Development of the adult endocrine pancreas during metamorphosis in the sea lamprey, Petromyzon marinus L. I. Light microscopy and autoradiography.

The results of this study were found to support the hypothesis put forth by Boenig (Z. Mikrosk-Anat. Forsch., 17:125-184, 1929) that the caudal pancreas of adult lamprey develops from the epithelium of the extrahepatic common bile duct in that the bile duct cells were found to undergo a great proliferation during the early stages of metamorphosis, with a large number of the cells incorporating 3H-thymidine. If the bile duct degenerated as suggested by Barrington (The Biology of Lampreys. Academic Press, London, pp. 135-169, 1972), this uptake would not be expected. The cranial pancreas was det

Somatostatin concentrations in the pancreatic-intestinal tissues of the sea lamprey, Petromyzon marinus L., at various periods of its life cycle.

1. Somatostatin concentrations were measured in homogenates of the pancreas-intestinal tissues from each period of the life cycle of Petromyzon marinus using radioimmunoassay. 2. Levels were very low in larva (4.0 pg/mg wet weight) and in the first three stages of metamorphosis, but increased from stage 4 onwards and reached a high in upstream-migrating adults (210.0 ng/mg). 3. These data correlate well with our previous morphological and immunohistochemical observations on the morphogenesis of somatostatin-containing D-cells during the life cycle and indicate that the increased concentration

Functional characterization and osmoregulatory role of the Na+-K+-2Cl- cotransporter in the gill of sea lamprey (Petromyzon marinus), a basal vertebrate.

The present study provides molecular and functional characterization of Na+-K+-2Cl- cotransporter (NKCC1/Slc12a2) in the gills of sea lamprey (Petromyzon marinus), the most basal extant vertebrate with an osmoregulatory strategy. We report the full-length peptide sequence for the lamprey Na-K-Cl cotransporter 1 (NKCC1), which we show groups strongly with and occupies a basal position among other vertebrate NKCC1 sequences. In postmetamorphic juvenile lamprey, nkcc1 mRNA was present in many tissues but was fivefold higher in the gill than any other examined tissue, and NKCC1 protein was only de

Development of the adult endocrine pancreas during metamorphosis in the sea lamprey, Petromyzon marinus L. II. Electron microscopy and immunocytochemistry.

The development of the adult endocrine pancreas was followed throughout metamorphosis in the sea lamprey using electron microscopy and immunocytochemistry. It was discovered that the caudal pancreas develops from the larval extrahepatic common bile duct through the process of transdifferentiation (dedifferentiation/redifferentiation). Early in metamorphosis the bile duct epithelial cells possess large vacuoles, resembling autophagic vacuoles, containing recognizable cell material. There is a loss of the large bundles of intermediate filaments characteristic of the larval bile duct epithelium.

Identifying spawning sites and other critical habitat in lotic systems using eDNA "snapshots": A case study using the sea lamprey Petromyzon marinus L.

Many aquatic species of conservation concern exist at low densities and are inherently difficult to detect or monitor using conventional methods. However, the introduction of environmental (e)DNA has recently transformed our ability to detect these species and enables effective deployment of limited conservation resources. Identifying areas for breeding, as well as the ecological distribution of species, is vital to the survival or recovery of a conservation species (i.e., areas of critical habitat). In many species, spawning events are associated with a higher relative abundance of DNA releas

Immunocytochemical study of growth hormone, prolactin, and thyroid-stimulating hormone in the adenohypophysis of the sea lamprey, Petromyzon marinus L., during its upstream migration.

Immunoreactivity to growth hormone (GH), prolactin (PRL), and thyroid-stimulating hormone (TSH) was found in the adenohypophysis of the sea lamprey, Petromyzon marinus L., during its upstream migration, by immunoperoxidase techniques with antisera to mammalian pituitary hormones. Cross-reactivity to GH and PRL was found in two different cell populations in the proximal pars distalis. Specific immunostaining for GH and PRL was absent in other parts of the lamprey pituitary. Immunoreactive TSH cells were located only in the rostral pars distalis, and corresponded in shape and size to the large b

Concentrations of insulin and thyroid hormones in the serum of landlocked sea lampreys (Petromyzon marinus) of three larval year classes, in larvae exposed to two temperature regimes, and in individuals during and after metamorphosis.

Serum concentrations of thyroxine (T4) triiodothyronine (T3), and insulin were measured in landlocked sea lampreys (Petromyzon marinus) of three larval year classes (II to IV), larvae in two fixed temperatures (13 and 21 degrees), individuals during metamorphosis (stages 3-7), and juveniles immediately following metamorphosis. There was no difference in serum insulin levels in the larval year classes but the concentrations of both T4 and T3 increased during the protracted larval life to reach a maximum in individuals of metamorphosing size (age) without initiating early (pro) metamorphic event

Sea Lamprey Alarm Cue Comprises Water- and Chloroform- Soluble Components.

A diversity of aquatic organisms manage predation risk by avoiding waters activated with conspecific alarm cues, a chemical mixture released from injuries. The sea lamprey (Petromyzon marinus) is a nocturnal migratory species that relies on its alarm cue to navigate around areas of predation risk when moving through river channels. Identification of the cue's chemistry would allow managers to harness this innate behavioral response to guide migrating sea lamprey to traps (invasive population in the Laurentian Great Lakes) or to fish passage devices where dams block migrations in their native r

Disturbances to energy metabolism in juvenile lake sturgeon (Acipenser fulvescens) following exposure to niclosamide.

