Koala retrovirus (KoRV) causes multiple disease phenotypes in koalas, including carcinogenesis. The study aimed to assess oncogene expression in spleen tissues from ten deceased koalas coinfected with different subtypes and peripheral blood mononuclear cells (PBMCs) from two subclinically coinfected koalas with KoRV-A and KoRV-B. Initially, KoRV subtyping involved amplifying endogenous KoRV-A, and exogenous KoRV-B, -C specific env gene fragments, followed by sequencing. Using quantitative real-time polymerase chain reaction (RT-qPCR), we examined five oncogenes (BCL2, BAX, BCL2L1, BCL3, and MY

A novel alphaherpesvirus was detected in a captive adult, lactating, female koala (Phascolarctos cinereus) admitted to James Cook University Veterinary Emergency Teaching & Clinical Hospital in March 2019, showing signs of anorexia and severe respiratory disease. Postmortem examination revealed gross pathology indicative of pneumonia. Histopathology demonstrated a chronic interstitial pneumonia, multifocal necrotising adrenalitis and hepatitis. Intranuclear inclusion bodies were detected by light microscopy in the respiratory epithelium of the bronchi, bronchioles, alveoli, and hepatocytes, bi

Koala Retrovirus (KoRV) has been associated with neoplasia in the vulnerable koala (Phascolarctos cinereus). However, there are conflicting findings regarding its association with secondary disease. We undertook a large-scale assessment of how the different KoRV subtypes and viral load are associated with Chlamydia pecorum infection and a range of disease pathologies in 151 wild koalas admitted for care to Currumbin Wildlife Hospital, Australia. Viral load (KoRV pol copies per ml of plasma) was the best predictor of more disease pathologies than any other KoRV variable. The predicted probabili

There is growing evidence that Chlamydia pecorum infection of the male koala reproductive tract causes inflammation and pathology of the urogenital tract. Previous studies have revealed that male koalas exhibiting severe clinical signs of urogenital chlamydial disease had an increased incidence of sperm DNA fragmentation and abnormal sperm morphology, suggestive of chronic exposure to C. pecorum infection and/or inflammation in the testis and epididymis, with residual pathology and lesions disrupting spermatogenesis and maturation of spermatozoa. This study specifically aimed to determine whet

Pneumonia has been reported in both free-ranging and captive koalas and a number of causative agents have been described. Between 2016 and 2019, 16 free-ranging and 1 captive koala (Phascolarctos cinereus) from the Mount Lofty Ranges of South Australia were identified with pyogranulomatous lobar pneumonia, which involved the left caudal lobe in 14/17 (82%) cases. Within lesions, numerous gram-positive or gram-variable, non-acid-fast filamentous bacteria were observed in association with Splendore-Hoeppli phenomenon. Culture yielded growth of anaerobic bacteria, which were unidentifiable by MAL

OBJECTIVE: Current haematology reference intervals (RIs) for koalas were developed in northern Australian koalas, using low numbers and/or individuals of unknown Chlamydia pecorum and koala retrovirus (KoRV) status. This study developed haematological RIs for wild, clinically healthy southern Australian koalas of known C. pecorum and KoRV infection status and investigated the effects of population, age and sex.
METHODS: Haematological RIs were determined for 138 clinically healthy South Australian koalas (Mount Lofty Ranges [MLR], n = 68; Kangaroo Island, n = 70) examined in April 2016 and Fe

Koala retrovirus (KoRV) infection, thought to be associated with lymphoid neoplasia, and Chlamydia pecorum-related ocular and urogenital disease are both highly prevalent in eastern Australian koala (Phascolarctos cinereus) populations. However, in South Australian koalas, little is known about KoRV infection and C. pecorum-associated disease. We report the first South Australian case of lymphoma in a KoRV-A-positive female koala also affected by severe reproductive chlamydiosis. The koala was from the Mount Lofty Ranges population and was presented with hindlimb lameness. Clinical examination

The koala, an endangered species in some parts of Australia, faces significant population decline, driven partly by Chlamydia pecorum, a pathogen causing blindness, infertility, and mortality. This study investigated C. pecorum infection dynamics in 285 koalas from Southeast Queensland, from two neighbouring study regions separated by a river into northern (n = 147) and southern study regions (n = 138). Ocular and urogenital swabs were screened for C. pecorum presence via quantitative PCR, while age, sex, body condition, and clinical signs, were assessed through veterinary examination. Multiva

Koala populations in some regions of eastern Australia are in critical condition. Our research aims to develop effective conservation strategies for these declining koalas threatened by chlamydia infection, predation, and climate change. To achieve this, we developed a mathematical model that includes populations of dingoes and koalas categorized as susceptible, infected, and confined. We conducted a bifurcation analysis within the ordinary differential equations (ODE) model to explore the occurrence of a Hopf bifurcation. This analysis aimed to identify conditions under which the system under

External signs of disease are frequently used as indicators of disease susceptibility. However, immune profiling can be a more effective indicator to understand how host responses to infection may be shaped by host, pathogen and environmental factors. To better inform wildlife health assessment and research directions, we investigated the utility of a novel multivariate immunophenotyping approach examining innate and adaptive immune responses in differing climatic, pathogen co-infection and demographic contexts across two koala (Phascolarctos cinereus) populations in New South Wales: the Liver

