Phase separation and protein aggregation in neurodegenerative diseases.

Neurodegenerative diseases such as Alzheimer's, Parkinson's, frontotemporal dementia, and ALS are characterized by amyloid protein aggregation involving intrinsically disordered proteins that are also capable of liquid-liquid phase separation (LLPS). LLPS, known to drive the formation of dynamic membraneless organelles essential for cellular functions, can play a role in limiting fibrillation process or aberrantly transition into solid aggregates under pathological conditions. Here we review how mutations, post-translational modifications, and environmental factors can modulate LLPS of protein

Optogenetics to biomolecular phase separation in neurodegenerative diseases.

Neurodegenerative diseases involve toxic protein aggregation. Recent evidence suggests that biomolecular phase separation, a process in which proteins and nucleic acids form dynamic, liquid-like condensates, plays a key role in this aggregation. Optogenetics, originally developed to control neuronal activity with light, has emerged as a powerful tool to investigate phase separation in living systems. This is achieved by fusing disease-associated proteins to light-sensitive oligomerization domains, enabling researchers to induce or reverse condensate formation with precise spatial and temporal

Contact temporal network during motility-induced phase separation.

Motility-induced phase separation (MIPS) is a paradigmatic nonequilibrium transition in active matter, determined by the Péclet number and packing fraction. We investigate the single-phase and phase-separated regimes of MIPS using a complex network approach, where networks are constructed from particle interactions over finite time windows. In the single-phase (gas-like) regime, the degree distributions P(k) exhibit Gaussian behavior and resemble those of random graphs. Plotting the location and height of the P(k) peak reveals a universal curve across different Péclet numbers at fixed packing

Liquid-Liquid Phase Separation in Hereditary Hearing Loss.

Hearing loss is one of the most prevalent sensory disorders affecting the human nervous system. Liquid-liquid phase separation (LLPS) is a physiological process that facilitates the reversible and dynamic assembly of biomolecular condensates. Increasing evidence suggests that LLPS plays a significant role in the pathogenesis of hereditary hearing loss. Nevertheless, there is a conspicuous lack of systematic investigations exploring the impact of LLPS abnormalities on the etiology of hereditary hearing loss. In this review, we examine the mechanisms by which dysfunctions in LLPS contribute to h

Aberrant Enhancer Regulation, Phase Separation, and Autoimmune Diseases.

In the intricate regulation of gene expression, multiple regulatory elements play crucial roles. Among these, enhancers stand out as important elements that can boost the activity of promoters and make target genes more likely to be transcribed. Recent studies have revealed that sequence variations and gene rearrangements in enhancer regions could disrupt enhancer-promoter interactions, contributing to disease susceptibility and developmental abnormalities. Enhancer dysregulation is associated with numerous pathological conditions, spanning malignancies, autoimmune diseases, cardiovascular pat

Phase separation of the microtubule-associated protein tau.

The aggregation and misfolding of the neuronal microtubule-associated protein tau is closely linked to the pathology of Alzheimer's disease and several other neurodegenerative diseases. Recent evidence suggest that tau undergoes liquid-liquid phase separation in vitro and forms or associates with membrane-less organelles in cells. Biomolecular condensation driven by phase separation can influence the biological activities of tau including its ability to polymerize tubulin into microtubules. In addition, the high concentrations that tau can reach in biomolecular condensates provide a mechanism

Deciphering Melatonin-Stabilized Phase Separation in Phospholipid Bilayers.

Lipid bilayers are fundamental building blocks of cell membranes, which contain the machinery needed to perform a range of biological functions, including cell-cell recognition, signal transduction, receptor trafficking, viral budding, and cell fusion. Importantly, many of these functions are thought to take place in the laterally phase-separated regions of the membrane, commonly known as lipid rafts. Here, we provide experimental evidence for the "stabilizing" effect of melatonin, a naturally occurring hormone produced by the brain's pineal gland, on phase-separated model membranes mimicking

Macroporous hydrogels derived from aqueous dynamic phase separation.

A method to generate injectable macroporous hydrogels based on partitioning of polyethylene glycol (PEG) and high viscous polysaccharides is presented. Step growth polymerization of PEG was used to initiate a phase separation and the formation of a connected macroporous network with tunable dimensions. The possibilities and physical properties of this new category of materials were examined, and then applied to address some challenges in neural engineering. First, non-degradable macroporous gels were shown to support rapid neurite extension from encapsulated dorsal root ganglia (DRGs) with unp

Autophagy regulation by phase separation, avidity, and wetting.

Autophagy enables cells to selectively degrade a wide range of macromolecules, and how this process achieves spatial precision within the densely packed cytosol is an active area of investigation. Recent advances suggest that phase separation provides a crucial organizational framework that converts autophagy into a spatiotemporally coordinated and self-organizing process. Biomolecular condensates formed by phase separation can create high-avidity binding platforms between autophagy receptors, scaffold proteins, and the cargo that stabilize transient molecular contacts. The formation of such c

Small Molecule Modulators of α-Synuclein Phase Separation.

Parkinson's disease (PD) is a progressive neurodegenerative condition that affects the brain's ability to control movement. The disease occurs when nerve cells of the brain become impaired. Alpha-synuclein protein (α-Syn) aggregation followed by fibrillation is a hallmark of PD. Recent studies showed that the liquid-liquid phase separation (LLPS) and droplet formation of α-Syn protein play a crucial role in the pathology of PD. We synthesized novel zwitterionic compounds (3), which inhibited α-Syn phase separation and subsequent formation of fibrillary aggregates in a concentration-dependent m

Neurological Diseases can be Regulated by Phase Separation.