Since the 1960s, invasive sea lamprey (Petromyzon marinus) populations in the Laurentian Great Lakes have been controlled by applying two chemicals, 3-trifluoromethyl-4-nitrophenol (TFM) and 2',5-dichloro-4'-nitrosalicylanilide (niclosamide, aka. Bayluscide®), to streams infested with larval sea lamprey. These "lampricide" applications primarily rely on TFM, and are often combined with 1-2% niclosamide, which increases treatment effectiveness. Niclosamide is also used alone to treat lentic habitats and in rivers with high discharge. However, little is known about niclosamide's possible adverse

Perspective: the variable lymphocyte receptor B system in the gut of the sea lamprey.

The evolutionary origin of vertebrate adaptive immunity has been a longstanding problem in evolutionary biology. The diversified proteins that orchestrate this complex system are present throughout the jawed vertebrates but are absent in extant jawless vertebrates and invertebrates. From work initially carried out in the sea lamprey (Petromyzon marinus) and subsequently extended to other lampreys and hagfishes, we now know that, instead of immunoglobulin domain-based receptors, jawless vertebrates have leucine-rich repeat-based variable lymphocyte receptors (VLRs). Like immunoglobulins (Igs) a

Characterization and Evolution of Germ1, an Element that Undergoes Diminution in Lampreys (Cyclostomata: Petromyzontidae).

The sea lamprey (Petromyzon marinus) undergoes substantial genomic alterations during embryogenesis in which specific sequences are deleted from the genome of somatic cells yet retained in cells of the germ line. One element that undergoes diminution in P. marinus is Germ1, which consists of a somatically rare (SR) region and a fragment of 28S rDNA. Although the SR-region has been used as a marker for genomic alterations in lampreys, the evolutionary significance of its diminution is unknown. We examined the Germ1 element in five additional species of lamprey to better understand its evolution

Using visible implant elastomer to study ammocoete populations with Cormack-Jolly-Seber models.

This study examined the efficacy of marking wild populations of lampreys with visible implant elastomer (VIE) for 6-18 months to examine ammocoete movements using Cormack-Jolly-Seber (CJS) open-population models. These methods were tested on two lamprey populations in different river systems. American brook lamprey Lethenteron appendix at Dyke Creek apparent survival (φ) was high in the summer and winter (c. 0·7), but declined after flow events in the spring and autumn. Sea lamprey Petromyzon marinus at Oquaga Creek φ in the top-ranked models varied with stream location and time. Estimates of

Effects of water pH on the uptake and elimination of the piscicide, 3-trifluoromethyl-4-nitrophenol (TFM), by larval sea lamprey.

Invasive sea lamprey (Petromyzon marinus) populations in the Great Lakes are controlled by applying the piscicide, 3-trifluoromethyl-4-nitrophenol (TFM), to infested streams with larval sea lamprey (ammocoetes). While treatment mortality is >90%, surviving lamprey, called residuals, can undermine control efforts. A key determinant of TFM effectiveness is water pH, which can fluctuate daily and seasonally in surface waters. The objectives of this research were to evaluate the influence of pH on the uptake, elimination, and accumulation of TFM by larval sea lamprey using radio-labeled TFM (14C-T

Occurrence of Ergasilus megaceros Wilson, 1916, in the sea lamprey and other fishes from North America.

Ergasilus megaceros (Copepoda: Ergasilidae) was recovered from the nasal fossae (lamellae) of the olfactory sac in 1 (1.8%) of 56 sea lampreys, Petromyzon marinus Linne, 1758, collected in May 2002 from the Cheboygan River, Michigan. Although the sea lamprey is a new host record for E. megaceros, this fish species may not be a preferred host because of its low prevalence. Ergasilus megaceros is the second ergasilid species reported from the sea lamprey in North America. This is the third report of an ergasilid species infecting the nasal fossae of fishes in North America, with E. rhinos being

Landmark discoveries in elucidating the origins of the hypothalamic-pituitary system from the perspective of a basal vertebrate, sea lamprey.

The hypothalamic-pituitary (HP) system, which is specific to vertebrates, is considered to be an evolutionary innovation that emerged prior to or during the differentiation of the ancestral jawless vertebrates (agnathans) leading to the neuroendocrine control of many complex functions. Along with hagfish, lampreys represent the oldest lineage of vertebrates, agnathans (jawless fish). This review will highlight our discoveries of the major components of the lamprey HP axis. Generally, gnathostomes (jawed vertebrates) have one or two hypothalamic gonadotropin-releasing hormones (GnRH) while lamp

Rainbow trout rapidly recover from exposure to niclosamide: A piscicide and molluscicide used to control sea lamprey and snail populations.

Niclosamide (2',5-dichloro-4'-nitrosalicylanalide) is a piscicide used to control invasive sea lamprey (Petromyzon marinus) in the Laurentian Great Lakes. It is also a molluscide used in tropical and sub-tropical freshwaters to control snail populations that are intermediate hosts to the blood flukes that causes schistosomiasis in humans. While the mechanism of niclosamide toxicity is known, its corresponding physiological effects on non-target fishes are not well-established. To better understand how niclosamide could adversely affect non-target fishes, rainbow trout (Oncorhynchus mykiss) wer

Periductal fibrosis and lipocytes (fat-storing cells or Ito cells) during biliary atresia in the lamprey.

Transmission and scanning electron microscopy were utilized to follow the degeneration of bile ducts of lampreys (Petromyzon marinus L.) during metamorphosis. The convoluted bile ducts of larval lampreys are surrounded by rich sinusoids, but this intimate biliovascular relationship is lost during metamorphosis because the bile duct degeneration is accompanied by the development of thick periductal fibrosis. Lipocytes, which are present not only in the parenchyma but also in the interstitial tissue of the liver, increase in number in the periductal fibrous tissue, and their processes are direct