OBJECTIVE: To describe outbreaks of sarcoptic mange caused by Sarcoptes scabiei in free-ranging koalas in Victoria (December 2008 to November 2015) and South Australia (October 2011 to September 2014).
METHODS: Koalas affected by mange-like lesions were reported by wildlife carers, veterinary practitioners or State Government personnel to the Faculty of Veterinary and Agricultural Sciences at The University of Melbourne and the School of Animal and Veterinary Sciences at The University of Adelaide. Skin scrapings were taken from live and dead koalas and S. scabiei mites were identified. Tissu

Three asymptomatic koalas serologically positive for cryptococcosis and two symptomatic koalas were treated with 10 mg/kg fluconazole orally, twice daily for at least 2 weeks. The median plasma Cmax and AUC0-8 h for asymptomatic animals were 0.9 μg/mL and 4.9 μg/mL·h, respectively; and for symptomatic animals 3.2 μg/mL and 17.3 μg/mL·h, respectively. An additional symptomatic koala was treated with fluconazole (10 mg/kg twice daily) and a subcutaneous amphotericin B infusion twice weekly. After 2 weeks the fluconazole Cmax was 3.7 μg/mL and the AUC0-8 h was 25.8 μg/mL*h. An additional three ko

The successful development and application of an assisted breeding program in any animal relies primarily on a thorough understanding of the fundamental reproductive biology (anatomy, physiology and behaviour) of the species in question. Surely, the ultimate goal and greatest hallmark of such a program is the efficacious establishment of a series of reliable techniques that facilitate the reproductive and genetic management of fragmented populations, both in captivity and in the wild. Such an achievement is all the more challenging when the reproductive biology of that species is essentially r

The hypothalamic-pituitary-adrenal axis responds to stress by releasing the adrenocorticotropic hormone which, in turn, stimulates the release of glucocorticoids (GCs). During acute stress events, the GCs' function is to maintain homeostasis. Short-term stress events trigger psychophysiological responses which are fundamental for survival in the natural world. Chronic stress occurs when the effect of stressors persists, and GCs secretion continues. The increase of GCs interacts with receptors in the brain triggering a negative feedback loop which inhibits the secretion of ACTH, consequently do

This study presents a retrospective analysis of clinical records data from koalas presenting for treatment following the large-scale bushfire event on Kangaroo Island 2019-2020. The aim of the study was to identify prognostic factors for koalas affected by bushfire. Koalas (n = 199) were grouped based on their burn status in combination with their burn bandaging requirement at triage; those with no burns, with burns that did not require bandaging and with burns that did require bandaging. Overall, 59.8% of koalas had positive outcomes, but this reduced to only 13% of koalas in the most severel

Gut microbiota is one of the major elements in the control of host health. However, the composition of gut microbiota in koalas has rarely been investigated. Here, we performed 16S rRNA gene sequencing to determine the individual and environmental determinants of gut microbiota diversity and function in 35 fecal samples collected from captive koalas. Meanwhile, blood immune-related cytokine levels were examined by quantitative reverse transcription-PCR to initially explore the relationship between the gut microbiota and the immune system in koalas. The relative abundance of many bacteria, such

Testis asymmetry, in which the testes in an individual differ in size, has recently been reported in koalas (Phascolarctos cinereus) in the Mount Lofty Ranges population of South Australia. We describe the morphology and histology of both testes from affected individuals in this population (n=56) and the parameters of koalas with normal-sized testes based on age and breeding season (n=56). Morphologic measurements included testis weight, length, width, and volume; histologic parameters included seminiferous tubule diameter, seminiferous epithelial height, and seminiferous tubule (interstitial

Koala populations are currently declining and under threat from koala retrovirus (KoRV) infection both in the wild and in captivity. KoRV is assumed to cause immunosuppression and neoplastic diseases, favoring chlamydiosis in koalas. Currently, 10 KoRV subtypes have been identified, including an endogenous subtype (KoRV-A) and nine exogenous subtypes (KoRV-B to KoRV-J). The host's immune response acts as a safeguard against pathogens. Therefore, a proper understanding of the immune response mechanisms against infection is of great importance for the host's survival, as well as for the developm

The koala (Phascolarctos cinereus) is recognised as a threatened wildlife species in various parts of Australia. A major contributing factor to the decline and long-term viability of affected populations is disease caused by the obligate intracellular bacteria, Chlamydia. Two chlamydial species infect the koala, Chlamydia pecorum and Chlamydia pneumoniae, and have been reported in nearly all mainland koala populations. Chlamydial infections of koalas are associated with ocular infections leading to blindness and genital tract infections linked to infertility, among other serious clinical manif

Chlamydiosis is a significant factor contributing to the decline of koala (Phascolarctos cinereus) populations in Australia but has not previously been reported in South Australia. We describe conjunctivitis in three wild koalas from South Australia, with Chlamydia pecorum identified by quantitative PCR.