Several neurological diseases arise from abnormal protein aggregation within neurones and this is closely regulated by phase separation. One such is motor neurone disease and aberrant aggregation of superoxide dismutase. Again these events are regulated by electrical forces that are examined.

Mesoscale Magnetostructural Phase Separation in Fe-deficient Fe5GeTe2.

2D Van der Waals ferromagnet Fe5-xGeTe2 (F5GT) is promising for spintronic applications due to its high Curie temperature, layered structure, and ability to host complex magnetic textures. However, the origin of its sample-dependent magnetic anisotropy remains unclear, hindering control of its magnetic behavior. Here, spatially resolved cryogenic scanning transmission electron microscopy (STEM) is used to correlatively map magnetism, lattice structure, and chemistry across atomic-to-micron scales. This is revealed that only mesoscale, not nanoscale, inclusions of a Fe-deficient secondary phase

Tau liquid-liquid phase separation in neurodegenerative diseases.

Aggregation of the microtubule-associated protein tau plays a major role in Alzheimer's disease and several other neurodegenerative disorders. An exciting recent development is the finding that, akin to some other proteins associated with neurodegenerative disease, tau has a high propensity to condensate via the mechanism of liquid-liquid phase separation (LLPS). Here, we discuss the evidence for tau LLPS in vitro, the molecular mechanisms of this reaction, and the role of post-translational modifications and pathogenic mutations in tau phase separation. We also discuss recent studies on tau L

Stimuli-Responsive Membranes through Sustainable Aqueous Phase Separation.

Polymeric membranes are used on huge scales for kidney dialysis, wastewater treatment, and drinking water production. However, almost all polymeric membranes are fabricated by a process reliant on the use of unsustainable, expensive, and reprotoxic dipolar aprotic solvents. In this work, we propose an aqueous phase separation approach for preparing porous membrane films. Poly(4-vinylpyridine) (P4VP), a pH-responsive polymer, is first dissolved at low pH where the polymer is charged and subsequently cast as a thin film. Switching to a high pH where the polymer is uncharged and insoluble results

Separation of lipid classes by solid phase extraction.

A rapid and reliable method for the separation of lipid classes is described using aminopropyl disposable columns. This method is a modification to an existing procedure that allows the separation of both neutral and acidic phospholipid fractions and a high recovery of the latter. Acidic phospholipids were eluted with a mixture of hexane-2-propanol-ethanol-0.1 M ammonium acetate-formic acid 420:350:100:50:0.5 containing 5% phosphoric acid after neutral phospholipids had been eluted with methanol. It was verified that extremely high recoveries of cholesterol (CH), triglycerides (TG), free fatty

Two-dimensional liquid chromatography normal-phase and reversed-phase separation of (co)oligomers.

Many samples contain compounds with various numbers of two or more regular structural groups. Such "multidimensional" samples (according to the Giddings' notation) are best separated in orthogonal chromatographic systems with different selectivities for the individual repeat structural groups, described by separation factors. Correlations between the repeat group selectivities characterize the degree of orthogonality and suitability of chromatographic systems for two-dimensional (2D) separations of two-dimensional samples. The range of the structural units in that can be resolved in a given ti

Electrically Fueled Liquid-Liquid Phase Separation for Active Transport.

Out-of-equilibrium liquid-liquid phase separation (LLPS) of biomacromolecules in living cells creates membraneless organelle, enabling diverse functions including active transport. This has inspired the development of synthetic nonequilibrium LLPS systems; however, their active states are often hampered by waste accumulation, and their emergent functional properties remain largely unexplored. Here, we report an electrically fueled dissipative LLPS that sustains active coacervation without waste generation and, most notably, enables the recruitment and directional transport of molecules and nan

Reordering-mediated nucleation dynamics in early stage phase separation.

Classical nucleation theories relying on quasi-equilibrium thermodynamics fail to capture the far-from-equilibrium structural reordering inherent to early stage condensed-cluster formation. We develop a framework that maps a continuous structural field onto a core-shell model, coupling local non-conservative reordering with mesoscopic phase-boundary propagation. By evaluating the reordering-mediated nucleation time, we analytically distinguish the static potential energy profile from the effective nucleation barrier. Our mean-field solution reveals a kinetic turnover effect: beyond a critical

Liquid-liquid phase separation of tau in Alzheimer's disease.

Tau is a microtubule-associated protein that plays a central role in neuronal stability and axonal transport. However, under pathological conditions, it undergoes structural and functional changes that lead to Alzheimer's disease (AD) and related tauopathies. Recent evidence highlights liquid-liquid phase separation (LLPS) as a critical mechanism underlying tau aggregation and the subsequent formation of neurofibrillary tangles (NFTs). This chapter explores the properties of tau, including its intrinsically disordered nature, isoforms, and post-translational modifications (PTMs), that predispo

Databases of liquid-liquid phase separation proteins and condensates.

Protein liquid-liquid phase separation (LLPS) and the formation of biomolecular condensates are increasingly recognized as important in both cellular physiology and pathology. However, the computational databases developed to catalog LLPS-associated proteins differ significantly in scope, curation strategies, evidence models, and annotation granularity. Here, we compare the main LLPS databases, discuss what each resource captures well, where role definitions and evidence models diverge, which metadata remain limited, and how these differences affect data interpretation and reuse. We argue that
