from Türkiye using serological and molecular methods, and RT-PCR-positive samples were further characterized by phylogenetic analysis. A total of 115 ruminant samples were co

[The newly discovered "atypical porcine pestivirus" (APPV): An old player in the "shaking piglets" disease complex?].

Congenital tremor (CT) or "shaking piglet" syndrome of newborn piglets is a well-known disease caused by different factors and resulting in different pathological alterations. In addition to non-infectious causes (like intoxication and genetic alterations), viral infections of the sow during gestation are of utmost importance. It has long time been known that classical swine fever virus, a virus belonging to the genus Pestivirus within the family Flaviviridae, induces CT. Very recently, a novel porcine pestivirus was discovered, which is also capable to induce the disease and was designated as

Adventitious pestivirus RNA in live virus vaccines against bovine and swine diseases.

Live virus vaccines against bovine and porcine diseases were examined for the presence of adventitious pestivirus RNA or pestiviruses by reverse transcription-polymerase chain reaction (PCR). Pestivirus RNA was detected in the live virus vaccines against Akabane disease, Ibaraki disease, infectious bovine rhinotracheitis, porcine parvovirus infection, transmissible gastroenteritis and Japanese encephalitis. Pestivirus RNA or pestivirus in the fetal bovine serum used to grow the host cells used to prepare the bovine and swine viral vaccines is a likely source of the contamination. Nucleotide se

Congenital infections with nonarbo togaviruses.

The present review deals with the similarities and differences of selected aspects of prenatal pestivirus infections of domestic animals and congenital rubella of man. Hog cholera virus, bovine virus diarrhoea virus and border disease virus are antigenically closely related, but unrelated to rubella virus. The nonarbo togaviruses are capable of producing congenital infections resulting in a wide spectrum of abnormalities. The infected foetus can die in utero, in the neonatal period, or it may be born with teratogenic defects. In addition, apparently healthy progeny can be delivered that develo
